Attitudes Towards Insects: An Attempt To Change Negative Attitudes To Neutral Or Positive, Christina Chapman Jul 2025

Attitudes Towards Insects: An Attempt To Change Negative Attitudes To Neutral Or Positive, Christina Chapman

Department of Entomology: Distance Master of Science Projects

Purpose and Summary

The purpose of this project was to examine whether negative attitudes towards insects could be changed in children after learning about them in a safe and enjoyable environment. At the start of this project, I was a science teacher in an urban middle school, where most students were neither fond of nor interested in insects. This experience shaped my project: I wanted to change attitudes from negative to positive, transforming “eww gross, kill it!” into “let it live; it has a job to do.” Through my studies in the entomology program, my belief that insects are highly …

The Effect Of Individual Variation On Epidemiological Dynamics: From Super-Spreaders To Landscapes Of Risk, Alexis Shannon Beagle Jul 2025

The Effect Of Individual Variation On Epidemiological Dynamics: From Super-Spreaders To Landscapes Of Risk, Alexis Shannon Beagle

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Individuals that make up a population always display some level of variation between members. At the smallest scale, individuals always maintain genetic variation, but variation continues to arise at other scales of biological organization from physiological or immunological variation within individuals to variation in behavior between individuals that can be both genetically and environmentally mediated. The role of individual variation is particularly important in the context of infectious diseases because there is strong evidence that infections are highly heterogenous, indicating that patterns of individual variation drive a significant portion of the epidemiological patterns we observe. Individual variation leads to heterogeneity …

Cazyme Gene Cluster Diversity In Human Gut Microbiome, Yi Xing Jul 2025

Cazyme Gene Cluster Diversity In Human Gut Microbiome, Yi Xing

Department of Food Science and Technology: Dissertations, Theses, and Student Research

In gut microbiome research, carbohydrate-active enzyme gene clusters (CGCs) have emerged as key functional units for understanding microbial glycan degradation. Unlike taxonomic or broad pathway annotations, CGCs offer gene-cluster-level resolution and capture substrate-specific microbial functions. However, their diversity and distribution in relation to host metabolic phenotypes, such as obesity, remain poorly characterized. This study tests the hypothesis that the composition and abundance of fiber-targeting CGCs vary between obese and healthy human gut microbiomes, reflecting distinct microbial carbohydrate utilization strategies. To examine this, we constructed a high-quality reference CGC dataset comprising 94,019 clusters from the Unified Human Gastrointestinal Genome and profiled …

Overcoming Maternal Antibody Interference In Swine Influenza: Advancing Dna Vaccine Strateies For Enhanced Immune Protection In Piglets, Danh Cong Lai Jul 2025

Overcoming Maternal Antibody Interference In Swine Influenza: Advancing Dna Vaccine Strateies For Enhanced Immune Protection In Piglets, Danh Cong Lai

School of Veterinary and Biomedical Sciences: Dissertations, Theses, and Student Research

Maternally derived antibodies (MDAs) play a vital role in protecting neonates from infectious diseases, but their presence at the time of vaccination can interfere with vaccine-induced immune responses, thereby reducing vaccine effectiveness. MDA interference has been documented in pigs vaccinated with whole inactivated virus (WIV) vaccines against swine influenza A virus (IAV). In this thesis, we evaluated the efficacy of a lipid nanoparticle encapsulated DNA (LNP-DNA) vaccine against swine IAV in the presence and absence of MDAs, comparing it to a WIV vaccine. In MDA-negative piglets, both the LNP-DNA and WIV vaccines induced strong immune responses and effectively prevented the …

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

With limited land and water resources available it has become critical to maximize output for agricultural products while maintaining sustainable farming practices. Ricefish co-culture can be used to maximize limited land and water resources for production of rice and fish concurrently and reduce the need of chemical inputs. While rice-fish co-culture has been practiced in Asia for centuries, very few studies have examined the feasibility and economic impact of rice fish co-culture in the U.S.A. Two studies were conducted in Stuttgart, Arkansas, U.S.A. from 2023-2024 to investigate the suitability of raising Koi carp in flooded rice paddies with a sushi …

Gmos And Public Perception: College Students’ Opinions And Knowledge Of Genetic Engineering, Serene M. Cheng Jun 2025

Gmos And Public Perception: College Students’ Opinions And Knowledge Of Genetic Engineering, Serene M. Cheng

RURALS: Review of Undergraduate Research in Agricultural and Life Sciences

Genetically modified organisms (GMOs) can be developed and implemented to address significant global challenges, including nutrient deficiencies, climate change, and crop disease. However, widespread opposition has prevented the success of GMOs. We believe this opposition is strongly connected to a lack of understanding about genetics in general, which has been reflected by previous studies.

In this study, we considered the relationship between GMO support and genetics knowledge by comparing results from a brief quiz and responses to prompts relating to GMOs. Using a survey of college students, we found that support for GMOs and general knowledge of genetics are positively …

A Battle Of Armies: Massive Attack Of Natural Enemies To The True Armyworm Mythimna Unipuncta (Haworth, 1809) (Lepidoptera: Noctuidae) In Forage Crops, Armando Falcon-Brindis, Raul T. Villanueva

Insecta Mundi

Understanding the interactions between plant-host-parasitoid plays a major role in insect pest management. Farmers in the southeastern United States rely on pasture systems for cattle milk and beef production, but the benefits of parasitoids and predators on major lepidopteran pests on pasture lands are still overlooked. Here, we aimed to identify and quantify the effect of parasitism on the true armyworm Mythimna unipuncta (Haworth, 1809) (Lepidoptera: Noctuidae) attacking teff grass Eragrostis tef (Zucc.) Trotter (Poaceae) and crabgrass Digitaria sanguinalis (L.) Scop. (Poaceae) in western Kentucky. In early August of 2023, 547 larvae of M. unipuncta were collected from experimental …

Integrating High Throughput Phenotyping Approaches To Characterize Plant Traits And Support Nutrient Management Research, Katie J. Bathke Jun 2025

Integrating High Throughput Phenotyping Approaches To Characterize Plant Traits And Support Nutrient Management Research, Katie J. Bathke

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

Over the past decade, considerable research has addressed the social, economic, and agronomic challenges posed by imbalanced nutrient applications, particularly nitrogen and, more recently, sulfur. In response, high throughput plant phenotyping, as one of the latest approaches, has contributed to the development of innovative techniques for assessing key plant phenotypes under stress over time in both field and greenhouse environments. However, there is a limited number of studies applying these techniques for nutrient management, particularly in scenarios where two abiotic stressors coexist, such as the combination of water stress and nitrogen deficiency, or concurrent nitrogen and sulfur deficiencies.

The Connection Between An Unequitable Distribution Of Care Responsibilities And A Widening Gender Nutrition Gap In Latin America, Alexis Nicole Bodlak May 2025

The Connection Between An Unequitable Distribution Of Care Responsibilities And A Widening Gender Nutrition Gap In Latin America, Alexis Nicole Bodlak

Honors Program: Senior Projects (Public)

The notion of care refers to activities that satisfy the basic needs of existence and reproduction. Within rural communities, care work such as the production and preparation of food contributes to food security at the household level. Care work supports the paid workforce, allowing workers to focus on commercial productive activities that generate economic income in rural communities. Women perform 6.3 to 29.5 more hours per week than men in unpaid care work, reducing their time for income-generating activities and personal nutrition management. Affecting their economic independence and ability to access and consume nutritious food. Studies show that in food-insecure …

Investigating Fatty Acid Influence On West Nile Virus In Mosquitoes And Birds, Hayley Moree May 2025

Investigating Fatty Acid Influence On West Nile Virus In Mosquitoes And Birds, Hayley Moree

Honors Program: Senior Projects (Public)

Vector-borne diseases, including Zika virus (ZIKV), Dengue virus (DENV), and West Nile virus (WNV), pose considerable global health threats, accounting for over 400 million infections and more than 700,000 deaths annually. Many vector-borne diseases are within the Flaviviridae family. Despite this, effective treatments have not been found for some flaviviruses. Flaviviruses exploit host cellular machinery to facilitate their replication and survival, reprogramming metabolic pathways, including lipid metabolism. Lipids play essential roles in the viral life cycle, providing structural components for the viral envelope, enabling membrane rearrangements for replication complexes, and supplying energy resources necessary for particle assembly.

Targeting lipid metabolism …

Mitigating Nitrogen Losses: The Role Of Enhanced Efficiency Fertilizers In Sustainable Corn Production, Dipesh Giri May 2025

Mitigating Nitrogen Losses: The Role Of Enhanced Efficiency Fertilizers In Sustainable Corn Production, Dipesh Giri

Department of Agronomy and Horticulture: Dissertations, Theses, and Student Research

Nitrogen (N) is essential for crop production, but its inefficient use often leads to environmental losses through nitrate leaching, ammonia volatilization, and nitrous oxide (N₂O) emissions. Enhanced efficiency fertilizers (EEFs), such as nitrification inhibitors (NIs) and urease inhibitors (UIs), have been developed to minimize these losses and improve nitrogen use efficiency (NUE). This research combined a multi-year field experiment and a controlled laboratory study to evaluate how dual-inhibitor fertilizers compare to conventional urea and single-inhibitor formulations in Nebraska cropping systems. The main objective was to understand how nitrogen inhibitors influence N availability, crop performance, and nitrogen losses across different environmental …

Implications Of Type Iv Pilus Retraction For Dna Uptake By Acinetobacter Baumannii, Yafan Yu May 2025

Implications Of Type Iv Pilus Retraction For Dna Uptake By Acinetobacter Baumannii, Yafan Yu

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Horizontal gene transfer (HGT) via natural competence allows bacteria to incorporate extracellular DNA (eDNA) into their genomes, facilitating genetic diversity and adaptation. In Acinetobacter species, natural competence depends on DNA binding and uptake mediated by type IV pili (T4P). T4P are dynamic extracellular filaments composed primarily of the major pilin subunit PilA, along with a few minor subunits. T4P drive a range of cellular functions including twitching motility, biofilm formation, and DNA uptake, with some functions dependent on pilus retraction and others not. However, how Acinetobacter T4P bind DNA and how T4P/eDNA interactions impact biofilm formation remain unclear. Here, I …

Assessing Sleep Quality In A Preclinical Model Of Low Back Pain Through Novel Noninvasive Methods, Lydia C. Saltz

Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research

Chronic low back pain (LBP) has widespread negative impacts on quality of life. Current treatments for chronic LBP fail to provide long-term efficacy and motivate the need to understand key drivers of poor patient outcomes. Mounting evidence indicates that sleep disturbances and chronic LBP have a strong reciprocal relationship. Women may be particularly vulnerable to the negative impacts of the sleep-pain axis, exhibiting greater prevalence in both conditions. However, the relationship between sleep, pain, and sex remains understudied. Preclinical models can be used to parse out characteristic sleep features that degrade with pain, which can be further broken out by …

Gene Editing Approaches To Improving Grain Amino Acid Profiles In Maize And Sorghum, Joseph Preston Hurst May 2025

Gene Editing Approaches To Improving Grain Amino Acid Profiles In Maize And Sorghum, Joseph Preston Hurst

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Maize grain is deficient in lysine. While the opaque2 mutation increases grain lysine, o2 is a transcription factor that regulates a wide network of genes beyond zeins, which leads to pleiotropic and often negative effects. Additionally, the drastic reduction in 19kDa and 22kDa alpha-zeins causes a floury kernel, unsuitable for agricultural use. Sorghum, closely related to maize, has a similar lack of lysine that can be corrected with prolamin alteration and also subject to the same deleterious kernel texture. CRISPR/cas9 gives researchers the ability to directly target genes of interest. In this research, gene editing was used to specifically target …
