The Interaction Of Promazine With Selected Macromolecules And A Membrane Utilizing Absorption And Emission Spectroscopy, Jess D. Keeler

The association of promazine-HCl with bovine serum albumin, ammonium detergent micelles, and hemoglobin-free-rabbit-erythrocyte membranes were measured using the changes in the absorption and the emission of the interactants plus the fluorescent probe 1-anilino-8-naphthalene sulfonate. The binding of a promazine to serum protein and membrane did not alter the emission of the drug. However, the binding did produce a measurable quenching of the membrane and protein fluorescence. The quenching reaction was pH sensitive for both the protein and the membrane. Promazine binding to serum protein produced a hyperchromic change in the drugs absorption spectrum which paralleled the quenching reaction. The binding …

Described are the results of harvesting the above-ground parts of an 18-year-old aspen stand and the observations made on the initial suckering of the new stand. The yields for the stand of 17.7 cunits per acre were greater than predicted yields of comparable material for the site at age 35. Suckering on the area the first area after cutting averaged 37,000 aspen stems per acre and 2.6 feet in height.

A Simple Microradiometric Technique For The Rapid Measurement Of Monoamine Oxidase Activity In Rat Testicular Minced And Teased-Tubular Preparations, Ronald L. Urry

Two radiometric techniques are described for the measurement of monoamine oxidase activity in testicular tissue preparations. These techniques utilize 5-HT-2-14C (serotonin) as the substrate and measure its conversion into 5-HIAA-14C (hydroxy-indole acetic acid). The first technique described uses a substrate concentration of 0.9 µM (0.05 µCi) in an incubation volume of 3.1 ml and 120 mg of teased-tubules. Samples are incubated 1 hour. This assay was found suitable for use with teased-tubule preparations but not for preparations containing interstitual cells. An inhibitor was present in the interstitual cell preparations which at the low substrate concentrations used …

The in vitro development of Eimeria magna was studied using various cell types and compared with corresponding development observed in the intestinal epithelial cells of rabbits at 3, 4 and 5 days after inoculation. Leighton tubes were inoculated with sporozoites and the cultures examined at various intervals after incubation with phase-contrast or interference-contrast microscopy. Mucosal scrapings from infected rabbits were also examined. Mature first- and second-generation schizonts were seen in Madin-Darby bovine kidney (MDBK) cells and epithelioid embryonic bovine liver cells, whereas no development beyond the sporozoite occurred in fibroblastic, embryonic, bovine liver cells, lamb trachea cells and aggregates of …

The Regulation Of Purine Biosynthesis In A Phosphoribosylpyrophosphate Aminotransferase-Deficient Mutant Of Salmonella Typhimurium, Wen-Cherng Tsai

The regulation of the alternate first enzyme of purine de novo synthesis (ribose-5'-phosphate aminotransferase) has been examined in pur-F 341, a purine-requiring mutant of Salmonella typhimurium deficient in the primary first enzyme (PP-ribose-P amidotransferase, EC 2.4.2.14). The ribose-5'- phosphate aminotransferase is feed-back inhibited by AMP, ADP, GDP and adenosine but not by ATP, GTP, GMP and free adenine. Pyrimidines (except CDP and UDP) and thiamine compounds show no significant inhibitory effect on ribose-5'-phosphate aminotransferase activity. The alternate enzyme seems to be also controlled at the gene-level by one or more intra-cellular purines (repression-derepression). The role of the aminotransferase in …

The purpose of this paper is to advocate the use of computer modeling as a tool in the study of homeotherm energetics. The relationships between measurable environmental variables and components of productivity were reviewed and discussed as a basis for data input in the models. Photoperiod, temperature, rainfall, and wind were suggested as measurable environmental parameters. The facets of productivity which can be predicted from these parameters are the timing, the energetic cost, and the energetic capacity for productivity.

Amino acid and protein metabolism were studied during growth and development of the alfalfa weevil. The amino acids were separated and identified by two-dimensional thin layer chromatography on cellulose plates and quantitative determinations were made with a ninhydrin spectrophotometric method. The radioactive isotope glucose-U-C14 was used to determine amino acid requirements and their rates of synthesis. A total of 23 amino acids were identified in the fourth instar larvae. Of these, 21 were free amino acids and 17 were protein amino acids. Limited concentrations of methionine, Y-aminobutyric acid and tryptophan in the insect were directly correlated with their dietary …

Levels of potassium in nectar of onion, Allium cepa L., varied from 3600 to 13,000 parts per million, about ten times higher than that of nectar of competing flora tested. An open-pollinated amphidiploid derived from the interspecific cross, A. cepa x A. fistulosum L., was the only onion with nectar potassium level as low as that of competing flora, and it was more attractive to honey bees, Apis mellifera L., than was the common onion. Addition of potassium salts to sugar solutions at the rate of 1500, 3000, 4500, 6000, or 7500 parts per million potassium clearly reduced uptake of …

L-asparaginase was isolated and purified 60 fold from Bacillus kaustophilus by heat treatment, ammonium sulfate fractionation, adsorption chromatography on glass beads and preparative electrophoresis. The purified sample from ammonium sulfate precipitation and adsorption chromatography on glass beads could be stored at 4 C for several weeks and could withstand freezing and thawing without loss of activity. Kinetic studies showed that this L-asparaginase has a temperature optimum of 74 C, an activation energy of 11,400 calories, a pH optimum of 9.2 in sodium carbonate-bicarbonate buffer and Michaelis constant for asparagine of 3.7 x 10-3 Mat 74 C and pH 9.2. …

This study takes a look at government aid to agriculture in Thailand and attempts to analyze its effectiveness. Only two crops have been aided directly by the government, rice and rubber, though others have undoubtedly benefitted by improvements in the country's infrastructure. The regressions performed on some simple production functions indicate that rice yields have been improved through government programs. Twenty four cattle, six each of healthy cows and calves, and cows and calves with brisket disease, were obtained, examined and slaughtered. The concentrations of calcium, chloride, cobalt, copper, iron, magnesium, molybdenum, phosphorus, potassium, sodium and zinc; and percent absolute dry matter and percent ash were determined in tissues selected from the following: cardiac, hepatic, renal, osseous, whole blood and blood serum. In addition, certain physical and biological parameters were recorded for each animal. The results were analyzed as a 2 x 2 factorial, segregating the effects of age and brisket disease, and the age-disease interaction.

The major objective of this study was to investigate various alternatives for lowering the cost of transporting sugar cane in Santa Cruz, Bolivia. These alternatives included both irrigation and non-irrigation possibilities. Production costs for sugar cane were determined by using studies completed by the USAID-Utah State University study team, budget studies of Senor Carlos Castro of the Guabira' sugar cane mill in Santa Cruz and a study undertaken by Senor Enrique Gomez, a graduate student at Utah State University.

Benefits and costs of implementing the specialized grazing systems on federal rangelands managed by the Bureau of Land Management (BLM) were studied. First only the direct benefits and costs were used to determine the internal rates of return on the Bureau of Land Management investments in grazing systems. Secondly, the effects of these systems on profits of private ranchers were determined. Finally the benefits and costs stream on the Bureau of Land Management and the changes in ranchers' profits were brought together to find out the net gain to society from investing the federal funds in specialized grazing systems.

This experiment was designed to measure in situ concentrations of NH4+ and NO3--N in a soil of the arctic coastal tundra plain, to determine if nitrification was taking place in this soil and to determine if the vascular plants growing in this soil could assimilate NH4+-N.

Population biology of the black-tailed jackrabbit population on a 250-square-mile area in Curlew Valley, northern Utah, was studied from 1962-70. During this period the fall population density index increased from 40.0 in 1962, to 60.6 in 1963, decreased progressively to a low of 21.2 in 1967, then increased the following 3 years to a high of 185.0 in 1970.

An inclined point frame was used to measure leaf area indices in an alpine tundra Kobresia stand and Deschampsia meadow on Niwot Ridge, Colorado. Measurements were made throughout the summer of 1971 at various heights in the canopy and for each species in the two communities. Maximum leaf area indices of 2.2 and 2.0 occurred in the Kobresia and Deschampsia sites, respectively. The Kobresia site was characterized by a single dominant species, Kobresia myosuroides, and a predominance of vegetation within 2.5 cm of mean ground level. Decision making and decision changes were studied in late adolescents regarding post-high school choices. A branching questionnaire was constructed and mailed to 1239 late adolescents who during the preceding year had indicated on the American College Test (ACT) that Utah State University (USU) was their first choice for college attendance, but whose names did not appear on the USU Registrar's list of enrolled freshmen, Fall 1969. 62% of the questionnaires were returned. Questions were asked concerning post-high school decision making and decision change:
