The sigB operon of Bacilus subtilis encodes (σB plus three additional proteins (RsbV, RsbW, and RsbX) that regulate σB activity. Using an anti-σB monoclonal antibody to monitor the levels of (σB protein, PSPAC to control the expression of the sigB operon, and a ctc-lacZ reporter system to monitor σB activity, we observed that the rsbV and rsbW products control (σB activity at the ctc promoter independently of their effects on (σB levels. σB is a secondary σ factor of Bacillus subtilis. RNA polymerase containing cr transcribes a subset of genes that are expressed after heat shock or the onset of the stationary phase of growth. Three genes (rsbV, rsbW, and rsbX), cotranscribed with the σB structural gene (sigB), regulate o dependent gene expression. RsbW is the primary inhibitor of this system with the other gene products acting upstream of RsbW in the σB regulatory pathway. Evidence is now presented that RsbW inhibits σB-dependent transcription by binding to crB and blocking …

The frequently inexorable fatality, pansexual communicability, and lengthy period of asymptomatic latency of HIV combine to form biological realities and ecological dangers that are individually and socially problematic at very deep levels. How can we think sociologically about the AIDS epidemic? Talcott Parsons’ concept of “sick role,”2 a venerable and productive staple of medical sociology, gives us little to work with here, precisely as sexually active, HIV-positive but undiagnosed and asymptomatic persons do not see themselves as “sick.” Nor, importantly, do others, including: sexual intimates, friends, family members, employers, and health professionals. Relative paleosol maturities are inversely proportional to the accumulation rates of the sediment upon which they formed, and are therefore excellent relative indicators of how much geologic time elapsed between any two horizons. An empirically-based model is advanced using paleosol maturities to estimate the relative geologic time separating any stratigraphic levels within the lower Eocene Willwood Formation. The revised Willwood time stratigraphy from this analysis helps evaluate the nature, tempo, and possible causes of three major episodes of mammalian appearance and disappearance. These faunal events are directly correlated with certain aspects of paleosol evolution in the Willwood Formation. That evolution …

Three new species of nematodes were encountered during a study of natural diversity of nematode associates of native species of Vitis L. in California. Achromadora walkeri sp. n. was found in rhizosoil of the native California grape Vitis californica Bentham and is characterized by the position of the amphid (within the vicinity of both dorsal and ventral teeth), a relatively long stoma, and the absence of a prerectum. The other 2 species were plant parasitic criconematids: Criconemoides featherensis sp. n., found in association with roots of V. californica, is characterized by possessing strongly retrose annuli posterior to the vulva, …

The ΣB-Dependent Promoter Of The Bacillus Subtilis Sigb Operon Is Induced By Heat Shock, Andrew K. Benson, W. G. Haldenwang

σB, a secondary sigma factor of Bacillus subtilis, was found to increase 5- to 10-fold when cultures were shifted from 37 to 48°C. Western blot (immunoblot) analyses, in which monoclonal antibodies specific for the sigB operon products RsbV, RsbW, and σB were used to probe extracts from wild-type and mutant B. subtilis strains, revealed that all three proteins increased coordinately after heat shock and that this increase was dependent on σB but not RsbV, a positive regulator normally essential for σB-dependent sigB expression. Development of water erοsiοn and surface water quality control practices requires information concerning the hydraulic characteristics of upland areas. The relatively small flow rates normally found within rills make measurement of hydraulic parameters difficult. Chemical tracing procedures, originally developed for stream and river systems, have been successfully used to measure rill flow properties. A chemical tracer of known concentration is added to the rill and by knowing the degree of dilutiοn at a downstream sampling point, flow rate can be calculated. The Niobrara Valley Preserve: An Inventory Of A Biogeographical Crossroads, Gail E. Kantak, Steven P. Churchill

Transactions of the Nebraska Academy of Sciences and Affiliated Societies

This paper synthesizes results of a biological inventory of a 219-km2 nature preserve in the central Niobrara Valley of Nebraska. The two-year inventory documented the presence of 80 species of mosses and liverworts, 581 of vascular plants, 130 of aquatic Invertebrates and 278 of vertebrate animals. Comparison with distribution records from four other counties in Nebraska shows that the Niobrara study site has higher vertebrate species diversity despite its smaller area. Although the study area is in the central grasslands of North America, many of the species here are more typical of forest biomes far to the east, north …

The mean larval paddlefish density was 60 times higher in the upper unchannelized section of the Missouri River in Nebraska compared to the lower unchannelized section, and was three times higher than the channelized section's density. Within the upper unchannelized section, 96.2% of the larvae were collected in the discharge of two tributaries, which lie in the lower one-third of the reach. Survival from larval to young-of-the-year stage (June through August) was highest during 1991 due, in part, to the recent drought. Reduced fluctuation of river stages occurred as a result of reduced runoff, which minimized the need to reduce …

Burbot harvest represented 5.1% of the fish-harvest in the tailwater of Gavins Point Dam in July 1956, one year after the dam was closed. Harvest increased from 510 in 1956 to 4,280 in 1958, but represented only 2% of the 1958 harvest. By 1961 burbot were no longer routinely caught in the tailwater, and their density decreased in the remainder of Nebraska's portion of the Missouri River. At present they are very rare and should be listed as endangered in Nebraska.

Woody Vegetation Of A Disjunct Bur Oak (Quercus Macrocarpa) Forest In East-Central Nebraska, Debra Ann Beightol, Thomas B. Bragg

Woody plant composition was assessed for three tree-size classes in two ravines of Oak Glen Wildlife Management Area, a disjunct oak forest in Seward County, Nebraska, using Importance Values (IV) obtained by the Point-Quarter method. Bur oak (Quercus macrocarpa Michx.) dominated the large-size class (>30 cm dbh) (IV = 258) but elms (Ulmus americana L. and U. rubra Muhl.) (IV = 130) dominated the forest in one ravine in the medium-size class (10-30cm dbh) and elm and hackberry (Celtis occidentalis L.) (IV = 114 and 27 respectively) dominated the small-size class (<10cm dbh). Bur oak in the small-size was low in abundance (IV = 9) and was absent from one of the study transects. These data on tree-size distribution in the extant forest suggests that the forest is likely to succeed from one presently dominated by bur oak, which appears to have characterized the presettlement forest, to one dominated primarily by elm or hackberry. Current management is likely to encourage a continuation of this succession.

Ten eastern Nebraska tallgrass prairie remnants, varying in size from one to 18 ha, were studied or visited during the growing seasons from 1979 to 1993. A total of 217 prairie plant species were recorded. Big bluestem (Andropogon gerardii), little bluestem (Andropogon scoparius), smooth brome (Bromus inermis subsp. inermis), New Jersey tea (Ceanothus herbaceus var. pubescens), false sunflower (Heliopsis helianthoides var. scabra), Kentucky bluegrass (Poa pratensis), and porcupine-grass (Stipa spartea) were prevalent in all the prairies although the specific floristic composition has been reported to vary …

In the springs of 1989 and 1990, we collected 15 lesser sandhill cranes from native grasslands along the Platte River. Macroinvertebrates constituted 89% of the diet, whereas plants constituted 11%. Scarab beetles were the predominant macroinvertebrate food. Other foods included earthworms, snail shells, sedge tubers, ground beetles, and crane fly larvae. Cranes exhibited differential use patterns on the study area; they extensively used areas composed of wet meadow and lowland grassland, but minimally used areas composed entirely of lowland grassland. Macroinvertebrate numbers and biomass were significantly different between areas extensively used by cranes and areas minimally used. Recommendations for management …

The saline wetlands associated with the Platte River have received very little attention from botanists since the 1890s, and several plants widespread in these habitats, such as rayless salt-marsh aster (Aster brachyactis), annual goldenweed (Haplopappus annuus) and thelypody (Thelypodium integrifolium) have been considered rare in the State. A floristic inventory of sixteen tracts of saline lowlands near the North Platte River in Garden, Morrill, and Scotts Bluff counties reveals the presence of 231 species of vascular plants (169 native and 62 introduced), including three Eurasian halophytes (Althaea officinalis, Najas marina, Spergularia marina) …

Transgenic pigs were generated that produced human protein C in their milk at up to 1 g/liter. The gene construct was a fusion gene consisting of the cDNA for human protein C inserted into the first exon of the mouse whey acidic protein gene. These results demonstrate that the mouse whey acidic protein gene contains regulatory elements that can direct cDNA expression at high levels in the pig mammary gland. Recombinant human protein C that was produced at about 380 pg/ml per hr in transgenic pig milk possessed anticoagulant activity that was equivalent to that of protein C derived from …

The purpose of this project was to conduct a systematic inventory of rare plant species on the Pine Ridge District of the Nebraska National Forest. The objective of the inventory was to locate rare plant occurrences and compile information on their location, status and any possible threats. Four areas within the Pine Ridge District were identified as priority areas for the inventory fieldwork. The four tracts included the Soldier Creek Wilderness Area, a tract encompassing the East and West Ash Creek drainages, a middle unit tract, and a tract which straddles U.S. Highway 385 and encompasses a number of timber …

Levels of human immunodeficiency virus (HIV) DNA, RNA, or p24 antigen and reverse transcriptase activity in T-cell cultures treated with 500 IU of recombinant alpha interferon (rIFNα) per ml were comparable to those in control cultures. Radioimmunoprecipitation analysis of proteins in lysates of IFN-treated T cells documented a marked accumulation of HlV proteins. Localization of gp120 by immunofluorescence showed a diffuse pattern in IFN-treated cells quite distinct from the ring pattern in untreated control cells. That large quantities of a120 in aberrant cell compartments might affect HlV morphogenesis was confirmed in infectivity studies: virions from IFN-treated cells were 100- to …
