Dynamic Analyses Of Lymphoblast Membranes Exposed To Alpha Interferon Using Flow Cytometry And Fluorescence Recovery After Photobleaching, E. Balint, A. Aszalos, P. S. Pine, P. M. Grimley
Scanning Microscopy
Interferons represent a major group of the biologic response modifiers which exert multipotent effects upon cell growth, cytodifferentiation and immune functions. Previous experimental studies with alpha interferon (IFN-𝝰) have suggested that modulation of transmembrane signaling could be a critical determinant in the bioregulatory diversity. To determine whether any initial changes at the plasma membrane would directly correlate with one or more actions of IFN-𝝰, we investigated cultures of Daudi lymphoblasts which are uniquely susceptible to growth inhibition. Complementary biophysical techniques were applied. In one approach, changes in plasma membrane ion flux were measured by flow cytometry, using a fluorescent dye …

Design Of The U2 Experiment Ground Support Equipment, Paul Panneton
Small Satellite Conference
Small satellites and sensor payloads aboard larger spacecraft can benefit from ground support equipment (GSE) which reduces the time and cost of developing and flight qualifying space instruments. User friendly microcomputers and a comprehensive system design make such GSE possible and, due to the availability of high speed processors with real-time operating systems, even sophisticated instruments can be accommodated. This was demonstrated by The Johns Hopkins University / Applied Physics Laboratory (JHU / APL) during its work with the U2 Experiment, part of the Strategic Defense Initiative Office's Delta 181 Mission. The U2 Experiment, an orbiting multi-sensor optical emissions instrument, …

Low Cost Molnyia Spacecraft For Navigation/Communications Missions, J Radbone
Small Satellite Conference
Although the Molniya orbit has been widely used by the Soviet Union for many years in support of a variety of communications, science and navigation missions, its advantages have never really been exploited in the west. The Molniya orbit offers improved communications at high latitudes particularly for mobile communications plays an important part in the NAVSAT navigation system constellation and offers scientists an opportunity to undertake scientific measurement over a variety of altitudes and hence environmental conditions with a single spacecraft. Surrey Satellite Technology have recently completed a study to assess the feasibility, and then to propose a preliminary design …

A Feasibility Study For A Remotely Controlled, Low-Power- Consumption Scanning Electron Microscope Suitable For Space Applications, John Chidester, Lynn Chidester
Small Satellite Conference
Experiments and instruments for operation in and on small satellites are constrained by the volume, weight, power, and ruggedness requirements of the satellite design and structure. It is anticipated that increasing numbers of "laboratory" style experiments will be performed in space. An application of current interest is the in-space observation of material surfaces exposed to the space environment. One such instrument for possible small satellite flight operational use, is a scanning electron microscope (SEM). The design factors and parameters of such an SEM, as well as the trade-offs and instrument limitations, will be discussed. The electronic control and image relay …

Design Of A Small Satellite For Use In Astronautics Education, Walter Daniel
Small Satellite Conference
The Naval Academy is pursuing a small satellite project to give midshipmen hardware and ground control experience. The concept is for a lightweight, gravity-gradient stabilized satellite to be deployed from a Get Away Special cannister; the payload would be two small, low-resolution imagers (one visible and one infrared). The 40 foot diameter ground station antenna being installed at the Naval Academy can receive a power of 10 milliwatts from a satellite that uses an omnidirectional antenna and is in a typical Space Shuttle orbit. Approximately 5 to 7 watts of power is constantly available to the payload of up to …

Surface Visualisation Of Tissue Interfaces By Scanning Electron Microscopy. Methods For Exposure Of The Basal Lamina And Associated Structures In Human Amnion, T. D. Allen, J. D. Aplin, S. Campbell
Surface Visualisation Of Tissue Interfaces By Scanning Electron Microscopy. Methods For Exposure Of The Basal Lamina And Associated Structures In Human Amnion, T. D. Allen, J. D. Aplin, S. Campbell
Scanning Microscopy
Tissue interfaces such as basal lamina have been traditionally investigated in transmission electron microscopy by sections cut vertical to the lamina, presenting information restricted to a single ultrathin plane. In order to overcome this limitation, a methodology for surface visualisation of the underside cell membranes of the amniotic epithelium, the upper and lower basal lamina surfaces, and their relationship to the stromal collagen has been devised. This involves alkaline, detergent or enzymatic loosening and/or removal of the epithelial monolayer prior to fixation, followed by dry fracture after critical point drying. In this way we have visualised large areas of all …
Preparation Of Isolated Blood Capillaries, R. C. Wagner, R. A. Buchanan, Ritchie Froehlich
Preparation Of Isolated Blood Capillaries, R. C. Wagner, R. A. Buchanan, Ritchie Froehlich
Scanning Microscopy
Blood capillaries have been isolated from various tissue sources yielding suspensions of capillary segments. These have provided opportunities to study the cellular properties of capillary endothelium under conditions uncomplicated by the presence of stromal tissues and in which measured parameters can be attributed to endothelial cells. Fresh capillary isolates have been used directly as experimental systems but the yield of endothelium is quite low. Amplification of endothelial biomass has been accomplished by using freshly isolated capillaries as explants for primary tissue culture. It has not been previously possible, however, to obtain large amounts of capillary endothelium from a single preparation …

Ultrastructural Characteristics Of The Fetal And Neonatal Rat Urinary Bladder, Samuel M. Cohen, Martin Cano, Takao Sakata, Sonny L. Johansson
Scanning Microscopy
The embryologic and neonatal development of the normal rat urinary bladder was investigated in Sprague-Dawley rats by light, transmission, and scanning electron microscopy from day 11 of gestation through 21 days of age. The epithelium at day 11 of gestation is composed of small, loosely-connected, rounded cells with occasional short microvilli on their surfaces. The large polygonal cells characteristic of the adult bladder begin to appear by day 15, but the microridges are not apparent until day 17. By day 20, the epithelium appears morphologically similar to the adult bladder. Several morphological features are observed at different times of gestation …
Phenotypic Changes In The Megakaryocyte-Platelet Lineage, Paula E. Stenberg
Phenotypic Changes In The Megakaryocyte-Platelet Lineage, Paula E. Stenberg
Scanning Microscopy
Morphologic and phenotypic changes occur during the maturation of megakaryocytes (MK) from the pluripotent stem cell to platelets. As the MK acquires organelles, it also acquires membrane glycoproteins and granule contents. Platelet membrane GP IIb/IIIa and platelet peroxidase are present from early stages of maturation to the final product of the megakaryocyte, the platelet, while Ia-like antigen appears to be expressed only during early stages of maturation. The MK synthesizes increasing amounts of lysosomal enzymes, GP Ib, and alpha granule proteins as it matures from the megakaryoblast stage to the mature cell. The platelet contains only vestiges of a protein …

Crystal Associated Diseases: Role Of Scanning Electron Microscopy In Diagnosis, K. P. H. Pritzker, P. -T. Cheng, M. D. Grynpas, D. P. Holmyard
Scanning Microscopy
As crystals are important etiologic agents for disease, their accurate identification in tissues and body fluids is of utmost importance. This paper surveys the roles of crystals in disease process and outlines current analytical techniques for crystal detection and identification in bone tissues. The value of multiple correlated techniques is demonstrated including scanning electron microscopy, x-ray energy spectroscopy and powder diffraction analysis. The current feasibility of utilizing intermediate voltage scanning transmission analytical electron microscopy to integrate these analytical techniques on the same tissue sample is emphasized.

Scanning Electron Microscopic Autoradiography Of Lung, K. E. Lauhala, C. L. Sanders, K. E. Mcdonald
Scanning Microscopy
Scanning electron microscopic (SEM) autoradiography of the lung is being used to determine the distribution of inhaled, alpha particle-emitting, plutonium dioxide particles. SEM autoradiography provides high visual impact views of alpha activity. Particles irradiating the bronchiolar epithelium were detected both on the bronchiolar surface and in peribronchiolar alveoli. The technique is being used to obtain quantitative data on the clearance rates of plutonium particles from bronchi and bronchioles.

Quantitative Analysis Of Particulate Burden In Lung Tissue, J. P. Mastin, L. E. Stettler, J. D. Shelburne
Scanning Microscopy
Numerous methods have been used in the preparation and analysis of the particulate matter deposited in human lungs. Preparation techniques include those for particle isolation and for in situ analysis. Analytical techniques include bulk and particle-by-particle analysis. In this paper, a general discussion of many of these methods is presented along with examples of how two specific techniques have been used. In one study, individual particles from the lungs of 75 randomly selected autopsy cases were analyzed using an automated scanning electron microscopy (SEM)/ energy dispersive X-ray microanalysis (EDX) system. An average of 613 million particles, of exogenous origin, per …

Studies Of Intestinal Lymphoid Tissue. Xi – The Immunopathology Of Cell-Mediated Reactions In Gluten Sensitivity And Other Enteropathies, Michael N. Marsh
Scanning Microscopy
Computerised image-analysis was used to quantitate small intestinal mucosae from celiac sprue and dermatitis herpetiformis patients, Gambian children with tropical-sprue-like malabsorption, first-degree celiac sprue relatives, and treated celiac sprue patients during challenge with a peptic-tryptic digest of gluten. A wide range of mucosal appearances was observed. Typically, 'flat' lesions (Type 2) revealed a reduced number of epithelial lymphocytes that were large and mitotically active. At the other extreme, mucosal architecture was relatively well preserved (Type 1) but surface epithelium contained an expanded population of small, non-mitotic lymphocytes, with or without crypt hyperplasia. Similar changes were observed in one-third of celiac …

Effects Of Macrocyclic Trichothecene Mycotoxins On The Murine Immune System, Brian J. Hughes
Biology
The macrocyclic trichothecenes are a unique group of toxins which have some antileukemic properties. In the first study, verrucarin A and roridin A were examined. Both mycotoxins were administered intraperitoneally at an equitoxic dose of 0.35 mg/kg to CD-1 mice. Lymphocyte proliferation was studied on days 2, 4, and 7 after animals were dosed with verrucarin A. After day 2, no differences in 3HThymidine (3H-TdR) incorporation were observed using concanavalin A (Con A), phytohaemagglutinin (PHA), pokeweed mitogen (PWM), or lipopolysaccharide (LPS). On day 4, DNA synthesis induced by Con A, PHA, and PWM increased significantly. On day …

1-Acyldihydropyridones As Synthetic Intermediates, Michael Andrew Foley
All Graduate Theses and Dissertations, Spring 1920 to Summer 2023
The most efficient and stereoselective total synthesis of (+/-)- lausbine II to date has been achieved. The key steps in this are the copper-mediated conjugate addition reaction of the Grignard reagent of 1-bromo-4-chlorobutane to a dihydropyridone and a stereoselective reduction of a quinolizidinone.
Methodology has been developed for the convenient synthesis of 1-acyl-2-substituted-1,2,5,6-tetrahydropyridines. This was accomplished by adding novel alkylzinc iodides to the 1-acyliminium ion derived from N-phenoxycarbonyl-4-methoxy-1,2,3,4-tetrahydropyridine.

Pastoral Development Strategies In The Sahel And East Africa: Can The Mistakes Be Corrected?, Ahmed Musa Haji Ahmed
All Graduate Plan B and other Reports, Spring 1920 to Spring 2023
There is much controversy on the definition given to those people who raise livestock on the natural vegetation. Differences in the degree of their dependence on livestock and livestock products for food, as well as differences in the patterns of their movements, create the controversy. "Pure-pastoralists", "nomadic pastoralists", "semi - pastoralists", "semi-nomadic", "transhumant", and "semisedentary" are but some of the many terms used in the literature to describe them (see, for example, Johnson, 1969; Jacobs, 1965; McGee, 1986; Horowitz, 1981). To avoid much of the confusion created by the use of these terms, we use the definition of pastoralists as …
