In 2014, the coal cleaning chemical 4-methylcyclohexane methanol (MCHM) spilled into the water supply for 300,000 West Virginians. Initial toxicology tests showed relatively mild results, but the underlying effects on cellular biology were underexplored. Treated wildtype yeast cells grew poorly, but there was only a small decrease in cell viability. Cell cycle analysis revealed an absence of cells in S phase within thirty minutes of treatment. Cells accumulated in G1 over a six-hour time course, indicating arrest instead of death. A genetic screen of the haploid knockout collection revealed 329 high confidence genes required for optimal growth in MCHM. These …

Background
The study of localized immune-related factors has proven beneficial for a variety of conditions, and one area of interest in the field of orthopaedics is the impact of implants and localized infections on immune response. Several cytokines have shown increased systemic concentrations (in serum/plasma) in response to implants and infection, but tissue-level cytokines have not been investigated as thoroughly.
Methods
This exploratory study investigated tissue-level cytokines in a cohort of patients (N = 17) in response to total knee arthroplasty and total knee revision to better understand the immune response to implants and localized infection (e.g., prosthetic joint infection). …

In recent months, the United States Postal Service (USPS) has taken center stage on a number of intersecting issues in our society: the pandemic; the upcoming election (through mail-in voting) and the controversy surrounding the appointment of Louis DeJoy to the position of Postmaster General. President Donald Trump has frequently made derogatory remarks regarding the Postal Service, calling it a “joke,” and has made repeated statements encouraging its privatization. Hydraulic fracturing has been booming in the last decade in the United States. While natural gas extraction and production has improved the national energy security, it has raised questions around the water security of those communities where extraction is taking place. Acute stroke causes complex, pathological, and systemic responses that have not been treatable by any single medication. In this study, using a murine transient middle cerebral artery occlusion stroke model, a novel therapeutic strategy is proposed, where blood replacement (BR) robustly reduces infarctions and improves neurological deficits in mice. Our analyses of immune cell subsets suggest that BR therapy substantially decreases neutrophils in blood following a stroke. Electrochemiluminescence detection demonstrates that BR therapy reduces cytokine storm in plasma and ELISA demonstrates reduced levels of matrix metalloproteinase-9 (MMP-9) in the plasma and brains at different time points post-stroke. The black-legged tick Ixodes scapularis is a common ectoparasite of animals and an obligate blood feeder, attaching to a host animal and taking a blood meal once per life stage (i.e. 3 times during its life). Unfortunately for the tick, blood represents a nutrient poor diet largely consisting of lipids and proteins but notably lacking in essential nutrients such a B vitamins (e.g., biotin, riboflavin, niacin) Other exclusive blood feeders such as the tsetse fly harbor symbiotic bacteria that provision B-complex vitamins to their host. Organomodified nanoclays (ONCs: i.e., smectite clays with different organic coatings), act as nanofillers in applications ranging from automotive, to aerospace, and biomedical implementations. Because of their large utilization, ONC industry is projected to be a U.S. $3.3 billion industry by 2023. However, emerging studies showed that ONCs and ONC-nanocomposites could harbor health risks upon pulmonary exposure and along their life cycle, namely during synthesis, handling, use, manipulation, and disposal. Organisms, such as fungi and insects, can cause millions of acres of agricultural and forest damage, while others provide billions of dollars in ecological services such as education, aesthetic enjoyment, pollination, and gardening. Plant breeding and biotechnology can potentially help establish a balance between the proliferation of detrimental pests and attraction of beneficial insects. Variation in plant physiological and morphological characteristics are extremely important in the ability of host tissues to support many different types of organisms. The chemical 4-methylcyclohexanemethanol (MCHM) spilled in the Elk River near Charleston, WV in January 2014, impacting the drinking water of 300,000 residents. Initial studies on MCHM following the spill attempted to characterize its toxicological effects in animals, but largely ignored characterization of its effects on cellular pathways and metabolism. In this study, I used this chemical as a novel stressor for the model eukaryote S. cerevisiae to explore the relationship of genotype and stress phenotypes. Initial exploration characterized the stress phenotype of MCHM in yeast through growth assays, cell cycle analysis, and transcriptomic data. MCHM exposure caused cells to arrest …

Huntington Disease (HD) is a fatal neurodegenerative disorder caused by an expanded polyglutamine domain (polyQ) in the first exon of the huntingtin protein (htt-exon1). The major hallmark of HD is the accumulation of aggregates into proteinaceous inclusion bodies. PolyQ expansion in huntingtin promotes self-assembly into a variety of toxic aggregates such as oligomers, fibrils, and amorphous aggregates. The resulting heterogeneous mixture of distinct species makes it difficult to assign a toxic function to specific aggregate structures. Progestins are synthetic hormones that are designed to mimic the biological actions of progesterone. They, however, possess other pharmacological actions and properties, in addition to their progestational activities. Medroxyprogesterone Acetate (MPA) is a progestin used globally in the hormonal contraceptive, Depo Provera®, by women in their reproductive prime and is a major compound found in hormone therapy (HT) formulations used by menopausal women. MPA is used by approximately 1 in 5 adolescents and adult women in the United States who are sexually active. Heart failure is a lifelong disability that for over half of those affected leads to mortality within five years after initial diagnosis. Left ventricular hypertrophy (LVH) is one of the most reliable independent predictors of heart failure. Pathological LVH is irreversible, but early diagnosis is often missed due to its asymptomatic nature. Obese Zucker rats (OZR), which develop obesity due to dysfunctional leptin signaling, naturally exhibit a LVH that mimics the obese human condition. Falls are a serious threat to older adults' quality of life. Evidence is lacking regarding the influence of diet on fall risk factors. This study aims to assess the relationship between diet, functional measures, and fall risk among older adults participating in a fall-prevention intervention. Cross-sectional analysis of baseline data from 192 participants with an average age of 70.9 years was conducted using Chi-square tests, t-test, Wilcoxon test, and nominal logistic analysis. Based on Dietary Screening Tool (DST) scores, 39.5% of participants were classified as nutritionally being “at-risk,” 46.1% were at “possible-risk,” and 14.4% were “not-at-risk.” Fall risk was assessed …

Improvement of lamb performance in low-input operations may prove vital for the sustainability of eastern US sheep production. Maintaining growth while improving carcass conformation using terminal sire crossbreeding systems may increase value in a pasture-based system. Therefore, the aim of this study was to compare effects of terminal sire breed on parasitism, growth, marketability of grazing lambs. Suffolk (SU; n = 2) and Texel (TX; n = 2) sires were mated to commercial ewes at the WVU Organic Farm. Rapid contemporary climate change is a potential threat to long-term persistence of montane wildlife species because they often have narrow thermal tolerances and have limited potential to shift their distributions. The Appalachian Mountain region in the eastern United States is a global biodiversity hotspot for woodland salamanders (genus Plethodon), many of which are high-elevation endemic species. Robust assessments of the vulnerability of high-elevation endemic salamanders to climate change, including delineation of future potential climate refugia, are needed to guide climate change adaptations strategies. Extended breastfeeding through infancy confers benefits on neurocognitive performance and intelligence tests, though few have examined the biological basis of these effects. To investigate correlations with breastfeeding, we examined the major white matter tracts in 4–8 year-old children using diffusion tensor imaging and volumetric measurements of the corpus callosum. We found a significant correlation between the duration of infant breastfeeding and fractional anisotropy scores in left-lateralized white matter tracts, including the left superior longitudinal fasciculus and left angular bundle, which is indicative of greater intrahemispheric connectivity. En echelon fractures and veins are among the most common and distinctive geological structures, yet their three-dimensional forms and relationships to surrounding structures are commonly unclear. X-ray computed tomography (CT) offers an unrivaled ability to examine structures within rocks in three dimensions, and it is applied here to a sample of drill core from the Marcellus Shale of southwestern Pennsylvania (USA). CT images yield qualitative and quantitative data on the transition from a pyrite-rich planar vein to an en echelon veinlet array, and on the heterogeneity of veinlets within the array. Othar Turner held the Fife and Drum Blues traditions of North Mississippi for decades. Since his passing in 2003, his granddaughter Sharde Thomas has taken over as the keeper of these traditions. The musical characteristics of Othar are still present in Sharde’s music today, however her interpretations have shaped these current traditions. This research discusses heritage and folklore theories of scholars Rodney Harrison, Martha Sims, and Martine Stephens as a lens to evaluate Sharde’s musical interpretations and compares them to Othar’s last recordings before his passing. A scarcity of field-based research investigating the relationships between E. coli (fecal matter) concentration, suspended particulate matter (SPM; including size distribution), physicochemical parameters (water temperature, pH, SPC, DO, chloride) and land use practices and the importance of these relationships, particularly for human health, policy makers, and water resource managers, provided the impetus for this research. A 22-site, nested-scale, experimental watershed study design was implemented to investigate E. coli concentrations and the aforementioned relationships in a contemporary mixed land use watershed in the Appalachian region of the eastern United States. Traumatic brain injury (TBI) can cause chronic psychiatric-like impairments that may be driven by inflammation in the brain. In the current study, inflammation was upregulated using a high-fat diet (HFD) to assess the role of inflammation in TBI-induced deficits. Rats were randomly assigned to receive an HFD or calorie-matched low-fat diet (LFD) for the duration of the experiment. With the current state of the field of forensic biology, collecting DNA from evidence samples may pose some difficulty. Traditional methods for collection of DNA from biological fluids and touch samples include cuttings, scrapings, and the double swab method. While all of these methods have benefits, they also have their drawbacks. In order to more efficiently collect DNA from an evidence sample, novel approaches need to be explored. This novel approach is the microbial vacuum (M-Vac®) Wet Vacuum System.

Over recent decades, the maker movement has expanded from the private sector, libraries, and higher education institutions to K-12 schools. While many studies provide implications of STEM and maker activities and their effects on learning in informal settings, few have framed such investigations within the context of a makerspace in K-12 schools. The hypothalamus plays a major role in regulating food-intake and energy-expenditure. Hypothalamic dysfunction implicated in neurodegenerative diseases is characterized by diet-induced neuroinflammation and energy dysregulation. Apple pomace, a waste byproduct of processing, is rich in polyphenols and soluble fibers, has the potential to ameliorate diet-inducing inflammation. The objective of this study was to determine the effects of Western diet and caloric substitution of Western diet with 10% apple pomace on brain fatty acid composition and gene expression related to hypothalamic function and health in growing female rats.
