Porosity was measured in Blue cheeses made from (i) homogenized butter oil - reconstituted nonfat dry milk (4% fat). (ii) homogenized butter oil reconstituted nonfat dry milk (14% fat) standardized to 4% fat with reconstituted nonfat dry milk , (iii} homogenized raw c ream (14% fat) standardized to 4% fat with raw skim milk , and (iv) homogenized pasteurized cream (14% fat) standardized to 4% fat with pasteurized skim milk. Cheeses made from (i) were the most porous and were significantly different from the other cheeses . In cheeses made from (i) and (ii) , about 40% of the holes …

The Use Of Transmission Electron Microscopy To Study The Composition Of Pseudomonas Fragi Attachment Material, Paula J. Herald, Edmund A. Zottola

Electron microscopy techniques were used to study the attachment matrix of Pseudomonas fragi used to demonstrate the presence of attachment fibrils of P. fragi adhered to stainless steel. Transmission electron microscopy (TEM) was used to examine thin sectioned cells strained with ruthenium red or alcian blue. Extracellular substances surrounding cells stained with ruthenium red or alcian blue had a mat- or spike-like morphology. This indicated that these substances were acidic mucopolysaccharides that may be involved in the attachment of P. fragi to food contact surfaces.

Immature and mature Granny Smith and Red Delicious apples (Malus domestica Borkh . ) were studied with scanning electron microscopy (SEM). Parenchyma cells were observed to form a net-like pattern in sections transverse to the stem-calyx of the apple. Bundles of about six intact cells were connected, creating large intercellular spaces. The intercellular spaces were different in shape from different perspectives to the cut surfaces ; round in stem-calyx transverse sections and elliptical in stem-calyx cross sections. Intrinsic characteristics of plant cell walls limiting susceptibility of structural carbohydrates to microbial attack in the ruminant's gastrointestinal tract are lignification of the cell wall, covalent bonding of phenolic acids to cell wall polysaccharides, the crystalline structure of cellulose and limited fibrolytic enzyme accessible space. The exact mechanism by which or degree to which each of these characteristics affect rate and/or extent of cell wall polysaccharide hydrolysis by gastrointestinal tract microbes is not well understood. Lignification and limited enzyme accessible space probably affect the extent of cell wall degradation by preventing contact between microbial enzymes and cell wall polysaccharides. Phenolic …

Amino acid compositions of stirred-curd Mozzarella, stretched Mozzarella, and process Cheddar cheeses were similar and did not change as the result of baking in a conventional oven. D-glutamic acid (D0Glue) and D-phenyl-alanine (D-Phe) were present at low concentrations in all cheese samples, the lowest concentrations having been found in unbaked stirred-curd Mozzarella cheese (2.7% D-Glu of total Glu present and <1.0% D-Phe of total Phe present). The highest concentrations were detected in unbaked stretched Mozzarella cheese (5.6% and 1.2%, respectively). Thechanges were not significant and were not the result of aking, indicating that the heat treatment during baking did n ot cause racemization of the amino acids.

Each cheese had a characteristic structure before baking. The structures of the Mozzarella cheeses were altered by baking in the conventional oven and also in a microwave oven and their original features such as curd granule junction and fat globule …

The post-storage quality of fruits depends on the ultrastructure of the outer tissues and on the fruit mineral composition (mostly on calcium). Pectins and their bonds with calcium play a central role in the tissues during ripening.

During senescence the middle lamella disappears from the cell walls, the fruits become soft and cell wall reticulation can be seen. There are changes within the cells too; plasmalemma separates from the wall, tonoplast from the cytoplasm, and plastids, mitochondria and nuclei are disorganized.

The hazards birds pose to aircraft has been of concern to the Air Force for more than 20 years. After losing several aircraft due to bird strikes in the early 1960's, the Air Force formed a team to evaluate bird hazards to Air Force aircraft. The team, from the Air Force Weapons Laboratory (AFWL) at Kirtland AFB NM, handed over this mission to the Bird Aircraft Strike Hazard (BASH) Team at HQ Air Force Engineering and Services Center (AFESC) at Tyndall AFB in 1975. In 1986 (October) the BASH team moved to Bolling AFB, Washington DC.

Silviculturists use a variety of techniques, including repellents, to reduce browse damage by white-tailed deer (Odocoileus virginianus) to valuable eastern hardwood seedlings. Systemic selenium, sodium selenite, was evaluated with captive white-tailed deer for its repellency in white ash (Fraxinus americana) and black cherry (Prunus serotina) seedlings. Selenium had no effect in reducing browsing of black cherry. However, there was a reduction (p < 0.05) in the white ash browsing level.

The oral mucosa is classified by function into lining, masticatory and specialized oral mucosa, with regional structural adaptation. In this review, the surface structures of the human oral mucosa have been studied in the scanning electron microscope (SEM). Regional variations in regard to keratinization, cell arrangements and microplications with related specific structures observed in SEM are described and correlated with the appearance of similar areas observed in the light microscope. Furthermore, human oral tissue and cell cultures have also been studied. Female reptiles have the ability to store sperm within their reproductive tracts for extended periods of time. Sites of sperm storage in lizards and snakes include both the anterior vagina and the infundibulum. Vaginal receptacles are found in between longitudinal folds (snakes) and in tubules formed by invaginations of the epithelium (lizards). Infundibular receptacles are alveolar or tubular in structure and are formed from invaginations into the lamina propria of the oviduct wall. The intrinsic and extrinsic blood supplies of the vertebral column of rats have been examined by corrosion cast with Mercox CL2B(R). The close functional and topographic relationship of vessels to supplied structures were exposed by fractionated maceration of the soft tissues. The segmental arteries diverge into an anterior, median and posterior group with various anastomoses between their branches. Vertebral bodies and intervertebral discs are preferably supplied by branches of the anterior and median section. The purpose of this study is to determine whether the structural organization of Pattern 2 marsupial enamel in the Koala is disposed to resist wear on the sectorial crests of the molar teeth. The orientation of wear on the crests is uniformly delineated by parallel scratches on their polished surfaces. Twin blades, a leading and a trailing edge of enamel are formed on each crest by wear into dentine on which the differential wear at enamel to dentine interfaces indicates that the direction of wear is labial to lingual.

Black stain may develop on the coronal surfaces of human teeth, and this type of stain is common in the Hong Kong Chinese population. The present study was undertaken to ascertain if the deposit conforms in composition to the black stain found elsewhere, and to describe its ultrastructure using the scanning electron microscope. Gram-stained smears were made from black stain on the teeth of 11 adult Hong Kong Chinese and studied microscopically. Effects Of Fractionated Irradiation On The Esophageal Mucosa: A Scanning And Transmission Electron Microscopic Study, M. Albertsson, C-H Hakansson, C. Mercke, H. Morner

The mucosa of rabbit esophagus was irradiated with daily fractions of 2 Gy to an accumulated dose of 20 Gy. Specimens were taken for scanning electron microscopy, transmission electron microscopy and light microscopy investigations. Examination was made 1-10 days after each fractionation schedule. Light microscopy showed dose-dependent edema of the irradiated mucosa which also could be seen and scored from SEM pictures. SEM investigations showed that this was accompanied by loosening of microridges and a slightly increased cell loss. By SEM, a varying amount of bacteria could be seen which did not make intimate contact with the surface cells.

Scanning electron microscopy of the placenta has a history of only twenty years. During that time, however, there have been dramatic advances in instrument technology coupled with the refinement of preparative techniques designed to reduce fixation artifacts to a minimum. As a result many of the early claims must be amended or suitably qualified, and this is one aim of the present review.

Syringomycin is a peptide toxin produced by the phytopathogen Pseudomonas syringae pv syringae. It is a major virulence factor produced by this organism and is involved in the development of a number of destructive plant diseases. Syringomycin preferentially stimulated the vanadate sensitive ATPase associated with the plasma membrane of red beet. The stimulation of ATPase activity was not due to a collapse of the ion gradients or to a detergent like breakdown of the membranes. Ionophores did not affect the stimulation of ATPase activity. Deoxycholate eliminated the ability of the toxin to stimulate ATPase activity, and the Zwittergent 3-14 …

Studies Of The Proteinase Activities Of Streptococcus Lactis And S. Cremoris, Fahad Ali Khayat

Twelve proteinase positive (Prt+) and twelve proteinase negative (Prt-) variant pairs of Streptococcus cremoris and three Prt+ and three Prt- variant pairs of S. lactis were assayed for proteolysis in milk using spectrophotometric o-phthaldialdehyde (OPA) and trinitrobenzene sulfonic acid (TNBS). Both methods separated Prt+ and Prt- cultures. The TNBS data range was wider between Prt+ and Prt- strains and thus greater differentiation was possible. The correlation of determination (R2) between OPA and TNBS methods was 0.88 with a coefficient of variation (CV) within of 2.2 and 3.0%. The data were compared to a cell mass increase method. In northeast Brazil grazing is a major use of much of the semiarid woodlands (caatinga). Animal production is limited by lack of dry season forage, primarily deciduous tree leaves. Management is constrained by the persistence of undesirable trees that sprout from the stump (coppice). This study evaluates the possibility of manipulating coppicing trees to improve caatinga management, particularly dry season forage production.

The season of cutting can influence coppicing. Trees were cut early and late in the wet and dry seasons. After two years, trees of all species cut in the late wet season produced less biomass than those cut …

The effect of ultrafiltration, diafiltration, and preacidification of milk on the partition of specific milk components between retentate and permeate was studied. Percent retention (for any component Y) was determined as:

[1 - (%Y in soln. permeate/%Y in soln. retentate)] X 100 where %Y in soln. = [%Y / (%Y + %H2O)]

Simultaneous samples of retentate and permeate were taken at several points during each process. Percent retention of total solids, fat, total protein, rennet clottable nitrogen, lactose, total calcium, ionic calcium, sodium, phosphorous, and riboflavin was determined at each sampling point. Five strains of Lactobacillus plantarum and four strains of Lactobacillus species isolated from fresh meat were examined for the presence of plasmid DNA. All strains examined contained between one and five plasmids ranging in molecular mass from 1.3 to 51.6 (Mdal). Plasmid-curing experiments suggest that maltose utilization is associated with a 51 Mdal plasmid in Lactobacillus sp. DB29 and 42 Mdal plasmids in Lactobacillus spp. DB27, DB28, DB31. Southern blot DNA-DNA hybridization showed homology between the maltose plasmid from Lactobacillus sp. DB29 and several plasmids from the other Lactobacillus spp.

The purpose of this research was to determine whether or not canning fruit using fluoridated water made a difference in the fluoride contents of the juice and the pulp. Four cities in Utah were chosen. Two cities were fluoridated (Brigham City and Helper) and two cities were not fluoridated (North Ogden and Price). Three homes were chosen within each city, and three jars of canned fruit (one jar each of apricots, cherries, and peaches) were selected from each home. Ten young adults were divided into an exercising group (n = 6) and sedentary group (n = 4). Smoking, ingestion of alcohol, drugs and oral contraceptives were prohibited during 9 weeks of study. Diets were prepared by university food service. Food consumed was recorded and nutrient intakes were assessed. Cheese was consumed in 84 to 112 gram portions every day for two separate 14 day periods. All other dairy products were prohibited in the diet except 240 ml of two percent milk per day. When cheese was consumed, daily diets contained ca 400 kcal, 100 mg cholesterol, and 700 mg …
