Forages as conserved silage or grass cannot supply enough nutrients and energy as required by lactating dairy cows. As a result, supplementation with grains is needed to provide animals with enough nutrients to be healthy and produce milk being profitable (NRC, 2001). High producing cows need protein supplementation from sources other than forages in order to maximize milk protein production, with emphasis on replenishing requirements for specific amino acids. Excessive protein in the diet or deficiency of an essential amino acid can reduce productivity and increase excretion of N to the environment, causing pollution. Research must be conducted to help …

Balancing diets for lysine (Lys) and methionine (Met) in dairy rations is becoming a regular practice in today’s industry. In order to be able to use rumen-protected AA (RPAA) in those diets a method to assess the efficacy and bioavailability needs to be proposed. The objective of the following studies was to propose the acceptance of the plasma free AA dose-response technique as a standardized methodology for evaluating RPAA in lactating dairy cows. Thirteen dose-response Latin square studies using ruminally-cannulated multiparous Holstein cows were conducted. As high tunnel production is relatively new to the Northeast, this research has helped understand knowledge-gaps in common warm and cold-season crop systems in the region. High tunnels increase crop yield and quality and extend the growing season, and are most commonly used for tomato in the warm-season and spinach in the cold-season.
Spinach was grown in unheated high tunnels throughout the cold-season to evaluate the effects of cultivar and planting date on yield and sweetness (Brix° levels). Cultivar and planting date had a significant effect on yield, where earlier planting dates resulted in higher yields. Cultivar had a significant …

As the American public school system is currently designed, the average student is likely to graduate high school without ever having received any formal instruction regarding the food system in which he or she takes part as a consumer. Such instruction was not necessary up until the current era of separation between consumers and food production. Through this research project, I sought to demonstrate the effectiveness of a single introductory lesson on the issues confronting the modern American food system, which I taught to two high school biology classrooms. Pasteurized donor human milk (PDHM) as feeding supplementation has been shown to prevent and lower rates of gastrointestinal infection and increase exclusive human milk consumption (EHM) in critically-ill, low birthweight and premature infants. Policies/ procedures for the use of PDHM in “well” newborns and low birth weight newborns with non-life-threatening illness, level I and II nurseries, respectively, have not been established. The objective of this research is to gather and summarize policies/procedures and experiences from four hospitals in the northeast US that use PDHM in level I/II nurseries. Iron acquisition is well studied in pathogens, and successful virulence is often attributed to iron acquisition by siderophore and heme uptake; however, the role of iron uptake in mutual symbiotic interactions is not as well understood. The mutual symbiosis between Vibrio fischeri and the Hawaiian bobtail squid, Euprymna scolopes, is a well-characterized system in which iron uptake has been implicated as a symbiotic factor. Four studies have implicated iron uptake in the symbiosis: 1) A TnLux reporter assay revealed that siderophore is more highly expressed by V. fischeri in the light organs of juvenile squid compared to V. fischeri in …

The selection of phenotypic traits in plant breeding has recently become augmented by the implementation of molecular markers. Marker-assisted breeding strategies are used to develop new cultivars of many important crop plants but have yet to be assimilated into breeding programs for many minor crops with untapped economic potential. In the following chapters, I discuss methods of marker development in crop plants, sex determination as a trait of economic interest in plant breeding, and the identification of two male-dominant, PCR-based markers in the commercially viable Kiwiberry species Actinidia arguta and A. kolomikta.

Historically, minority women have made up a disproportionate percentage of the low-income population who receive safety net benefits. However, there has been no previous research that assesses how alternative poverty measures could impact these isolated groups of women. This study aims to determine which groups of people would receive the largest benefit if state and federal agencies used the supplemental poverty measure (SPM) rather than the official poverty measure (OPM) to determine eligibility for various safety net programs. Research has shown that direct-fed microbials (DFM) and enzyme supplementation can impact the dry matter intake (DMI), milk production, and milk composition. However, limited research has evaluated the impact of DFM and enzyme supplementation on colostrum quality and the uptake of the Immunoglobulins A and G (IgA and IgG) by calves. In this study, 36 multiparous Holstein cows were blocked by expected calving date and randomly assigned to 1 of 3 treatments 3 weeks prior to calving and remained on these treatments until week 8 postpartum. This thesis examines the feeding of kelp meal (KM), most commonly produced from the species Ascophyllum nodosum to lactating dairy cattle. In the first experiment KM was fed at incremental levels to lactating jersey cattle over the winter season in Lee, New Hampshire. The objective of this study was to investigate the effects of incremental amounts (0, 57, 113, and 170 g/d) of KM on animal performance and milk iodine concentration in dairy cows fed high-forage diets. While animal performance was not improved, milk iodine output increased linearly in cows fed KM. Trends and quadratic effects were observed for nutrient …

In cows, colostrum is composed of several antibodies and nutrients to provide immunity and energy to the calf. Feeding calves high quality colostrum has been shown to improve calf health, leading to reduced mortality in calves and greater milk production in cows. The addition of direct-fed microbials (DFM) to cow diets has been theorized to improve feed efficiency and milk production, with studies showing mixed results. This project examined approaches to establishing a functionally diverse forage crop “cocktail”, a mixture of 15 annual and perennial forage crop species with contrasting functional traits, within a hayfield for the purpose of increasing hayfield forage plant diversity and productivity. The forage crop cocktail was intended to enhance the diversity of the resident hayfield, as well as contribute to enhancements in overall hayfield productivity via biological interactions among the cocktail and resident hayfield species, rather than through external chemical inputs. Symbiotic associations with subcuticular bacteria (SCB) have been identified and studied in numerous echinoderms, including the SCB of the brooding brittle star, Amphipholis squamata. These SCB, however, have not been studied using current next generation sequencing technologies. Previous studies on the SCB of A. squamata placed these bacteria in the genus Vibrio (γ-Proteobacteria), but subsequent studies suggested that the SCB are primarily composed of α-Proteobacteria. Started in Fall 2015, Project OASIS (Optimizing Aquaponic Systems to Improve Sustainability) is an interdisciplinary capstone project with the goal of designing a sustainable and affordable small-scale aquaponic system for use in developing nations to tackle the problems of malnutrition and food insecurity. Aquaponics is a symbiotic relationship between fish and vegetables growing together in a recirculating system. The project’s goals were to minimize energy consumption and construction costs while using universally available materials. The computational fluid dynamics (CFD) software OpenFOAM was used to create transient and steady-state models of fish tanks to visualize velocity profiles, streamlines, and particle movement. …

Previous studies have suggested that horseshoe crabs prefer warm water, suggesting that they may be able to detect changes in water temperature. The overall goal of this study was to test this hypothesis. Our specific objectives were to: 1) find out if horseshoe crabs can detect temperature changes; 2) determine the magnitude of temperature change they can detect, and; 3) determine whether their temperature receptors are located internally or externally. Background and Aims
The importance of the uptake of nitrogen in organic form by plants and mycorrhizal fungi has been demonstrated in various ecosystems including temperate forests. However, in previous experiments, isotopically labeled amino acids were often added to soils in concentrations that may be higher than those normally available to roots and mycorrhizal hyphae in situ, and these high concentrations could contribute to exaggerated uptake.
Methods
