Initial Characterization Of Coagulin Polymerization And A Novel Trypsin Inhibitor From Limulus Polyphemus, Maribeth Ann Donovan

In the final step of coagulation in the horseshoe crab, coagulogen (M$\sb{\rm w}$ = 19,000, from sedimentation equilibrium) is converted by discrete proteolysis into its clottable form, coagulin. Coagulogen does not have a strong tendency to self-associate. In 0.1 M ammonium bicarbonate, 50 mM Tris-HCL (pH 8.1), trypsin generated coagulin rapidly associates to form a stable, solid gel. Polymerization is pH-dependent, with reversible depolymerization occurring in acid (midpoint pH 5.4). Sedimentation velocity analysis reveals that stable intermediates remain at pH 2.5, with 30-31 s being the predominant form along with monomer (1.89 s) and larger forms ($>$60 s). Near …

Information about the ligand environment of the iron binding sites in a cyanide adduct of transferrin was obtained by an analysis of powder-type ENDOR and ESEEM spectra. The low-spin cyanide adduct of transferrin is formed only in the C-terminal site of the protein and is characterized by a rhombic EPR spectrum. Earlier work demonstrated that three cyanide groups are necessary to form the adduct, but it wasn't clear whether these groups were coordinated directly to the metal or to cationic sites on the protein. The ENDOR spectra of the $\sp{13}$CN adduct showed one set of $\sp{13}$C ENDOR resonances which probably …

The genetic structure of a two hectare stand of eastern white pine (Pinus strobus L.) was studied by relating foliar allozyme composition of individual trees to tree location and age. Using starch gel electrophoresis, the trees were found to have variants for six of the seven enzymes analyzed. Trees of similar age and with common allozymes were found in clusters, suggesting that genetic structure existed in the stand. The genetic structure may have originated during establishment of several seedlings from common parents when openings were created in the canopy. In an attempt to develop a comprehensive approach to the study of the adoption of energy systems, this dissertation first critically examines a variety of approaches to this phenomenon, including market models of adoption, the adoption of innovations literature, and the social psychology of attitudes and behavior. Market models are seen as generally reductionistic and the argument is made that they fail to take into consideration a variety of non-cost considerations. The assumption here is that while price considerations play a role in this process, decisions are also affected by values and attitudes which are not directly incorporated into market …

Researchers have been unable to discern the relationship between energy expenditure and obesity because there has been no reliable, unobtrusive method to capture daily activity. Single-plane accelerometers have been developed, using microcomputer technology, which may provide a means for measuring expenditure in human subjects. The Caltrac Personal Activity Computer$\sp\circler$ has shown high test-retest reliability and a strong relationship to several measures of expenditure in some laboratory tests. In Experiment 1, 40 university women (mean age = 19.15 yr) wore Caltracs during waking hours for either three or six consecutive days, with the devices programmed to count activity regardless of individual …

This dissertation explores the development of student- and process-centered writing classrooms within the context of a traditionally structured, curriculum-centered high school. The focus of the study is on how teachers and students experience and address contradictions between assumptions implicit in writing process pedagogy and assumptions implicit in the structure and organization of the high school.

An eight-month ethnography of two high school English classrooms, the study is a descriptive narrative of the classroom and school events punctuated by the reflective comments of teachers and students in intensive and extensive interviews. Carex section Scirpinae is a small group of North American sedges that possess a dioecious breeding system, unispicate inflorescences and pubescent perigynia. Historically, ten different taxa had been recognized at the specific or infraspecific levels based primarily on morphological characters. Taxonomic problems in the group were related to the lack of a comprehensive monographic treatment. Floristic treatments were limited and focused only on a few regional taxa. Descriptions were often incomplete, lacking descriptions of staminate material or complicated by the inconsistent use of the terms phyllopodic and aphyllopodic. Campylobacter jejuni is a major cause worldwide of human gastroenteritis. In this study, the pathogenicity of the organism was examined in fertile hens' eggs and the mortality data and histopathological findings induced by both the organism and bacteria-free filtered broth were similar. The absence in chick embryo tissues of either bacteria or an inflammatory infiltrate suggested a toxic etiology. A protein was obtained from the filtrate by gel electroelution and this was sensitive to heat, pH changes and trypsin treatment and was lethal for the fertile hen's egg. Transcription of the glucose-repressible ADH (ADH2 locus) in Saccharomyces cerevisiae is controlled by two regulatory pathways. The general transcriptional factors CCR4, CRE1, and CRE2 constitute the first pathway while the second pathway is comprised of the trans-activators ADR1 and CCR1. Both ADR1 and CCR1 act through upstream activation sequences (UAS) found in the 5$\sp\prime$-regulatory region of the ADH2 structual gene. In contrast, the action of CCR4, CRE1, and CRE2 is likely to be at sequences near the TATAA element of ADH2.

The CCR4 locus was precisely mapped on the left arm of chromosome I where it had previously been localized. …

Kelp blades (Laminaria spp.) with colonies of Obelia geniculata were collected over a 28 month period from Cape Neddick, ME to document the epifaunal community and to determine the possible mechanisms allowing four nudibranch species to coexist. Regression analyses correlated the abundances of epifauna with habitat parameters. The feeding biology of nudibranchs was described. Density manipulations of nudibranchs in pair-wise behavioral interactions tested the effects of interference on the location of nudibranchs in the colony.

The relationship of both family structure and process variables to both mental illness and family violence are examined in this study. A non-clinical sample of 100 university student families and a clinical sample of 100 in-patient families at a psychiatric hospital are utilized. Both non-clinical and clinical samples are utilized with subjects of similar age. In a series of four experiments, subjects examined the perceived rigidity of rotating eight-vertex polyhedra. The requirement for polyamines in the proliferation of cells was first demonstrated in bacteria (3). While significant progress has been made in this field using animal cell cultures, only preliminary studies have been reported with plant tissues. Serafini-Fracassini et al. (9) showed a marked increase in polyamine synthesis early during the G 1 phase, concomitant with the synthesis of RNA, following the break of dormancy in Helianthus tuberosus tuber tissue. Carrot cell cultures provide a model experimental system for the analysis of biochemical and molecular events associated with morphogenesis in plants (3, 4, 5, 14). Among the biochemical changes accompanying somatic embryogenesis in this tissue is an increased biosynthesis ofpolyamines (1, 2, 7, 10, 11, 13). A variety of inhibitors of polyamine biosynthetic enzymes have been used to analyze the role of polyamines in somatic embryogenesis. Heart electrical activity (ECGs), gill closer muscle potentials (EMGs), and blood pressures in the heart and the branchiocardiac canals, were measured in adult horseshoe crabs (Limulus polyphemus) during various activities. During ventilation, hyperventilation, and swimming, large transient increases in pressures (10-35 cm H2O) occur in the branchiocardiac canals, which carry blood from the gills to the heart. These pulses of positive pressure are related to, and apparently caused by, gill plate closing. During quiescent periods, with no ventilatory activity, there are no pressure pulses in the canals, but the pressure is still greater than zero. We found covariation of heart …

The biochemical mechanisms by which octopamine, catecholamines and the peptide proctolin exert their actions on Limulus cardiac muscle were investigated. Amines produced long-lasting increases in the amplitude of contractions evoked by electrical stimulation. At 10(−5) mol l-1, the apparent order of potency for amine-induced increases in evoked contraction amplitude was dopamine approximately equal to octopamine greater than norepinephrine approximately equal to epinephrine. At this dose, amines produced long-lasting increases in the levels of cyclic AMP (octopamine greater than dopamine approximately equal to norepinephrine approximately equal to epinephrine), but not of cyclic GMP, in Limulus cardiac muscle.
