This thesis contains the results of an investigation of Cd, Cu and Ni in the colloidal size-range in seawater. The main objectives are: 1) to evaluate an Osmonics cross-flow filtration system for submicron sampling of Cd, Cu and Ni; and 2) to directly assess the concentration of Cd, Cu and Ni in truly dissolved (<1,000 NMW), colloidal (1,000 NMW-1 μm) and particulate (>1 μm) size-fractions in the Gulf of Maine.

A 1,000 nominal molecular weight (NMW) spiral wound cross-flow filtration (CFF) membrane has been evaluated for sampling Cd, Cu and Ni in <1,000 NMW and >1,000 NMW fractions of prefiltered seawater. Mass balance results averaged 93 ± 6% (Cd), 81 …

The Maine green sea urchin fishery has experienced dramatic growth since its inception in 1987. Landings have increased from 1.4 million pounds in 1987 to 41.6 million pounds in 1993 (NMFS, Fisheries Statistics). The reproductive gonads, referred to as roe or 'uni' are shipped fresh from Maine to japan, where the uni is a delicacy item fetching high unit prices on the Japanese fish market. The green sea urchin from Maine, Strongylocentrotus droebachiensis, is the third highest priced sea urchin product imported in Japan (Anon., 1989). The International Commission for the Conservation of Atlantic Tunas (ICCAT) and federal management under the U.S. National marine Fisheries Service (NMFS) has certainly been contentious. Atlantic bluefin tuna have been managed at the international level by ICCAT for the past 25 years. NMFS is the organization responsible for implementing ICCAT recommended conservation and management measures in the United States. In an attempt to halt an apparent decline in the abundance of bluefin tuna in the western Atlantic, ICCAT has recommended strict management measures including catch quotas, minimum sizes, "no-sale" provisions, and spawning area protection since 1981. Survival, growth, and starvation times were studied in summer flounder Paralichthys dentatus larvae hatched in the laboratory at 12.5 and 21*C. The observations spanned the time interval from hatching throughout the period of feeding on rotifers. Survival and growth in length and weight were strongly dependent on water temperature and delay of the initial feeding. At either temperature, the percentage of summer flounder larvae surviving beyond the rotifer phase increased if food was made available at the time of mouth opening. At 12.5*C, hatching started 85 h after fertilization. All feeding delays resulted in 2 separate periods of mortality, which …

An ecosystem-level experiment was conducted to identify the nutrient most limiting to productivity and biomass in the marine lagoons of the northeast United States. Mesocosms containing a complex of species characteristic of shallow coastal marine environments were enriched with P alone, N alone, or combined N plus P, at loadings typical of highly enriched natural lagoons. The mesocosms showed significant responses to enrichment with N alone but not P alone, indicating limitation by N. Enrichment with N alone caused increased water column concentrations of chlorophyll a and particulate nitrogen (PN), increased water column daytime net production (NP), and …

The Galician shelf off NW Spain (43N° 9W°) exhibits mesoscale spatial and temporal changes in biological productivity associated with upwelling. Spatial heterogeneity results from local geomorphic and land‐sea interactions superimposed on the large scale atmospheric processes that produce upwelling. Wind‐induced upwelling events, commonly of short (i.e., week) duration, are more common in the summer than in the winter. A series of cruises, including some time series sampling, and satellite imagery analysis showed that surface upwelling was more common and persistent on the northern coast compared with the western coast off the coastal embayments, the Rias Bajas. In 1979 the US Fish and Wildlife Service published and adopted a classification of wetlands and deepwater habitats of the United States. The system was designed for use in a national inventory of wetlands. It was intended to be ecologically based, to furnish the mapping units needed for the inventory, and to provide national consistency in terminology and definition. We review the performance of the classification after 13 years of use. The definition of wetland is based on national lists of hydric soils and plants that occur in wetlands. Our experience suggests that wetland classifications must facilitate mapping and inventory …

Subcellular locations and chaperone functions of Hsp60 and Hsp70 with flagellin were investigated in Borrelia burgdorferi. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis and Western blot (immunoblot) analysis of fractionated cells showed Hsp60 to be present in the soluble fractions and the Triton X-100 detergent-soluble membrane fraction at growth temperatures ranging from 20 to 37°C. The relative amount of Hsp60 associated with the membrane increased with growth temperature. Hsp70 was found in soluble fractions at growth temperatures between 28 and 37°C, but at 20°C it was also present in the Triton X-100-insoluble membrane fraction. The activation of phospholipase D (PLD) is a receptor-mediated event that has been implicated in signal transduction and membrane traffic in eukaryotic cells. Little is known about the biochemical and molecular properties of signal-activated PLDs, and none has been isolated. Here we report that phosphatidylinositol 4,5-bisphosphate (PIP₂) potently stimulates brain membrane PLD activity in vitro in a highly specific manner. PIP2 increases 10-fold the maximal activity of a partially purified PLD with an EC50 of < 0.5 mol %. Other acidic phospholipids, including phosphatidylinositol 4-phosphate, phosphatidylinositol, phosphatidylserine, and phosphatidic acid, are completely or nearly ineffective. Neomycin, a high affinity ligand of PIP₂, inhibits membrane-bound PLD but has no effect on the activity of a detergent-solubilized or partially purified enzyme. The addition of PIP …

Riparian forests are in a unique position in the landscape since they form a transition between uplands and aquatic systems. These ecosystems may be highly susceptible to nitrogen (N) saturation since they may be subject to high inputs of N from upland areas. We measured potential net N mineralization and nitrification, soil inorganic N levels, microbial biomass carbon (C) and N content, and the N content of litter as indicators of N saturation in two riparian zones on the eastern and western sides of a stream. We sought to validate an in vitro system which could predict the minimal effect dose of antiretroviral agents. Mixtures of uninfected CEM cells and CEM cells chronically infected with human immunodeficiency virus (HIV) type 1 MN were exposed to 2',3'-didehydro-3'-deoxythymidine (D4T) in vitro in a hollow-fiber model which simulates the plasma concentration-time profile of D4T in patients. Drug concentration was adjusted to simulate continuous intravenous infusion, or an intravenous bolus administered twice daily. The effect of the dosing regimen was measured with viral infectivity, p24 antigen, and reverse transcriptase or PCR for unintegrated HIV DNA. Dose deescalation studies on a …
