Observations Of Lens Epithelium In Cell Cultures, Walter J. Geeraets

The following investigation had been conducted to evaluate lens epithelial cells in vitro as a potential suitable model for future studies of chronic low dose ionizing radiation effects on this cell type in vitro. Since there is a great range of dose requirements to produce in vivo radiation cataract in man, monkeys, and rabbits, and an equally wide range in time from radiation exposure until the first minimal lens changes can be biomicroscopically detected, it may be assumed that in vitro comparison of cells derived from these three species may provide some explanation for the in vivo differences.

What I want to discuss are not the potential therapeutic or diagnostic uses of these newly available compounds, which are really very speculative, but certain features of the physiological control system in which these compounds are thought to play key roles. Any rational system of therapeutics or prophylaxis involving these releasing factors, or their derivatives, must be based on adequate understanding of (1) the physiological roles of these compounds, (2) how these roles may be altered by disease, and (3) the effects specific disturbances of the control system(s) in which they operate can be expected to have on reproductive processes.

The subject of my discussion is prenatal diagnosis of genetic disorders of a metabolic rather than a chromosomal nature. Whereas the chromosomal defects are the result of either the transmission of a translocated chromosome from a single parent to its offspring or an error in meiosis or mitosis, almost all metabolic disorders are inherited in an autosomal recessive fashion. Therefore, except in rare instances, the only clue to the possible presence of an inborn error of metabolism (IEM) in the fetus is that a previous child of the parents has had the disorder. The proper use of mechanical ventilators is related to many problems; however, the primary one is the knowledge of the physician, nurse, inhalation therapist, and other paramedical personnel with a given ventilator. It is important to remember that there are certain inherent problem with all ventilators. These problem must be carefully searched for and avoided.

Public education, although beset with criticism from its inception, is in the midst of weathering its most severe storm. One need only note the newspaper headlines for a week to see the myriad of demands being placed upon the schools. On one hand, there is increased demand for productivity--what Talcott Parsons, in his book, The Social System, would call instrumental education (education for usefulness)--and on the other, there is the insistence upon strengthening humanism in teaching and learning--again drawing on Talcott Parsons--called expressive education (education for self-fulfillment) (1). In recent years renal homotransplantation has become more frequently used in the treatment of terminal renal disease in man. By carefully selecting donors with the aid of tissue compatibility tests and employing immunosuppressive drugs, these transplants have become increasingly more successful. It is true, however, that even with good tissue match-up and drug treatment rejection episodes occur, frequently within the first two-postsurgical weeks. The majority of these rejection crises may be reversed by increasing drug dosages and occasionally applying x-irradiation to the area of the transplant. In other cases rejection by the recipient is responsible for cessation of function of …

The access of drugs to variofis sites of action in the body is impeded by a succession of membranes. Likewise, the removal of a drug or foreign substance from the body is similarly dependent on the ability of the substance under consideration to permeate biological barriers. A con-committant problem generally arises concerning the overall time course of drug action: that of drug storage through binding or similar processes.
