Background

Metastases play a role in about 90% of cancer deaths. Markers of epithelial-mesenchymal transition (EMT) measured in primary tumor cancer cells might provide diagnostic information about the likelihood that cancer cells have detached from the primary tumor. Used together with established diagnostic tests of detachment—lymph node evaluation and radiologic imaging—EMT marker measurements might improve the ability of clinicians to assess the patient’s risk of metastatic disease. Translation of EMT markers to clinical use has been hampered by a lack of valid analyses of clinically-informative parameters. Here, we demonstrate a rigorous approach to estimating the sensitivity, specificity, and prediction increment …

Characterizing Cdom Spectral Variability Across Diverse Regions And Spectral Ranges, Brice K. Grunert, Colleen B. Mouw, Audrey B. Ciochetto Jan 2018

Characterizing Cdom Spectral Variability Across Diverse Regions And Spectral Ranges, Brice K. Grunert, Colleen B. Mouw, Audrey B. Ciochetto

Graduate School of Oceanography Faculty Publications

Satellite remote sensing of colored dissolved organic matter (CDOM) has focused on CDOM absorption (aCDOM) at a reference wavelength, as its magnitude provides insight into the underwater light field and large-scale biogeochemical processes. CDOM spectral slope, SCDOM, has been treated as a constant or semiconstant parameter in satellite retrievals of aCDOM despite significant regional and temporal variabilities. SCDOM and other optical metrics provide insights into CDOM composition, processing, food web dynamics, and carbon cycling. To date, much of this work relies on fluorescence techniques or aCDOM in spectral ranges unavailable to current …

The literature suggests that small genomes promote invasion in plants, but little is known about the interaction of genome size with other traits or about the role of genome size during different phases of the invasion process. By intercontinental comparison of native and invasive populations of the common reed Phragmites australis, we revealed a distinct relationship between genome size and invasiveness at the intraspecific level. Monoploid genome size was the only significant variable that clearly separated the North American native plants from those of European origin. The mean Cx value (the amount of DNA in one chromosome set) for …

The ability to predict animal movement based on environmental change is essential for understanding the dynamic nature of their spatial ecology, and in turn the effectiveness of conservation strategies. We used a large marine predator that displays partial migration (the tiger shark Galeocerdo cuvier) as a model to test the role of oceanic conditions in predicting the space-use of different size classes. By using generalized additive mixed models (GAMMs), we revealed that environmental variables (sea surface temperature, primary productivity, thermal fronts, and bathymetry) had much greater predictive power for the movements of large, migratory tiger sharks than for small, …

Cellulose synthases (CESAs) are glycosyltransferases that catalyze formation of cellulose microfibrils in plant cell walls. Seed plant CESA isoforms cluster in six phylogenetic clades, whose non‐interchangeable members play distinct roles within cellulose synthesis complexes (CSCs). A ‘class specific region’ (CSR), with higher sequence similarity within versus between functional CESA classes, has been suggested to contribute to specific activities or interactions of different isoforms. We investigated CESA isoform specificity in the moss, Physcomitrella patens (Hedw.) B. S. G. to gain evolutionary insights into CESA structure/function relationships. Like seed plants, P. patens has oligomeric rosette‐type CSCs, but the PpCESAs diverged independently and …

On microevolutionary timescales, adaptive evolution depends upon both natural selection and the underlying genetic architecture of traits under selection, which may constrain evolutionary outcomes. Whether such genetic constraints shape phenotypic diversity over macroevolutionary timescales is more controversial, however. One key prediction is that genetic constraints should bias the early stages of species divergence along “genetic lines of least resistance” defined by the genetic (co)variance matrix, G. This bias is expected to erode over time as species means and G matrices diverge, allowing phenotypes to evolve away from the major axis of variation. We tested for evidence of this signal in …

Accumulation of fibrillar amyloid β protein in blood vessels of the brain, a condition known as cerebral amyloid angiopathy (CAA), is a common pathology of elderly individuals, a prominent comorbidity of Alzheimer disease, and a driver of vascular cognitive impairment and dementia. Although several transgenic mouse strains have been generated that develop varying levels of CAA, consistent models of associated cerebral microhemorrhage and vasculopathy observed clinically have been lacking. Reliable preclinical animal models of CAA and microhemorrhage are needed to investigate the molecular pathogenesis of this condition. Herein, we describe the generation and characterization of a novel transgenic rat (rTg-DI) …

Selling Anthropocene Space: Situated Adventures In Sustainable Tourism, Amelia Moore

Marine Affairs Faculty Publications

The Anthropocene is a proposed technical term for a new geological timeframe, but it is also a conceptual tool with the potential to redefine the stakes of contemporary environmental politics. One facet that is often overlooked is that the Anthropocene is a concept with commercial potential, even if the term itself has not been widely adopted. This article presents an investigation of the commercial potential of the Anthropocene idea through the lens of self-described sustainable tourism ventures in The Bahamas. These examples demonstrate some of the ways in which Anthropocene imaginaries participate in the recreation, redesign, and rebranding of specific …

Results from live cell imaging of fluorescently tagged Cellulose Synthase (CESA) proteins in Cellulose Synthesis Complexes (CSCs) have enhanced our understanding of cellulose biosynthesis, including the mechanisms of action of cellulose synthesis inhibitors. However, this method has been applied only in Arabidopsis thaliana and Brachypodium distachyon thus far. Results from freeze fracture electron microscopy of protonemal filaments of the moss Funaria hygrometrica indicate that a cellulose synthesis inhibitor, 2,6-dichlorobenzonitrile (DCB), fragments CSCs and clears them from the plasma membrane. This differs from Arabidopsis, in which DCB causes CSC accumulation in the plasma membrane and a different cellulose synthesis inhibitor, isoxaben, …

Spatial Ecology Of Carcharias Taurus In The Northwestern Mid-Atlantic Coastal Ocean, D. E. Haulsee, M. W. Breece, L. M. Brown, Bradley M. Wetherbee, D. A. Fox, M. J. Oliver

Biological Sciences Faculty Publications

The sand tiger shark Carcharias taurus is a highly migratory coastal species with declining populations worldwide. This species exhibits many behaviors that make coastal sharks difficult to manage, including aggregatory behavior, sexual segregation, and large-scale migrations through shallow coastal waters with many opportunities for human interactions. Sand tigers from the Western North Atlantic subpopulation are known to seasonally inhabit Delaware Bay and surrounding coastal waters. This region has been recommended as a habitat area of particular concern for the Western North Atlantic sand tiger population, and increased understanding of their movements and habitat requirements will facilitate management efforts. We developed …

Can Darapsa Myron (Lepidoptera: Sphingidae) Successfully Use The Invasive Plant Ampelopsis Brevipedunculata As A Food Resource?, Alex K. Baranowski, Evan L. Preisser

Biological Sciences Faculty Publications

Although biological invasions are generally thought to negatively impact native fauna, native herbivores that can utilize invasive plants may benefit. The East Coast of the United States has been invaded by the vitaceous plant Ampelopsis brevipedunculata. The invaded range of A. brevipedunculata overlaps with that of the native Vitis labrusca, a closely-related species that is a host plant for the native moth Darapsa myron (Lepidoptera: Sphingidae). We reared D. myron larvae on either V. labrusca or A. brevipedunculata to assess whether development and survival differed on the two plant species. Larval growth and survival to pupation was only …

User-Based Photometer Analysis Of Effluent From Advanced Nitrogen-Removal Onsite Wastewater Treatment Systems, Bianca N. Ross, George W. Loomis, Kevin P. Hoyt, José A. Amador

Natural Resources Science Faculty Publications

Advanced nitrogen-removal onsite wastewater treatment systems (OWTS) are used to reduce total nitrogen (N) levels in domestic wastewater. Maintaining system performance requires regular monitoring and in situ rapid tests can provide an inexpensive option for assessing treatment performance. We used a portable photometer to measure ammonium and nitrate concentrations in final effluent from 46 advanced N-removal OWTS, sampling each site at least three times in 2017. To assess photometer accuracy, we compared measurements made using the photometer with those determined by standard laboratory methods using linear regression analysis and a two-tailed t test to compare regression parameters to those for …

Comparing Fishers' And Scientists' Estimates Of Size-At-Maturity And Maximum Body Size As Indicators Of Overfishing, Elizabeth Layli Mclean, Graham E. Forrester Jan 2018

Comparing Fishers' And Scientists' Estimates Of Size-At-Maturity And Maximum Body Size As Indicators Of Overfishing, Elizabeth Layli Mclean, Graham E. Forrester

Natural Resources Science Faculty Publications

We tested whether fishers’ local ecological knowledge (LEK) of two fish life-history parameters, size at maturity (SAM) at maximum body size (MS), was comparable to scientific estimates (SEK) of the same parameters, and whether LEK influenced fishers’ perceptions of sustainability. Local ecological knowledge was documented for 82 fishers from a small-scale fishery in Samaná Bay, Dominican Republic, whereas SEK was compiled from the scientific literature. Size at maturity estimates derived from LEK and SEK overlapped for most of the 15 commonly harvested species (10 of 15). In contrast, fishers’ maximum size estimates were usually lower than (eight species), or overlapped …
