R-Ketorolac Targets Cdc42 And Rac1 And Alters Ovarian Cancer Behaviors Critical For Invasion And Metastasis, Yuna Guo

Cdc42 (cell division control protein 42) and Rac1 (Ras-related C3 botulinum) are attractive therapeutic targets in ovarian cancer based on their established importance in tumor cell migration, adhesion and invasion. Despite a predicted benefit, targeting GTPases has not yet been translated to clinical practice. Based on lead identification through high-throughput screening and computational shape homology approaches, R-ketorolac is identified as a Cdc42 and Rac1 regulator, an activity that is distinct from the anti-inflammatory, cyclooxygenase inhibitory activity of S-ketorolac. In this dissertation, R-ketorolac is established as an allosteric inhibitor of Cdc42 and Rac1. Studies on immortalized human ovarian adenocarcinoma cells (SKOV3ip), …

Drinking during pregnancy can result in fetal alcohol spectrum disorders (FASD), an umbrella term that encompasses a wide array of symptoms ranging from neurocognitive deficits to physical malformations. The mechanism for FASD is not yet fully understood, however recently, activation of the neuroimmune system has been suggested to play a role. Neuroinflammation, characterized by increased pro-inflammatory cytokine production and glial cell activation, has been observed following exposure to alcohol during the neonatal period. Furthermore, neuroimmune activation has been implicated in alcohol-induced neuronal death and/or damage. Background: Post-placental intrauterine device (IUD) insertion has the potential to reduce rapid repeat pregnancies. As the procedure becomes more widely adopted in the US, it is important to understand womens perceptions about the procedure. Studies examining patient-centered outcomes are currently lacking. Objectives: The objectives of this study include: 1) To describe women's experiences with post-placental IUD insertion through postpartum semistructured interviews. 2) To establish a mean pain score for the procedure using a 100mm visual analogue scale (VAS). 3) To assess procedural pain using a 4-point Likert verbal rating scale (VRS). S. aureus is the dominant cause of skin and soft tissue infections (SSTIs) in humans. The importance of S. aureus as the primary cause of skin infections has increased exponentially over the last few decades since the emergence of resistance to β-lactam antibiotics. Furthermore, the rise of methicillin-resistant S. aureus (MRSA) strains has led to hospitals stays, increased financial burden and increased mortality. A significant proportion of MRSA infections have been attributed to community-acquired strains (CA-MRSA), specifically the USA300 isolates, which can cause deadly disease in otherwise healthy individuals. Skin cancer is the one of the most diagnosed cancers in the United States with increasing incidence of 6% every year. In 2015, it is estimated 73,870 new cases of melanoma will be identified and 9,480 individuals will die of their disease. While melanoma only accounts for approximately 2.4% of all cancer related deaths and is the 5th leading diagnosed cancer (US) it is the one of the most common cancers in young adults, age 25-29, particularly for young women. Radiation therapy is one of the most effective means for treating cancers. An important calculation in radiation therapy is the estimation of dose distribution in the treated patient, which is key to determining the treatment outcome and potential side effects of the therapy. Biological dose — the level of biological damage (e.g., cell killing ratio, DNA damage, etc.) inflicted by the radiation is the best measure of treatment quality, but it is very difficult to calculate. Shiga toxin-producing Escherichia coli (STEC) strains are important food-borne pathogens that have been linked to human illness. In the United States, STEC O157:H7 strains have been attributed to the majority of disease and outbreaks; however, the frequency of disease associated with STEC non-O157 strains have also increased and is often under recognized due to diagnostic limitations. In 2012, the Food Safety and Inspection Service (FSIS) implemented mandatory testing for non-O157 STEC serogroups O26, O45, O103, O111, O121, and O145. Shiga toxin-producing Escherichia coli (STEC) have been identified by the USDA as a serious threat to the nations health stemming from contaminations in the food supply, specifically, the beef chain. I have developed an assay that is able to screen samples for STEC in a rapid and multiplexed format. Multiplex oligonucleotide ligation-PCR (MOL- PCR) is a nucleic acid based assay patented at Los Alamos National Laboratory (LANL) that uses flow cytometry and multiplex microsphere arrays for detection of nucleic acid based signatures. We report rediscovery of the Chihuahuan Dwarf Crayfish Cambarellus chihuahuae at the desert spring Ojo Solo in Ejido Rancho Nuevo in Chihuahua, México. This species was reported to be extinct. Morphological evaluation of 12 voucher specimens confirmed the population as C. chihuahuae. Crayfish were abundant upon rediscovery in September 2012 and remained so throughout bimonthly monitoring from February 2014 to April 2015. Despite abundance at Ojo Solo, the species is critically imperiled, with four of its five native habitats dry and the remaining one in decline; however, a population was established at Ojo Caliente, a nearby natural refuge habitat we …

In the Open Air: Rediscovering Self and the Environment through Site-Specific Dance and Alternative Teaching Methods examines the accessibility of modern and contemporary dance today. It reflects on personal research, teaching values and the human/nature interface as motives for bringing dance out of the theatre and into nontraditional environments. I examine methodologies for creating and teaching dance in nature and nontraditional settings. To support this research I have studied Anna Halprin and her approach of taking dance outdoors as a way of promoting community and self-discovery. I also review why Halprins work matters to me. This study examined what New Mexico dentists have reported regarding the knowledge and confidence levels of dental hygienists in providing patient education on oral/systemic periodontal risk factors. Further examined were New Mexico dentists views are on structured SPT protocols that include dentist/dental hygienist patient education on major risk factors for oral/systemic disease, and referrals and consultation with primary physicians. Members of the New Mexico Dental Association (n-700) were asked to participate in a 20-question online survey. Survey questions focused primarily on the level of risk assessment for periodontal disease that New Mexico dentists report dental hygienists discussing with periodontal patients. …

Calcium leached from NUKON fiberglass in containment following a Loss of Coolant Accident (LOCA) could lead to the formation of chemical precipitates that in turn cause head-loss increase due to filtering through a fibrous debris accumulation at the sump pump screens. Experiments conducted on the bench scale show that the level of fiberglass destruction does not affect the concentration of calcium leached. Reduced-scale experiments were conducted on three solution inventory scales (0.5 L, 31.5 L, 1136 L) with three different flow conditions, and two fiberglass concentrations to investigate calcium release from NUKON fiber. Objectives: Patients who use substances or those who are on opioid maintenance therapy could be at risk of inadequate nutrition. These inadequacies could translate to adverse outcomes during pregnancy. The objective of this study was to determine differences in dietary macro and micronutrient intake in pregnant women on OMT compared to healthy controls. Methods: Participants from a parent prospective cohort study ENRICH' were classified into two groups: OMT users and healthy controls.
