Isolation Of The Virulence Gene(S) In The New, Live, Oral Vaccine Candidate Salmonella Typhimurium Sr-11 Fad, James H. Allen

A DNA fragment containing the defective virulence gene(s) in the live oral vaccine candidate Salmonella typhimurium SR-11 Fad was isolated. Genomic DNA fragments from the vaccine candidate, constructed by Tn10d::cam transposon mutagenesis, were probed with the 1.4 Kbp chloramphenicol resistance gene contained in Tn10d::cam and analyzed by Southern hybridization. Two of the fragments, an 8.0 Kbp Hind III fragment and a 4.5 Kbp Pst I fragment, were isolated, cloned into E. coli HB 101, and mapped. The Pst I fragment, containing approximately 2.9 Kbp of flanking genomic DNA, was subcloned into the sequencing vector pBluescript …

Growth Rates Of Planktonic And Newly Settled American Lobsters Homarusamericanus, Mary-Jane James-Pirri, J. Stanley Cobb

Growth rates, as estimated by the RNA:DNA ratio, were determined for planktonic postlarvae and for recaptured and wild newly settled benthic stages (fifth and sixth instars) of the American lobster Homarus americanus. The mean growth rate of planktonic postlarvae in 1994 was 0.522 ± 0.247 mg protein d-1. This was significantly higher than planktonic growth rates observed in 1991 (0.449 ± 0.121 mg protein d-1) but not in 1992 (0.460 ± 0.144 mg protein d-1). The percentage of poorly nourished planktonic postlarvae, those with growth rates <0.220 mg protein d-1, ranged from 3 to 13% …

A new indirect fluorescent-antibody (IFA) assay with antigen produced in vitro in the human promyelocytic leukemia cell line HL60 was used to identify the first recognized case of human granulocytic ehrlichiosis in Rhode Island. This IFA assay was used to detect granulocytic ehrlichiae in white-footed mice and in a dog inhabiting the area surrounding the patient’s residence. Host-seeking Ixodes scapularis ticks found in the same habitat also were infected. I. scapularis ticks collected from other locations were fed on dogs and New Zealand White rabbits to assess the competency of these species as hosts of granulocytotropic Ehrlichia. Tick-induced …

The ability of naturally infected and cured mice to resist reinfection with tick-transmitted Borrelia burgdorferi was tested over a 1-year period. All of the mice were resistant to reinfection when they were challenged at 1.5 months after cure. The majority of animals were resistant to reinfection for up to 10.5 months after cure, but this resistance was lost at 1 year after cure. Both protected and unprotected animals showed a diverse array of antibodies on Western immunoblots. Protection was not associated with the killing of spirochetes in ticks, and naturally infected mice produced no antibodies to outer surface protein A …

A-77003, a human immunodeficiency virus type 1 (HIV-1) protease inhibitor, is effective for both acute and chronic infection in vitro and was evaluated clinically by continuous intravenous infusion administration. The minimum effective dose (the concentration required to completely inhibit viral replication) was determined in vitro in a population of uninfected (99%) and HIV-infected (1%) cells exposed to A-77003 by continuous infusion in hollow-fiber bioreactors. The production of infectious HIV and release of p24 antigen from infected cells were completely inhibited in cultures exposed to A-77003 at or above a concentration of 0.5 μM. In the mid seventies, Robert Ader and Nicholas Cohen (1975) conducted and published a series of experiments that are generally recognized as a major turning point for the behavioral and physical sciences. In classical conditioning experiments, Ader and Cohen were able to weaken the immune system reaction against foreign material in rats. The experiments demonstrated that the "self-regulating and autonomous" immune system was vulnerable to behavioral factors. These experiments seemed to galvanize a community of scientists who had hypothesized like scientists and philosophers before them the role of emotional states in influencing physiological processes.

We sought to validate an in vitro system which could predict the minimal effect dose of antiretroviral agents. Mixtures of uninfected CEM cells and CEM cells chronically infected with human immunodeficiency virus (HIV) type 1 MN were exposed to 2',3'-didehydro-3'-deoxythymidine (D4T) in vitro in a hollow-fiber model which simulates the plasma concentration-time profile of D4T in patients. Drug concentration was adjusted to simulate continuous intravenous infusion, or an intravenous bolus administered twice daily. The pharmacodynamics of dosage regimens of piperacillin alone or in combination with tazobactam against piperacillin-resistant or -susceptible bacteria were studied in an in vitro model of infection. Experiments were conducted by using a fixed daily exposure of 12 g of piperacillin, given as 3 g alone or in combination with tazobactam at 0.375 g every 6 h, or the same total dose of the combination given as 4 g of piperacillin plus 0.5 g of tazobactam every 8 h. This research involved three independent sample with over 600 heterosexually active individuals and examines the applicability of the Transtheoretical Model of Behavior Change to contraceptive and condom use behavior. The pilot study involved 123 college men and women. Measures representing two of the major constructs from the model, stages of change and decisional balance, were developed for general contraceptive and condom use. The second investigation was a measurement study conducted in collaboration with the Centers for Disease Control and Prevention (CDC), and consisted of 238 impoverished women at high risk for HIV infection or transmission. Escherichia coli F-18, a normal human fecal isolate, is an excellent colonizer of the streptomycin-treated mouse large intestine. E. coli F-18 Col-, a derivative of E. coli F-18 which no longer makes the E. coli F-18 colicin, colonizes the large intestine as well as E. coli F-18 when fed to mice alone but is eliminated when fed together with E. coli F-18. Random sequences of E. coli F-18 DNA were cloned into pRLB2, a par-B-stabilized derivative of pHC79. In Vitro Postantibiotic Effect Following Repeated Exposure To Imipenem, Temafloxacin, And Tobramycin, Betty J. Mcgrath, Randall Marchbanks, Deborah H. Gilbert, Michael N. Dudley

The postantibiotic effect (PAE) following three consecutive 2-h exposures to imipenem, temafloxacin, and tobramycin was determined in Pseudomonas aeruginosa. A PAE and a bactericidal effect were consistently observed for imipenem following each cycle of drug exposure and regrowth. In contrast, the PAE increased with repeated exposure with temafloxacin (1.8 to > 5 h), but disappeared with tobramycin by the third exposure (0.9 to 0 h). These data show that the in vitro PAE may change within a strain following multiple cycles of drug exposure and bacterial regrowth.

The effect of dose or dose interval on the pharmacodynamics of simulated high-dose intravenous ciprofloxacin therapy on infection due to Pseudomonas aeruginosa and Staphylococcus aureus was studied in an in vitro hollow-fiber model of infection. Simulated doses of 1,200 mg of ciprofloxacin per day as either 400 mg every 8 h or 600 mg every 12 h against P. aeruginosa resulted in selection of ciprofloxacin-resistant bacteria. The results with one test strain that was isolated from a patient prior to administration of intravenous ciprofloxacin demonstrated selection of a gyrA mutant in the model, as had occurred in vivo. Salmonella choleraesuis (a pig pathogen), Salmonella typhimurium (a virulent strain in mice), and three strains of Escherichia coli (including a human enterohemorrhagic strain, a human urinary tract isolate, and a human fecal isolate) grew as well in vitro utilizing the lipids derived from mouse cecal mucus as the sole source of carbon and nitrogen as they did in mouse crude cecal mucus. Further analysis of the total lipid extracts of mucus dialysates showed that the acidic lipid fraction supported growth nearly as well as the total lipid fraction. Chapter 1 reviews the 1iterature on the debate over flexibility and its relevance to the medical instruments and supply industry.

Chapter 2 briefly introduces the methodology used in this research.

Chapter 3 reviews factors influencing the development of the industry.

Chapter 4 profiles the medical supply industry in the US today.

The disease prevalence in American oysters, Crassostrea virginica, from coastal areas in Rhode Island was studied. Oysters were collected by hand or tongs from the Pawcatuck River (3 stations), Narrow River (2 stations), Charlestown Pond (3 stations) and Green Hill Pond (3 stations), during July/August 1991, November 1991 , March 1992, and May 1992. Of the forty oysters collected at each of the four sites, thirty were processed for histologic examination. The remaining 10 oysters of each site were used to determine the condition index.

The ability of Escherichia coli K-12(K88ab) to adhere to immobilized porcine small intestine mucus was examined. E. coli K-12(K88ab) but not the isogenic E. coli K-12 strain was found to adhere readily to immobilized crude mucus but not to bovine serum albumin. The adhesion of E. coli K-12(K88ab) was inhibited in a specific fashion by anti-K88 antiserum. Adhesion was also inhibited by pretreatment of receptor-containing crude mucus preparations with sodium metaperiodate or proteolytic enzymes. Removal of glycolipids from crude mucus by chloroform methanol extraction did not affect the ability of E. coli K-12(K88ab) to bind to mucus preparations. Adsorption of …

Escherichia coli F-18, isolated from the feces of a healthy human, makes type 1 fimbriae and is an excellent colonizer of the streptomycin-treated mouse large intestine. Recently, it was shown that the inability to produce type 1 fimbriae had no effect on the ability of E. coli F-18 to colonize the streptomycin-treated mouse large intestine, suggesting the possibility that E. coli F-18 does not express type 1 fimbriae in vivo. However, we show here that E. coli F-18 does express type 1 fimbriae in mouse cecal mucus in vivo and, in fact, appears to express substantially more type 1 fimbriae …

Over the past several years there has been a growing awareness of water pollution problems in Greenwich Bay, a subembayment of Narragansett Bay which contains valuable shellfish beds. Studies have shown that water quality problems in the bay are due largely to nonpoint source pollution, which is conveyed by stormwater runoff. Nonpoint source pollution can be treated with a variety of source-control and structural techniques to manage stormwater, known as best management practices, or BMP's. High performance developed to isolate phospholipids and liquid chromatographic methods were and purify naturally occurring synthetic experimental Anti-AIDS liponucleotides on a preparative scale. Separations were optimized on packed with an analytical size method development column, the same media used in the preparative separations. A phospholipid class separation, on a silica stationary phase, was developed for the isolation of lecithin in chicken egg yolk. The collected fractions were analyzed for purity by analytical HPLC with UV and light scattering detection. Liponucleotides have been synthesized and required HPLC for purification. The methodology was based upon conventional phospholipid species separations on reversed-phase supports. …
