Egg Plastron Of The Bog Copper Butterfly Tharsalea (Epidemia) Epixanthe (Bsd. & Le C. [1835]) (Lycaenidae: Lycaeninae), David M. Wright
The Taxonomic Report of the International Lepidoptera Survey
The egg of the Bog Copper butterfly, Tharsalea (Epidemia) epixanthe, has a prominent highly-sculptured chorionic surface. Trapped within the chorion is a labyrinth of air spaces which has been proposed as a plastron for gas exchange while the egg is submerged in water. Data derived from scanning electron microscopy (SEM) confirms the plastron should function as predicted. Furthermore, the insulating air spaces should prevent water loss of the diapausing first instar larvae while overwintering.

Host innate immune response follows severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection, and it is the driver of the acute respiratory distress syndrome (ARDS) amongst other inflammatory end-organ morbidities. Such life-threatening coronavirus disease 2019 (COVID-19) is heralded by virus-induced activation of mononuclear phagocytes (MPs; monocytes, macrophages, and dendritic cells). MPs play substantial roles in aberrant immune secretory activities affecting profound systemic inflammation and end-organ malfunctions. All follow the presence of persistent viral components and virions without evidence of viral replication. To elucidate SARS-CoV- 2-MP interactions we investigated transcriptomic and proteomic profiles of human monocyte-derived macrophages. While expression of the …

Department of Electrical and Computer Engineering: Faculty Publications
Plasmonic nanopatch antennas that incorporate dielectric gaps hundreds of picometers to several nanometers thick have drawn increasing attention over the past decade because they confine electromagnetic fields to grossly sub-diffraction-limited volumes. Substantial control over the optical properties of excitons and color centers confined within these plasmonic cavities has already been demonstrated with far-field optical spectroscopies, but near-field optical spectroscopies are essential for an improved understanding of the plasmon–emitter interaction at the nanoscale. Here, we characterize the intensity and phase-resolved plasmonic response of isolated nanopatch antennas by cathodoluminescence microscopy. Furthermore, we explore the distinction between optical and electron beam spectroscopies of …

Molecular Characterization Of Western Corn Rootworm Pyrethroid Resistance, Dariane Souza, Blair D. Siegfried, Lance Meinke, Nicholas J. Miller
Department of Entomology: Faculty Publications
Background: Western corn rootworm (WCR) pyrethroid resistance has been confirmed in the western US Corn Belt. Toxicological and biochemical studies indicated that multiple mechanisms of resistance might be involved in the resistance trait, such as enhanced metabolism and/or kdr target-site mutation(s) in the voltage-gated sodium channels. To characterize the mechanisms of WCR pyrethroid resistance at the molecular level, pairwise comparisons were made between RNA-Seq data collected from pyrethroid-resistant and -susceptible WCR populations. Gene expression levels and sodium channel sequences were evaluated.
Results: Seven transcripts exhibited significantly different expression (q ≤ 0.05) when comparing field-collected pyrethroidresistant (R-Field) and -susceptible (S-Field) WCR …

Roman L. Hruska U.S. Meat Animal Research Center: Reports
Polymorphisms in μ-calpain (CAPN1) that beneficially associate with beef tenderness are reported to antagonistically associate with calving day in beef heifers and post-partum interval to estrus in beef cows. We, therefore, hypothesized that a molecular breeding value for slice shear force, calculated based on CAPN1 and calpastatin (CAST) genotypes, would demonstrate an antagonistic relationship between genomically predicted slice shear force and ordinal calving date in replacement beef heifers. A secondary objective of this study was to evaluate the association of a polymorphism in diacylglycerol O-acyltransferase (DGAT1) with reproductive traits in beef heifers. One hundred eighty-seven MARC III heifers (¼ Angus, …

Greenhouse Gas Emissions And Drought Projections For Nebraska, Adam Liska
Adam Liska Papers
This lecture will focus on three issues. The first is a presentation of data from the first greenhouse gas emissions inventory for the State of Nebraska, which will be published in the coming weeks. In 2016, emissions from beef cattle made up 23% of net state emissions, and coal for electricity made up 23.7% of net state emissions. The second issue to be addressed are the impacts of the 2012 drought on agriculture in Nebraska and the region. The drought of 2012 indicates that future droughts in the 21st century in the region can be a dominant influence on …

Roman L. Hruska U.S. Meat Animal Research Center: Reports
Cattle are the main reservoir of Enterohemorrhagic Escherichia coli (EHEC), with O157:H7 the distinctive serotype. EHEC is the main causative agent of a severe systemic disease, Hemolytic Uremic Syndrome (HUS). Argentina has the highest pediatric HUS incidence worldwide with 12–14 cases per 100,000 children. Herein, we assessed the genomes of EHEC O157:H7 isolates recovered from cattle in the humid Pampas of Argentina. According to phylogenetic studies, EHEC O157 can be divided into clades. Clade 8 strains that were classified as hypervirulent. Most of the strains of this clade have a Shiga toxin stx2a-stx2c genotype. To better understand the molecular bases …

This pilot study describes a nascent first-year honors colloquia series using human-centered design (HCD). An interdisciplinary team of instructors redesigned the course with the intention of engaging the whole student in transformative learning and creating a curriculum that addresses problems and opportunities focused on the needs, contexts, emotions, and behaviors of all students, faculty, administrators, and community involved in the series. Authors describe the HCD process, observing the challenges faced by faculty in realizing its design principles, and student (n = 98) reflections on a two-part prototype involving innovation and entrepreneurship emphasizing “wicked” problems and resolutions. Students were asked to …
Exploration Of Lived Experiences Of Science Teachers Of English Language Learners: A Transcendental Phenomenological Study, Uma Ganesan
The Nebraska Educator: A Student-Led Journal
English language learners (ELLs) are a talented pool of culturally and linguistically diverse students who are persistently increasing both in absolute size and percentage in the U.S. school population; however, they are underrepresented in science, technology, engineering, and mathematics (STEM) fields in college as well as in the workforce (National Academies of Sciences, Engineering, and Medicine, 2018). Education and scientific communities have found it challenging to improve students’ participation in STEM fields (Martinez et al., 2011). Exploring science teachers’ experiences could aid in improving academic achievement of ELLs and promoting educational equity. The purpose of this transcendental phenomenological study is …

What Really Matters For Instructors Implementing Equitable And Inclusive Teaching Approaches, Tracie Marcella Addy, Philip M. Reeves, Derek Dube, Khadijah A. Mitchell
To Improve the Academy: A Journal of Educational Development
Supporting instructor implementation of equitable and inclusive teaching approaches is a critical area of focus in educational development. However, there is limited empirical evidence on factors that either support or hinder instructors’ implementation of inclusive teaching. The results of this national survey study reveal several predictors of instructors’ utilization of inclusive teaching approaches and reported obstacles faced. For this sample, knowledge of inclusive teaching was a statistically significant predictor of implementation, as was being from a non-STEM discipline. Responses highlighted promising approaches, several of which can inform the efforts of educational developers.

To Improve the Academy: A Journal of Educational Development
This study seeks to explore the physical spaces centers for teaching and learning (CTLs) occupy; with an emphasis on gaining a better picture of what CTL spaces look like; where they are located; how they developed: and what these spaces represent. We gathered visual, empirical, and qualitative data to take the first steps towards developing a shared vision of not only of our physical spaces, but also as a jumping off point for further analysis of the CTL as a meaningful place.

Where Are The Students In Efforts For Inclusive Excellence? Two Approaches To Positioning Students As Critical Partners For Inclusive Pedagogical Practices, Alison Cook-Sather, Tracie Marcella Addy, Anna Devault, Nicole Livitskiy
To Improve the Academy: A Journal of Educational Development
Most educational development for inclusive excellence does not draw directly on the experiences and perspectives of students. This article presents two different approaches to positioning undergraduate students as critical partners in developing inclusive pedagogical practices. Co-authored by the directors of and student partners who participated in each approach, the article defines inclusive excellence and inclusive teaching and provides selected examples of partnership work that strives for equity and inclusion. It then describes our different approaches, discusses potential benefits of launching student-faculty partnership work through these approaches, and offers recommendations for developing pedagogical partnership efforts for inclusive excellence at other institutions.

Analysis Of Single-Site Cysteine Mutation, I412c, In Human A Glycine Receptor States To Further Refine Structure And Allostery, Leah Engquist
Honors Program: Senior Projects (Public)
The glycine receptor (GlyR) is the major inhibitory receptor in the brain and spinal cord. A member of the pentameric ligand gated ion channel superfamily, crystal structures are available but there are still unresolved areas, specifically the C-terminal tail and TM3-TM4 intracellular loop. Further refinement can provide deeper understanding of the molecular mechanism and allow the creation of novel therapeutics to modulate its function. We propose to insert a single cysteine mutation, I412C, into a Cys null background (C41S/C290A/C345S) to study non- conducting states (resting, desensitized) or with F207G/A288G mutations to study the open state. Purified, reconstituted GlyR is crosslinked …

Wildlife Damage Management Technical Series
Domestic cats (Felis catus) are a common household pet in the United States, with an estimated 25.4% of households owning cats (American Veterinary Medical Association 2018). While an increasing number of cat owners keep their pet cats exclusively indoors, a portion of society maintains that domestic cats are entitled to a free-ranging lifestyle and may even consider unowned domestic cats to be wildlife. Although wildlife managers recognize the beliefs of many concerned stakeholders, including advocates who use strong emotional appeals on behalf of cats, it remains that free-ranging and feral domestic cats are an invasive species spread by humans (Lowe …

Pregnancy status is a key parameter used to assess reproductive performance of a species as it represents a starting point for measuring vital rates. Vital rates allow managers to determine trends in populations such as neonate survival and recruitment; two important factors in ungulate population growth rates. Techniques to determine pregnancy have generally involved capture and restraint of the animal to obtain blood samples for determining serum hormone levels. Non-invasive pregnancy assessment, via feces, eliminates any hazards between handler and animal, as well as removes handling-induced physiological biases. Using noninvasive fecal sampling, we conducted hormone validations, investigated pregnancy rates, and …

Stem rust caused by Puccinia graminis f. sp. tritici Eriks. is an important disease of common wheat globally. The production and cultivation of genetically resistant cultivars are one of the most successful and environmentally friendly ways to protect wheat against fungal pathogens. Seedling screening and genome-wide association study (GWAS) were used to determine the genetic diversity of wheat genotypes obtained on stem rust resistance loci. At the seedling stage, the reaction of the common stem rust race QFCSC in Nebraska was measured in a set of 212 genotypes from F3:6 lines. The results indicated that 184 genotypes (86.8%) had different …

Heme is an essential cofactor required for a plethora of cellular processes in eukaryotes. In metazoans the heme biosynthetic pathway is typically partitioned between the cytosol and mitochondria, with the first and final steps taking place in the mitochondrion. The pathway has been extensively studied and its biosynthetic enzymes structurally characterized to varying extents. Nevertheless, understanding of the regulation of heme synthesis and factors that influence this process in metazoans remains incomplete. Therefore, we investigated the molecular organization as well as the physical and genetic interactions of the terminal pathway enzyme, ferrochelatase (Hem15), in the yeast Saccharomyces cerevisiae. Biochemical and …

Irrigation has a great impact on global food security as it contributes to the majority of the world’s agricultural food supply. It is essential to judiciously utilize water resources through efficient irrigation management since the majority of U.S. groundwater aquifers are rapidly depleting. Thus, quantification of the relationships between water depletion and environmental factors is important for understanding crop response to varying levels of water stresses that depletion can cause. The objectives of this research were to: 1) investigate the relationship between root zone water depletion (Drw) and canopy temperature differential (ΔT) at different ranges of Drw …

With rates of psychiatric illnesses such as depression continuing to rise, additional preclinical models are needed to facilitate translational neuroscience research. In the current study, the raccoon (Procyon lotor) was investigated due to its similarities with primate brains, including comparable proportional neuronal densities, cortical magnification of the forepaw area, and cortical gyrification. Specifically, we report on the cytoarchitectural characteristics of raccoons profiled as high, intermediate, or low solvers in a multiaccess problem-solving task. Isotropic fractionation indicated that high-solvers had significantly more cells in the hippocampus (HC) than the other solving groups; further, a nonsignificant trend suggested that this …

Ammonia (NH3) is the most abundant alkaline component and can react with atmospheric acidic species to form aerosols that can lead to numerous environmental and health issues. Increasing atmospheric NH3 over agricultural regions in the US has been documented. However, spatiotemporal changes of NH3 concentrations over the entire US are still not thoroughly understood, and the factors that drive these changes remain unknown. Herein, we applied the Atmospheric Infrared Sounder (AIRS) monthly NH3 dataset to explore spatiotemporal changes in atmospheric NH3 and the empirical relationships with synthetic N fertilizer application, livestock manure production, and climate factors across the entire US …

Dead neonatal mice are currently used as bait for delivery of toxin to invasive brown treesnakes (Boiga irregularis) on Guam; once deployed in the field the mice are highly attractive to the snakes but only for about four days. An artificial bait containing a mixture of fats mimicking those in skin of the mice is also highly attractive to the snakes and remains attractive 2–3 times longer. The artificial bait, however, costs more than the mice, and is more difficult to attach to the capsules of a novel aerial bait delivery system. This paper describes a reformulation of the bait …
