Biochemical Characterization Of The Association Between Glycolytic Enzymes And Clathrin Coated Vesicles Using Centrifugation, Electrophoretic, And Ligand Blotting Techniques., Roxann Ranee Karkhoff-Schweizer Jan 1989

Biochemical Characterization Of The Association Between Glycolytic Enzymes And Clathrin Coated Vesicles Using Centrifugation, Electrophoretic, And Ligand Blotting Techniques., Roxann Ranee Karkhoff-Schweizer

Theses and Dissertations

Clathrin coated vesicles (CCVs) are complex organelles reported to bind specific glycolytic enzymes. The purpose of this study was to further biochemically characterize the interactions of selected enzymes with CCV's by conventional and new techniques.Purified brain CCVs were observed to retain, in varying degrees, endogenous glycolytic enzymes, with glyceraldehyde 3-phosphate dehydrogenase (GAPDH) and pyruvate kinase (PK) having the highest specific activities. Furthermore, Scatchard and Hill plot analyses of saturation binding data suggested that the binding of pure aldolase (ALD) and GAPDH involved heterogenous binding sites or cooperativity. Moreover, increasing concentrations of added ALD could not completely release bound GAPDH from …

Basement Membrane Heterogeneity: A High Resolution Sem Study Of Renal Basement Membrane Ultrastructure Following Enzyme Microdissection., Walter Jesse Berger Iii Dec 1988

Basement Membrane Heterogeneity: A High Resolution Sem Study Of Renal Basement Membrane Ultrastructure Following Enzyme Microdissection., Walter Jesse Berger Iii

Theses and Dissertations

The purpose of this study is to utilize high resolution SEM to examine topographical features of acellular renal cortical basement membranes (BMs) in the rat. Alterations in ultrastructure of protease-digested samples are evaluated in terms of the concept of BM heterogeneity, and an attempt is made to correlate observed changes with known components of BMs.Renal cortical blocks were rendered acellular by sequential detergent extraction, and digested either with 150 $\mu$g/ml Streptomyces griseus protease (SGP) or with 5 mg/ml pepsin and conventionally prepared for SEM and correlative TEM.With both proteases, Bowman's capsular BM (BCBM), tubular BM (TBM) and glomerular BM (GBM) …

Induced Resistance To Ixodid Tick Infestation: Analysis And Isolation Of Antigens., Fred William Olsen Jr. Dec 1988

Induced Resistance To Ixodid Tick Infestation: Analysis And Isolation Of Antigens., Fred William Olsen Jr.

Theses and Dissertations

Ticks are both vectors and reservoirs for significant pathogens of man and livestock. They are currently controlled through use of acaracides, but have rapidly developed resistance to every new acaracide produced to date. Alternative methods of control are needed. One proposed alternative is immunologic control, achieved through vaccinations which induce resistance to tick infestation. This proposal stems from the knowledge that cattle and laboratory animals actively acquire resistance to infestation. Immunization with crude extracts of different ixodid life cycle stages or organs has induced limited levels of resistance compared to acquired resistance. Defined tick antigens are needed to study the …

The Effects Of Hydrocortisone And Caffeine On Memory For Word Lists, Daniel L. Scharf Dec 1988

The Effects Of Hydrocortisone And Caffeine On Memory For Word Lists, Daniel L. Scharf

Theses and Dissertations

Hydrocortisone has frequently been hypothesized to increase arousal. This hypothesis was primarily based on the findings of increased adrenal size in animals who had been exposed to stressful conditions and the enhancement or impairment in cognitive abilities of humans depending on dose. There has been a lack of studies directly looking at the arousal hypothesis of the effects of hydrocortisone. The purpose of this study was to directly assess the effects of hydrocortisone on levels of arousal by manipulating both state and trait arousal. This study used caffeine as a comparison drug that alters state arousal because of its widely …

Napoleon And Russia And The Teaty Of Tilsit: Its Implications For Europe, Paul A. Hefti Dec 1988

Napoleon And Russia And The Teaty Of Tilsit: Its Implications For Europe, Paul A. Hefti

Theses and Dissertations

The Treaty of Tilsit of 1807 between Russia and France was a minor slowing of the flow of the Napoleonic Wars that dominated Europe in the early nineteenth century. It united the two nations against Britain, but, in larger terms, it polarized continental Europe into two separate (or distinct) conflicting spheres. The treaty and its effects provided a foreshadowing of post-World War I Europe and the subsequent twentieth-century concept of peaceful coexistence.

The major personalities involved in making the treaty were Alexander I of Russia and Napoleon Bonaparte of France. Their meeting on a boat in the Niemen River had …

Vasopressin's Effect On Sex Differences In Cognitive Functions: Verbal And Spatial Abilities And Verbal-Interference Tasks., Karen Kay Knutson Aug 1988

Vasopressin's Effect On Sex Differences In Cognitive Functions: Verbal And Spatial Abilities And Verbal-Interference Tasks., Karen Kay Knutson

Theses and Dissertations

The effects of a vasopressin analogue, desmopressin acetate (DDAVP) upon cognitive functioning performance was investigated in this study. Forty male and forty female college students were first assessed using the Vocabulary and the Block Design subtests of the Wechsler Adult Intelligence Scale-Revised (WAIS-R) to determine their verbal and spatial-visualization ability levels. Then they were treated intranasally with either DDVAP (60 ug), or an equal volume of saline. Finally, subjects were tested using these cognitive measures: The Paper Folding Test, the Stroop Color and Word Test and twelve lists of semantically unrelated words.Analyses of variance of the test data identified several …

Lysosomal Enzymes In Undifferentiated And Differentiated Hl-60 Cells, Julie J. Ackerman Aug 1988

Lysosomal Enzymes In Undifferentiated And Differentiated Hl-60 Cells, Julie J. Ackerman

Theses and Dissertations

The human promyelocytic leukemia cell line HL-60 has been studied extensively since the time of it's discovery and has been proven to be a valuable tool for the study of the differentiation process. The capacity of the HL-60 cell line to differentiate to macrophage-like cells, which may be immunologically competent, has proven to be invaluable in the study of these cells as a model for macrophage function. A number of hydrolytic enzymes are characteristically found in macrophages. In an effort to further characterize diferentiated HL-60 cells as macrophages, the location and concentration of a number of these enzymes was determined. …

A Study Of The Effects Of Polyethylene Glycol On Interactions Between Glycolytic Enzymes And F-Actin, And Among Glycolytic Enzymes, Julie L. Walsh May 1988

A Study Of The Effects Of Polyethylene Glycol On Interactions Between Glycolytic Enzymes And F-Actin, And Among Glycolytic Enzymes, Julie L. Walsh

Theses and Dissertations

Many studies of the protein interactions that compose the microtrabecular lattice have been done in dilute solutions. However, solutions containing inert polymers such as polyethylene glycol (PEG) more effectively mimic the crowded interior of the cell. Therefore, the interactions of D-glyceraldehyde-3-phosphate ketolisomerase (TPI, EC 5.3.1.1), D-phosphoglycerate 2,3-phosphomutase (PGM, EC 5.4.2.1), ATP:3-phospho-D-glycerate 1-phosphotransferase (PGK, EC 2.7.2.3), 2-phospho-D-glycerate hydro-lyase (enolase, EC 4.2.1.11), D-fructose-1,6-bisphosphate D-glyceraldehyde-3-phosphate lyase (aldolase, 4.1.2.13), D-glyceraldehyde-3-phosphate:NAD$\sp{+}$ oxidoreductase (GAPDH, EC 5.3.1.1), D-glucose-6-phosphate ketol-isomerase (GPI, EC 5.3.1.9), (S)-lactate:NAD+ oxidoreductase (LDH, EC 1.1.1.27), and ATP:Pyruvate O$\sp2$-phosphotransferase (PK, EC 2.7.1.40) with each other, with ATP:D-fructose-6-phosphate 1-phosphotransferase (PFK, EC 2.7.1.11) and with F-actin have been …

Ethnogeography Of Middle Missouri River Indians: 1738-1889, Kenneth C. Dagel May 1988

Ethnogeography Of Middle Missouri River Indians: 1738-1889, Kenneth C. Dagel

Theses and Dissertations

The purpose of this thesis is to present the ethno- geography of the Mandan and Hidatsa Indians, and to a lesser extent, the ethnogeography of the Arikara and Lakota during the period 1738-1889. It provides not only a listing of place-names, but it also includes an explanation of why the locations associated with those names were so important, and remain important to these tribes.

The primary hypothesis of this research is that toponyms of Native American origin were generated because of perceptual and/or pragmatic reasons. Place-names, obtained from historical journals and secondary sources, were classified using a standard toponym typology. …

Pragmatics And Grammar: Motivation And Control, Robert A. Dooley Jan 1988

Pragmatics And Grammar: Motivation And Control, Robert A. Dooley

Work Papers of the Summer Institute of Linguistics, University of North Dakota Session

From the introduction: "Much of what has been done as "pragmatics" has dealt with utterance interpretation, that is, with the influence of context on meaning. Relatively little has been said about the interaction between context and form. As a result, there is much we do not know about how pragmatic phenomena relate to the grammar of a language. The present paper makes three points in this regard: (a) it is not anomalous for there to be pragmatic phenomena encoded in the syntax and controlled by the grammar; (b) it is not anomalous for there to be pragmatic phenomena encoded in …
