Human papillomavirus (HPV) is the most common sexually transmitted infection in the United States; yet, despite the availability of safe and effective HPV vaccines, only half of eligible adolescents have completed the vaccine series. School-entry requirements are one proven strategy to increase vaccination rates among children and youth and reduce the burden of HPV-related cancer. This study investigated community perceptions of an HPV vaccine school-entry mandate in Virginia and the consequences of a low threshold exemption option included in the legislation. We conducted 40 interviews with community members including 15 interviews with parents, 19 with healthcare providers, and 6 with …

Sphingosine Kinase 2 Depletion Dysregulates Hepatic Redox Metabolism And Enhances Inflammation In Early Metabolic Dysfunction-Associated Steatohepatitis (Mash), Kaitlyn Georgene Jackson
Theses and Dissertations
Metabolic dysfunction-associated steatotic liver disease (MASLD) currently affects one in three people worldwide; its prevalence is growing alongside other cardiometabolic disorders. Aberrations to redox and sphingolipid metabolism contribute to MASLD pathologic progression. Sphingosine-1-phosphate (S1P) is an active lipid, meaning that S1P participates in biochemical processes within the cell. S1P is generated by two distinct isoenzymes, sphingosine kinase (SphK)1 and SphK2. The localization of the SphK enzyme dictates S1P function. While cytosolic SphK1-generated S1P is well-characterized, nuclear and mitochondrial SphK2-generated S1P is less known. Moreover, SphK2 depletion effects on cellular redox processes and inflammation are not well-defined.
This study defines the …
The Role Of Hdac6 In Periodontal Inflammation, Hannah S. Lohner
The Role Of Hdac6 In Periodontal Inflammation, Hannah S. Lohner
Theses and Dissertations
Periodontal disease is a polymicrobial infection-induced chronic inflammatory disease and results in irreversible tissue damage and bone loss. While bacteria play a large role in the initiation of this disease, excessive and prolonged inflammatory immune responses are fundamental for its progression, making host directed therapy (HDT) a possible and potentially valuable treatment. Histone Deacetylase 6 (HDAC6) is an enzyme that removes acetyl groups from both histone and non-histone proteins, giving it a potential role in regulating inflammation initiated by pathogenic oral bacteria. Previous studies indicated that HDACs act as a bivalent regulator in inflammatory responses; however, the role that HDAC6 …
Using The Microbiome To Assess The Health Status Of The Individuals Recovered From The East Marshall Street Well, Arian Karim
Using The Microbiome To Assess The Health Status Of The Individuals Recovered From The East Marshall Street Well, Arian Karim
Master of Science in Forensic Science Directed Research Projects
In 1994, during the construction of the Kontos Building on the Medical College of Virginia Campus, a well on East Marshall Street was discovered to contain artifacts and human remains of African descent. Upon archaeological analysis by the Smithsonian, the ancestral remains can be dated back to the mid-19th century, when grave robbing and the use of cadavers for anatomical purposes [BS1] in medical schools were common. The human oral microbiome is the second largest human microbiota, containing over 600 species of organisms. Dysbiosis of the oral microbiome can lead to the overgrowth of pathogenic species, resulting in illness and …
The Determination Of Ethanol And Phosphatidylethanol (Peth) In Donated Packed Red Blood Cells, Jordan E. Keller
The Determination Of Ethanol And Phosphatidylethanol (Peth) In Donated Packed Red Blood Cells, Jordan E. Keller
Master of Science in Forensic Science Directed Research Projects
This research investigates the safety and reliability of blood donations by examining the presence of ethanol and phosphatidylethanol (PEth) in packed red blood cells (pRBCs). As blood donation is integral to healthcare, ensuring the safety of blood products is paramount. Current protocols focus on screening for infectious diseases, such as hepatitis C and human immunodeficiency virus (HIV), but do not routinely asses alcohol consumption, which can significantly impact patient outcomes. As a direct indicator of alcohol consumption, ethanol can affect blood quality, interfere with medical treatments, and present legal issues, such as false conclusions or assumptions. Ethanol detection methods include …

Mindfulness, Moral Outrage, And The Climate Crisis: A Novel Approach Toward Motivating Pro-Environmental Behaviors, Erika Blair
Mindfulness, Moral Outrage, And The Climate Crisis: A Novel Approach Toward Motivating Pro-Environmental Behaviors, Erika Blair
Theses and Dissertations
The consequences of anthropogenic climate change are severe, continuously accelerating, and pose serious threats to the human and natural world. It is imperative to address the climate crisis from a psychosocial perspective and incite lasting, individual-level behavioral change. Various theoretical models have been adapted or developed to explain pro-environmental behaviors. However, the predictive powers of the most commonly used models have proven to be limited, and the underlying psychological processes of pro-environmental behavior constitute a significant gap in the literature. The present study identifies two psychological processes that show significant promise in motivating pro-environmental behaviors: moral outrage and mindfulness. Moral …
Mathematical Modeling Of Phage-Bacteria Population Dynamics, John Lawrence D. Palacios
Mathematical Modeling Of Phage-Bacteria Population Dynamics, John Lawrence D. Palacios
Theses and Dissertations
Bacteriophages are viruses that infect and replicate within bacteria. Lytic phages cause the bacterial cell to burst, killing the bacteria. These types of phages can be used to treat patients with antibiotic-resistant bacterial infections. As a step in developing successful treatment protocols, we aim to understand the population dynamics of phages and bacteria using an in vitro model. We model the dynamics using the Campbell model, which consists of a delay differential equation (DDE), as a base model. We extended the model by including the emergence of phage resistance. We then compared the DDE model with a parallel ordinary differential …

Interactions Between Hiv And Opioids On Antiretroviral Accumulation, The Blood Brain Barrier, And The Inflammatory Response In The Brain., Kara Rademeyer
Theses and Dissertations
The complex mechanisms related to HIV infection, neurodegeneration, and chronic neuroinflammation collectively describe neuroHIV (Hauser et al. 2007; Chang et al. 2014; Smith et al. 2014). Specifically, opioid abuse, poor penetration of antiretroviral (ARV) drugs, chronic inflammation and neuronal injury/degeneration are all implicated in neuroHIV (Fantuzzi et al. 2003; Letendre et al. 2004; Verani et al. 2005; Duncan and Sattentau 2011; Hong and Banks 2015; Simoes and Justino 2015; Olivier et al. 2018; Murphy et al. 2019; Osborne et al. 2020). For the first time, we demonstrate that morphine, fentanyl, and methadone in vivo alter the brain accumulation of ARVs, …
Activity Levels Of Different Cell Types In An Epileptogenic Model Of Cortical Microgyria: A Cfos Investigation, Peter F. Lamothe
Activity Levels Of Different Cell Types In An Epileptogenic Model Of Cortical Microgyria: A Cfos Investigation, Peter F. Lamothe
Theses and Dissertations
A large percentage of individuals with intractable epilepsies have an accompanying cortical malformation, the underlying cellular mechanisms of which are not fully understood. In an animal model for one such malformation, polymicrogyria, epileptogenesis occurs most easily in a region of tissue just adjacent to the microgyria termed the paramicrogyral region (PMR). Previous studies implicate somatostatin containing interneurons (SOM) as a potential contributor to this pathology, and show increased excitation of SOM in the PMR. We hypothesis that SOM are more active in the PMR when compared to SOM within the homologous region of the control cortex. In addition to this …

Access To Hiv Prevention, Care, And Treatment Services In The Us: The Role Of Primary Care, Jessica S. Kiernan
Access To Hiv Prevention, Care, And Treatment Services In The Us: The Role Of Primary Care, Jessica S. Kiernan
Theses and Dissertations
This dissertation examines the role of access to primary care in benefitting HIV outcomes through increased access to HIV prevention, care, and treatment services, among populations disproportionately affected by HIV in the US. Over 30,000 individuals are newly diagnosed with HIV annually in the US and diagnoses are concentrated in specific populations. At an individual-level, ~ 70% of new diagnoses are among gay and bisexual men and of those diagnoses, 70% are among Non-Hispanic Black and Hispanic gay and bisexual men. At a community-level, over 50% of new HIV diagnoses occur in less than 2% of US counties (48), Washington, …
The Hepatic Immunological Pattern Shaped By Dominant-Subdominant Cellular Interactions Creates A Collective Function Beyond The Function Of Each Cellular Constituent To Orchestrate Progression Of Hepatocellular Carcinoma, Nicholas J. Koelsch
Theses and Dissertations
Abundance of data on the role of inflammatory immune responses in the progression or inhibition of hepatocellular carcinoma (HCC) has failed to offer a curative immunotherapy for HCC. This is largely because of taking reductionist approaches and missing the collective function of the hepatic immune system by focusing on specific immune cell types. To this end, we propose that focusing on the dominant-subdominant patterns of the immune cells would allow understanding of the mechanism by which a collective immune function emerges. To identify the collective immune function through a systems immunology perspective, we performed high-throughput analysis of snRNAseq data collected …
The Impact Of Hiv-1 Tat And Morphine On Spatial Distribution Of Drugs In The Brain., Austin M. Jones
The Impact Of Hiv-1 Tat And Morphine On Spatial Distribution Of Drugs In The Brain., Austin M. Jones
Theses and Dissertations
Despite combination antiretroviral therapy effectively suppressing HIV within the periphery, the central nervous system (CNS) remains affected by the virus. Approximately half of people living with HIV will experience HIV-associated neurocognitive disorders (HAND) or neuroHIV. Concurrent opioid use exacerbates neuroHIV by promoting neuroinflammation, viral replication, and potentially altering the antiretroviral concentrations within the brain.
Using a transgenic mouse that expresses the HIV-1 Tat protein, we examined the effects of Tat and morphine on antiretroviral accumulation and distribution and the effects of Tat on morphine accumulation within the brain using infrared matrix-assisted laser desorption electrospray ionization with mass spectrometry imaging (IR-MALDESI-MSI). …
Studying Gene Function In Streptococcus Sanguinis Using Rna-Sequencing Analysis, Ahmed Ismail
Studying Gene Function In Streptococcus Sanguinis Using Rna-Sequencing Analysis, Ahmed Ismail
Theses and Dissertations
The oral cavity hosts hundreds of bacterial species, amongst them are the Streptococci family. More specifically, S. sanguinis is a benign bacteria in the oral cavity to protect the tooth surface from S. mutans. which is a pathogen causing dental caries. While S. sanguinis is harmless in the oral cavity, it becomes pathogenic once entering the bloodstream and causing Infective Endocarditis (IE). To further study S. sanguinis and eventually develop an effective drug to treat IE caused by S. sanguinis, a gene expression study was conducted. In order to study gene expression, RNA-sequencing was conducted, and appropriate methods to remove …
Transcriptional Coregulation Among Diabetes-Associated Pathways By Genome Organization, Musaddiq K. Lodi
Transcriptional Coregulation Among Diabetes-Associated Pathways By Genome Organization, Musaddiq K. Lodi
Theses and Dissertations
There are currently 96 million individuals affected by diabetes in the United States alone. Crucial information such as biomarkers, regulatory networks, and metabolic pathways in diabetes still remain an active area of research. The primary focus of this study is on a phenomena known as transcription factor co-regulation. Transcription factors (TFs) are DNA-binding proteins that can control the rate of transcription of genetic information from DNA to mRNA, hence misregulation of one of them can lead to misregulation of many of its target genes and associated pathways. In this study I propose a framework to quantify transcriptional coherence in co-regulated …

Factors Associated With Turnover Intention Of Medical Laboratory Professionals During The Covid-19 Pandemic: A Mixed Methods Approach, Keandra Walthall
Theses and Dissertations
The COVID-19 pandemic disrupted and strained healthcare systems worldwide. Frontline healthcare workers have experienced increased rates of psychological distress and burnout during the COVID-19 pandemic, which have led to high turnover intention and workforce shortages. Medical Laboratory Professionals have endured similar experiences during the COVID-19 pandemic; however, there has been limited research exploring these factors for this group of professionals. This study utilized a convergent mixed methods approach to investigate the impact of psychological distress and burnout on the turnover intentions of Medical Laboratory Professionals in the United States during the COVID-19 pandemic, as well as to explore motivational factors …
Proposing An Rna Interference (Rnai)-Based Treatment For Human Immunodeficiency Virus (Hiv) By Analyzing The Post-Transcriptional Gene Targeting Of Sars-Cov-2, Hepatitis C Virus, And A549 Lung Cancer Cells, Arjun Jagdeesh
UROP Posters
Human Immunodeficiency Virus (HIV) is a retrovirus that infects CD4+ T cell lymphocytes in humans, leading to the development of Acquired Immunodeficiency Syndrome (AIDS) if left untreated. While current treatment methods, including antiretroviral combination treatments, effectively limit HIV replication, HIV can evade these treatments due to its high mutation rate. Long-term antiretroviral treatment can also be toxic to patients, meaning patients would benefit from a new mechanism of HIV treatment. RNA interference (RNAi) is an antiviral pathway found in mammals, plants, and insects that involves a small-interfering RNA that is incorporated into a protein complex called the RNA-induced Silencing Complex …
