Evidence from investigations of genetic differences among human beings shows that genetic diseases are often the results of genetic mutations. The most common form of these mutations is single nucleotide polymorphism (SNP). A complete map of all SNPs in the human genome will be extremely valuable for studying the relationships between specific haplotypes and specific genetic diseases. Some recent discoveries show that the DNA sequence of human beings can be partitioned into long blocks where genetic recombination has been rare. Because of the species diversity, varied site conditions and growth rates, it is really challenging to manage the central Appalachian hardwoods. Examining the harvesting techniques and interactions among stand, harvest, and machines is becoming a concern to the researchers in the region. Experiments were established in fall 2003 at West Virginia University to evaluate the effects of incorporation of composted poultry litter (CPL) on subsoil physical and chemical properties, Kentucky bluegrass ( Poa Pratensis L.) establishment, root depth, and weed pressure. Twenty cm of topsoil was removed and CPL was incorporated to a depth of 12.7 cm at 10%, 20%, and 40% v/v prior to seeding or sodding. Composted plots were compared to N-fertilized (454 Kg ha-1) and control plots. Positive linear increases in total water content (thetaT), organic matter, pH, and CEC were observed following compost incorporation with a decrease in …

Metallothioneins (MT) are cysteine-rich intracellular proteins that sequester epoxides, such as the genotoxic styrene metabolite, styrene oxide (SO). Covalent adduct formation by the ring-opening of SO and its homologue, 1-phenylpropylene oxide (trans), with MT forms stable complexes that potentially may serve as biomarkers for epoxide exposure. MT was reacted with SO and trans-phenylpropylene oxide (PPO) separately in water. Mass spectra of adducts were obtained using electrospray ionization with positive-ion detection in an ion trap mass spectrometer. MS/MS analysis established the covalent nature of the complexation. Histone acetylation is modulated through the action of histone acetyltransferases and deacetylases, which play key roles in the regulation of eukaryotic gene expression. The RPD3 group of histone deacetylases constitutes the first class of histone deacetylases (HDACs) in eukaryotes.;In yeast and mammalian cells, it was found that RPD3 histone deacetylases are involved in the ageing and development. This research is an investigation of how remote sensing can be used to supplement traditional methods of corridor management planning. Cultural resource inventory for corridor and interpretive management practices is an important component of applications for the designation of a selected route as Scenic Historic American Byways through the National Scenic Byways Program. A case study of a section of the National Road near Wheeling, West Virginia, showed that contemporary and historic aerial photography can facilitate the identification of possible impacts on a route’s historic integrity. During the seventeenth century a vibrant group of Native Americans, the Monytons, controlled southern West Virginia and northeastern Kentucky. The effects of contact with Europeans destabilized their societies as far inland as the Ohio Valley. This began the process which eventually pushed most Monytons from the region. The Five Nations Iroquois, pressured by social changes farther north, attacked the Monytons further drawing people out of the Ohio Valley. A growing southern Indian slave trade also contributed to the decay of Monyton dominance in the region. A central point in this thesis is that Monytons formed and reformed traditions to deal …

A review of the literature indicates there is a need for alternative assessment practices for elementary students. Seven fourth grade students in a small rural school will learn how to construct an electronic portfolio that will exhibit their progress and achievement. The contents of the portfolio must include student participation in the selection as well as provide evidence of self-reflection. This study researches a computer-based portfolio, which uses HyperStudio software. This program will allow students to create sound, video, graphics, and animation during the construction process. Apoptosis has been implicated in mediating the process of muscle loss during muscle disuse and with aging. However, the physiologic role of apoptosis in muscle remodeling is largely unknown. The purposes of this dissertation were to examine the role of apoptosis in different muscle remodeling conditions including muscle denervation, hindlimb suspension, muscle unloading following overload, endurance treadmill training, and muscle overload in mice, rats, or quails. Recent surveys have indicated that populations of fishes and crayfish are small in the Stony River, Grant Co., WV. The Stony River is influenced by thermal enrichment and acid mine drainage (AMD) within a 13 km stream reach, downstream of Mt. Storm Lake. In an attempt to determine the influence of each anthropogenic disturbance on these affected taxa, studies of their distribution and survival were undertaken. A crayfish trapping survey was conducted along a gradient of thermal and AMD influences in the Stony River system. This paper presents the findings of an investigation to determine whether of not there is a difference in hospitalized adolescents and non-hospitalized adolescents. The participants were 60 adolescents between the ages of 12 to 18 years. Their fears, stressors, and coping skills were evaluated by using three instruments: a 35-item self-report test on coping; a 108-item self-report test on fears; and a 15-item self-report test on stress. The results revealed that on specific measures hospitalized adolescents exhibited a statistically significantly less degree of emotional upset during hospitalization than did the community sample. Consumer confidence impacts agricultural markets. In response, many food industries have adopted cost-effective programs to reduce risk and offer third party verification of product quality. The purpose of this project was to implement the West Virginia Dairy Quality Assurance Program (WVDQAP) which promotes best management practices to produce high quality milk and meat products. This study surveyed dairy producers throughout the state, to establish the most effective method of program implementation. Students have been responding to art using critical inquiry, but there has been relatively nothing written in regard to poetic expression and non-verbal responding in the teaching of art interpretation. The genes of the endothelin system are the potential targets for regulation by prostaglandin F2α (PGF2α) and have been studied in greater detail. The effects of a luteolytic dose of PGF2α on the mRNA encoding endothelin converting enzyme-1 (ECE1), pre-pro endothelin-1 (ET-1) and the ET receptors ETA, ETB, and in bovine corpus luteum (CL) collected during the early, mid or late luteal phases were examined. The effects of treatment with PGF2α on ECE-1 protein, big ET-1 and the biologically active mature ET-1 were examined. Most importantly, the direct activity of ECE-1 was determined. Low grain prices and recurrent periods of drought have elicited interest in early weaning. Calves can efficiently utilize feed for weight gain when it is fed directly, rather than through their dams. This study evaluated calf response in three weaning systems (experiment 1) and profiles of body energy reserves in early-weaned beef cows (experiment 2). Treatment x year, and calf sex x year interaction (P < 0.05) were observed for calf weight gain. These interactions underscored the need for constant readjustments in management systems that incorporate early weaning in their programs. In experiment 2, body weight and condition score of cows were affected marginally (P = 0.06) by early weaning, whereas ribfat and rumpfat differed (P < 0.05) between earlyweaned and control cows. These findings indicated that subjective methods (body weight and condition score) should be used in combination with more objective measurements (ribfat and/or rumpfat) for accurate evaluation of energy status of beef cows. Cows of various ages responded differently to early weaning. Three to five-year-old cows exhibited greater response to early weaning as opposed to 2-year-olds and cows > 5 years of age, indicating that profit potential of the cow-calf operation can be improved by altered management of middle-aged cows.

Roads and culverts have many effects on stream salamanders. We examined 120 culvert crossings in the Shavers Fork and Dry Fork watersheds in eastern West Virginia. Complete barriers to salamander passage occurred at 55.0% of the sites visited and partial barriers at 34.2%. Analyses showed that 20.6% of the total stream length in the Dry Fork watershed and 18.4% in the Shavers Fork watershed were isolated by barriers. We conducted salamander sampling at 16 streams and used Akaikeís Information Criterion to select the best a priori models explaining salamander diversity, richness, and abundance. Roads benefited disturbance tolerant species while negatively …

I investigated whether seedlings of non-agricultural plants exposed to varying levels of UV-B exposure had altered growth, physiological, and tissue chemistry responses under changing environmental conditions. In a field experiment, using a modulated UV-B lamp bank system, elevated UV-B did not significantly alter the total biomass of the 6 plant species that were studied, and only Quercus rubra showed a trend towards some sensitivity to UV-B. In two species—Platanus occidentalis and Setaria faberii—I found an increase in the production of UV-B absorbing compounds under elevated UV-B while two grasses—S. faberii and Elymus virginicus—had significantly decreased C:N ratios under elevated UV-B. …

Therefore, it stands to reason that production of protective mucosal immunity is a desirable outcome following vaccination for those infections. However, only a few mucosal vaccines have been developed to date, in part because of a general lack of understanding of mucosal immunity. Cytochromes P450 comprise a ubiquitous superfamily of heme-thiolate enzymes, considered among the most versatile biological catalysts known. Structure-function relationships of P450s are of particular interest to those attempting to elucidate such aspects of xenobiotic metabolism as substrate selectivity and regiospecific oxidations. We studied two members of the P450 1A subfamily, P450 1A1 and 1A2, found in highest abundance in lung and liver tissues, respectively. Rx is a paired-like homeodomain transcription factor that is expressed in the developing eyes, ventral forebrain, hypothalamus, posterior pituitary, and the pineal gland. In adults, Rx is primarily expressed in the neural retina, ciliary bodies, pineal gland, hypothalamus, and posterior pituitary. Rx mutations in multiple species are associated with anophthalmia. During the course if this work, we identified a human patient with anophthalmia and sclerocornea that carries mutations in the RX gene. CYP1A1 metabolizes polycyclic aromatic hydrocarbons (PAHs), such as those in cigarettes, to reactive intermediates which interact with DNA and lead to cancer. Silica is a lung carcinogen. Epidemiology studies of silica and lung cancer are not all positive. A possible explanation for these inconsistencies may be that silica is a modifier of PAH metabolism and, thus, cigarette smoke carcinogenesis. We hypothesize that crystalline silica exposure alters CYP1A1 expression, thereby modifying lung cancer risk. Prechondrocytes exhibit a change in their actin cytoskeletal arrangement when subjected to loading. This study looks at the implications of the altered solid-fluid interactions due to this change. A 2-D Finite element model of a cell was developed. A solid-fluid interaction solution procedure was validated and used to determine fluid flow/pressure in a cell when subject to deformation. These values were then used to calculate fluid flow induced stresses on individual actin fibers. The results suggest that prechondrocytes alter their cytoskeletal structure to reduce stresses as a result of fluid flow due to deformation of the cell.

American ginseng (Panax quinquefolius L.) is a long-lived understory herb considered rare within its range. Harvesting pressures and habitat degradation/destruction are thought to be the primary causes of population decline although others factors such as browsing by white-tailed deer (Odocoileus virginianus Zimm.) have not yet been explored. To understand the effects of deer browsing on natural ginseng populations, my research focused on four specific objectives. Twelve rats were exposed to two variable-time schedules that differed in their rates of food delivery. The "imposed" schedule was normally in effect, but lever pressing occasionally switched to the "alternate" schedule for periods averaging 30 s. Increases in food frequency afforded by switching to the alternate schedule were manipulated across conditions. All rats were exposed to the same absolute food frequency increases, but the programming of these increases was varied among the rats by holding particular food delivery rates constant across conditions. Managing Classroom Behavior Of Head Start Children Using Response Cost And Token Economy Procedures, Jennifer D. Tiano

The efficacy of three behavior management techniques used in a Head Start classroom was examined. The three techniques included: (a) techniques currently used by the teacher, (b) response cost, and (c) the Level System (token economy). The current study used an ABACA single subject withdrawal design with follow-up where all conditions were implemented until stability was reached. Classroom behavior was evaluated by both behavioral observation and teacher report. Children's behavior, teacher skill, number of time-outs, and satisfaction with behavior management techniques (i.e., teacher, parent, and child) were examined. The purpose of this investigation is to obtain the geotechnical characteristics of the site and to aid in the creation of a proposed engineered wetland at Montrose, West Virginia. The tests performed in the laboratory to obtain the soil properties were: water content, Atterberg limits (liquid and plastic limits), grain size (sieve analysis and hydrometer), specific gravity, field density, and hydraulic conductivity. Tests were performed on soil samples taken from the site during installation of monitoring wells. Results from tests were used in the classification of soil. Brook trout (Salvelinus fontinalis) in the Appalachian Mountains are an important game species in West Virginia, yet angling mortality on these populations in headwater streams is poorly understood. The objectives of this study were to: (1) examine relative changes in body composition of brook trout as they fluctuate throughout the year, and to identify critical periods of survival to determine if brook trout reach critical levels of resource depletion over winter, and (2) evaluate whether angler harvest affects brook trout populations in spring in headwater streams.
