Meaning construction is a complex concept that encapsulates language and thought as a whole system in which sense and meaning have a unique relationship highly marked by the social and cultural environment of the reader. Hence, Kichwa-Spanish bilinguals’ reading practices in the Southern Andes of Ecuador cannot be discussed as two separate processes but as an integrated structure embedded in a speech community. UNLABELLED: Human papillomaviruses (HPVs) are a significant public health concern due to their widespread transmission, morbidity, and oncogenic potential. Despite efficacious vaccines, millions of unvaccinated individuals and those with existing infections will develop HPV-related diseases for the next two decades. The continuing burden of HPV-related diseases is exacerbated by the lack of effective therapies or cures for most infections, highlighting the need to identify and develop antivirals. The experimental murine papillomavirus type 1 (MmuPV1) model provides opportunities to study papillomavirus pathogenesis in cutaneous epithelium, the oral cavity, and the anogenital tract. The websites of disability resource centers at six universities are used in a discourse analysis to forefront ways in which the Whiteness of disability is upheld. The main research question, built using a DisCrit (Disability/Critical Race theory) lens, is: how do the institutional discourses of disability resource centers reproduce or challenge particular identities for college students with disabilities? The research sub-question explored in this dissertation that built off of this wider scope is: how are the politics of intersectionality addressed in such discourses? Shoulder pain and dysfunction may significantly impact quality of life. If conservative measures fail, advanced disease is frequently treated with shoulder arthroplasty, which is currently the third most common joint replacement surgery following the hip and knee. The main indications for shoulder arthroplasty include primary osteoarthritis, post-traumatic arthritis, inflammatory arthritis, osteonecrosis, proximal humeral fracture sequelae, severely dislocated proximal humeral fractures, and advanced rotator cuff disease. Several types of anatomic arthroplasties are available, such as humeral head resurfacing and hemiarthroplasties, as well as total anatomic arthroplasties. Aquatic and riparian food webs are inextricably linked. In deserts, they provide critical energy subsidies to surrounding terrestrial food webs, but these vital subsidies are not guaranteed; streams in arid lands are especially sensitive to perturbations. In this dissertation, I investigated the role of an oft-cited threat to stream communities in the lower Colorado River basin: non-native crayfish. I contributed to methodologies in passive integrated transponder use and length-weight relationships in crayfish to aid future research. I leveraged stable isotope analysis of δ15N, δ13C, and δ2H to understand crayfish role in the food webs. …

Transcriptional Silencing Of Cdk18 And Its Role In Lung Carcinogenesis Of Brg1-Mutant Lung Cancers, Loryn M. Phillips

BRG1 is mutated in 10% of lung cancers, making this mutation clinically relevant. The downstream effects of BRG1 included significantly affecting the cell cycle control and chromosomal replication pathway. CDK18, a cyclin-dependent kinase, was determined to be the gene with significantly decreased expression (p < 0.0001) in the cell cycle control and chromosomal replication pathway. CDK18 is active during the S-phase of the cell cycle and required for genomic stability. Studies were conducted to determine the epigenetic mechanisms that contributed to the repression of CDK18. Histone methylation contributes to the repression of CDK18, and histone deacetylation globally regulates expression of CDK18, while cytosine methylation was found to not contribute to CDK18 expression. Studies were performed to elucidate CDK18 functional role lung carcinogenesis and the genome-wide impact of CDK18 repression. Repression of CDK18 affects cell migration, proliferation, cell-cell signaling, and anchorage-independent growth that contribute to cancer initiation and progression.

Intriguing photophysical properties of color centers in diamond make them ideal candidates for many applications from imaging and sensing to quantum networking. In the first part of this work, we have studied the silicon vacancy (SiV) centers in diamond for nanoscale imaging applications. We showed that these centers are promising fluorophores for Stimulated Emission Depletion (STED) microscopy, owing to their photostable, near-infrared emission and favorable photophysical properties. In the second part, we built a femtotesla Radio-Frequency (RF) magnetometer based on the diamond nitrogen vacancy (NV) centers and magnetic flux concentrators. Understanding what motivates non-clinical healthcare workers to seek feedback in the workplace is critical in supporting such behavior within organizations. This study aims to answer the following research question “Why do non-clinical healthcare workers seek feedback in the workplace?” To conduct this study, I analyzed 13 semi-structured interviews with non-clinical healthcare workers. Findings confirmed the presence of four self-motives (self-assessment, self-improvement, self-enhancement, and self-verification). Additionally, study findings highlighted the presence of other-oriented motives, when individuals seek feedback due to their concern or care for others (i.e., patients, teams, and supervisors). Sadism is an elusive construct within psychology. Multiple types are studied without clear psychometric or theoretical distinctions, and operationalizations of these respective sub-constructs lack validity. This study explores the empirical distinction between two sadism types: consensual sexual sadism (i.e. in the context of BDSM) and trait sadism. Trait sadism is widely synonymized with “everyday sadism”, but here conceptualized as a higher-order construct encompassing both everyday and a novel “prosocial sadism”. I develop and pilot the BDSM Identities and Behaviors (BIB) checklist in a sample of BDSM practitioners. I then compare those practitioners to non-practitioners on trait sadism and dark triad …

Despite the nearly ubiquitous nature of sleep in the animal kingdom, behavioural research on sleep has focused on a few model organisms and widespread behaviours studied in artificial laboratory settings. In this study, we examine the ontogeny of sleeping perch selection in four closely related species of arboreal lizard in situ. The study species each occupy similar habitats and share "solitary" ecology wherein potential lizard competitors are absent, thus providing four partially independent replicates with which to test hypotheses. We find that structural niche, analysed as perch height, varies by age and sex. Males sleep higher than females and adults …

Medical students in the Quantitative Medicine Block completed an Evidence Based Practice (EBP) assignment worth 10% of the final Block grade. The question formulation segment was worth 50% of the assignment score. Students received instructions with an designated subject and some parameters for developing their own EBP questions using the rubric. For space surveillance applications, neuromorphic imaging is being studied as it may perform sensing and tracking tasks with less power and downstream datalink demand. The read-out of the event-based camera is made to only be sensitive to changes in the signals it receives from the photodetector, which results in a datastream of events indicating where and when changes in illumination occur. This is in contrast to the conventional framing camera, which produces images by essentially counting the electrons produced by light incident on each pixel’s photodetector. Multiple sclerosis (MS) is a neurodegenerative disease in which the central nervous system’s myelin sheath is degraded by auto-immune cells. Relapse remitting multiple sclerosis (RRMS) is the most common MS phenotype, accounting for ~85% of new diagnoses. Disease modifying therapies (DMTs) are used in RRMS to reduce the frequency of relapses and prevent disability progression. The present study performed a cost effectiveness analysis of three B-cell depleting monoclonal antibody DMTs: ocrelizumab, ofatumumab and rituximab, in comparison to commonly utilized interferon beta-1a (IFN-B1a). Objective: Bodyweight interval training (BW-IT) using a social media platform may appeal to those who do not wish to exercise in public or do not have access to specialized exercise equipment. We investigated the ability of a 6-week BW-IT program to improve cardiovascular and metabolic markers. Methods: Fourteen adults (30.7 ± 10.3 yrs.) with obesity (body mass index 35.5 ± 5.4 kg/m2) participated. IMPORTANCE: Emotional and behavioral dysregulation during early childhood are associated with severe psychiatric, behavioral, and cognitive disorders through adulthood. Identifying the earliest antecedents of persisting emotional and behavioral dysregulation can inform risk detection practices and targeted interventions to promote adaptive developmental trajectories among at-risk children.
OBJECTIVE: To characterize children's emotional and behavioral regulation trajectories and examine risk factors associated with persisting dysregulation across early childhood.

Background:
Engaging diverse populations in cancer genomics research is of critical importance and is a fundamental goal of the NCI Participant Engagement and Cancer Genome Sequencing (PE-CGS) Network. Established as part of the Cancer Moonshot, PE-CGS is a consortium of stakeholders including clinicians, scientists, genetic counselors, and representatives of potential study participants and their communities. Participant engagement is an ongoing, bidirectional, and mutually beneficial interaction between study participants and researchers. PE-CGS sought to set priorities in participant engagement for conducting the network's research.

SIGNIFICANCE STATEMENT: Pathogenic structural genetic variants, also known as genomic disorders, have been associated with pediatric CKD. This study extends those results across the lifespan, with genomic disorders enriched in both pediatric and adult patients compared with controls. In the Chronic Renal Insufficiency Cohort study, genomic disorders were also associated with lower serum Mg, lower educational performance, and a higher risk of death. A phenome-wide association study confirmed the link between kidney disease and genomic disorders in an unbiased way. Systematic detection of genomic disorders can provide a molecular diagnosis and refine prediction of risk and prognosis.

Transferrin receptor (TFRC) is the major mediator for iron entry into a cell. Under excessive iron conditions, TFRC is expected to be reduced to lower iron uptake and toxicity. However, the mechanism whereby TFRC expression is maintained at high levels in iron-enriched cancer cells and the contribution of TFRC to cancer development are enigmatic. Here the work shows TFRC is induced by adenomatous polyposis coli (APC) gene loss-driven β-catenin activation in colorectal cancer, whereas TFRC-mediated intratumoral iron accumulation potentiates β-catenin signaling by directly enhancing the activity of tankyrase. SIGNIFICANCE STATEMENT: Pathogenic structural genetic variants, also known as genomic disorders, have been associated with pediatric CKD. This study extends those results across the lifespan, with genomic disorders enriched in both pediatric and adult patients compared with controls.
