Ranaviruses are emerging pathogens that can cause morbidity, mortality and population declines in ectothermic hosts; however, there is no standardized approach to diagnostics. Here, we compared the inter-assay variation and intra-assay precision among 2 commonly used quantitative PCRs (qPCRs), a conventional and a nested PCR assay (used as a gold standard), using laboratory-propagated ranavirus (FV3 and CMTV) and field-collected samples. A qPCR assay (‘Leung’) detected viral DNA in dilutions 2 orders of magnitude lower than other assays regardless of the viral lineage of the cultured isolate (FV3/CMTV). Plastic pollution is a pervasive and growing problem. To estimate the effectiveness of interventions to reduce plastic pollution, we modeled stocks and flows of municipal solid waste and four sources of microplastics through the global plastic system for five scenarios between 2016 and 2040. Implementing all feasible interventions reduced plastic pollution by 40% from 2016 rates and 78% relative to “business as usual” in 2040. Even with immediate and concerted action, 710 million metric tons of plastic waste cumulatively entered aquatic and terrestrial ecosystems. Mediterranean coasts are affected by multiple mounting pressures. In Cyprus, marine fish farming has grown rapidly in the past decade and is concentrated in the west side of Vasiliko Bay. The east coast of this bay has ports, a power station, a desalination unit, a cement factory, a major new oil terminal, and gas storage facilities. The bay is earmarked to create the largest hydrocarbon processing, storing, and transport facility in the region. Most clinicians receive little or no palliative care (PC) education. Similarly, patients and their families receive little or no information on PC. Our study explored education in PC, while examining for its impacts on service delivery and utilization from the perspective of health care professionals (HCPs), patients, and their families. An ethnographic approach was utilized to gather data from 41 participants. Spradley’s ethnographic analytical framework guided data analysis. Two themes identified were inadequate HCPs’ knowledge base and impact of service-users’ inadequate health education. Roundhouses are ubiquitous features of Iron Age landscapes across North West Europe, yet the way they were used internally is not well understood. We demonstrate how spatial analyses of steroid lipid biomarkers advances our understanding of household activities, living conditions and animal management associated with a well-preserved 5th century BCE roundhouse from Scotland's first Iron Age wetland village, Black Loch of Myrton, especially when combined with more traditional archaeological approaches. Faecal steroids (5ß-stanols and bile acids) are well preserved within the wetland roundhouse floor deposits. Diffuse faecal inputs are identified within these deposits, limiting the resolution of faecal source discrimination …

Climate change and coastal urbanization are driving the replacement of natural habitats with artificial structures and reclaimed land globally. These novel habitats are often poor surrogates for natural habitats. The application of integrated greening of grey infrastructure (IGGI) to artificial shorelines demonstrates how multifunctional structures can provide biodiversity benefits whilst simultaneously serving their primary engineering function. IGGI is being embraced globally, despite many knowledge gaps and limitations. It is a management tool to compensate anthropogenic impacts as part of the Mitigation Hierarchy. Background: the aim of this study was to assess the 2-year pulp survival of deep carious lesions in teeth excavated using a self-limiting protocol in a single-blind randomized controlled clinical trial. Methods: At baseline, 101 teeth with deep carious lesions in 86 patients were excavated randomly using self-limiting or control protocols. Standardized clinical examination and periapical radiographs of teeth were performed after 1- and 2-year follow-ups (REC 14/LO/0880). Results: During the 2-year period of the study, 24 teeth failed (16 and 8 at T12 and T24, respectively). Final analysis shows that 39/63 (61.9%) of teeth were deemed successful (16/33 (48.4%) …

Purpose: This study compared the antimicrobial efficacy of calcium silicate sealers (BioRoot RCS and Total Fill BC) and conventional sealers (AH Plus and Tubli-seal) against planktonic bacteria and a nutrient-stressed multispecies biofilm. Methods: Antimicrobial properties of freshly mixed sealers were investigated using the direct contact test (DCT) and a nutrient-stressed multispecies biofilm comprised of five endodontic strains. Antimicrobial activity was determined using quantitative viable counts and confocal laser scanning microscopy (CLSM) analysis with live/dead staining. We revise the Afrotropical Agabus raffrayi species group, motivated by the discovery of new diversity in Kenya and South Africa. Whilst Agabus is mainly a holarctic genus, the Agabus raffrayi group is restricted to high altitude regions of eastern Africa and temperate parts of South Africa, from where we describe the southernmost Agabus in the world. This paper proposes an interactive approach to aid the compositional process using the brain-computer music interface (BCMI). In particular, the system developed in this paper focuses on creating an interactive compositional process inclusive to non-musicians and those with motor disabilities, in addition to providing a new method of composing. This paper references and compares two BCMI systems that have been developed for compositional purposes and draws upon these ideas to form the proposed system in this paper. Such system would aid the compositional process based on note velocity, chords and key changes.

The river Teign in Devon has come under scrutiny for failing to meet environmental quality standards for ecotoxic metals due to past mining operations. A disused mine known as Bridford Barytes mine, has been found to contribute a significant source of Zn, Cd and Pb to the river. Recently, studies have been focused on the remediation of such mine sites using low-cost treatment methods to help reduce metal loads to the river downstream. With sea level rise accelerating and coastal populations increasing, the requirement of coastal managers and scientists to produce accurate predictions of shoreline change is becoming ever more urgent. Waves are the primary driver of coastal evolution, and much of the interannual variability of the wave conditions in the Northeast Atlantic can be explained by broadscale patterns in atmospheric circulation. Two of the dominant climate indices that capture the wave climate in western Europe’s coastal regions are the ‘Western Europe Pressure Anomaly’ (WEPA) and ‘North Atlantic Oscillation’ (NAO). This paper introduces GeMS, a system for music composition informed by Synthetic Biology. GeMS generates music with simulations of genetic processes, such as transcription, translation and protein folding, with which biological systems render chains of amino acids from DNA strands. The system comprises the following components: the Miranda Machine, the Rhythmator and the Pitch Processor. The Miranda Machine is an abstract Turing Machine-like processor, which manipulates a sequence of DNA symbols according to a set of programming instructions. Background: Over 13,000 new cases of non-Hodgkin’s lymphoma (NHL) are diagnosed in the UK, with approximately 4,900 attributable deaths each year. Diffuse Large B-cell Lymphoma (DLBCL) is the most common NHL comprising one third of adult NHL cases. R-CHOP (rituximab, cyclophosphamide, doxorubicin, vincristine, prednisolone) is accepted as the international standard first-line regimen, but improvement in first line treatment is needed. Dysregulated B-cell receptor (BCR) signalling has been identified as a feature of DLBCL. Final 5-Year Findings From The Phase 3 Helios Study Of Ibrutinib Plus Bendamustine And Rituximab In Patients With Relapsed/Refractory Chronic Lymphocytic Leukemia/Small Lymphocytic Lymphoma, Gam Fraser, S Rule, Et Al

We report final analysis outcomes from the phase 3 HELIOS study (NCT01611090). Patients with relapsed/refractory chronic lymphocytic leukemia/small lymphocytic lymphoma without deletion 17p (n = 578) were randomized 1:1 to 420 mg daily ibrutinib or placebo plus ≤6 cycles of bendamustine plus rituximab (BR), followed by ibrutinib or placebo alone. Median follow-up was 63.7 months. Median investigator-assessed progression-free survival was longer with ibrutinib plus BR (65.1 months) than placebo plus BR (14.3 months; hazard ratio [HR] 0.229 [95% confidence interval (CI) 0.183-0.286]; p < .0001). Despite crossover of 63.3% of patients from the placebo plus BR arm to ibrutinib treatment upon disease progression, ibrutinib plus BR versus placebo plus BR demonstrated an overall survival benefit (HR 0.611 [95% CI 0.455-0.822]; p = .0010; median not reached in either arm). Long-term follow-up data confirm the survival benefit of ibrutinib plus BR over BR alone. Safety profiles were consistent with those known for ibrutinib and BR.
