The use of X-ray microanalysis in human pathology may require the use of cryoprepared tissue. Often it is impossible to carry out freezing of the tissue in an optimal way, and in addition, it is difficult to carry out experiments in living patients. The use of in vitro systems and cell cultures allows separation of the process of tissue removal and the freezing procedure, and also makes testing of pharmacological or toxic substances possible. In experiments with animal tissue it was shown that incubation in a physiological buffer induced significant changes in the concentrations of Na, K, and Cl. In …

To directly assess the physiological roles of sarcoplasmic reticulum (SR) and miitochondria (MT), we have utilized energy dispersive electron probe microanalysis (EPMA) on ultrathin freeze-dried cryosections from isolated papillary muscles, rapidly frozen at precise time points of the contractile cycle. Using this approach, we can detect redistribution of subcellular Ca2+ during the cardiac contractile cycle. Changes in Ca2+ of less than 1.0 mmol/kg dry wt can be detected. By determining the variability of the Ca2+ measurements in preliminary experiments, we have also demonstrated that it is possible to optimize experimental design, i.e., to predict the number of …

The X-ray microanalysis of thin biological samples which are usually supported on a thin organic film or are self-supporting specimens, has required the use of standards which contain the elements of interest. Spectra from the standards are used to calculate the factors for converting X-ray data recorded on the specimen into elemental concentrations. A method is discussed here, in which these factors are evaluated from formulae. The most important physical process to be evaluated is that of characteristic X-ray production in the specimen. The bremsstrahlung production must also be evaluated if the Hall or continuum normalisation (CN) method of quantitation …

The purpose of this paper is to promote the wise management of ferruginous hawk habitat to enable the species to maintain or increase its population levels. Twenty-seven recommendations are presented, supported by reviews of population status, nesting chronology, nest site characteristics, food habits, and spatial considerations. This is followed by discussion of the impacts of human activities on the ferruginous hawk, such as urbanization, cultivation, grazing, land conversion, poisoning and small mammal control, mining, fire and fire management, and other activities.

The Bureau of Reclamation conducted failure investigations to identify the cause(s) of a catastrophic rupture of prestressed concrete pipe under impressed current cathodic protection, which occurred on Reach 3 of the Jordan Aqueduct near Salt Lake City, Utah, on August 7, 1984. The multidisciplinary analyses included corrosion, design, petrographic, structural, and metallurgical investigations. From the onset of the failure, the issue was one of reconciling cathodic overprotection and defective (longitudinally cracked) prestressing wire. The report contains a summary of the findings from Reclamation investigations.

In 1991, the Soil Landscape Climate Program (SLCP) was organized as part of the Solid Earth Sciences Program at NASA. Part of the research to be conducted in the SLCP included studies of the systematics of carbon storage and flux in the terrestrial environment, specifically terrestrial soils. This report summarizes the results of the research funded through the SLCP that supported our efforts focussed on the nature of carbon behavior in arid environments, where the majority of the carbon is present as inorganic carbon stored as pedogenic carbonate in desert calcic soils.

Minerals and nutrients for desert tortoise food plants from the northeastern Mojave Desert are generally in the normal range for semiarid environments, except that sodium values are low for plants and soil. Annual forbs are often higher in nutritive quality than other plant classes.

Alternatives range from no change from past management practices, which in some instances have resulted in less than favorable ecological conditions on National Forest Rangelands and riparian resources, to managing these resources to achieve the "Potential Natural Community" in terms of vegetative cover types and condition. McMurry Oil Company (McMurry) proposes to drill 40 natural gas wells and construct associated well pads, access roads, gathering and sales pipeline systems; expand an existing compressor station; and include a wareyard at their existing office site.

The Bureau of Land Management (BLM) and the Animal and Plant Health Inspection Service (APHIS) recognize that native animals are resources of value and interest to the American people. Animal damage control (ADC) management may be required to minimize depredations to livestock and wildlife species; to protect threatened and endangered species; to maintain viable populations of native wildlife species; to preserve ecologically unique areas; to minimize rodent and other wildlife damage to cropland, grassland, and forestland; and, to suppress animal-borne diseases. ADC functions as a supplement to, not a substitute for, standard husbandry practices and techniques.

Water released at night from roots into upper portions of the soil profile in the process of hydraulic lift may contribute to reducing spatial soil nutrient heterogeneity. A manipulative field experiment was conducted in a semiarid shrub stand to determine if circumvention of hydraulic lift, by nighttime illumination of the shrub canopy, would result in greater soil nutrient heterogeneity than if the hydraulic lift process was allowed to operate. Nutrient-enriched patches were superimposed on the existing soil heterogeneity and after 40 days, the patches and interspaces were sampled for ions of different mobility and for root mass. Stratospheric ozone reduction is occurring and will continue to increase in magnitude into the next century. Yet, the consequences for terrestrial ecosystems of the increased solar W-B (280-320 nm) radiation resulting from total column ozone reduction are not understood. Based on studies of higher plant response to UV-B, several possible consequences for ecosystems include decreased primary production, altered plant species composition, and altered secondary chemistry with implications for herbivory, litter decomposition and biogeochemical cycles. However, like the assessment of increased atmospheric CO2, extrapolation from studies with isolated plants to ecosystem function is very tenuous at best. Very few UV-B studies …

As a result of those comments the EA has been rewritten and a copy of the revised EA is attached.

Low Temperature Embedding Of Chemically Unfixed Biological Material After Cryosorption Freeze-Drying, L. Edelmann

After freeze-drying of bulk specimens in a newly developed cryosorption freeze-dryer (CFD) a special accessory is used to infiltrate the specimens in Lowicryl HM20 and to polymerize them at low temperature by ultra-violet (UV) irradiation within the CFD chamber in flat embedding moulds. The accessory allows polymerization in a dry, oxygen free environment without the risk of evaporation of volatile components of the resin which may lead to unsatisfactory polymerization. First results demonstrate the quality of structure preservation of biological material not treated with any chemical fixative.

The purpose of this paper is to provide an overview of the significant economic considerations is the analysis of the management, use, and enhancement of riparian zones in the interior West. Applied demonstration work has been done, including basic biological research relating to the functioning of riparian ecosystems and to the effects of domestic livestock grazing on the performance of the riparian system. The literature relating to that work is reviewed here, with an emphasis on implications for economic analysis. Attention is given to the subject of the costs and benefits of riparian-zone management, use, and enhancement strategies from the …
