The formation of the myelin sheath is a crucial step during development because it enables fast and efficient propagation of signals within the limited space of the mammalian central nervous system. During the process of myelination, oligodendrocytes actively interact with the extracellular matrix (ECM). These interactions are considered crucial for proper and timely completion of the myelin sheath. However, the exact regulatory circuits involved in the signaling events that occur between the ECM and oligodendrocytes are currently not fully understood. I examined the abundance, composition, and distribution of fish communities in the lower Roanoke River, a hydropeaking system in North Carolina. Fishes were sampled at before and after peaking events over three years; 2007 to 2009. I evaluated trends in species richness, diversity, and assemblage composition. There were no significant differences in either richness or diversity suggesting consistent trends in richness and diversity throughout the study. I used non-metric multidimensional scaling (NMDS) to create a community composition model. Fish composition was noticeably greater post-peaking and changed minimally across time and event. Aging is an intrinsic process that is independent of obvious disease. In contrast to normal aging, age-related diseases are conditions that typically manifest at advanced ages, are associated with explicit pathology and cause disability and premature death. We used Drosophila as a model to investigate the molecular-genetic mechanisms associated with aging and age-related disease. Age-related locomotor impairment (ARLI) is a serious condition for the elderly and greatly impacts their quality of life. Toward identifying genes and mechanisms that influence ARLI, we performed a forward genetic screen using Drosophila mutants. This screen identified a loss of function mutant in PDK1, a …

Beginning with a former grocery store building, “An Affordable Living Community,” explores the possibilities of redesigning big box buildings. Here, the building is transformed into an affordable living community - a place for people to live, work, learn, and interact. The renovation creates a place for not only its residents, but also the surrounding neighborhoods. The building provides the challenges of breaking the generic, window-less facade, as well as bringing light into the building. This model would encourage the health, learning, and support of its community.

Worldwide, bacterial vaginosis (BV) is the most common vaginal disorder in women of childbearing age. BV is of major clinical importance due to its ability to significantly affect pregnancy outcome and enhance the transmission and acquisition of HIV. BV is characterized by a dramatic shift in the vaginal microflora; in most BV cases, the predominant bacterial species is Gardnerella vaginalis. It has been demonstrated that G. vaginalis forms an adherent biofilm on the vaginal epithelium of women with BV. Furthemore, evidence suggests that the high rate of recurrence associated with BV is related to incomplete eradication of the biofilm. The …

Dystonia is a movement disorder characterized by patterned, repetitive, and sustained muscle contractions that cause ineffective and often painful movements. The overall goal of this project was to understand the physiological mechanisms of dystonia in a rodent model as a basis for developing innovative treatments for secondary dystonias. The first half of the project was focused at developing essential techniques for systematically investigating the movement disorder in these animals. IT’S ALWAYS SOMEBODY’S PARIS: AN EXAMINATION OF PLACE IN NONFICTION WRITING By Jessica E. Sculthorpe This thesis examines the importance of place in nonfiction writing, using both the author’s personal experience as a student in Paris and the writings of other Americans in Paris, including members of the Lost Generation. The first two chapters examine the author’s experience as a young student in Paris. The third and fourth chapters contain the author’s reflection on the process of writing the thesis and an examination of the ways in which other writers have written about Paris in their own nonfiction writing.

This thesis project examined the interactive fluid dynamics between a blood pump and the univentricular Fontan circulation. 2-D particle image velocimetry (PIV) measurements were conducted on an idealized total cavopulmonary connection (TCPC) with an axial pump prototype in the inferior vena cava (IVC). Fluid velocity profiles were examined under various physiologic conditions for Fontan patients. The velocity profiles for all cases demonstrated the shunting of flow from the IVC toward the right pulmonary artery. A rotational component in the pump outflow was observed forcing flow to the periphery as compared to the flow profile without a pump present in the …

Few therapeutic alternatives exist for patients with a failing single ventricle physiology. To address this need, this thesis project investigated two new therapeutic alternatives, which sought to positively augment the Fontan hemodynamics. The first modality introduced a non-invasive method of external pressure application to the lower extremities. A clinical study (n=2) was conducted, and results indicated an increase in flow as a consequence to an increase in transmural pressure in the lower extremities. The second modality investigated a minimally invasive blood pump. Numerical analyses of the pump were performed to examine hydraulic performance under physiologic conditions. While there is currently active debate over possible therapeutic applications of marijuana and cannabis-based compounds, consistently their primary drawbacks have been the psychoactive properties, dependence, and abuse potential. Prolonged administration of ∆9-tetrahydrocannabinol (THC), the primary psychoactive constituent in marijuana, demonstrates both tolerance and physical withdrawal in both preclinical and clinical studies. Repeated THC administration also produces CB1 receptor adaptations in the form of reduced activation of receptors, along with a downregulation of membrane surface receptors, in many brain regions involved in THC-associated behaviors. The Virginia Museum of Fine Arts (VMFA) of Richmond, Virginia, is completing its largest expansion and reinstalling over 6000 artworks, including the Ancient American art collection, to be displayed in the museum’s first permanent gallery space for Ancient American art. In preparation for expansion, the VMFA issued its “Interpretive Plan Guiding Principles,” identifying visitor motivations for viewing the collections. As collection accessibility is central to the museum’s mission statement, all galleries are to provide visitors with the tools to engage with artworks. Minkisi (sing. nkisi) were sacred objects that housed ancestral spirits and were used for divination, healing and social justice by the Kongo people of Central Africa. When the Kongo were brought as slaves to the New World, they contributed significantly to the development of African American artistic and spiritual culture. In the Caribbean, aspects of minkisi have been retained in the creolized spiritual beliefs of Haitian Vodou, Cuban Palo Monte Mayombe and Brazilian Candomble. Cocaine dependence is a major health concern worldwide, but despite this high rate of abuse there are currently no approved therapies for cocaine dependence. Replacement pharmacotherapies are one possible approach for treating cocaine dependence, and identification of such therapeutics for cocaine abuse is the long-term goal of this research. Cocaine binds to, and inhibits uptake at the dopamine (DAT), serotonergic (SERT) and noradrenaline (NET) uptake transporters, but studies have shown that cocaine produces its strong behavioural and positive reinforcing effects through inhibition of the DAT. The human figure, allegory, myth, and the appropriation of other artist’s compositions are elements in my work. I aim to update traditional stories to conform to contemporary times and culture. In addition, I am striving to create a new method to visually express figurative storytelling. Breaking from the traditional flat painting surface, I use multiple shaped panels. The surface is broken into different shaped panels at varying distances from each other and from the wall. Glioblastoma multiforme (GBM) is an aggressive central nervous system (CNS) cancer characterized by enhanced tumor cell motility, pernicious invasion into the normal brain, extensive tumor-induced angiogenesis, and adaptive resistance to current therapeutic paradigms. One of the difficulties associated with GBM is the ability of the tumor cells to infiltrate normal CNS tissue. Body size is important in determining the outcome of competition and predator-prey interactions. Size structure of a population (i.e. relative proportion of large and small conspecifics) may be particularly important in organisms with prolonged breeding periods and rapid growth where populations may have multiple cohorts at different stages of development competing for one resource. Both the consumptive and nonconsumptive effects of predators can also be size-dependent and can alter competitive interactions. In risk analysis, Benchmark dose (BMD)methodology is used to quantify the risk associated with exposure to stressors such as environmental chemicals. It consists of fitting a mathematical model to the exposure data and the BMD is the dose expected to result in a pre-specified response or benchmark response (BMR). Most available exposure data are from single chemical exposure, but living objects are exposed to multiple sources of hazards. Furthermore, in some studies, researchers may observe multiple endpoints on one subject. Statistical approaches to address multiple endpoints problem can be partitioned into a dimension reduction group and a dimension preservative group. …
