Sex dimorphisms in immunity are well-documented and have significant clinical implications. While the immunomodulatory effects of sex hormones have been extensively documented, recent recognition of the nuanced contributions of the sex chromosome complement (XX vs. XY) and the gut microbiome to immune sex dimorphisms underscores the need for a more comprehensive understanding. Within this context, the work herein focuses on unraveling the individual and collaborative impacts of the sex chromosome complement and gut microbiota on humoral immune activation. To better understand wood turtle (Glyptemys insculpta) population trends, two standardized population survey protocols were recently developed for use by state monitoring programs. The protocols were independently developed, tested, and deployed for population monitoring in the eastern and upper midwestern United States. Phosphodiesterase-6 (PDE6) serves as a pivotal component in the phototransduction pathways of both cone and rod photoreceptors. In cones, PDE6 consists of tetrameric subunits: inhibitory (γ') and catalytic (α'). The catalytic subunit, PDE6α', contains a C-terminal prenylation motif. Deletion of this motif is associated with achromatopsia (ACHM), a form of color blindness. The mechanisms underlying the disease and the roles of PDE6 lipidation in vision remain elusive. Meanwhile, rod PDE6 is composed of α and β catalytic subunits and γ inhibitory subunits, with alterations in the C-terminal "prenylation motif" of PDE6β linked to retinitis pigmentosa (RP) pathology. Higher education is experiencing a mental health epidemic (Venit, 2022). There is unprecedented student demand for psychological services that colleges are unable to meet (Lipson et al., 2019a), leading to high rates of mental illness-related attrition (Koch et al., 2018). Traumatic brain injury (TBI) is a significant health concern across the United States as well as the broader globe, affecting millions of individuals on an annual basis. Surface mining can impact land cover, forests, and water quality. Current reclamation strategies include revegetation with herbaceous species due to the ease and speed of cover establishment. Herbaceous and woody biomass crops have been used in various studies to reclaim surface mines and act as an alternative to nonrenewable energy sources. The objectives of this study were to quantify the state of vegetation growth on former surface mines in West Virginia over a 9-year period and identify suitable acreage for bioenergy production. In this dissertation, I utilize the post-structural philosophy of Gilles Deleuze and Félix Guattari as a lens for investigating the proof process. Deleuze and Guattari were both post- structural philosophers who, like many in this tradition, troubled traditional notions related to stable identities, meaning, language, and mathematics. For Deleuze, sense and meaning is not the result of a sterile, transcendent effect or condition that is associated with propositions and states of affairs. Childhood adversity has long-lasting negative effects across the lifespan including increased mortality risk. The love and support individuals receive from others, also known as social support, has shown to be a protective factor against ACEs. However, little research has investigated the amplifying effects of social conflict and strain that often accompanies social relationships. Utilizing data from the Midlife Development in the U.S. (MIDUS) study, I tested whether higher levels of social support would buffer the negative effects of adverse childhood experiences on mortality risk, and whether higher levels of social strain would amplify these associations. In recent history, natural, meandering streams have been straightened and dredged to reduce flooding. While this practice can be effective in reducing flooding locally, it often results in the degradation of stream water quality and aquatic ecosystems. A straighter channel inherently increases the stream gradient, which could increase flow velocity, shear stress, and potentially downstream sediment yield. Studies have shown that straightened, channelized streams often begin to return to a meandering pattern 35-50 years post-channelization. An excretory/secretory (E/S) product, common to many helminths, is neutrophil inhibitory factor (NIF) which negatively affects neutrophil migration and activity. Due to differences in neutrophil accumulation between parasite resistant St. Croix STC and parasite susceptible Suffolk (SUF) sheep, we hypothesized that Hc-NIF may inhibit migration of SUF and not STC neutrophils. Previous studies have demonstrated that Hc produces NIF but is not commercially available. To test our hypothesis, neutrophils were cultured from STC and SUF sheep in the presence of Ancylostoma caninum-derived NIF (Ac-NIF) and measure chemotaxis to Interleukin-8 (IL-8), HcLA or H. contortus third-stage excretory/secretory (E/S) products. The neural mechanisms that control the onset of puberty are not completely understood. In livestock, the onset of puberty relies on an increase in gonadotropin releasing hormone (GnRH) and luteinizing hormone (LH). Kisspeptin, neurokinin B, and dynorphin neurons (KNDy) and their receptors, Kiss1r, NK3R, and KOR, respectively, are important regulators of puberty. Background: Nephrotoxin exposure may worsen kidney injury and impair kidney recovery if continued in patients with acute kidney injury (AKI).

Objective: To determine if tiered implementation of a clinical decision support system (CDSS) would reduce nephrotoxin use in cardiac surgery patients with AKI.

Methods: We assessed patients admitted to the cardiac surgery intensive care unit at a tertiary care center from January 2020 to December 2021, and August 2022 to September 2023. The relative effort of target and alternative responses during treatments using differential reinforcement of alternative behavior may impact the likelihood that a previously reduced target response will reemerge following a context change (i.e., “renewal”). Ecosystem services provided by dung-associated beetles, such as reducing pasture fouling, parasite suppression, supporting nutrient recycling, and increasing water infiltration, contribute to animal and pasture health. However, parasiticides used to manage cattle parasites, particularly flies and helminths, in West Virginia, can adversely affect these non-target beetles. We assessed the exposure and impact of some parasiticides used in WV including Clarifly® (active ingredient [a.i.] diflubenzuron), Cydectin® (a.i. moxidectin), LongRange® (a.i. eprinomectin), and Safe-guard® (a.i. fenbendazole). Background: Clostridium difficile infection (CDI) is a prevalent infection often contracted in hospitals. CDI is also increasingly originating from community settings. In addition to the health risks associated with CDI, diagnosis of a hospital-acquired CDI can have a significant financial impact on hospital systems. When hospital acquired, hospital systems are no longer reimbursed by payers. Secondary to the increasing prevalence of community-acquired CDI, mechanisms for early detection and isolation are warranted.

Microtubules are ubiquitous cytoskeletal filaments essential in the function of eukaryotic cells. These polymers, comprised of αβ-tubulin heterodimers, are critical for cell division, intracellular transport, and formation of the axonemes of cilia and flagella. This functional diversity of microtubules is attributed to the incorporation of various α- and β-tubulin isotypes encoded by different genes. Indeed, mutations in human tubulin genes have been linked to various tissue-specific diseases, suggesting a unique function for each tubulin isotype. Study 1. Including exogenous enzymes in poultry diets can liberate nutrients that are typically inaccessible to monogastric animals. Phytase feeding research has traditionally been conducted by rearing broilers in raised wire cages, whereas commercially grown broilers are reared on built-up litter. Therefore, cage-based studies may not accurately represent commercial bird performance response to phytase. T helper (Th) 17 cells mediate the pathogenesis of MS and its animal model, experimental autoimmune encephalomyelitis (EAE), in part by infiltrating the CNS and interacting with astrocytes, a resident glial cell. In population modeling, dynamic processes are frequently represented by systems of differential equations, allowing for the analysis of various phenomena. The first application explores modeling cloned hematopoiesis in chronic myeloid leukemia (CML) via a nonlinear system of differential equations. By tracking the evolution of different cell compartments, including cycling and quiescent stem cells, progenitor cells, differentiated cells, and terminally differentiated cells, the model captures the transition from normal hematopoiesis to the chronic and accelerated-acute phases of CML. In Chapter 2, a study was designed to evaluate the effects of varying dietary branched-chain amino acids (BCAA) ratio in corn and soybean meal-based turkey starter diets. The BCAA leucine, isoleucine, and valine are considered indispensable amino acids needed for energy and stimulation of muscle protein synthesis via the mammalian target of rapamycin (mTOR) pathway. Imbalanced BCAA ratios can lead to antagonism and degradation of limiting BCAA, particularly in diets with excess leucine. Synaptic zinc signaling modulates synaptic activity and is present in specific populations of cortical neurons, suggesting that synaptic zinc contributes to the diversity of intracortical synaptic microcircuits and their auditory tuning properties. Stimulus-specific adaptation is a hallmark of sensory processing in which a repeated stimulus results in diminished successive neuronal responses, but a deviant stimulus will still elicit robust responses from the same neurons. Recent work has established that synaptically released zinc is an endogenous mechanism that shapes neuronal responses to sounds in the auditory cortex. Memory recall and forgetting is a complex process. Memories can be completely forgotten (e.g., Ebbinghaus, 1885/1964; Murre, 2015) or repressed and recovered at a later time (e.g., Key et al., 1996; Foster-Lee et al., 1999). Instances of sexual assault may be disclosed to a close acquaintance or medical professional at the time of the event or kept secret until the time of reporting to authorities (e.g., Pica et al., 2020a).
