The spatial organization and dynamics of proteins and lipids within the cell membrane is important for the regulation of cell signaling, adhesion, and cell communication. Within the bone marrow niche, communication between hematopoietic stem/progenitor cells (HSPCs) and niche cells is essential for regulating their proliferation, differentiation, and survival. Our previous work has ascertained that HSPCs utilize a polarized domain on the plasma membrane that serves as the contact site with osteoblasts, which are important members of the bone marrow niche. The focus of the work in this dissertation is to improve the efficiency of a gene therapy for the treatment of chronic pain. The introduction, Chapter 1, is intended to orient the reader to the underlying physiological principles and anatomical structures involved in general sensory and pain transmission in the peripheral and central nervous systems. This thesis is an in-depth exploration of the situation of women in the border town of Mezquital, Mexico. The local experiences of women in Mezquital are situated within the larger theoretical debates of economic policy in Latin America, gender, labor and development. The theoretical analysis sets the stage to facilitate a process aimed at assisting the local women of Mezquital in creating alternative income generating opportunities to meet their specific economic needs and to highlight their talents. Guided by a social-ecological approach to health, this study explored a new approach to sex education--sex-positive sexuality education (SPSE). A collective case study was completed on three organizations that utilize a sex-positive approach to sexuality education--Good Vibrations, Scarleteen.com, and The National Sexuality Resource Center's Summer Institute. Good Vibrations is an adult sex toy retailer, Scarleteen.com is an adolescent sexual health website, and the Summer Institute is an academic institute for scholars and practitioners of sexuality studies. Computer architecture design is in a new era where performance is increased by replicating processing cores on a chip rather than making CPUs larger and faster. This design strategy is motivated by the superior energy efficiency of the multi-core architecture compared to the traditional monolithic CPU. Dietary niches have widespread effects on individuals life histories, behaviors, and morphologies. Capuchin monkeys inhabit a complex dietary niche that often entails hunting of relatively large vertebrate prey, tool-use, and extraction of embedded resources that other closely related and sympatric species do not exploit. In this dissertation I examine, a) how juvenile capuchins overcome the challenges of reliance on a difficult-to-acquire diet, b) at what age juveniles achieve maximum foraging return rates for difficult-to-acquire foods, and c) what nutritional benefits capuchins obtain from exploitation of these foods. Where animals spend time can provide important clues to their ecological needs, but this information is very difficult to obtain for small volant animals. The research described in Chapter 1 was an attempt to discern how L. nivalis utilize the landscape in relation to the distribution of Agave havardiana in Big Bend National Park, Texas. I found that although the landscape use of Leptonycteris nivalis is centered on habitat with high concentrations of blooming A.havardiana, it is not restricted to those areas, and furthermore adults and juveniles may differ in their behavior. The Effect Of Increased Growth Temperature On Mid-Day And Mid-Night Co2 Fluxes In Populus Deltoids, Rachel Zulick

As global temperatures rise, understanding the effects of increased temperature on daily carbon dioxide fluxes is necessary to predict changes to the global carbon cycle. Using tunable diode laser spectroscopy, I measured CO2 fluxes and changes in isotopic composition of root respiration, leaf respiration and photosynthesis over a 12-hour night to day transition. I found that after a 10°C increase in growth temperature for four weeks, root CO2 fluxes acclimated to the higher temperature (i.e. they were not significantly different from controls). In contrast, both photosynthesis and respiration of leaves were higher at the elevated growth temperature. Though the root …

Marsupials are the closest relatives to eutherians. They are born immune-incompetent. Androgen deprivation therapy, one of the standard treatments for prostate cancer, induces apoptosis as well as autophagy in androgen-responsive PCa cells. As modulation of autophagy is a new paradigm for enhancing the therapeutic efficacy of various cancer therapies, we sought to determine the functions of autophagy during androgen deprivation. In this study, we confirmed that androgen removal or inhibition induces autophagy in two different hormone sensitive prostate cancer cells. The genus Flavivirus comprises viral species with positive sense, single stranded RNA genomes, most of which are transmitted by mosquitoes and ticks. During replication, RNA viruses have higher mutation rates than DNA viruses, because their virally encoded RNA dependent RNA polymerase has no proofreading activity. These viruses have been shown to exist within and between hosts as swarms of closely related mutant genomes that can be examined through sequence analysis. Fetal alcohol spectrum disorder results from developmental exposure to ethanol. Although many organ systems are targeted by this teratogen, the central nervous system is exquisitely sensitive. Children with this disease often present with behavioral disorders as well as impairments in learning and memory. Ethanol exposure affects developmental processes throughout pregnancy; however, the third trimester-equivalent has demonstrated heightened sensitivity. My dissertation evaluated whether the palaces built in Florence, Italy during the Renaissance are a form of costly signaling. Costly signaling theory was developed to explain why organisms have attributes and behaviors that appear to defy basic Darwinian logic by having costs that are not offset by obvious benefits. The theory proposes that such attributes and behaviors persist because they are reliable signals of information about the signalers. Signal audiences use the information content of signals to rank signalers and to modify their interactions with signalers in ways that benefit signalers. Myogenic tone is an important regulator of blood flow and may contribute to peripheral vascular resistance and blood pressure. Myogenic tone is a luminal pressure-induced constriction of the vasculature that is mediated by a vascular smooth muscle cell (VSMC) plasma membrane potential (Em) depolarization and Ca2+ influx. Ca2+ sparks, which are ryanodine receptor (RyR) mediated Ca2+ release events, oppose myogenic tone by activating large-conductance Ca2+-activated K+ channels (BKCa) to hyperpolarize VSMC Em. Respiratory infections with the virulent type A strains of Francisella tularensis (Ft) cause pneumonic tularemia, an illness which results in high case fatalities in the absence of timely antibiotic treatment. Although immunization is an effective countermeasure, vaccines against pulmonary tularemia are currently unavailable. A better understanding of protective immune components is therefore needed to develop licensable vaccines against pulmonary tularemia. While the indispensability of cell-mediated immunity in protection against this gram-negative intracellular bacterium is well-documented, the protective role of antibodies (Abs) is unclear. Estrogen (17β-estradiol, E2) plays an important role in regulating an array of functions in both male and female reproductive physiology. In the mammary gland, E2-induced proliferation, ductal outgrowth and subsequent branching morphogenesis is required for proper development of the breast. In males, E2 is required for proper testicular development, spermatogenesis, and sperm maturation in the epididymis, but can also negatively regulate these functions with inappropriate exposure.
