Investigating The Lyoprotective Properties Of An Antifreeze Protein From A Broadleaf Desert Shrub, Alexzandria Sheppard

Lyophilization, or freeze-drying, is an important preservation method for protein-based products, such as protein pharmaceuticals. However, the process introduces both freezing and drying stresses that can harm proteins. Here, we have studied the efficacy of AnAFP, a dehydrin-like antifreeze protein isolated from the broadleaf desert shrub Ammopiptanthus nanus, in reducing enzyme damage during freeze-drying, or lyophilization. We have shown that AnAFP can protect the model enzyme lactate dehydrogenase (LDH) from both freezing and dehydration damages during lyophilization with similar or better efficacy than 10% w/v sucrose and superior to that of another known antifreeze protein.

Investigating The Mechanisms Of Freeze Tolerance In Lemna Minor, Tamara Bouchard

Freezing presents a significant abiotic stress to aquatic plants, yet little is known about the mechanisms by which species like Lemna minor (duckweed) survive subzero temperatures in nature. This study investigates whether L. minor employs freeze-avoidance or freeze-tolerance strategies and explores the potential role of its microbiome in enhancing cold resilience. Using both field-collected fronds from naturally frozen ponds and controlled freezing experiments in the lab, I assessed survival through viability staining and post-thaw survival. Results showed that L. minor can survive natural ice encasement and that prior cold acclimation significantly improves tissue survival under induced freezing. Trypan blue staining …

The US education system has seen a recent push for more secondary school curriculum to include STEM (Science, Technology, Engineering, and Mathematics) materials and activities. Middle and high school is a pivotal time when academic and personal experiences affect the development of student attitudes towards STEM (Christensen et al., 2015; Moreno et al., 2016). Teachers often utilize hands-on activities to promote student engagement with science curriculum (Brooks & Poles, 2021; Christensen et al., 2015). Background: Skin-to-skin (STS) contact, also known as kangaroo care, is a critical intervention for preterm infants, especially for those in the neonatal intensive care unit (NICU). It promotes neurodevelopmental outcomes, physiological stability, and parental bonding. However, STS often falls below recommended daily levels due to a variety of berries. Exploring aids like belly bands may help address these barriers and promote improved outcomes.

There are nearly 4,100 species of plant-parasitic nematodes that pose a serious threat toglobal food security, and it is estimated that about $100 billion of crops are damaged annually due to these plant parasitic nematodes. Also, animal-parasitic nematodes pose a major global health challenge, impacting approximately 3.5 billion individuals annually, particularly in tropical and developing areas. PDEs are significant determinants of cellular levels of cyclic nucleotides because the process of lowering active intracellular cyclic nucleotide second messengers (cAMP and cGMP) in cells is mediated through PDEs. Viscosity Stratified Turbulence: Insights From Direct Numerical Simulations, Quasilinear Reduction And Resolvent Analysis, Pulkit Kumar Dubey

This dissertation focuses on wall-bounded shear-driven turbulence with temperature-dependentviscosity. Direct numerical simulations and quasilinear reduction have been used to establish the asymmetric variation of turbulent statistics in the wall-normal direction. A comparison of energy spectra in the cold and hot halves of the channel reveals that large scale motions are amplified in the more viscous half while small scale motions are amplified in the less viscous half. Interestingly, the threshold wavenumber distinguishing these scales remains independent of stratification. This observation has been verified via a low-order reconstruction of the turbulent kinetic energy profile using resolvent analysis. Fourier modes that selectively …

The legacy effects of crop diversity on maize (Zea mays L.) tissue nutrient composition, weed community structure, and intensity of weed-crop competition were assessed through a field experiment at two sites in the northeastern United States. Fields were conditioned with crop diversity gradients from summer 2016 to spring 2019. The crop diversity gradients ranged from a single cultivar to sixteen intercropped cultivars (four species, four cultivars per species) and were established in organic annual and perennial cropping systems. Following the three-year conditioning phase, maize was planted across the entire experiment, and each conditioning-phase diversity treatment was split into weed-free, ambient-weed, …

Snowpacks are changing in northeastern North America as the regional climate warms, yet the relative influence of changes in precipitation compared to changes in ablation on snowpacks is poorly understood. We use 56 years of weekly snow water equivalent (SWE) measurements from three locations within a study site which vary in elevation and aspect, paired with adjacent daily climate measurements, to investigate relationships between climate and snowpack onset, maximum, and disappearance.
