5-fluorouracil is a chemotherapeutic agent which is used in the treatment of disseminated colon and breast cancers. As an antimetabolite it has been found to inhibit the mechanism in which the normal substrate reacts with the enzyme and a cofactor in nucleic acid synthesis. A number of mechanisms have been summarized describing the conversion of deoxyuridylate (dUMP) to deoxythymidylate (dTMP), elucidating the role of the involved enzyme, thymidylate synthetase, and the co-factor, N('5), N('10)-methylenetetrahydrofolate. The effect of replacing the hydrogen bonded to the 5-position of the pyrimidine ring by a fluorine atom is also mechanistically described. A molecular orbital study …

The Limulus serum is capable of precipitating various components of the lipopolysaccharide molecule from R mutants of Salmonella minnesota. Precipitation-inhibition studies using 2-keto-3-deoxyoctonate (KDO) have supported the hypothesis that this acidic residue is the immunodeterminant group in the core region of the lipopolysaccharide molecule. Limulus also contains a lectin which interacts with the lipid A region of lipopolysaccharide; the proposed binding site in this region is the glucosamine backbone of the lipid A molecule.

A detailed study of the marine flora of Tunisia was conducted by assessing the seasonal variation of seaweed populations along a 35 m transect at one site (La Marsa), as well as by qualitatively describing the species composition throughout the country, and comparing the floristic affinities of the Tunisian flora with other Mediterranean and northeastern Atlantic areas. The occurrence and distribution of the Tunisian flora were correlated with a variety of parameters, including water temperature, salinity, substrata and currents. A Model System For The Concentration Of Reovirus From Aqueous Environments, David Marion Dziewulski

A filter adsorption-elution method for the routine isolation of reovirus from model aqueous systems was developed. Reovirus was found to be stable within a pH range of 3 to 10, with a minimum loss of viability. A slight hydrogen ion excess minimized virus aggregation phenomena and promoted efficient adsorption of virus to filter surfaces. Adsorption of virus at pH 6.0 to all filters examined was enhanced by 50 mM MgCl(,2). The use of AlCl(,3) as an enhancing salt was contraindicated since reovirus was not recoverable from 0.5 mM AlCl(,3) solutions. Elution of virus was most effective when a 3% beef …

Self-Incompatibility In Cheiranthus Allionii, Lee Anne Chaney

The presence of a self-incompatibility (SI) system in the polyploid crucifer, Cheiranthus allionii = Siberian wallflower, was suggested by results of greenhouse screening and confirmed by results of controlled pollinations in the field. Sporophytic control of the system was indicated by a stigmatic barrier to incompatible pollen tube growth, seen in scanning electron micrographs, and by differences in reciprocal cross-compatibilities. Strength of the SI system varied, but no completely self-compatible plants were found. Marked segregation for SI level within self- and cross-progenies of a plant with weak SI suggested that weak SI might be due to a single gene. The …

The surface slope induced pressure gradient, mean current velocity profile, and bottom stress are investigated both theoretically and experimentally for a tidal channel in a well-mixed estuary. The theory focuses on the use of eddy viscosity models. Surface slope estimates based on tide predictions, current velocity profile measurements, and stress estimates from near bottom turbulence measurements are used to investigate the dynamic balance in the tidal channel, and to evaluate the eddy viscosity models. This dissertation describes the productive, sexual, and public roles of married women living in the province of New Hampshire and the two Massachusetts counties which bordered it, Essex County to the south and York County (now Maine) to the north and east. Because there are no female diaries for the period and few letters, the study depends upon an accumulation of evidence from court records, account books, probate inventories, church records, paintings, embroideries, gravestones, genealogical records, captivity narratives, sermons, and the letters and diaries of husbands and sons.

Land treatment of wastewater involves the use of plants and the soil to remove unwanted constituents. Removal of phosphorus is particularly important to avoid excessive biological activity in water systems receiving the treated wastewater. This research examines the phosphate adsorption - desorption behavior of two soils: Charlton silt loam, a typical acid soil from New England which is being used in experimental wastewater treatment, and Tujunga coarse sandy loam from a wastewater land treatment facility located at Manteca, California, which has failed to remove phosphate from wastewater efficiently.

Part 1. Some Chemical Constituents of the Brown Alga, Agarum cribosum. Agarum cribosum was examined for major chemical constituents. Halogenated terpenes found in red algae were not present in this seaweed. The sterol content was determined by GC/MS analysis and six sterols were identified: fucosterol, 24-methylenecholesterol, cholesterol, 24-ketocholesterol, saringosterol, and desmosterol. The presence of desmosterol in brown algae had not been previously confirmed. A seventh sterol could not be identified from its mass spectrum. The fatty acid content was also determined by GC/MS analysis. The major fatty acids were C(,16:0), C(,16:1), C(,20:4) and C(,22:5). The sugar mannitol was isolated.

Ornithine decarboxylase activity was stimulated in high-density HeLa cell cultures by dilution of or replacement of spent culture medium with fresh medium containing 10% (v/v) horse serum. After stimulation, ornithine decarboxylase activity reached a peak at 4-6 h. then rapidly declined to the low enzyme activity characteristic of quiescent cultures, where it remained during the remainder of the cell cycle. The stimulation of ornithine decarboxylase was eliminated by the addition of 0.5 (mu)M-spermine or spermidine or 10 (mu)M-putrescine to the HeLa cell cultures at the time of refeeding with fresh medium. The metal ion complexing chemistry of organic ligands commonly found in drinking water sources is the object of the studies in this dissertation. These ligands, called humics as a class, were modelled in some cases by a soil-derived humic material called fulvic acid. The role of natural humics in the transport and speciation of dissolved metal ions in drinking water sources, particularly health hazards like Cd('2+), is investigated. Toxic cyanobacteria bloom in eutrophic, freshwater lakes and ponds and may cause environmental, health and recreational problems during or after the massive growth of these microorganisms Microcystis aeruginosa is commonly involved in freshwater blooms and one of its toxins (microcystin) causes liver damage in birds and mammals. This study has determined the specific site of action of microcystin and characterized the hepatic damage at the ultrastructural level. Histological changes in centrilobular regions of liver tissue were noted after intraperitoneal administration in mice. Hepatic sinusoidal epithelial and hepatocyte plasma membranes ruptured with the release of cellular components that pooled with blood. …
