Although human exposure to Gram-negative Vibrio vulnificus (V. vulnificus) lipopolysaccharide (LPS) has been reported to result in septic shock, its impact on the central nervous system’s innate immunity remains undetermined. The purpose of this study was to determine whether V. vulnificus MO6-24/O LPS might activate rat microglia in vitro and stimulate the release of superoxide anion (O2−), a reactive oxygen species known to cause oxidative stress and neuronal injury in vivo. Brain microglia were isolated from neonatal rats, and then treated with either V. vulnificus MO6-24/O LPS or Escherichia coli O26:B6 LPS for 17 …

Biomedical and Pharmaceutical Sciences Faculty Publications

To study the mechanism of tea polyphenols (TP)-induced apoptosis of breast cancer cells. Proliferation of MCF-7 and SK-BR-3 cells was evaluated by MTT assays. Cellular ultrastructure was examined by electron microscopy. Apoptosis was detected by TUNEL. PCNA, Cyclin D1, Cyclin E and Survivin expression was measured by Western blot. Cell proliferation was significantly inhibited by TP. Spindle and round cells were loosely distributed with increased particles after TP treatment. Increased cell size, frequent nuclear atypia and a collapse of apoptosis were observed. The nucleus was pushed towards one side, while the cytoplasm was rich in free ribosome. The membrane of …

The American Heart Association (AHA) has advocated that women and men not consume more than 100 and 150 kcal/day, respectively, from added sugars. These levels are currently exceeded by over 90% of the adult population in the United States. Few data exist on longer-term metabolic effects when sucrose and High Fructose Corn Syrup (HFCS), the principal sources of added dietary sugars, are consumed at levels typical of the general population. Sixty five overweight and obese individuals were placed on a eucaloric (weight stable) diet for 10-weeks, which incorporated sucrose- or HFCS-sweetened, low-fat milk at 10% or 20% of calories in …

Targeting Diseased Tissues By Phlip Insertion At Low Cell Surface Ph, Oleg A. Andreev, Donald M. Engelman, Yana K. Reshetnyak Mar 2014

Targeting Diseased Tissues By Phlip Insertion At Low Cell Surface Ph, Oleg A. Andreev, Donald M. Engelman, Yana K. Reshetnyak

Physics Faculty Publications

The discovery of the pH Low Insertion Peptides (pHLIPs®) provides an opportunity to develop imaging and drug delivery agents targeting extracellular acidity. Extracellular acidity is associated with many pathological states, such as those in cancer, ischemic stroke, neurotrauma, infection, lacerations, and others. The metabolism of cells in injured or diseased tissues often results in the acidification of the extracellular environment, so acidosis might be useful as a general marker for the imaging and treatment of diseased states if an effective targeting method can be developed. The molecular mechanism of a pHLIP peptide is based on pH-dependent membrane-associated folding. …

Encouraging Family Forest Owners To Create Early Successional Wildlife Habitat In Southern New England, Bill Buffum, Christopher Modisette, Scott R. Mcwilliams Feb 2014

Encouraging Family Forest Owners To Create Early Successional Wildlife Habitat In Southern New England, Bill Buffum, Christopher Modisette, Scott R. Mcwilliams

Natural Resources Science Faculty Publications

Encouraging family forest owners to create early successional habitat is a high priority for wildlife conservation agencies in the northeastern USA, where most forest land is privately owned. Many studies have linked regional declines in wildlife populations to the loss of early successional habitat. The government provides financial incentives to create early successional habitat, but the number of family forest owners who actively manage their forests remains low. Several studies have analyzed participation of family forest owners in federal forestry programs, but no study to date has focused specifically on creation of wildlife habitat. The objective of our study was …

Marketing Archaeology, William H. Krieger Feb 2014

Marketing Archaeology, William H. Krieger

Philosophy Faculty Publications

In the 19th century, ‘scientific archaeologists’ split from their antiquarian colleagues over the role that provenience (context) plays in the value of an artifact. These archaeologists focus on documenting an artifact’s context when they remove it from its original location. Archaeologists then use this contextual information to place these artifacts within a particular larger assemblage, in a particular time and space. Once analyzed, the artifacts found in a site or region can be used to document, to understand, and explain the past. Given the central place of context for archaeological excavation, archaeologists have done everything in their power to combat …

Cross-Conservation Of T-Cell Epitopes: Now Even More Relevant To (H7n9) Influenza Vaccine Design, Anne S. De Groot, Lenny Moise, Rui Liu, Andres H. Gutierrez, Frances Terry, Ousmane A. Koita, Ted M. Ross, William Martin

Institute for Immunology and Informatics Faculty Publications

A novel avian-origin H7N9 influenza strain emerged in China in April 2013. Since its re-emergence in October–November 2013, the number of reported cases has accelerated; more than 220 laboratory-confirmed cases and 112 deaths (case fatality rate of 20–30%) have been reported. The resurgence of H7N9 has re-emphasized the importance of making faster and more effective influenza vaccines than those that are currently available. Recombinant H7 hemagglutinin (H7-HA) vaccines have been produced, addressing the first problem. Unfortunately, these recombinant subunit vaccine products appear to have failed to address the second problem, influenza vaccine efficacy. Reported unadjuvanted H7N9 vaccine seroconversion rates were …

Assessing Conduct Disorder: A New Measurement Approach, Racheal Reavy, L. A.R. Stein, Kathryn Quina, Andrea L. Paiva

Psychology Faculty Publications

The Delinquent Activities Scale (DAS) was used to develop indicators of conduct disorder (CD) in terms of symptom severity and age of onset. Incarcerated adolescents (N = 190) aged 14 to 19 were asked about their delinquent behaviors, including age the behavior was first performed, as well as substance use and parental and peer influences. Assessments were performed for the 12 months prior to incarceration and at 3-month postrelease follow-up. Evidence supports the utility of the DAS as a measure of CD diagnosis, including concurrent incremental validity. Furthermore, CD severity (symptom count) was significantly associated with two peer factors: …

Choppi: A Web Tool For The Analysis Of Immunogenicity Risk From Host Cell Proteins In Cho-Based Protein Production, Chris Bailey-Kellogg, Andres H. Gutiérrez, Leonard Moise, Frances Terry, William D. Martin, Anne S. De Groot

Institute for Immunology and Informatics Faculty Publications

Despite high quality standards and continual process improvements in manufacturing, host cell protein (HCP) process impurities remain a substantial risk for biological products. Even at low levels, residual HCPs can induce a detrimental immune response compromising the safety and efficacy of a biologic. Consequently, advanced-stage clinical trials have been cancelled due to the identification of antibodies against HCPs. To enable earlier and rapid assessment of the risks in Chinese Hamster Ovary (CHO)-based protein production of residual CHO protein impurities (CHOPs), we have developed a web tool called CHOPPI, for CHO Protein Predicted Immunogenicity. CHOPPI integrates information regarding the possible presence …

Effect Of Temperature On Feeding Period Of Larval Blacklegged Ticks (Acari: Ixodidae) On Eastern Fence Lizards, Eric L. Rulison, Roger A. Lebrun, Howard S. Ginsberg Jan 2014

Effect Of Temperature On Feeding Period Of Larval Blacklegged Ticks (Acari: Ixodidae) On Eastern Fence Lizards, Eric L. Rulison, Roger A. Lebrun, Howard S. Ginsberg

Plant Sciences and Entomology Faculty Publications

Ambient temperature can influence tick development time, and can potentially affect tick interactions with pathogens and with vertebrate hosts. We studied the effect of ambient temperature on duration of attachment of larval blacklegged ticks, Ixodes scapularis Say, to eastern fence lizards, Sceloporus undulatus (Bosc & Daudin). Feeding periods of larvae that attached to lizards under preferred temperature conditions for the lizards (WARM treatment: temperatures averaged 36.6°C at the top of the cage and 25.8°C at the bottom, allowing behavioral thermoregulation) were shorter than for larvae on lizards held under cool conditions (COOL treatment temperatures averaged 28.4°C at top of cage …
