Temporal And Vertical Distributions Of Ip25 And Other Lipid Biomarkers In Sea Ice From Resolute Bay, Nunavut, Canada, Ashleigh Elizabeth Ringrose
Other Faculty of Science and Engineering Theses
IP25 (Ice Proxy with 25 carbon atoms) is a highly branched isoprenoid biomarker, specifically produced by marine diatoms in Arctic sea ice. Temporal and vertical IP25 concentrations were measured in sea ice from a second location in the Canadian Arctic, Resolute Bay, Nunavut. Sea ice samples were collected as part of the Arctic ICE project in collaboration with scientists of the University of Manitoba from April to June 2011. Comparisons were made between other established lipid biomarkers of diatom origin and general organic production, along with previously established temporal and vertical distributions in sea ice collected from the Amundsen Gulf, …

The Characterisation Of The Freezing Damage Response During Flowering In European And Middle Eastern Wheat Cultivars (Triticum Aestivum L), Mohammed Hamdan Edan Al-Issawi
Other Faculty of Science and Engineering Theses
Wheat (Triticum aestivum L.) is occasionally exposed to low temperature during flowering and huge economic losses can occur especially in some key production countries such as Australia. Although it is generally predicted that there will be a rise in global temperature there are still predicted to be risks associated with low temperature for temperate crops. Post head emergence frost damage remains a major constraint to increasing wheat production. Five Iraqi varieties (Abu-Ghariab, Fatah, Sham6, IPA95 and IPA99) were screened for their frost hardiness (LT50). Abu-Ghariab was chosen for further investigations along with the European cv. Claire because the acclimated frost …

Advancing The Separation Sciences Through The Delivery Of New Materials, Technology And Methodology., Brett Paull
Other Faculty of Science and Engineering Theses
Separation science is a multi‐faceted discipline, underpinning almost all other fields of science and technology. Its scope encompasses fundamental and cutting edge processes and technologies, based upon exploitation of the physical distribution of chemical and biochemical species between solid, liquid or gaseous ‘phases’, facilitating their separation, purification, and analysis. Separation science plays a particularly pivotal element within the majority of modern analytical methods, methods which continue to support all manner of cutting edge scientific endeavour, including, for example, the current ‘‐Omics’ scientific revolution.
Molecular Characterisation Of Clinical And Environmental Isolates Of Vibrio Parahaemolyticus, Hadaf Mahdi Kadhim
Molecular Characterisation Of Clinical And Environmental Isolates Of Vibrio Parahaemolyticus, Hadaf Mahdi Kadhim
Other Faculty of Science and Engineering Theses
The halophilic bacterium Vibrio parahaemolyticus is widely distributed as a natural inhabitant of marine and estuarine environments. Some strains cause gastroenteritis in humans. Clinical isolates are thought to possess virulence factors that are absent from the majority of environmental isolates. Studies were undertaken to differentiate clinical (virulent) and environmental (mainly avirulent) forms of Vibrio parahaemolyticus. Initially, identification and confirmation of a total of 55 V. parahaemolyticus isolates (23 clinical and 32 environmental) was carried out by using selective media, biochemical and nutritional tests. Identity was confirmed by specific polymerase chain reaction (PCR) targeting the toxR gene. In an attempt to …
The Effect Of Hyperbaric Oxygen Therapy On Osteoclast And Osteoblast Function, Hadil Al-Hadi
The Effect Of Hyperbaric Oxygen Therapy On Osteoclast And Osteoblast Function, Hadil Al-Hadi
Other Faculty of Science and Engineering Theses
Bone remodelling, the process by which the skeleton adapts to environmental changes, is dependent on the actions of osteoclasts that resorb bone and osteoblasts which make new bone matrix. Aberrant remodelling underpins bone loss in several debilitating skeletal diseases such as osteoporosis, metastatic breast cancer and multiple myeloma. Changes in remodelling activity can also arise as a consequence of therapeutic intervention for instance intravenous bisphosphonate treatment is associated with osteochemonecrosis of the jaw and localised osteoradionecrosis is a common side effect of radiotherapy. Hyperbaric oxygen is often used as an adjunctive therapy in the treatment of these disorders. HBO involves …

The Effect Of Environmental Factors On The Physiology, Yield And Oil Composition Of Safflower (Carthamus Tinctorius L.), Shiren Jalal Mohamed
Other Faculty of Science and Engineering Theses
This study investigated the effects of drought, nitrogen fertilizer and elevated CO2 and its interaction with nitrogen fertilizer on the physiology, growth, and production of the oil crop safflower (Carthamus tinctorius L.) in a semi-controlled glasshouse environment. Three levels of water stress were imposed: rosette (mid-season drought), stem elongation (terminal drought) and rosette to maturity (mid-season + terminal drought). Results indicated that all drought treatments imposed reduced stomatal conductance, but after the relief of mid-season drought plants recovered and as a result there were no significant differences from control in terms of yield components (branch and capital number) and seed …

Molecular Approaches For Studying Root Herbivores, Cm Benefer, Rp Blackshaw
School of Geography, Earth and Environmental Sciences
The use of molecular techniques in insect ecology has expanded rapidly, allowing ever more challenging questions to be addressed. Compared to their aboveground equivalents, root herbivore molecular ecology has received less attention, despite essentially the same ecological questions being of importance in both the above- and belowground ecosystems. Studies so far have concentrated on economically important taxa, using mitochondrial and nuclear DNA sequencing and a variety of markers to investigate the species identity and relationships, population dynamics and dispersal, distribution, feeding behaviour and interactions with other organisms. Although this has proved useful for elucidating these aspects of their ecology, there …

Critical Comparison Of Intravenous Injection Of Tio2 Nanoparticles With Waterborne And Dietary Exposures Concludes Minimal Environmentally-Relevant Toxicity In Juvenile Rainbow Trout Oncorhynchus Mykiss., D Boyle, Ga Al-Bairuty, Tb Henry, Rd Handy
School of Biological and Marine Sciences
A critical comparison of studies that have investigated tissue accumulation and toxicity of TiO2-NPs in fish is necessary to resolve inconsistencies. The present study used identical TiO2-NPs, toxicological endpoints, and fish (juvenile rainbow trout Oncorhynchus mykiss) as previous studies that investigated waterborne and dietary toxicity of TiO2-NPs, and conducted a critical comparison of results after intravenous caudal-vein injection of 50 μg of TiO2-NPs and bulk TiO2. Injected TiO2-NPs accumulated only in kidney (94% of measured Ti) and to a lesser extent in spleen; and injected bulk TiO2 was found only in kidney. No toxicity of TiO2 was observed in kidney, …

An Assessment Of Current Methodologies For Mitigating The Potential Effects Of Anthropogenic Underwater Sound On Marine Life, And Recommendations For Best Practice, Ross Craig Compton
School of Biological and Marine Sciences Theses
Underwater sound from anthropogenic sources such as seismic surveys, marine renewable device installations and naval exercises has the potential to disturb and cause injury to a variety of marine species. There is particular concern for the potential effects upon marine mammals, which utilise sound to communicate, hunt and navigate. Observed effects include behavioural changes and reduced sighting rates, with unknown consequences for individuals or populations over time. Potential effects on marine mammals include sound induced damage to auditory systems, altered diving behaviour potentially resulting in decompression sickness, stranding and death. The aim of the thesis was to develop a framework …
Macrophage Subset Sensitivity To Endotoxin Tolerisation By Porphyromonas Gingivalis., Andrew D. Foey, St John Crean
Macrophage Subset Sensitivity To Endotoxin Tolerisation By Porphyromonas Gingivalis., Andrew D. Foey, St John Crean
School of Biomedical Sciences
Macrophages (MΦs) determine oral mucosal responses; mediating tolerance to commensal microbes and food whilst maintaining the capacity to activate immune defences to pathogens. MΦ responses are determined by both differentiation and activation stimuli, giving rise to two distinct subsets; pro-inflammatory M1- and anti-inflammatory/regulatory M2- MΦs. M2-like subsets predominate tolerance induction whereas M1 MΦs predominate in inflammatory pathologies, mediating destructive inflammatory mechanisms, such as those in chronic P.gingivalis (PG) periodontal infection. MΦ responses can be suppressed to benefit either the host or the pathogen. Chronic stimulation by bacterial pathogen associated molecular patterns (PAMPs), such as LPS, is well established to induce …
Inclusion Of Lupin Meal And Effect Of A Commercial Feed Supplement (Synergen™) In Diets For Carp, Cyprinus Carpio, Ayub Anwar
School of Biological and Marine Sciences Theses
Abstract Plant proteins are mainly used to formulate diets for carp. Soybean meal (SBM) is one of the most nutritious of all plant protein sources in carp feeds. However, an increase in the use of soybean meal for human consumption and animal feed in both developed and developing countries has resulted in an increase market price of soybean meal globally. Therefore, using other inexpensive plant protein sources in carp feeds would be beneficial to reduce feed cost and contribute to food security as well as to sustain aquaculture production. Anti-nutritional factors (ANFs) are believed to be the most important factors …

Wap Four-Disulfide Core Domain Protein 2 Mediates The Proliferation Of Human Ovarian Cancer Cells Through The Regulation Of Growth- And Apoptosis-Associated Genes, Yao Chen, Xia Mu, S Wang, L Zhao, Et Al
School of Biomedical Sciences
The WAP four-disulfide core domain protein 2 (WFDC2) is frequently overexpressed in epithelial ovarian cancer cells and has been proposed as a potential biomarker. The biological function of WFDC2 in tumor progression remains unclear. In this study, the stable expression of short hairpin RNA (shRNA) against WFDC2 in the human ovarian SKOV3 cell line was established. Cell proliferation in vitro was determined by MTT assay. Cell cycle and apoptosis were analyzed by FACS. The expression of genes related to cell proliferation and survival was detected by real-time RT-PCR and western blotting. In vivo tumor growth assay was performed by establishing …

Exogenous Application Of Molybdenum Affects The Expression Of Cbf14 And The Development Of Frost Tolerance In Wheat., Mohammed Al-Issawi, Hail Z. Rihan, Wondwossen Abate Woldie, Stephen Burchett, Michael P. Fuller
School of Biological and Marine Sciences
Wheat is able to cold acclimate in response to low temperatures and thereby increase its frost tolerance and the extent of this acclimation is greater in winter genotypes compared to spring genotypes. Such up-regulation of frost tolerance is controlled by Cbf transcription factors. Molybdenum (Mo) application has been shown to enhance frost tolerance of wheat and this study aimed to investigate the effect of Mo on the development of frost tolerance in winter and spring wheat. Results showed that Mo treatment increased the expression of Cbf14 in wheat under non-acclimating condition but did not alter frost tolerance. However, when Mo …

A Novel Application Of Motion Analysis For Detecting Stress Responses In Embryos At Different Stages Of Development., Oliver Tills, Tabitha Bitterli, Phil Culverhouse, John I. Spicer, Simon Rundle
School of Biological and Marine Sciences
Motion analysis is one of the tools available to biologists to extract biologically relevant information from image datasets and has been applied to a diverse range of organisms. The application of motion analysis during early development presents a challenge, as embryos often exhibit complex, subtle and diverse movement patterns. A method of motion analysis able to holistically quantify complex embryonic movements could be a powerful tool for fields such as toxicology and developmental biology to investigate whole organism stress responses. Here we assessed whether motion analysis could be used to distinguish the effects of stressors on three early developmental stages …

The Effects Of Environmental Conditions On Quorum Sensing And Community Interactions In Coral-Associated Bacteria, Emma Ransome
The Effects Of Environmental Conditions On Quorum Sensing And Community Interactions In Coral-Associated Bacteria, Emma Ransome
School of Biological and Marine Sciences Theses
The coral holobiont contains diverse communities of bacteria that play a role in the maintenance of coral ecosystems, however little is known about the structure and conservation of the host-bacterial relationship. Declines in coral ecosystems have been partly attributed to outbreaks of disease in tropical and sub-tropical regions, which have been linked to increasing temperatures. Bacteria are thought to play a role in some of these diseases, however little is understood about the mechanisms behind disease progression or the series of events involved in the shifts of coral-associated bacteria from conserved, potentially beneficial communities to those including potential pathogens. Investigations …

Peninsula Medical School
Loss of the Merlin tumour suppressor causes abnormal de-differentiation and proliferation of Schwann cells and formation of schwannoma tumours in patients with neurofibromatosis type 2. Within the mature peripheral nerve the normal development, differentiation and maintenance of myelinating and non-myelinating Schwann cells is regulated by a network of transcription factors that include SOX10, OCT6 (now known as POU3F1), NFATC4 and KROX20 (also known as Egr2). We have examined for the first time how their regulation of Schwann cell development is disrupted in primary human schwannoma cells. We find that induction of both KROX20 and OCT6 is impaired, whereas enforced expression …

Interdisciplinary Study Of Hydrodynamic And Biogeochemical Processes Of A Large-Scale River Plume, Robert Mcewan
Interdisciplinary Study Of Hydrodynamic And Biogeochemical Processes Of A Large-Scale River Plume, Robert Mcewan
School of Biological and Marine Sciences Theses
This research has utilised the Massachusetts Institute of Technology gen- eral circulation model (MITgcm) along with observations taken as part of the River In uences on Shelf Ecosystems (RISE) study to investigate the dynamic processes associated with the Columbia River plume at different temporal and spatial scales. Firstly, a high resolution ( x= y=25 m) investigation of the near-field plume was undertaken using the fully non-hydrostatic mode of the MITgcm. This resulted in the reproduction of a detailed inner plume as well as a series of radiated internal waves. In addition to first mode internal waves, second order waves were …
An Ecotoxicological Assessment Of The Impacts Of Chronic Exposure To Metals And Radionuclides On Marine Mussels: Relating Genotoxicity To Molecular And Organism-Level Effects, Lorna Jane Dallas
School of Biological and Marine Sciences Theses
Metals and radionuclides are environmentally relevant contaminants, yet their potential impacts on marine organisms have not been adequately evaluated. This is especially true for exposures of longer duration and/or lower contaminant concentration (i.e. chronic) which are often more representative of real world scenarios. In this context, a suite of biomarkers at different levels of biological organisation were investigated in an ecologically relevant bivalve species, Mytilus galloprovin- cialis after exposure to nickel (a metal), zinc pyrithione (an organometal) and tritiated water (a radionuclide). These contaminants were chosen based on their differing properties, and hence, mechanisms of action. All three contaminants produced …

Signalling And Behaviour Of Globodera Pallida In The Rhizosphere Of The Trap Crop Solanum Sisymbriifolium, Ayano Sasaki-Crawley
School of Biological and Marine Sciences Theses
Potato cyst nematodes (PCN), Globodera rostochiensis and G. pallida, are economically important pests of potato (Solanum tuberosum) crops in potato growing regions worldwide. Integrated management is under threat, with effective nematicides increasingly being withdrawn on environmental and health grounds. Alternative strategies are urgently needed and trap cropping could be one of them. The non-tuber-bearing Solanum sisymbriifolium is regarded as an effective trap crop for PCN with strong hatching ability and immunity to PCN infection and has been used in the UK and The Netherlands. However, its mode of action is unknown. In order to shed light on the mode of …
