A problem of classification places adolescents and emerging adults with special health care needs among the most at risk for poor or life-threatening health outcomes. This preliminary proof-of-concept study was conducted to determine if phenotypes of health care transition (HCT) for this vulnerable population could be established. Such phenotypes could support development of future studies that require data classifications as input. Mining of electronic health record data and cluster analysis were implemented to identify phenotypes. While early childhood circumstances influence the accumulation of skills and abilities, parental and policy responses can magnify or mitigate the effects of childhood circumstances. This work explores the two ideas mentioned above.
Chapter 1 briefs the historical and philosophical roots of the study of human capital and discusses the economic approach to the understanding of child wellbeing before the age of five. Because childhood circumstances influence the accumulation of skills, Chapter 2 estimates the impact of temperature shock during pregnancy on early childhood health. Volatile organic compounds (VOCs) are of great interest and the subject of much research, as they can be used in various applications. Preconcentrating this class of analytes is necessary as they can be elusive with simple sampling techniques. VOCs are often captured using sorbents in thermal desorption tubes as preconcentrates prior to analysis, commonly gas chromatography mass spectroscopy (GCMS). There are gaps in knowledge in behavior of materials used as sorbents i.e., if they will retain an analyte of interest appropriately. The effectiveness of perinatal exercise (i.e., exercise by mothers immediately prior to, throughout, and after pregnancy) to attenuate high-altitude pulmonary hypertension and associated health consequences in the offspring exposed to chronic hypoxia in early life was investigated. Four groups were used to compare the effects of perinatal exercise to offspring exposed to low altitude (i.e., normoxic) conditions or simulated 4500 meters of high altitude (i.e., hypoxic) as well as their respective, sedentary controls. Offspring in the hypoxic groups had lower body weights and tail lengths as well as higher hematocrits, right ventricular systolic pressures, and right heart weights. The hypoxic …

Arsenite exposure leads to the retention of UV-induced DNA damage, thus burdening translesion synthesis (TLS). Rad18 is an essential factor in initiating TLS through PCNA monoubiquitination and is implicated in homologous recombination. It contains two functionally and structurally distinct zinc fingers that are potential targets for arsenite binding. Results from this study reveal arsenite binding to both zinc fingers of Rad18 and a corresponding loss of domain function. Importantly, arsenite inhibited Rad18 RING-dependent PCNA monoubiquitination and polymerase eta recruitment to DNA damage. Further analysis demonstrated multiple effects of arsenite, including the reduction in the nuclear localization and UV-induced chromatin recruitment …

A staggering amount of data has been collected and analyzed since the beginning of the COVID-19 pandemic. Some of that data, however, particularly computed tomography (CT) scans of lungs, are difficult to analyze computationally. Segmentation of COVID-19 lesions provides researchers with insights into where lesions form, their volume, and in the case of time course data, how lesions grow as disease progresses. Spatial information, which has received little attention in the literature, has the potential to provide valuable insight to within-host viral and immune dynamics. Spatially resolved transcriptomics technologies investigate gene expression in the context of a tissue. While imaging-based technologies enable gene expression quantification at a single-cell level, only two NGS-based technologies are currently capable of single-cell resolution. Both Stereo-seq and Seq-Scope involve two rounds of sequencing: one to sequence the location of spatial barcodes and the other to obtain sequence of cDNA. Thus, the objective of the research study is to develop a technology that can capture transcripts from every cell of a tissue without the need for two rounds of sequencing. Background: Racial and ethnic health disparities in prostate cancer prevalence, incidence, mortality, quality of life, screening, and treatment constitute the largest of all cancer disparities. These health disparities are associated with higher morbidity and mortality rates in minority populations. Treatment with androgen deprivation therapy (ADT) has been the most widely used therapeutic modality to reduce the progression of prostate cancer to a worse disease stage and relieve potential obstructive symptoms. However, ADT is associated with possible toxic metabolic and cardiovascular (CVS) adverse events that may occur within six months of ADT initiation. This thesis is an application of epidemiological models for infectious disease transmission and the use of partially observed Markov process (POMP) for model fitting. It focuses on COVID-19 pandemic in the state of New Mexico. The analysis covered March 2020 to June 2021. Daily data of COVID19 cases and deaths and a daily index of eleven statewide government non-pharmaceutical intervention (NPI) policies were collected from six public sources and were validated. These data were integrated through the Susceptible-Exposed-Infected-Removed (SEIR) model. Cities have little knowledge about their parking infrastructure despite clear evidence that abundant parking has environmental, economic, and transportation consequences. The objective of this thesis is to illustrate the need to accurately estimate the area dedicated to vehicle infrastructure (parking lots and streets) within urban areas. I focused on a method that estimated the vehicle infrastructure area within the Downtown Core of Albuquerque, New Mexico, by cross-referencing geospatial cadastral data with minimum parking requirements. Traumatic brain injury (TBI) is one of the most suffered injuries with detrimental effects on the individual’s health, personality, and behavior. Overlap exists between TBI sequelae and symptoms of psychopathy. Both conditions are especially prevalent in incarcerated populations which makes studying their interrelation critical. Two studies examined the relationship between history of TBI (TBI+ vs. TBI-) and psychopathy (via the Hare Psychopathy Checklist-Revised; PCL-R) in adult incarcerated men and women at correctional facilities in New Mexico and Wisconsin. The current study aimed to profile styles of eating among dieting and non-dieting college-aged women, and to explore the extent to which dieting attitudes were associated with both nutritionally healthy eating and psychological well-being. A latent profile analysis was conducted using indicators of self-reported intuitive eating, binge eating, and two psychometric measures of dietary restraint; various measures of eating restraint were utilized to potentially capture distinct characteristics of dieting, such as self-discipline and disinhibition (Polivy et al., 2020; Stice et al., 2010). Due to the marginalized status of the population they serve, collegiate recovery programs are generally underfunded and employ one staff member who typically holds other job tasks in addition to those of collegiate recovery leader. The consequence of the varied nature by which collegiate recovery programs are founded and implemented is inconsistency, which can threaten the longevity of the field. The model minority myth (MMM) is predicated on stereotypical perceptions of Asian Americans as subservient high-achievers who comply with the ideologies of meritocracy, whiteness, and capitalism. However, South Asian American (SAA) activists and community organisers, the focus of this study, are working to confront and abolish racist, heterosexist, and other exclusionary injustices, policies, and practices. This dissertation seeks to understand the historical influences of the MMM, the challenges SAA activists and organisers face, and the communication strategies they use to negotiate the MMM through their activism. Experimental and computational studies pinpoint rate-limiting step(s) in metastasis governed by Rac1. Using ovarian cancer cell and animal models, Rac1 expression was manipulated, and quantitative measurements of cell-cell and cell-substrate adhesion, cell invasion, mesothelial clearance, and peritoneal tumor growth discriminated the tumor behaviors most highly influenced by Rac1. The experimental data were used to parameterize an agent-based computational model simulating peritoneal niche colonization, intravasation, and hematogenous metastasis to distant organs. Increased ovarian cancer cell survival afforded by the more rapid adhesion and intravasation upon Rac1 overexpression is predicted to increase the numbers of and the rates at which tumor cells …

American Indians (AIs) in New Mexico have lower cancer screening rates compared to other populations and are more likely to be diagnosed with cancer at an advanced stage of the disease as reported by Li et al. (Archives of Internal Medicine 163(1):49-56, 2003). AIs also have the lowest 5-year cancer survival rates compared to any ethnic/racial group in the USA as reported by Clegg et al. (Arch Intern Med 162:1985-1993, 2002) and Edwards et al. (Cancer 97:1407-1427, 2005). Academic searching is integral to research activities: (1) searching to retrieve specific information, (2) to expand our knowledge iteratively, (3) and to collate a representative and unbiased selection of the literature. Rigorous searching methods are vital for reliable, repeatable and unbiased searches needed for these second and third forms of searches (exploratory and systematic searching, respectively) that form a core part of evidence syntheses. Despite the broad awareness of the importance of transparency in reporting search activities in evidence syntheses, the importance of searching has been highlighted only recently and has been the explicit focus of reporting guidance (PRISMA-S). Objective: The purpose of the present study was to determine whether six weeks of home-based high-intensity interval training versus six weeks of moderate-intensity walking improves cognition, depression, and anxiety in women that are overweight or obese. Design: A randomized control trial design. Subjects: Twelve sedentary women characterized as overweight or obese were randomized into either a six-week home-based high-intensity interval training (HIIT, n = 6, 26.6 ± 8.9 years, 37.4 ± 4.9% body fat) group or a six-week moderate-intensity walking (Walk, n = 6, 22.5 ± 3.7 years, 40.2 ± 4.1% body fat) group. Would you make it to the future? For the last five years, I have started my Assisted Reproductive Technology (ART) lecture in Family Law with this question. Students take the query seriously. They ponder their lived experiences such as home training, medical history, education, financial well-being, personality traits, work ethic, and social graces when determining if they would be the “model DNA” someone might select in a future society.
