Outside Things, Jacob Sorenson May 2011

Outside Things, Jacob Sorenson

Theses and Dissertations

My thesis is a description of the issues encountered during the process of research and construction of the objects leading to, and consisting of, the work shown in Outside Things. The work is my attempt to gain personal insight into the complicated relationship between nature and culture. Through abstract furniture objects, floral patterns, and robots I explore the relationship between actual landscape and the constructed man-made bio-mimicry that convolutes the definition of Nature.

Craft Cyborg, Laina Seay May 2011

Craft Cyborg, Laina Seay

Theses and Dissertations

By merging the ancient associations that clay has with the human form and prosthetic science I question the relevancy and role of the human body in the future. As prosthetics heighten the awareness of the body through absence these additive limbs further this relevancy by presence. With greater advances in genetic engineering and plastic surgery biology will no longer dominate and these ridged clay extensions could become flesh.

Analysis Of The Mechanism By Which Ykl-40 Promotes Glioma Cell Migration, Bahiya Osrah May 2011

Analysis Of The Mechanism By Which Ykl-40 Promotes Glioma Cell Migration, Bahiya Osrah

Theses and Dissertations

This thesis elucidates the crucial role of YKL-40 in enhancing glioma cell migration and invasion in vitro. Increased levels of YKL-40 are specifically associated with the increased invasive capacity of glioma multiforme (GBM) tumors and lower survival rate of GBM patients. In order to examine the effects of YKL-40 on the migration and invasion of GBM cells, we overexpressed YKL-40 in three different glioma cell lines. The overexpression of YKL-40 significantly enhanced glioma cells migration and invasion in vitro and also increased ERK phosphorylation, which is believed to enhance glioma cell survival, and invasiveness. Although receptors for YKL-40 are still …

Prenatal Alcohol Exposure Reduces Dendritic Spine Density Across Sensory Cortices, Francis Oppong May 2011

Prenatal Alcohol Exposure Reduces Dendritic Spine Density Across Sensory Cortices, Francis Oppong

Theses and Dissertations

Dendritic spines are the major site of excitatory synapses in cortex, and factors that reduce dendritic spine numbers will produce serious cortical processing deficits, such as has been demonstrated for mental retardation and other psychiatric disorders. Prenatal alcohol exposure also has detrimental effects on brain development that lead to Fetal Alcohol Spectrum Disorder (FASD), which results in reduction of dendritic spine numbers in the hippocampus, prefrontal cortex and somatosensory cortex. FASD also is associated with temporal processing disorders involving sequential auditory stimuli that would be processed in auditory cortical areas. However, it is unknown if the reduction of spine density …

Analysis Of Nfi-X3 And Stat3 Interaction And Its Functions, Etsegenet Tizazu May 2011

Analysis Of Nfi-X3 And Stat3 Interaction And Its Functions, Etsegenet Tizazu

Theses and Dissertations

YKL-40 is a secreted protein that is highly up-regulated in malignant glioblastoma (GBM). Its expression is correlated with the invasive nature of GBMs and poor diagnosis of patients (Nigro et al., 2005). Previous research has shown that in astrocytes and GBM cells, YKL-40 expression is regulated by two transcription factors, NFI-X3 and STAT3, which form a complex with each other (Singh et al., 2011). Here, we show that the N-terminal domain of NFI-X3 is sufficient and required for its interaction with STAT3. We also show that the DNA-binding domain of NFI-X3 is required to induce YKL-40 expression. Thus, the interaction …

Computational Modeling Of Mulitsensory Processing Using Network Of Spiking Neurons, Hun Ki Lim May 2011

Computational Modeling Of Mulitsensory Processing Using Network Of Spiking Neurons, Hun Ki Lim

Theses and Dissertations

Multisensory processing in the brain underlies a wide variety of perceptual phenomena, but little is known about the underlying mechanisms of how multisensory neurons are generated and how the neurons integrate sensory information from environmental events. This lack of knowledge is due to the difficulty of biological experiments to manipulate and test the characteristics of multisensory processing. By using a computational model of multisensory processing this research seeks to provide insight into the mechanisms of multisensory processing. From a computational perspective, modeling of brain functions involves not only the computational model itself but also the conceptual definition of the brain …

Meaning Maintenance In Close Relationships, Daryl R. Van Tongeren May 2011

Meaning Maintenance In Close Relationships, Daryl R. Van Tongeren

Theses and Dissertations

The human motivation for meaning has been a longstanding topic of inquiry, and recent research has begun to investigate how individuals maintain a sense of meaning in life. Given the importance of social connectedness in human life, as well as the role of relationships in providing a sense of meaningfulness, the present investigation explored meaning maintenance within the domain of close relationships. Specifically, a theoretical model for interpersonal meaning maintenance is presented, which proposes that moral actions—particularly forgiveness—are oriented toward restoring relationship quality, and, in turn, meaning in life, in the wake of interpersonal transgressions. The tenets of this model …

Turning To The Bottle For Answers: Identifying Racial Differences In Predictors Of Risk Drinking Among College Women, Melody Mickens May 2011

Turning To The Bottle For Answers: Identifying Racial Differences In Predictors Of Risk Drinking Among College Women, Melody Mickens

Theses and Dissertations

Epidemiological data suggest that alcohol use and related problems have increased among college women. The current study examined psychosocial predictors of risk drinking in a sample of college women (N=360), whether race moderated this relationship. Potential predictors included: daily smoker; premenstrual syndrome (PMS) symptom severity; age at first alcohol use, negative affect, parental history of alcohol problems and minority status. Analyses found that somatic PMS symptom severity score, age of first alcohol use, daily smoking, age of first alcohol use and non-minority status were related. Findings suggest that minority group membership was associated with low risk drinking, while somatic PMS …

Cumulative Risks To Eastern Oysters, Crassostrea Virginica In The James River, Va, Vrushali Lele May 2011

Cumulative Risks To Eastern Oysters, Crassostrea Virginica In The James River, Va, Vrushali Lele

Theses and Dissertations

In an effort to apply Cumulative Risk Assessment (CRA) as developed by the U.S. EPA, the present study investigates the cumulative risks to Eastern oysters due to multiple stressors such as salinity, temperature and oxygen and carbon dioxide. I also compared the effectiveness of the Hazard Quotient Method (HQ) in CRA. Ambient conditions in the James River, VA were obtained from the Virginia DEQ database and respiratory responses were estimated using values from the literature. The multiple environmental stresses are evaluated using a probabilistic analysis that combines the environmental conditions. It was concluded that salinity was the most influential stressor …

Defining The Bacterial Flora Of Periodontal Pockets In Chronic Periodontitis Patients, Rafael Rodriguez May 2011

Defining The Bacterial Flora Of Periodontal Pockets In Chronic Periodontitis Patients, Rafael Rodriguez

Theses and Dissertations

PURPOSE: The purpose of this study is to describe the subgingival bacterial biodiversity in untreated chronic periodontitis patients through the use of next generation 16S rRNA molecular analysis, and to determine similarities or differences between deep and shallow pockets within the same patients. METHODS: The analysis involved paired subgingival plaque samples from 24 subjects diagnosed with Generalized Moderate to Severe Chronic Periodontitis. One sample was selected from a single site having a probing depth >5 mm (i.e. Deep Site), and the other from a site with a probing depth <3mm (i.e. Shallow Site) within each subject. Bacterial DNA amplification of the V4-V6 region of the 16S rRNA was performed. The amplicons were sequenced via 454 Roche Genome Sequencer FLX System. The identified sequences were evaluated, and then compared to calculated false discovery rates. RESULTS: A total of 119 independent microbial genera were identified within the samples analyzed. Seven genera were identified to be statistically significant (p<0.05) in their association to deep or shallow sites following t-test and boot strap randomization: Actinomyces (p=0.004), Methylobacterium (p=0.028), Veillonella (p=0.028), and Rothia (p=0.038), and Streptococcus (p=0.033) in Shallow sites; while Mycoplasma (p=0.007) and Fusobacterium (p=0.016) were associated with deep sites. However, taking into account the calculated false discovery rates, it is suggested that none of the 119 microbial genera identified in this study were significantly associated with either deep nor shallow sites. CONCLUSION: The microbial genera identified within this study to be associated with deep and shallow sites follows the traditional pattern anticipated from the literature. However, the calculated false discovery rates suggest that these results may have occurred by chance and not due to a true difference.

Discriminative Stimulus Properties Of 3-Substituent Rimonabant Analogs, D. Matthew Walentiny Apr 2011

Discriminative Stimulus Properties Of 3-Substituent Rimonabant Analogs, D. Matthew Walentiny

Theses and Dissertations

Cannabinoid agonists (e.g., THC) dose-dependently decrease locomotor activity and body temperature and produce antinociception and catalepsy. Drugs that produce this tetrad of effects within a limited dose range are likely to function as CB1 receptor agonists. A structure activity relationship study from our laboratory investigating analogs of the CB1 antagonist rimonabant revealed that certain alterations in the 3-substituent of rimonabant’s pyrazole core conferred agonist-like properties in the tetrad. Interestingly, these effects were present in CB1 -/- mice, and were not reversed by rimonabant in wild-type mice. The present study evaluated two novel 3-substituent rimonabant analogs, O-6629 and O-6658 in the …

Phytoremediation Of Pcb Contaminated Soil: Effectiveness And Regulatory Policy, Nathan Pinsker Apr 2011

Phytoremediation Of Pcb Contaminated Soil: Effectiveness And Regulatory Policy, Nathan Pinsker

Theses and Dissertations

The purpose of this paper was to combine available peer-reviewed literature on PCB phytoremediation and make direct comparisons using ANCOVA statistics in order to determine if and what plants are presently a viable option for the remediation of PCB contaminated soils. Pumpkin (Curbita pepo cv Howden) consistently had the top root and shoot concentrations, as well as total plant accumulation. Their consistency shows that they can be reliable in the field and the most viable option. Tall fescue and sedge were also top contenders. Due to the small sample size for many plants and accounting for several confounding variables, very …

Can Interior Design Erase Architecture? The Integration Of A Pet Care Facility Into The Fan, Erin Brunner Apr 2011

Can Interior Design Erase Architecture? The Integration Of A Pet Care Facility Into The Fan, Erin Brunner

Theses and Dissertations

Many urban pet care and boarding facilities tend to rest on the outskirts of the community, in quasi-industrial and commercial areas. Far from the homes of the people who would use their services, the locations of these facilities are inconvenient for most. Students and the employed have trouble making time in their hectic schedules to give their pets the attention and activity they need to be healthy, and often, pets are left alone, sedentary, for a substantial portion of the day. But what if there was a pet care facility that was within walking distance from home that could provide …

Distinct T Cell Clones Are Associated With Graft-Versus-Host Disease (Gvhd), And Potentially Graft-Versus-Tumor (Gvt), Responses Following Allogeneic Stem Cell Transplantation, Jennifer Berrie Apr 2011

Distinct T Cell Clones Are Associated With Graft-Versus-Host Disease (Gvhd), And Potentially Graft-Versus-Tumor (Gvt), Responses Following Allogeneic Stem Cell Transplantation, Jennifer Berrie

Theses and Dissertations

In patients undergoing hematopoietic stem cell transplantation (HSCT) with HLA-identical donors, genetic polymorphisms result in a mismatch between donors and recipients in their minor histocompatibility antigens (mHAgs), and tumors may also express tumor-associated antigens (TAA) that may not be abundantly present in the donors. Donor T cells can recognize such mHAgs and TAAs as foreign antigens and generate an objective response against hematologic malignancies in a graft-versus-tumor (GVT) effect. However, a major side effect of HSCT occurs when donor T cells are alloreactive against the recipients’ normal cells, leading to graft-versus-host disease (GVHD). The ability to identify T cell clones …

Sulfatide Is Required For Organization Of The Paranode In The Myelinated Axon In The Peripheral Nervous System, Eva Kwong Apr 2011

Sulfatide Is Required For Organization Of The Paranode In The Myelinated Axon In The Peripheral Nervous System, Eva Kwong

Theses and Dissertations

Myelin facilitates the timely, efficient conduction of action potentials along axons. Made by Schwann cells (SCs) in the PNS, myelin is unique in that it is composed of a high percentage of lipids, particularly galactolipids. Sulfatide, one such galactolipid, is made by cerebroside sulfotransferase (CST) and has been shown to play a role in organizing paranodal domains in myelinated axons. However less is known regarding the involvement of sulfatide in the establishment and maintenance of the node, of particular interest as it is responsible for the potentiation of action potentials along the axon. Using immunohistochemical and, to a lesser extent, …

Role Of Two-Component System Response Regulators In Virulence Of Streptococcus Pneumoniae Tigr4 In Infective Endocarditis, My Trinh Apr 2011

Role Of Two-Component System Response Regulators In Virulence Of Streptococcus Pneumoniae Tigr4 In Infective Endocarditis, My Trinh

Theses and Dissertations

Streptococci resident in the oral cavity have been linked to infective endocarditis (IE). While viridans streptococci are commonly studied and associated with IE, less research has been focused on Streptococcus pneumoniae. Two-component systems (TCSs), consisting of a histidine kinase (HK) protein and response regulator (RR) protein, are bacterial signaling systems that may mediate S. pneumoniae TIGR4 strain virulence in IE. To test this hypothesis, TCS RR mutants of TIGR4 were examined in vivo through use of rabbit models. There were 14 RR proteins identified and 13 RR mutants synthesized because SP_1227 was found to be essential. The requirement of the …

Processing And Classification Of Physiological Signals Using Wavelet Transform And Machine Learning Algorithms, Abed Al-Raoof Bsoul Apr 2011

Processing And Classification Of Physiological Signals Using Wavelet Transform And Machine Learning Algorithms, Abed Al-Raoof Bsoul

Theses and Dissertations

Over the last century, physiological signals have been broadly analyzed and processed not only to assess the function of the human physiology, but also to better diagnose illnesses or injuries and provide treatment options for patients. In particular, Electrocardiogram (ECG), blood pressure (BP) and impedance are among the most important biomedical signals processed and analyzed. The majority of studies that utilize these signals attempt to diagnose important irregularities such as arrhythmia or blood loss by processing one of these signals. However, the relationship between them is not yet fully studied using computational methods. Therefore, a system that extract and combine …

Resolution, Jason Robinson Apr 2011

Resolution, Jason Robinson

Theses and Dissertations

This paper is an exploration of the concepts and motivations behind the video installation and performance that made up my MFA Thesis Exhibition. My primary interest as a filmmaker is in the “invisible art” of editing. In my practice I employ iconic imagery such as old home movies and classic Hollywood films, as my source material. This “found footage” is deconstructed, drastically altered, and reassembled into a new movie that tells my story while still maintaining the memory of the original images. My process and philosophy concerning the salvage and repurposing of video is analyzed and discussed. I also examine …

Targeting The Endocannabinoid System To Reduce Nociception, Lamont Booker Apr 2011

Targeting The Endocannabinoid System To Reduce Nociception, Lamont Booker

Theses and Dissertations

Pain of various etiologies (e.g., visceral, inflammatory) can be a debilitating disorder that presents a problem of clinical relevance. While it is known that ∆9-tetrahydrocannabinol (THC) the primary psychoactive constituent found in marijuana produces analgesia in various rodent models of pain, its pharmacological properties are overshadowed by its psychomimetic effects. THC is the primary phytocannabinoid found in marijuana though other prevalent constituents such as the phytocannabinoids (e.g., cannabidiol (CBD), cannabinol (CBN), cannabichromene (CBC), tetrahydrocannabivarin (THCV)) may possess antinociceptive actions without the psychomimetic effects associated with THC. Indeed, these phytocannabinoids act upon the endocannabinoid system (ECS) that is comprised of the …

Parasitic Interiors, Michael Kanasink Apr 2011

Parasitic Interiors, Michael Kanasink

Theses and Dissertations

This project is an experiment in the adaptive reuse of an Art Deco style manufacturing facility in Richmond, VA. The building has fallen into disrepair and has been inhabited by two different owners over the years, but the structure remains very much as it did when it was erected in 1946. I will propose adapting to the defunct space, a forward thinking school based on hands-on, creative learning. This school will challenge accepted pedagogies and serve as an exapmple of future learning. I have always looked as adaptive reuse interior design as a parasitic form of designing. The new program …

Electrophysiological Characterization Of Enteric Neurons Isolated From The Immortomouse, Edward G. Hawkins Apr 2011

Electrophysiological Characterization Of Enteric Neurons Isolated From The Immortomouse, Edward G. Hawkins

Theses and Dissertations

The availability of murine genetic models is extremely advantageous to studying gastrointestinal function, but the benefits afforded by studying enteric neurons in mice has been hindered by their accessibility. Fetal (E13) and 2 day post-natal (P2) enteric neuron cell lines (IM-FEN and IM-PEN, respectively) were recently developed from the H-2Kb-tsA58 immortomouse. Our goal was to identify the electrophysiological properties of these cell lines and clarify their utility as a model of enteric neurons. IM-PEN cells stained positively for the neuron specific markers βIII-tubulin and PGP9.5 and were negative for the glial cell marker S100. Detection of mRNA for TRPA1, TRPV1, …

Based On A True Story, Mary Elkins Apr 2011

Based On A True Story, Mary Elkins

Theses and Dissertations

Trying to remember is a form of forgetting. Memory fades, changes meaning, and disappears over time. While trying to find other ways to preserve stories about my family, it occurred to me that I could recreate what I remember in clay. I am creating collections of physical mementos of the memories that fill my head, focusing mainly on my childhood. Remembering is in itself an act of forgetting, and thus this is my memory preservation kit. I am recording memories of my family for posterity in clay before I have a chance to forget.

Speaking Subjects: Beckett’S Not I, Rushdie’S The Satanic Verses, And Coetzee’S Foe, Jake Khoury Apr 2011

Speaking Subjects: Beckett’S Not I, Rushdie’S The Satanic Verses, And Coetzee’S Foe, Jake Khoury

Theses and Dissertations

In repositioning Beckett’s Not I in relation to Rushdie and Coetzee, I show that The Satanic Verses and Foe suggest approaches to language similar to Beckett’s play, insofar as each text interrogates the ability of the marginalized speaking subject to maintain control of his or her voice, finding that the speaking subject’s voice is constantly infused with the voices of others. Additionally, I demonstrate Beckett’s relevance to the postcolonial environment and delineate convergences and divergences in how Rushdie and Coetzee formulate the voices, bodies, and identities of marginalized and postcolonial speaking subjects.

On Things And Margins, Will Machin Apr 2011

On Things And Margins, Will Machin

Theses and Dissertations

This thesis is an exploration into an alternate ground for discourse. It is an exploration of the edges of the possibilities of words, the centers of margins, and the question of the lives of objects in this time of abundant cast-offs. The truths and limits of an emerging globalized multi-polarity of culture, are put in play. The secret lives of things are brought into the same room as words. The potential for radical landscape is considered.

Involvement Of P53 In The S-Phase Checkpoint During Nucleotide Deficiencies, Cortney Heyer Apr 2011

Involvement Of P53 In The S-Phase Checkpoint During Nucleotide Deficiencies, Cortney Heyer

Theses and Dissertations

Several classes of antimetabolites have been developed for the treatment of cancer, including numerous inhibitors of nucleotide biosynthesis. N-(phosphonacetyl)-L-aspartate (PALA) and hydroxyurea (HU) are two antimetabolites that inhibit nucleotide biosynthesis; PALA inhibits de novo pyrimidine synthesis and HU inhibits the conversion of ribonucleotide diphosphates to deoxyribonucleotide diphosphates. Due to the similar mechanisms, it was thought that cancer cells would respond similarly to HU and PALA treatment. However, studies in this dissertation revealed strikingly different responses to either HU or PALA treatment in HCT116 cells. A cytoprotective S-phase arrest was activated upon HU treatment while PALA treatment failed to activate the …

Embody - Encode, Mary Catherine Brooks Apr 2011

Embody - Encode, Mary Catherine Brooks

Theses and Dissertations

According to Walter Murch, sound designer, “The clearest example of encoded sound is speech. The clearest example of embodied sound is music.” In image, object and performance I have been exploring the embodiment of the encoded through the graphic representation of symbols (illustrate), the manifestation of symbols through material construction (embody), the activation of symbolized meaning though performance (enact), and the dematerialization of form and democratic dispersal of information through shared experience (engage). This methodological approach aims to transform metaphysics into physical being as a mode for researching energetics and abstract communication by bringing theory into practice where it can …

Adam10 Exacerbation Of Allergic Disease Is Potentially Explained By Its Role In Cd23 Exosomal Sorting., Joel Mathews

Theses and Dissertations

CD23, the natural negative regulator of IgE, has been shown to be involved in asthma progression through its regulation of IgE. To investigate if its sheddase, ADAM10, is also involved in asthma progression, three mouse models were utilized; an IgE/mast cell dependent model, an IgE dependent, mast cell independent model and a mast cell and IgE independent model. Experimental asthma was then induced in mice which were selectively deficient for ADAM10 in B cells (ADAM10-/-) and compared to WT controls. The ADAM-/- mice had decreased signs of asthma, including eosinophilia, AHR and IgE synthesis in the IgE dependent model compared …
