The Pic Protease Of Enteroaggregative Escherichia Coli Promotes Intestinal Colonization And Growth In The Presence Of Mucin, Susan M. Harrington, Jalaluddin Sheikh, Ian R. Henderson, Fernando Ruiz-Perez, Paul S. Cohen, James P. Nataro Jan 2009

The Pic Protease Of Enteroaggregative Escherichia Coli Promotes Intestinal Colonization And Growth In The Presence Of Mucin, Susan M. Harrington, Jalaluddin Sheikh, Ian R. Henderson, Fernando Ruiz-Perez, Paul S. Cohen, James P. Nataro

Cell and Molecular Biology Faculty Publications

Enteroaggregative Escherichia coli (EAEC) is increasingly being recognized as a cause of diarrheal disease in diverse populations. No small animal model is currently available to study this pathogen. We report here that conventional mice orally inoculated with prototype EAEC strain 042 generally became colonized, though the abundance of organisms cultured from their stool varied substantially among individual animals. In contrast, mice whose water contained 5 g/liter streptomycin consistently became colonized at high levels (ca. 108 CFU/g of stool). Neither conventional nor streptomycin-treated mice developed clinical signs or histopathologic abnormalities. Using specific mutants in competition with the wild-type strain, we …

Cell and Molecular Biology Faculty Publications

Different Escherichia coli strains generally have the same metabolic capacity for growth on sugars in vitro, but they appear to use different sugars in the streptomycin-treated mouse intestine (Fabich et al., Infect. Immun. 76:1143-1152, 2008). Here, mice were precolonized with any of three human commensal strains (E. coli MG1655, E. coli HS, or E. coli Nissle 1917) and 10 days later were fed 105 CFU of the same strains. While each precolonized strain nearly eliminated its isogenic strain, confirming that colonization resistance can be modeled in mice, each allowed growth of the other commensal strains to higher numbers, …

Epigenetics, Oxidative Stress, And Alzheimer Disease, Nasser H. Zawia, Debomoy K. Lahiri, Fernando Cardozo-Pelaez Jan 2009

Epigenetics, Oxidative Stress, And Alzheimer Disease, Nasser H. Zawia, Debomoy K. Lahiri, Fernando Cardozo-Pelaez

Biomedical and Pharmaceutical Sciences Faculty Publications

Alzheimer disease (AD) is a progressive neurodegenerative disorder whose clinical manifestations appear in old age. The sporadic nature of 90% of AD cases, the differential susceptibility to and course of the illness, as well as the late age onset of the disease suggest that epigenetic and environmental components play a role in the etiology of late-onset AD. Animal exposure studies demonstrated that AD may begin early in life and may involve an interplay between the environment, epigenetics, and oxidative stress. Early life exposure of rodents and primates to the xenobiotic metal lead (Pb) enhanced the expression of genes associated with …

Validity Of Stage Assessment In The Adoption And Maintenance Of Physical Activity And Fruit And Vegetable Consumption, Sonia Lippke, Jochen P. Ziegelmann, Ralf Schwarzar, Wayne F. Velicer

Cancer Prevention Research Center Faculty Publications

Objective: Stage assessments are examined to develop and test refined measurements that can be used for classifying individuals.

Design: Stages were assessed in 1,850 persons in terms of their physical activity and dietary behaviors.

Main Outcome Measures: Stages for both behaviors were compared to behavior and other test variables. Misclassification, sensitivity, specificity, receiver-operation-curves, and discontinuity patterns were computed. Discontinuity patterns were tested with trends across stages and planned contrasts between adjacent stages.

Results: In comparison to previous studies, sensitivity (70% to 80%) and specificity (80% to 87%) were high. When using lower level criteria (such as …

Development And Validation Of A Weight-Related Behaviors Questionnaire, Susan M. Schembre Jan 2009

Development And Validation Of A Weight-Related Behaviors Questionnaire, Susan M. Schembre

Open Access Dissertations

Research shows the prevalence of obesity is increasing. The greatest increase occurs between 20 and 29 years of age. Obesity results from a long-term positive energy balance attributable to eating and exercise behaviors. Research shows effective behavior change interventions impact cognitive constructs which explain specific behaviors. Objectives were to develop and validate a weight-related behaviors questionnaire (WRBQ) for young adults with individual eating and exercise behavior scales and explore the impact of the scales to explain patterns of energy intake and exercise participation. The sample (N-721) consisted of 18 to 24 year old females (69.5%) and males (30.5%) with a …

An Examination Of The Relationships Among Cardiorespiratory Fitness Status, Body Mass Index, Appetite And Grelin, Jennifer Arts Jan 2009

An Examination Of The Relationships Among Cardiorespiratory Fitness Status, Body Mass Index, Appetite And Grelin, Jennifer Arts

Open Access Master's Theses

Problem Statement: Obesity and its related metabolic disorders rank among the leading causes of illness and mortality worldwide. The prevalence of obesity has led researchers to focus on body weight regulation and the balance between energy intake and energy expenditure.

Background: Studies have suggested that ghrelin is involved in both short and long term energy balance. Previous research has examined the relationships among BMI, ghrelin and appetite but few studies have investigated the role that fitness status plays in relation to these.

Methods: Data from two studies that were previously conducted in the Energy Metabolism Laboratory were combined for a …

Methicillin-Resistant Staphylococcus Aureus: Pharmacoepidemiologic Assessment Of Common Medications, Aisling R. Caffrey Jan 2009

Methicillin-Resistant Staphylococcus Aureus: Pharmacoepidemiologic Assessment Of Common Medications, Aisling R. Caffrey

Open Access Dissertations

Background. Methicillin-resistant Staphylococcus aureus (MRSA) is a prevalent organism, causing serious infections which are difficult and expensive to treat. Despite the implementation of evidence-based infection control practices and development of newer efficacious antibiotics, MRSA infection and colonization rates continue to increase. Treatments for MRSA have received considerable research attention over the past 10 years, yet several gaps in knowledge remain. In particular, controversy regarding the effectiveness of linezolid as compared to vancomycin in treating MRSA infections persists, and few risk factors associated with mupirocin resistance have been identified. Methods. In the first manuscript of this dissertation, we sought to estimate …

Eutrophication And The Macroscope, Scott W. Nixon Jan 2009

Eutrophication And The Macroscope, Scott W. Nixon

Graduate School of Oceanography Faculty Publications

It is important to view eutrophication as an increase in the supply of organic matter to an ecosystem rather than as a simple problem of nutrient pollution. This emphasizes that eutrophication is a fundamental change in the energetic base that may propagate through the system in various ways and produce a variety of changes. Some of these changes may be desirable (e.g., increased secondary production) and some may not (e.g., hypoxia). Defining eutrophication in terms of changing nutrient concentrations or chlorophyll levels or species composition confuses symptoms with the underlying phenomenon. While nutrient enrichment is the most common cause of …

Influence Of Vitamin E On Innate And Humoral Components Of The Immune System Of Pregnant Ewes And Their Lambs, Saritha Anugu Jan 2009

Influence Of Vitamin E On Innate And Humoral Components Of The Immune System Of Pregnant Ewes And Their Lambs, Saritha Anugu

Open Access Master's Theses

The effect of parenteral administration of vitamin E during the pregnancy and lactation period of ewes and their lambs on innate and humoral components of the immune system was investigated in a 3 part study. 1.) Pregnant/Lactating ewes: Fifteen pregnant Dorset sheep were supplemented with vitamin E (30 IU d-α-tocopherol /Kg body weight (BW), E, n=l0) or placebo (emulsified base, P, n=5) from the last trimester of gestation through weaning. Blood and colostrum were analyzed every 2 wk for α-tocopherol, selenium, glutathione peroxidase activity, lysozyme activity, total IgG and IgG antibodies against tetanus toxoid. 2.) Lambs (0-8wks): Lambs born to …

Fertilizing The Land, Lagoons, And Sea: A First Look At Human Impacts On The Nile Delta Fishery, Egypt, Autumn Jean Oczkowski Jan 2009

Fertilizing The Land, Lagoons, And Sea: A First Look At Human Impacts On The Nile Delta Fishery, Egypt, Autumn Jean Oczkowski

Open Access Dissertations

The primary focus of this dissertation is to look more closely at the influence of anthropogenic nutrients on the recovery of Egypt's Nile Delta fishery since the mid-1980's. Up to half of Egypt's fish production has come from four large coastal lagoons, which receive large amounts of nitrogen (N) and phosphorous (P) rich agricultural drainage. In Chapter 1, I provide some of the first evidence supporting a previously theoretical model that links increasing nutrient concentrations to fishery yields. Using data compiled from the literature, fish landings in the four lagoons increased with increased nutrients up to a peak at approximately …

Therapeutic Treatments For Children With Recurrent Abdominal Pain: A Multiple Baseline Pilot Study, Christine Barrett Sieberg Jan 2009

Therapeutic Treatments For Children With Recurrent Abdominal Pain: A Multiple Baseline Pilot Study, Christine Barrett Sieberg

Open Access Dissertations

Chronic pain in children is a significant problem affecting about 15-20% of children (Goodman & McGrath, 1991), yet it is a neglected area in pediatric research and clinical service. Recurrent Abdominal Pain (RAP) is an example of chronic pain in children and one of the most common complaints of childhood and is associated with many adverse outcomes. Research has also identified parental anxiety and depression as potent risk factors for RAP (Garber et al., 1990). Despite the high prevalence rates of RAP, only 12 intervention studies from 1974-2006 have been conducted in this area (Spirito & Kazak, 2006). The primary …

The Invasive Colonial Ascidian Didemnum Vexillum On Georges Bank - Ecological Effects And Genetic Identification, Nicole L. Lengyel, Jeremy S. Collie, Page C. Valentine

Graduate School of Oceanography Faculty Publications

Since the discovery of the invasive colonial tunicate Didemnum vexillum Kott, 2002 on Georges Bank in 2002, research has focused on investigating the spread of the tunicate invasion, evaluating its potential impact on the benthic community, identifying it to species level, and determining its region of origin. The percent cover of Didemnum vexillum, measured from bottom photographs, ranges from 0-100% in individual photos and between 0-79% when averaged within photo transects. Individual photos represent an area of the seabed measuring ~ 0.39 m2 while photo transects range from ~ 700-1000 meters in length. Hydroids are the second most abundant …
