Effect Of Various Nitrogen Sources On The Metabolism Of Miserotoxin In Astragalus Miser Dougl. Ex Hook., Robert Parker

Wasatch milkvetch [Astragalus miser Dougl., var. oblongifolius (Rydb.) Cronq.] was highest in miserotoxin, a poisonous nitro compound, when potassium nitrate was used as the nitrogen source in nutrient cultures. Ammonium nitrate, added to Hoagland solutions to provide four and one-half times normal nitrogen, lowered percent miserotoxin in the plants. Plants grew abnormally in the greenhouse, producing little growth and becoming periodically senescent. High greenhouse temperatures, up to 45 C, were probably responsible for the abnormal growth.

Attempts to inoculate and fix nitrogen in Wasatch milkvetch with Rhizobium bacteria were unsuccessful in the greenhouse.

A Comparison Of The Ecological Life Histories Of Delphinium Occidentale S. Wats. And Delphinium Barbeyi Huth, Jere Randolph Holman

Two species of the holartic genus Delphinium (D. occidentale S. Wats. and D. barbeyi Huth) were studied in the field and laboratory to determine the factors influencing the life stage development and distribution of these plants.

Delphinium occidentale, more widely distributed geographically and altitudinally than D. barbeyi, is found as a member of aspen and sagebrush communities. Sites are mesic in early spring but turn xeric by midsummer. Delphinium barbeyi, more limited in distribution geographically and altitudinally, appears to be limited to subalpine sites.

Adult biological properties are similar for both species. Morphological and taxonomic differences …

Uroporphyrinogen decarboxylase which catalyses the formation of coproporphyrinogen from uroporphyrinogen is located in the soluble fraction of tobacco leaves and was purified 72 fold through ammonium sulphate precipitation and calcium phosphate gel adsorption.

Kinetic studies indicated that the apparent Michaelis constant was 1 x 10-6 for uroporphyrinogen III (pH 6.5; 37° C). Uroporphyrinogen III served as a much better substrate than uroporphyrinogen I under the standard conditions of this study.

Enzyme activity was inhibited by thiol reagents and heavy divalent cations.

Both chloride and fluoride salts inhibited the formation of coproporphyrinogen from uroporphyrinogen.

The association of promazine-HCl with bovine serum albumin, ammonium detergent micelles, and hemoglobin-free-rabbit-erythrocyte membranes were measured using the changes in the absorption and the emission of the interactants plus the fluorescent probe 1-anilino-8-naphthalene sulfonate. The binding of a promazine to serum protein and membrane did not alter the emission of the drug. However, the binding did produce a measurable quenching of the membrane and protein fluorescence. The quenching reaction was pH sensitive for both the protein and the membrane. Promazine binding to serum protein produced a hyperchromic change in the drugs absorption spectrum which paralleled the quenching reaction. The binding …

The Regulation Of Purine Biosynthesis In A Phosphoribosylpyrophosphate Aminotransferase-Deficient Mutant Of Salmonella Typhimurium, Wen-Cherng Tsai

The regulation of the alternate first enzyme of purine de novo synthesis (ribose-5'-phosphate aminotransferase) has been examined in pur-F 341, a purine-requiring mutant of Salmonella typhimurium deficient in the primary first enzyme (PP-ribose-P amidotransferase, EC 2.4.2.14). The ribose-5'- phosphate aminotransferase is feed-back inhibited by AMP, ADP, GDP and adenosine but not by ATP, GTP, GMP and free adenine. Pyrimidines (except CDP and UDP) and thiamine compounds show no significant inhibitory effect on ribose-5'-phosphate aminotransferase activity. The purpose of this paper is to advocate the use of computer modeling as a tool in the study of homeotherm energetics. The relationships between measurable environmental variables and components of productivity were reviewed and discussed as a basis for data input in the models. Photoperiod, temperature, rainfall, and wind were suggested as measurable environmental parameters. The facets of productivity which can be predicted from these parameters are the timing, the energetic cost, and the energetic capacity for productivity.

Amino Acid And Protein Metabolism During Development Of The Alfalfa Weevil Hypera Postica (Gyllenhal), Darrel J. Gunderson

Amino acid and protein metabolism were studied during growth and development of the alfalfa weevil. The amino acids were separated and identified by two-dimensional thin layer chromatography on cellulose plates and quantitative determinations were made with a ninhydrin spectrophotometric method. The radioactive isotope glucose-U-C14 was used to determine amino acid requirements and their rates of synthesis. A total of 23 amino acids were identified in the fourth instar larvae. Of these, 21 were free amino acids and 17 were protein amino acids. Limited concentrations of methionine, Y-aminobutyric acid and tryptophan in the insect were directly correlated with their dietary …

Chemical Differences Between Nectar Of Onion And Competing Plant Species And Probable Effects On Attractiveness To Pollinators, Gordon D. Waller

Levels of potassium in nectar of onion, Allium cepa L., varied from 3600 to 13,000 parts per million, about ten times higher than that of nectar of competing flora tested. An open-pollinated amphidiploid derived from the interspecific cross, A. cepa x A. fistulosum L., was the only onion with nectar potassium level as low as that of competing flora, and it was more attractive to honey bees, Apis mellifera L., than was the common onion. Addition of potassium salts to sugar solutions at the rate of 1500, 3000, 4500, 6000, or 7500 parts per million potassium clearly reduced uptake of …

Twenty four cattle, six each of healthy cows and calves, and cows and calves with brisket disease, were obtained, examined and slaughtered. The concentrations of calcium, chloride, cobalt, copper, iron, magnesium, molybdenum, phosphorus, potassium, sodium and zinc; and percent absolute dry matter and percent ash were determined in tissues selected from the following: cardiac, hepatic, renal, osseous, whole blood and blood serum. In addition, certain physical and biological parameters were recorded for each animal. The results were analyzed as a 2 x 2 factorial, segregating the effects of age and brisket disease, and the age-disease interaction.

This experiment was designed to measure in situ concentrations of NH4+ and NO3--N in a soil of the arctic coastal tundra plain, to determine if nitrification was taking place in this soil and to determine if the vascular plants growing in this soil could assimilate NH4+-N.

The purpose of this study was focused in two specific objectives: (1) to determine attitudes of professionals, namely pediatricians, general practitioners, and orthodontists toward the use of the oral pacifier; and (2) to determine the attitudes of primiparous mothers toward the use of the pacifier.

The mothers were chosen from the r1cKay-Dee Hospital in Oqden, Utah. They were divided into three groups: (1) 17 mothers with three-month- old babies, (2) 15 mothers with six-month-old babies, and (3 ) 15 mothers with nine-month-old babies, for a total of 47 mothers.

Paper waste discarded by families of five persons in Cache County was studied for two seven-day periods. The sample consisted of 19 families comprised of a father who was employed full-time, a mother, and three children living at home. A background questionnaire was administered to each family for the purpose of describing the sample.

Sample families were given (1) plastic bags for storing of paper wastes and (2) bathroom tissue, the unused portion of which was collected with the other paper discards. The weight of all paper discards was tabulated for each family and an average was tabulated for families …

A procedure was developed for the extraction of rennet from fresh frozen vells. Frozen vells were minced in a Model VCM-25 Hobart Vertical Cutter/Mixer. Dry sodium chloride was added until the salt concentration in the moisture of the tissue was 10%. Salted tissue was placed in galvanized steel cells and heated in a water bath to 49, 54, 57 and 60 C for 0, 15, 30, 45 and 60 minutes. An assay for catheptic activity of lamb gastric tissue extract has been proposed which involves the use of a pH 3.5 hemoglobin substrate following activation of zymogens at pH 2.0, 25C for 30 min.; and inactivation at pH 8.0, 40C for 30 min thereby eliminating the effects of pepsin and rennin. Cathepsin was isolated and purified by ammonium sulfate fractionation, acetone precipitation and gel filtration. The purified cathepsin represented approximately 50 fold increase in specific activity over the original extract and a recovery of 15% of the original activity. Degree of purity was monitored by isoelectric focusing in polyacrylamide gels. …

A specific method has been developed for the extraction and measurement of staphylococcal nuclease in cheese in which Staphylococcus aureus has grown. Ten grams of cheese sample were homogenized with ninety milliliters of pH ten buffer for three minutes. Ammonium sulfate fractionation was used and a forty to eighty percent fraction was collected and concentrated using ultrafilters. The nuclease activity was determined using a toluidine blue deoxyribonucleic acid agar slide method and a spectophotometric method. Attempts were made to adapt the microtiter hemagglutination inhibition assay technique for the assay of enterotoxin A. The presence of a potent hemagglutinin in crude and partially purified preparations and the instability of sensitized erythrocytes prevented its use for routine analysis of enterotoxin from culture media and foods.

A capillary tube immunological assay was developed in which 1 μ g of enterotoxin/ml was detected in less than 1 hr . Interfacial reaction of antisera and enterotoxin solutions in a 1 mm internal diameter capillary tube allowed rapid detection and serological typing of enterotoxins.

The catalytic necessity of tyrosine residues in uridine diphospho- glucose pyrophosphorylase [E.C. 2.7.7.9] was investigated. Chemical modification of the pyrophosphorylase by N-acetylimidazole indicated that tyrosine residues were essential for activity. Approximately 23 of 112 tyrosines per molecule of 475,000 Daltons could be 0-acetylated. Solvent perturbation difference spectroscopy supported this number of exposed tyrosine side chains and in conjunction with chemical modification indicated that at least 11 to 12 tyrosyl residues per protein molecule are fully exposed. it her subst rate, uridine t riphosphate or uridine diphosphoglucose, afforded significant protection against inactivation by N-acetylimidazole.
