Glioblastoma multiforme (GBM) is an aggressive cancer that affects millions of patients per year. Conventional therapies combining chemotherapeutic agents with radiation can only extend survival by a few months; therefore, there is a dire need for an effective means of treating this deadly disease. Melanoma differentiation-associated gene-7/interleukin-24 (mda-7/IL-24), currently in the early stages of FDA pre-IND drug trials, has proven to be an effective cancer specific cytokine, able to trigger the onset of mitochondrial dysfunction and/or autophagy. Decades of work has led researchers to believe that risk for complex behavioral phenotypes, such as alcohol use disorders, is likely influenced by multiple genes of small effect acting in conjunction with each other and the environment. Currently, the field of psychiatric genetics is developing methodologies for the identification of genetic risk variants that predispose individuals to the development of complex behavioral disorders. Several challenges related to the complex and polygenic nature of these phenotypes, must be considered. The chick chorio-allantoic membrane (CAM) contains a complex vascular network commonly used to study angiogenesis. The application of chemical factors and oxygen barrier films onto this tissue can easily influence the process of angiogenesis. In this study, oxygen barrier film patches (Krehalon, polyvinylidene chloride, 12 μm thick, O2 transmission rate = 2.19 cm3•ml/100 in2•day•atm) were applied to areas of the CAM. Holes were made in the film and alginate beads incubated in various chemical factors were placed in the holes. Mitf and Otx transcription factors have been identified as essential to the development of the retinal pigmented epithelium (RPE), but the relationship between these factors and their specific role in the RPE developmental pathway have not been clearly defined. The role of the two Mitf transcription factors (Mitfa and Mitfb) and two Otx transcription factors (Otx1a and Otx2) in zebrafish RPE development was explored in these experiments. The loss of Mitf activity in mitfa, mitfb, or double mitf null mutant fish lines had no effect on RPE pigmentation or development. The loss of Otx2 activity through morpholino knockdown produced a …

An agent based model was developed to examine the growth of a tumor in a healthy cell population. Response to radiation and impact of mutation and bystander effects were studied. In the growth model, the cancer cells proliferated outward becoming invasive. The mass of cancer cells developed a necrotic core. Various treatment protocols of radiation were compared. Timing of treatments was critical to the success of therapy. The event of mutation was rare. When mutation occurred, either unsuccessful treatment or re-growth could result. Multiple rounds of radiation potentially led to increased mutation. Low levels of the bystander effect had little …

The α7 nicotinic acetylcholine receptor (nAChR) subtype is abundantly expressed in the central nervous system (CNS) and in the periphery. Positive allosteric modulators (PAMs) of the α7 increase the response to an agonist and are divided into two types depending on whether they also decrease desensitization of the receptor (type II) or not (type I). Therefore, this study aims to investigate whether the enhancement of endogenous α7 nAChR function will result in a beneficial effect in nociceptive, inflammatory and chronic neuropathic pain models. The human pathogen Trypanosoma cruzi is an intracellular parasite and the etiological agent of Chagas disease. Protective immune responses to T. cruzi are highly dependent on T helper 1 and CD8+ T cells which produce interferon-gamma. A deficiency in these responses has severe consequences on the ability to control infection. Our investigation into the role of the Th1 transcription factor, T-bet, during murine T. cruzi infection revealed that T-bet is required for resistance. Contrary to our expectations, T-bet was not required for the development of Th1 immunity during infection, as T-bet-deficient mice still developed interferon-gamma-producing T cells. Accelerated tumor cell repopulation during radiation therapy is one of the leading causes for low survival rates of head-and-neck cancer patients. The therapeutic effectiveness of radiotherapy could be improved by selectively targeting proliferating tumor subvolumes with higher doses of radiation. Positron emission tomography (PET) imaging with 3´-deoxy-3´-[18F]-fluorothymidine (FLT) has shown great potential as a non-invasive approach to characterizing the proliferation status of tumors. This thesis focuses on histopathological validation of FLT PET imaging specifically for image-guidance applications in biologically adaptive radiotherapy. Despite substantial research literature on the positive relationship between Black racial identity and psychological wellbeing, Black-White biracial individuals are underrepresented in studies that provide evidence for this relationship. In obesity research, it is assumed that the population is homogeneous. While this approach has yielded important insights, testing this supposition might reveal information that could impact our understanding of the phenomena and its treatment. People agree that forgiveness is a virtue in essentially all countries. However, different cultures have different ideas about how willing one should forgive and under what circumstances. Although the study occurred in the USA, I recruited both foreign-extraction and Virginia born-and-raised female college students (N=102) to participate a six-hour REACH forgiveness intervention, promoting their forgiveness through psychoeducational groups. In my thesis, I investigated whether students of foreign extraction and Virginia-born students would respond similarly to the intervention. Surfaces with static contact angle greater than 150 degrees are typically classified as superhydrophobic. Such coatings have been inspired by the lotus leaf. As water flows over a superhydrophobic surface, "slip effect" is produced resulting in a reduction in the skin-friction drag exerted on the surface. Slip flow is caused by the entrapment of a layer of air between water and the surface. Superhydrophobicity could be utilized to design surfaces for applications such as energy conservation, noise reduction, laminar-to-turbulent-transition delay, and mixing enhancement. The bacterial ribosome is essential to cell growth yet little is known about how its proteins attain their mature structures. Recent studies indicate that certain Staphlyococcus aureus bacteriophage protein sequences contain specific sites that may be cleaved by a non-bacteriophage enzyme (Poliakov et al. 2008). The phage cleavage site was found to bear sequence similarity to the N-terminus of S. aureus ribosomal protein L27. Previous studies in E. coli (Wower et al.1998; Maguire et al. 2005) found that L27 is situated adjacent to the ribosomal peptidyl transferase site, where it likely aids in new peptide formation. Understanding the impacts of explosive contamination on vegetation is key to understanding explosives behavior in the environment. I quantified shrub growth responses to 1,3,5-trinitroperhydro-1,35-triazine (RDX) soil contamination across three life stages and I hypothesized that RDX would have the greatest impact on seed germination. Morella cerifera seeds were germinated on soils amended with RDX up to 1500 mg RDX kg-1 dry soil. Juvenile and adult individuals were exposed for 6 weeks to soil amended with RDX up to 750 and 1500 mg RDX kg-1 dry soil, respectively. PDMS is commonly described as a soft polymer with very appealing chemical and physical properties such as optical transparency, low permeability to water, elasticity, low electrical conductivity, and flexible surface chemistry. PDMS microfluidic device fabrication is done easily with the use of soft lithography and rapid prototyping. Mechanical and chemical debridement plays an important role in reducing intracanal microorganisms. Effective root canal irrigation depends on both the root canal irrigant and irrigation system. The objective of this study was to evaluate the debridement efficiency of four root canal irrigation systems, Endovac®, PiezoFlow™, EndoActivator® and traditional needle irrigation using saline as an irrigant. Colonizing vegetation was assessed over three growing seasons in both tidal and non-tidal environments. It uses pilgrimage accounts from medieval miracle story collections and to gather information about individual travel patterns. The study uses GIS to analyze gendered mobility patterns, and to investigate whether there were noticeable differences in the distance which men and women traveled and the geographical area of the country they originated. Filamentous bacteriophages were engineered to express foreign genes with the ultimate purpose of displaying transmission control anti-malarial peptides as in phage display. It was hypothesized that expression of foreign genes would be possible using the phage’s promoters. This hypothesis was tested by assuming that promoters for the phage major coat protein (MCP) gene would also promote the expression of any foreign gene inserted downstream of the MCP gene. As proof of principle, the bacteriophages Pf3, Pf1, and M13 were engineered in this way to successfully synthesize Enhanced Green Fluorescent Protein (EGFP). Radiation therapy is a widely used tool in cancer therapy and is frequently offered as the first line of treatment for cancers of the breast. While radiotherapy is often initially effective in killing tumor cells or suppressing their growth, there are factors that confer tumor cell resistance to irradiation. Development of resistance may lead to disease recurrence despite the use of surgery, chemotherapy and radiation therapy.
