Context.—: Autopsy pathologists, including medical examiners, provide valuable public health support for infectious disease deaths through surveillance for deaths of public health concern including emerging infections, identifying causative organisms for unexplained deaths, and providing insights into the pathology and pathogenesis of novel or unusual infections. However, autopsy poses biosafety risks to workers within and outside the laboratory. The highest rates of laboratory-acquired infections occur in autopsy workers.
Objective.—: To design and construct an appropriately biosafe autopsy laboratory.
Design.—: We conducted a biosafety risk assessment for autopsy workers using the process developed by the US Centers for Disease Control and Prevention …

Characterizing And Assessing The Hydrological Connection Of Sawyer Fen To Nearby Bluewater Creek In The Zuni Mountains, New Mexico, Luke Collis
Water Resources Professional Project Reports
Wetlands can hydrologically connect to nearby surface waters allowing for interaction with other landscape elements through spatial and temporal variation. The hydrologic connection of wetlands to surface waters is an important issue due to policies and regulations of the Clean Water Act (CWA) which emphasize the physical connection that wetlands have with nearby surface waters. The goal of this research was to quantify the hydrological connection of Sawyer Fen to a nearby Bluewater Creek in the Zuni Mountains, western New Mexico. Data were collected in the summer through the winter of 2019 at seven locations including Sawyer Fen, Bluewater Creek …
How Do Dairy Feedlot Size And Land Use Practices Affect Groundwater Quality Over Time? A Preliminary Study In New Mexico, Nancy J. Mcduffie
How Do Dairy Feedlot Size And Land Use Practices Affect Groundwater Quality Over Time? A Preliminary Study In New Mexico, Nancy J. Mcduffie
Water Resources Professional Project Reports
Agricultural activities in general and dairy operations in particular, have been identified as a potentially significant source of nitrate contamination in groundwater. The late 1990s was a period of rapid growth for New Mexico’s dairy industry. The New Mexico State University Cooperative Extension Service reports that the industry grew from 105 producers and 80,000 cows statewide in 1990 to 175 producers and 310,000 cows in 2003, to 145 producers and 323,000 cows in 2015. New Mexico now ranks ninth in the nation in milk production by volume, fifth in the nation for cheese production, and has the largest number of …
Project C.O.P.E Sexual Health Intervention On Sexual Negotiations Skills Among Black And Hispanic Adolescents In An Under-Resourced Northeastern Urban Community, Irene N. Cantu
Individual, Family, and Community Education ETDs
Decades of sexual health interventions have focused on the disparities youth of color face; however, inequalities in health outcomes and resources remain. This study evaluated Project C.O.P.E. (Communities Organizing for Prevention and Empowerment), a sexual health intervention in an under-resourced northeastern urban community. The goal of the study was to determine if the intervention, personal domains (ethnic identity and family support), or sexual health domains (Viral Hepatitis [VH] knowledge and human immunodeficiency virus [HIV] knowledge) had an impact on sexual negotiation skills in Black and Hispanic adolescent participants (n =510; Mage =16.78 ± 1.14). Between group analyses found significant …
Prediction Of Self-Diffusion Constants In Model And Real Systems Using Machine Learning, Joshua P. Allers
Prediction Of Self-Diffusion Constants In Model And Real Systems Using Machine Learning, Joshua P. Allers
Chemical and Biological Engineering ETDs
Understanding diffusion of chemical compounds is important for the design and optimization of many chemical engineering and energy processes. Recent modifications to the Darken equation allow for accurate prediction of Maxwell-Stefan (MS) diffusion in mixtures. Still, there are few practical applications due to the requirement of individual self-diffusion constants. A reliable predictive model for self-diffusion constants would be highly valuable when used in conjunction with the modified Darken equation. Here, we show that Machine Learning (ML) can be used to develop generalized models for self-diffusion in Lennard Jones (LJ) systems and real systems of pure solutions. The use of Artificial …

T cell acute lymphoblastic leukemia (T-ALL) is an aggressive hematologic malignancy with inferior outcome compared with that of B cell ALL. Here, we show that Runt-related transcription factor 2 (RUNX2) was upregulated in high-risk T-ALL with KMT2A rearrangements (KMT2A-R) or an immature immunophenotype. In KMT2A-R cells, we identified RUNX2 as a direct target of the KMT2A chimeras, where it reciprocally bound the KMT2A promoter, establishing a regulatory feed-forward mechanism. Notably, RUNX2 was required for survival of immature and KMT2A-R T-ALL cells in vitro and in vivo. We report direct transcriptional regulation of CXCR4 signaling by RUNX2, thereby promoting chemotaxis, adhesion, …

In order to achieve the global elimination of cervical cancer as a public health problem, close surveillance of progress in public health and clinical activities and outcomes across the three pillars of vaccination, screening and treatment will be required. Surveillance should ideally occur within an integrated system that is planned, funded, and regularly evaluated to ensure it is providing timely, accurate and relevant feedback for action. In this paper, we conceptualise the main public health surveillance objectives as process and outcome measures in each of the three pillars. Process measures include coverage/participation measures for vaccination, screening and treatment alongside the …

Disparities in sleep health are important but underrecognized contributors to health disparities. Understanding the factors contributing to sleep heath disparities and developing effective interventions are critical to improving all aspects of heath. Sleep heath disparities are impacted by socioeconomic status, racism, discrimination, neighborhood segregation, geography, social patterns, and access to health care as well as by cultural beliefs, necessitating a cultural appropriateness component in any intervention devised for reducing sleep health disparities. Pediatric sleep disparities require innovative and urgent intervention to establish a foundation of lifelong healthy sleep. Tapping the vast potential of technology in improving sleep health access may …

The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike protein (S) plays critical roles in host cell entry. Non-synonymous substitutions affecting S are not uncommon and have become fixed in a number of SARS-CoV-2 lineages. A subset of such mutations enable escape from neutralizing antibodies or are thought to enhance transmission through mechanisms such as increased affinity for the cell entry receptor, angiotensin-converting enzyme 2 (ACE2). Independent genomic surveillance programs based in New Mexico and Louisiana contemporaneously detected the rapid rise of numerous clade 20G (lineage B.1.2) infections carrying a Q677P substitution in S. The variant was first detected in …

The transmembrane protein heart of glass1 (HEG1) directly binds to and recruits Krev interaction trapped protein 1 (KRIT1) to endothelial junctions to form the HEG1-KRIT1 protein complex that establishes and maintains junctional integrity. Genetic inactivation or knockdown of endothelial HEG1 or KRIT1 leads to the upregulation of transcription factors Krüppel-like factors 4 and 2 (KLF4 and KLF2), which are implicated in endothelial vascular homeostasis; however, the effect of acute inhibition of the HEG1-KRIT1 interaction remains incompletely understood. Here, we report a high-throughput screening assay and molecular design of a small-molecule HEG1-KRIT1 inhibitor to uncover acute changes in signaling pathways downstream …

Molecular Mechanisms Of Circular Rna-Mediated Regulation Of Synaptic Gene Expression - Implications For Psychiatric Disorders, Alexander K. Hafez
Biomedical Sciences ETDs
Circular RNAs (circRNAs) are highly stable non-coding RNAs, derived from the covalent back-splicing of precursor mRNA molecules. Although circRNAs are highly enriched in the brain, the mechanism of action of brain expressed circRNAs and their relevance for psychiatric disorders remains largely unknown. Here we show that circHomer1, a neuronal-enriched circRNA abundantly expressed in the frontal cortex, derived from the Homer protein homolog 1 (HOMER1) gene, is significantly reduced in the prefrontal cortex and induced pluripotent stem-cell-derived neuronal cultures from patients with schizophrenia and bipolar disorder and is associated with the age of onset of schizophrenia. Furthermore, circHomer1 …

Vascular access connection configurations during tandem extracorporeal membrane oxygenation (ECMO) and therapeutic plasma exchange (TPE) may impact exchange kinetics. In these tandem procedures, typically the TPE inlet line is proximal to the TPE return line with respect to blood flow in the ECMO device, maximizing the opportunity for replacement fluid homogenization within the ECMO circuit. However, if TPE inlet and return line connections are switched, recirculation-a phenomenon in which replacement fluid leaving the TPE return line is prematurely drawn into the TPE inlet line prior to satisfactory homogenization within the ECMO circuit-will occur. Such recirculation could diminish TPE efficacy in …

BACKGROUND: Female Mexican Immigrants (FMIs) experience high rates of depression compared with other populations. For this population, depression is often exacerbated by social isolation associated with the experience of immigration. Aim 1. To measure whether a culturally situated peer group intervention will reduce depression and stress associated with the experience of immigration. Aim 2. To test whether an intervention using a "women's funds of knowledge" approach results in improved resilience, knowledge and empowerment. Aim 3. To investigate whether a culturally situated peer group intervention using a women's funds of knowledge approach can give participants a sense and experience of social …

BACKGROUND: Rho-family GTPases, including Ras-related C3 botulinum toxin substrate 1 (Rac1) and cell division control protein 42 (Cdc42), are important modulators of cancer-relevant cell functions and are viewed as promising therapeutic targets. Based on high-throughput screening and cheminformatics we identified the R-enantiomer of an FDA-approved drug (ketorolac) as an inhibitor of Rac1 and Cdc42. The corresponding S-enantiomer is a non-steroidal anti-inflammatory drug (NSAID) with selective activity against cyclooxygenases. We reported previously that R-ketorolac, but not the S-enantiomer, inhibited Rac1 and Cdc42-dependent downstream signaling, growth factor stimulated actin cytoskeleton rearrangements, cell adhesion, migration and invasion in ovarian cancer cell lines and …

Post-conviction DNA testing is often the last option a convicted person may have to establish that they are not guilty of a crime. New Mexico's post-conviction DNA statute requires convicted persons who seek DNA testing to claim innocence and establish that the identity of the perpetrator was an issue at trial. These requirements are currently included in many state post-conviction DNA statutes; however, some states have amended their statutes to remove these unnecessary requirements. Convicted persons who claimed self-defense, or another affirmative defense may be denied post-conviction DNA testing because of inability to claim "innocence" and because the identity of …

While current treatment regimens for acute leukemia can dramatically improve patient survival, there remains room for improvement. Due to its roles in cell differentiation, cell survival, and apoptotic signaling, modulation of the cyclic AMP (cAMP) pathway has provided a meaningful target in hematological malignancies. Several studies have demonstrated that gene expression profiles associated with increased pro-survival cAMP activity or downregulation of various pro-apoptotic factors associated with the cAMP pathway are apparent in acute leukemia patients. Previous work to increase leukemia cell intracellular cAMP focused on the use of cAMP analogs, stimulating cAMP production via transmembrane-associated adenylyl cyclases, or decreasing cAMP …

Pyramidal neurons from medial prefrontal cortex (mPFC) project into striatum, thalamus, hypothalamus, and went as far as dorsal raphe nucleus (midbrain) and locus coeruleus (pons). Neural activity in mPFC is reciprocally modulated by monoamine systems arising in these distant brain regions: noradrenergic, dopaminergic and serotonergic. By deregulating these systems genetically, we previously reported that functional projections from mPFC are similarly regulated by these monoaminergic systems using brain-wide tracing based on manganese-enhanced magentic resonance imaging (MEMRI) followed by statistical parametric mapping (SPM). Here we report that a single experience of acute threat alters mPFC projections in wild type mice, and this …
