This paper summarizes studies that were performed to evaluate the effectiveness and utility of protocols used for RNA isolation and detection. In addition, DNA probes were prepared and evaluated. Methodologies that were evaluated include the maxi-prep and mini-prep plasmid preparation techniques; isolation of RNA by extraction with organic solvents and by centrifugation through a cesium chloride gradient; and comparison of the detection of RNA-DNA hybrids using radioactively-labeled probes and digoxigenin-labeled probes. The maxi-prep and mini-prep plasmid preparation protocols both produced good quality DNA. The maxi-prep method was superior to the mini-prep method in terms of total quantity of DNA produced, …

To better understand the mechanisms governing the proliferation of cardiac myocytes it is important to identify the factors controlling this phenomenon, and to characterize their actions. The ventricular myocyte from the adult newt, Notophthalmus viridescens, is well suited to this type of study, due to its large chromosomes, ease of culture maintenance, and ability to undergo mitosis with cytokinesis. DNA synthesis was quantified in vitro in myocytes treated with platelet-derived growth factor (PDGF), transforming growth factor-beta (TGF-$\beta$), fibroblast growth factors (FGFs), 12-0-tetradecanoylphorbol-13-acetate (TPA), heparin, or conditioned medium from ventricular myocytes or nonmyocytes (primarily endothelial cells). The existing headache literature neglects to examine the possible differences between tension headache subjects and controls with regard to cognitive appraisals or coping styles. Also, despite the traditional view that tension headaches occur in close proximity to a stressor, there is limited empirical evidence supporting this idea (Ad Hoc Committee, 1962; Gannon, Haynes, Cuevas & Chavez, 1987). In addition, the results from studies on the efficacy of the traditional muscle-contraction model of tension headaches are ambiguous (Chapman, 1986; Hursey, Holroyd, Penzien & Holm, 1985).Lazarus and Folkman (1984) suggest that a person's ideas will have the greatest influence on appraisal when …

Twenty-six male and female spouses of patients served by a small hospice in the upper Midwest served as subjects for this dissertation. At the time of referral of their terminally ill spouse they completed measures of Sense of Coherence and social support. Their spouses died from an anticipated death, cancer, and they were contacted by phone within the first three months of bereavement. At that time, measures of grief, depression, anxiety, and anger were obtained.This first hypothesis, that social support would influence bereavement outcome (spouses with more social support would experience less grief, depression, anxiety, and anger than spouses with …

5,424 Japanese prisoners were captured by Allied forces during World War II and brought to the United States for internment. No scholarly account has been written of these men. This thesis will explain why there were so few, where they were interned, and the conditions of their internment.

The topic was researched at the National Archives and Records Administration in Washington, D. C., through interviews with veterans who served in the Pacific Campaign or had contact with the Japanese prisoners, through newspaper and magazine articles of the time, and through secondary sources of the Pacific Campaign.

The present study was designed to investigate the relationship between locus of control and alcoholism. The questions which this study sought to address included a comparison of locus of control in inpatient and outpatient white male treatment groups with a nonalcoholic control group. In addition changes in locus of control during alcohol treatment were also assessed.

A total of 129 white men were placed into one of three groups. Thirty-one men from the community who scored within the nonalcoholic range on the Michigan Alcoholism Screening Test (MAST) made up the control group. Two groups of men in treatment for alcoholism …

The purpose of this study was to investigate curricular patterns for general education requirements in elementary teacher education during the period 1970-1990 and to examine the general education of elementary education seniors at the University of North Dakota in 1987-88 within the framework of changing views of the purposes of general education in teacher education.

Students were selected randomly from a list of seniors majoring in elementary education, elementary/early childhood education and elementary/special education during spring semester of 1988. This qualitative study focuses on five prospective female teachers' attitudes toward mathematics. The meanings each informant ascribes to her experiences surrounding mathematics are described in this work. Each informant was interviewed weekly during her senior mathematics methods course and observed and interviewed weekly throughout her student teaching experience. Interviews were open-ended and was the central focus of methodology. A field diary was used to record the reflective portions of field notes.

Experiences were carefully detailed and the informants directly quoted throughout the portraits and themes. The themes that emerged were: (1) meaning in mathematics, (2) narrow view of mathematics, (3) …

Eric Overmyer's play On the Verge focuses on three women who leave their world in the year 1888 and enter Terra Incognita, a fanciful world that propels then into the future until they land in the year 1955. This study examined the relationships between previous school and life experiences of pre-service elementary school teachers at the University of North Dakota and their attitudes toward science and science teaching.

The study incorporated both qualitative and quantitative methodologies. The first six pre-service teachers from one of the science methods class who volunteered were selected for taped private interviews. Based on this information a quantitative survey (Sampson Survey I) was designed for discovering the relationship between past experiences of pre-service teachers and their current attitudes toward science and teaching science. Pulsed electromagnetic fields (EMF) and electric fields have been demonstrated to promote osteogenesis and wound healing. Pulsed EMF's have been approved since 1979 by the FDA, and are highly effective in the treatment of non-union fractures. Increased linear growth, cellular proliferation, cAMP and uptake of tritiated thymidine have been documented on short term exposure. Yet the mechanisms have been difficult to quantify. Little has been published in the literature about the effects of a magnetic fields upon bone growth. A Study Of The Relationship Of Maternal And Child Risk Factors To Development And Low-Income Status In Grand Forks County., Kay Elizabeth Campbell

The purpose of this study was to investigate the correlation between maternal and child risk factors and the variables of development, low-income status, and multiple births. The sample consisted of 193 children referred to the North Dakota Early Childhood Tracking System in Grand Forks County from 1988 to mid-1991. The referral criteria were based on established, biological, and environmental risk factors. The correlation between development and risk factors was not significant. Low-income status was significantly correlated to maternal risk factors (p $<$.001), short term respiratory problems (p $<$.05), neonatal risk factors (p $<$.05), and single parenthood (p $<$.001). Multiple births were significantly correlated to neonatal risk factors (p $<$.05) and prematurity (p $<$.001).

Spousal Bereavement In The Second Year: Differences In Bereavement Outcome Among The Rural And Urban Elderly., Laurie Ann Brustad

Twenty-five bereaved rural elderly subjects (10 males, 15 females) were compared with twenty-five bereaved urban elderly subjects (16 females, 9 males) in the second year after the death of a spouse. All subjects were drawn from a Hospice population. The rural elderly were found to have higher levels of depression, unresolved grief, and poorer health than the urban elderly as measured by the Multiscore Depression Inventory, the Beck Depression Inventory, the Wahler Physical Symptoms Inventory, and the Texas Revised Inventory of Grief. Multiple regression analyses revealed that several variables influenced scores on the outcome measures. Depression scores were best predicted …

Biological denitrification controls nitrate distribution in the Elk Valley aquifer, which underlies approximately 725 square kilometres of northeastern North Dakota and provides drinking water for farms and rural communities in the area. Increased irrigation has raised concerns over potential contamination from agricultural practices. The purpose of this study was to establish the role of biological denitrification in minimizing concentration and distribution of nitrate and to determine the effect irrigation practices have on groundwater flow and nitrate distribution in the saturated zone.

The aquifer is unconfined and consists of up to 20 metres of glacial outwash and deltaic shale-rich sand deposits. …
