Fibroblast And Epidermal Cell-Type I Collagen Interactions: Cell Culture And Human Studies, Charles J. Doillon, Frederick H. Silver, Robert M. Olson, Chandrakala Y. Kamath, Richard A. Berg
Scanning Microscopy
Fibroblast and epidermal cell-type I collagen sponge interactions were studied in cell culture as well as in humans. In cell culture, fibroblasts were observed to migrate and proliferate throughout a type I collagen sponge containing either hyaluronic acid (HA) or fibronectin (FN). Fibroblasts accumulated in the center of the pores in sponges containing HA and appeared to surround themselves with newly synthesized extracellular matrix. In sponges containing FN, fibroblasts attached to and elongated along the collagen fibers of the sponge. In the absence of FN or HA protein synthesis of fibroblasts appeared to be inhibited by the presence of the …

The Role Of Scanning Electron Microscopy In Periodontal Research, A. Carrassi, S. Abati, G. Santarelli
Scanning Microscopy
During recent years a great amount of research has led to a better understanding of the etiology, pathogenesis and pattern of progression of periodontal diseases. Scanning electron microscopy (SEM) has contributed to this improvement, mainly with respect to histology of periodontal tissues, the description of the morphology and distribution of bacteria on the exposed root surface, analysis of the host-parasite interactions on the gingival pocket wall, and morphological evaluation of root treatment. This review deals with all these topics. Unusual types of SEM research are also described and discussed. Uncommon sample preparation techniques for SEM in periodontal research are described. …
Ion And Water Retention By Permeabilized Cells, Carlton F. Hazlewood, Miklos Kellermayer
Ion And Water Retention By Permeabilized Cells, Carlton F. Hazlewood, Miklos Kellermayer
Scanning Microscopy
Nonionic detergents, Triton X-100 and Brij 58, removed, within 2-5 minutes, lipid membrane of suspended thymus lymphocytes and monolayer H-50 cells grown in culture. Studies of hydration, ionic asymmetry, and ionic and protein release kinetics were conducted on these membraneless cellular preparations. The hydration of nuclei isolated by Triton X-100 procedures appears to be influenced strongly by the monovalent ionic concentration of the buffer bathing the organelles. The putative monovalent ionic concentration of the cellular aqueous phase (i.e., 150 meq/L) caused nuclei to swell and coalesce. Monovalent ionic concentrations of 30 meq/L or less caused minimal changes in volume and …

Scanning Electron Microscopy Of Oral Mucosa In Vivo And In Vitro: A Review, Kristina Arvidson, Roland C. Grafström, Anders Pemer
Scanning Microscopy
The oral mucosa is classified by function into lining, masticatory and specialized oral mucosa, with regional structural adaptation. In this review, the surface structures of the human oral mucosa have been studied in the scanning electron microscope (SEM). Regional variations in regard to keratinization, cell arrangements and microplications with related specific structures observed in SEM are described and correlated with the appearance of similar areas observed in the light microscope. Furthermore, human oral tissue and cell cultures have also been studied. These systems offer usable and complementary models for performing similar studies in vitro under controlled experimental conditions. We now …
The Role Of The Cytoskeleton In Endothelial Repair, Avrum I. Gotlieb, Michael K. K. Wong, Patricia Boden, Alan Choo Fone
The Role Of The Cytoskeleton In Endothelial Repair, Avrum I. Gotlieb, Michael K. K. Wong, Patricia Boden, Alan Choo Fone
Scanning Microscopy
The injured endothelium undergoes rapid repair of areas of cell desquamation in order to maintain the structural integrity of the endothelial surface. Endothelial repair involves a series of processes which include endothelial cell spreading, translocation, and proliferation. These processes are well defined events which occur sequentially in time. Spreading and translocation are mediated by the cell cytoskeleton - F-actin microfilaments and microtubules and associated centrosomes. The regulation of these processes is complex and is likely due to soluble factors present at the site of injury which are released from activated endothelial cells, platelets, the subendothelial substratum, activated serum factors, and …

Ultrastructural Analysis Of Enamel Formation During In Vitro Development Using Chemically-Defined Medium, P. Bringas Jr., M. Nakamura, E. Nakamura, J. Evans, H. C. Slavkin
Scanning Microscopy
To test the hypothesis that enamel biomineralization is regulated by sequential expression of an intrinsic genetic program, we designed experiments to determine if a serumless, chemically-defined medium is permissive for position-dependent ameloblast differentiation and subsequent enamel tissue-specific biomineralization in vitro. In the absence of serum or other exogenous growth factors, Swiss Webster strain mouse embryonic (15-and 16-days gestation) mandibular first molar tooth organs (cap stage) developed within 21 days in vitro into well-defined molar tooth organs expressing dentine and enamel biomineralization. Analysis of data obtained from von Kossa histochemistry for calcium salt formation, as well as ultrastructural information obtained …

Fluoride Effects On The Growth And Morphology Of Two Thermophilic Cyanophytes Synechcoccus Lividus And Phormidium Laminosum, Ibrahim A. Arif
Biology
The effects of various concentrations of fluoride on growth of two thermophilic cyanophytes, Synechococcus lividus (unicellular) and Phormidium laminosum (filamentous) were studied. Growth of both algae was inhibited by fluoride in the concentrations tested. As the concentrations of fluoride increased, an extended adaptation lag phase was encountered. Synechcoccus lividus was more sensitive to fluoride than was Phormidium laminosum, being inhibited in total growth as well as growth rate. Fluoride effects on growth were compared to chloride and to chloride plus fluoride. The growth of S. lividus was stimulated by chloride alone and in combination with fluoride at all concentrations used; …
