The Modification Of Hepatic Microsomal Drug Metabolism By Phenobarbital And D-Amphetamine, Robert Thomas Louis-Ferdinand Jan 1970

The Modification Of Hepatic Microsomal Drug Metabolism By Phenobarbital And D-Amphetamine, Robert Thomas Louis-Ferdinand

Open Access Dissertations

This investigation was carried out to provide a mechanistic explanation for the modification of hepatic microsomal drug metabolism produced by amphetamine and phenobarbital. The influence of phenobarbital administration on the activity of microsomal ribonuclease during induction of drug-metabolizing enzymes, was determined. Administration of phenobarbital (100 mg/kg) for six days resulted in a significant (P ≤ 0. 05) increase in hepatic microsomal oxidative demethylation. Conversely, hepatic microsomal ribonuclease activity of phenobarbital-treated rats was significantly reduced to less than 5% of control values. When phenobarbital (50 mg/kg) was administered for 5 days ribonuclease activity was inhibited to about one-half of control values. …

The Influence Of 1, 3-Butylene Glucol And Fatty Acid Esters On Rat Reproduction And Development, Patricia King Adams

Open Access Master's Theses

The purpose of this study was to investigate the safety for human dietary consumption of three synthetic compounds which are potential sources of dietary energy, namely 1, 3-Butylene Glycol (BG), 1, 3-Butylene Glycol Mono-Oleate (BGMO) and 1, 3-Butylene Glycol Mono-Stearate (BGMS), by feeding these compounds to parents and three generations of rats. Previous animal feeding studies had indicated that BG was low in toxicity, but the effects of BG on reproduction had not been adequately established. No relative studies were known of the physiological and toxicological effects of the BGMO and BGMS compounds. Control and test diets were composed of …

Learning Associated With The Amphetamine-State: Role Of Brain Amines, Mark Roffman Jan 1970

Learning Associated With The Amphetamine-State: Role Of Brain Amines, Mark Roffman

Open Access Master's Theses

Behavior is under the control of external and internal stimuli. The emission of behavior is therefore most efficient when the stimulus-conditions similar to those under which the behavior was acquired are reinstated. Amphetamine was shown to produce a "stimulus-state" which controlled behavior. A decrement in response strength occurred when a response acquired under the influence of amphetamine was emitted in the absence of amphetamine or in the presence of no-drug. The response strength recovered when the amphetamine-state was reinstated. Rats were trained to jump to a wooden platform from an electrifiable grid-floor, in order to avoid shock. A buzzer was …

Effect Of Acute Hypoxia Or Chronic Social Isolation On Brain Sensitivity And Barbiturate Metabolism, Irwin Paul Baumel Jan 1970

Effect Of Acute Hypoxia Or Chronic Social Isolation On Brain Sensitivity And Barbiturate Metabolism, Irwin Paul Baumel

Open Access Dissertations

The mechanisms by which hypoxia or social isolation interact with drug action were investigated by the use of several parameters including drug-induced narcosis, convulsions, change in body temperature and various biochemical parameters.

Acute Hypoxia

Hypobaric hypoxia (364 mm Hg, 10% O2) enhanced. the depressant effects of barbital, pentobarbital and chloral hydrate in mice and rats. Mice were far more sensitive than rats, barbital narcosis being affected to the greatest extent. Reduced oxygen (10%) at normal pressure also potentiated pentobarbital narcosis.

Hypothermia during hypoxia was greater in mice than in rats. Mice exposed to hypoxia at 30°C instead of …

Alteration Of Proline Hydroxylase Activity By Corticosteroids, Kenneth Robert Cutroneo Jan 1970

Alteration Of Proline Hydroxylase Activity By Corticosteroids, Kenneth Robert Cutroneo

Open Access Dissertations

The biochemical mechanism by which glucocorticoids affect collagen metabolism was investigated. Triamcinolone, hydrocortisone and methylprednisolone significantly decreased liver proline hydroxylase activity in vivo. This was not a manifestation of an antianabolic effect on protein. Triamcinolone inhibited liver proline hydroxylase activity in a dose dependent manner in vivo. An observed elevation of liver hydroxylase activity in adrenalectomized animals was decreased to control level by hydrocortisone treatment.

Subdermal implants of three types of sponges (polyurethane, cellulose and polyvinyl) were used to induce granuloma growth as a model system of inflammation. The anti-inflammatory activity of corticosteroids was correlated with inhibition of …

The Ocean Resource: A Scenario For Its Control, Ronald Poitras Dec 1969

The Ocean Resource: A Scenario For Its Control, Ronald Poitras

Marine Affairs Theses and Major Papers

As man expands his horizons on planet earth, he is beginning to turn increasingly to the oceans as a source of resources, and at a pace commensurate with the growth of his needs and his technical ability. Traditionally the deep sea has always been a "no man's land," an unlimited source of space to be traversed and used infrequently. A scarcity of natural resources and an abundance of human extensions (i.e. technology) are together making deep ocean seclusion a thing of the past. Increasing use of a particular resource is a limited space sooner or later implies an increasing need …

Characterization Of The Effect Of Methylchloroform Inhalation On Hepatic Drug Metabolism In The Rat, Arnold Merwin Olshan Jan 1969

Characterization Of The Effect Of Methylchloroform Inhalation On Hepatic Drug Metabolism In The Rat, Arnold Merwin Olshan

Open Access Master's Theses

Male rats were exposed to methylchloroform (1,1,1-tricholoethane) vapors (concentration about 2500 PPM) for 24 hours and the effects of this treatment on drug responses and hepatic drug metabolism studied 24 hours later to determining the effect of methylchloroform on inducible hepatic drug-metabolizing enzymes. Exposure to methylchloroform vapor significantly decreased the sleeping time of the rat in response to hexobarbital (120 mg/kg, i.p.) and zoxazolamine (80mg/kg, i.p.). Rat blood and liver methylchloroform content measured immediately following exposure suggested that the effects of methylchloroform were caused by a systemic action. Experiments with 9000 x G supernatants indicated that the metabolism of hexobarbital …

Pharmacology Of Some 1-Alkyl-3-Benzoylpyridinium Halides, Angela Caretta Springfield

Open Access Master's Theses

Several 1-alkylbenzoylpyridinium halides containing alkyl side chains of one to fifteen carbons were screened for pharmacological activity. A comparison of potency was based on the ability to produce mortality in 50 percent of mice (LD 5D). The following parameters were investigated in a select number of compounds representative of the series: acute intraperitoneal LD50 in rats; EDSO (hypotensive) in rats; effects on the autonomic nervous system of anesthetized cats and dogs as indicated by alteratio11s in blood pressure; effects en the nictitating membrane in anesthetized cats; direct effects upon blood vessels employing hind quarter perfusion in intact rats; effects on …

Investigation Of The Role Of Dopamine In Maintenance Of Arterial Hypertension, Harold Llewellyn Crossley Jan 1969

Investigation Of The Role Of Dopamine In Maintenance Of Arterial Hypertension, Harold Llewellyn Crossley

Open Access Master's Theses

The relationship between urinary dopamine levels and arterial blood pressure was studied in normal, unilaterally nephrectomized, and renal hypertensive male albino rats. Experimental hypertension was produced by a unilateral nephrectomy followed by compression of the contralateral renal artery. The two operations were spaced three weeks apart to allow for recovery. Nine weeks after-the-second operation; mean systolic blood pressure of the renal hypertensive control group increased to 170 mm Hg. Mean systolic blood pressure of the diethyldithiocarbamate (DDC)-treated renal hypertensive group decreased abruptly after two weeks of treatment and remained at 105 mm Hg throughout the nine week period. Mean systolic …
