Attenuation Of Mtorc1-Driven Secretion Of Lipoproteins And Triacylglycerides By Short Chain Fatty Acids: Mechanistic Insight Into The Pathogenesis Of Hypertriglyceridemia, Joseph L. Roberts
Department of Nutrition and Health Sciences: Dissertations, Theses, and Student Research
The mechanistic target of rapamycin complex 1 (mTORC1) is a serine/threonine kinase that drives several anabolic processes including lipid synthesis, protein synthesis, and adipogenesis. mTORC1 is highly active in the livers of obese rodents, in overnutrition, and is implicated in the development of obesity related metabolic disorders, including the overproduction of atherogenic lipoproteins. Direct inhibition of mTORC1 is not a viable treatment strategy because it prevents feedback inhibition of the insulin-signaling cascade, leading to increased lipid synthesis and secretion of lipoproteins. Thus, therapeutic approaches that drive catabolic pathways are considered promising mechanisms for overcoming mTORC1-driven anabolism. Human liver HepG2 cells …

Getting To The Heart Of Our Students: First-Year Students And Their Wellness, Shannon Ford
Department of Educational Administration: Dissertations, Theses, and Student Research
Wellness is a topic everyone is talking about these days. While a keyword among conversations, wellness within literature is not broken down but simply a theme. Therefore, I wanted to gain a better understanding of how students across college campuses view and perceive their personal wellness. To do this, I conducted a phenomenological mixed methods study, which explored how first-year students perceive wellness.
Through administering the 36-question Perceived Wellness Inventory survey (Adams, Bezner & Steinhardt, 1997) and conducting a focus group, three themes emerged: behaviors versus knowledge, feelings, and support. These three themes supported existent wellness literature and added areas …

Although recent research revealed an impact of westernization on diversity and composition of the human gut microbiota, the exact consequences on metacommunity characteristics are insufficiently understood, and the underlying ecological mechanisms have not been elucidated. Here, we have compared the fecal microbiota of adults from two non-industrialized regions in Papua New Guinea (PNG) with that of United States (US) residents. Papua New Guineans harbor communities with greater bacterial diversity, lower inter-individual variation, vastly different abundance profiles, and bacterial lineages undetectable in US residents. A quantification of the ecological processes that govern community assembly identified bacterial dispersal as the dominant process …
Characteristics Of Trailer Thermal Environment During Commercial Swine Transport Managed Under U.S. Industry Guidelines, Y Xiong, Angela Green, Richard S. Gates
Characteristics Of Trailer Thermal Environment During Commercial Swine Transport Managed Under U.S. Industry Guidelines, Y Xiong, Angela Green, Richard S. Gates
Department of Animal Science: Faculty Publications
Transport is a critical factor in modern pork production and can seriously affect swine welfare. While previous research has explored thermal conditions during transport, the impact of extreme weather conditions on the trailer thermal environment under industry practices has not been well documented; and the critical factors impacting microclimate are not well understood. To assess the trailer microclimate during transport events, an instrumentation system was designed and installed at the central ceiling level, pig level and floor-level in each of six zones inside a commercial swine trailer. Transport environmental data from 34 monitoring trips (approximately 1–4 h in duration each) …

Sphingolipids Are Required For Efficient Triacylglycerol Loss In Conjugated Linoleic Acid Treated Adipocytes, Wei Wang, Michael E. Fromm
Department of Animal Science: Faculty Publications
Conjugated linoleic acid (CLA) reduces adiposity in human and mouse adipocytes. This outcome is achieved through a variety of biological responses including increased energy expenditure and fatty acid oxidation, increased inflammation, repression of fatty acid biosynthesis, attenuated glucose transport, and apoptosis. In the current study, profiling of 261 metabolites was conducted to gain new insights into the biological pathways responding to CLA in 3T3-L1 adipocytes. Sphinganine and sphingosine levels were observed to be highly elevated in CLA treated adipocytes. Exogenous chemicals that increased endogenous ceramide levels decreased lipid levels in adipocytes, and activated AMP-activated protein kinase (AMPK) as well as …

Effects Of Poverty On Children’S Education, Jasmin Valdez
Nebraska College Preparatory Academy: Senior Capstone Projects
Poverty has been affecting children in elementary school for many generations. It affects their lives by decreasing their chances at a better life outside of poverty through their education. Through research, I found that children living in poverty do not do as well in school than those living with a higher income. Poverty affects them through their family, social, psychological, and health aspects

Comparative Analysis Of Aquaponic Grow Beds, Thomas Malachi Mcguire, Geoffrey Addison Popken
Comparative Analysis Of Aquaponic Grow Beds, Thomas Malachi Mcguire, Geoffrey Addison Popken
Department of Environmental Studies: Undergraduate Student Theses
The global agricultural system has become a major source of stress on the earth’s natural resources. The current food production and distribution system is not secure and will not sustain future populations as global food production pressures intensify. It will be important to explore other avenues of food production in order to build a more sustainable system in the future. Aquaponics eliminates soil from the equation of food production by combining aquaculture(fish farming) and hydroponics(growing plants in water saturated media). Aquaponics may provide a source of relief for areas without suitable soil or dense human populations. This experiment looks to …

Micrornas Are Absorbed In Biologically Meaningful Amounts From Nutritionally Relevant Doses Of Cow’S Milk And Chicken Eggs And Affect Gene Expression In Peripheral Blood Mononuclear Cells, Cell Cultures, And Mouse Livers, Scott Baier
Department of Nutrition and Health Sciences: Dissertations, Theses, and Student Research
Throughout the twenty-first century, evidence has been continually increasing to show the importance of epigenetic regulation in health. While the term “epigenetics” can be applied to many different processes, the focus of this dissertation will be on microRNAs and chromatin structure. Ultimately, both of these forms of epigenetic regulation can be used to fine tune gene expression based on environmental cues. The first three chapters of the dissertation focus on microRNA bioavailability, stability, and function from two commonly consumed food products: cow’s milk and chicken eggs. This important work has been the first of its kind to demonstrate the bioavailability …
“At-Risk” Food Deserts In Lincoln, Nebraska, Samantha Corr
“At-Risk” Food Deserts In Lincoln, Nebraska, Samantha Corr
Department of Environmental Studies: Undergraduate Student Theses
This investigational study conducted in Lincoln, Nebraska seeks to explore the link between geographic and economic barriers to food access. Food access is a problem in the United States. 29 percent of U.S. ZIP codes do not contain a grocery store or supermarket. Families also face barriers to food economically, as low income individuals are less able to afford food. Through the use of ArcGIS 10.2.2 for Desktop software this project seeks to visually represent the areas in which access to food is limited both geographically and financially. Creating one mile buffer zones around each grocery store in Lincoln, Nebraska …
Stability And Biological Activity Of Dietary Micrornas, Katherine Howard
Stability And Biological Activity Of Dietary Micrornas, Katherine Howard
Department of Nutrition and Health Sciences: Dissertations, Theses, and Student Research
MicroRNAs play important roles in gene regulation by binding to complimentary sites at the 3’ untranslated region of target mRNA molecules. Binding results in inhibition or degradation of target mRNA. Many bovine and chicken microRNA are homologous with human counterparts enabling gene regulation. A recent study in our lab provided undisputable evidence that endogenous milk microRNAs are bioavailable in humans; resulting in regulation of human gene expression. Based on these findings, we wanted to explore the possibility that other exogenous food borne microRNAs are able to be absorbed through the diet. My thesis surrounds two aims: 1) assessing the stability …
Climate Effects On Mammalian Body Size And Grassland Composition Inferred From Late Quaternary Grazers In The Great Plains Of North America, Tom Baldvins
Department of Earth and Atmospheric Sciences: Dissertations, Theses, and Student Research
Several mammalian taxa exhibited a diminution during the Pleistocene-Holocene transition. Two hypotheses have been proposed to explain this phenomenon: (1) human hunting and (2) climate change along with its effects on grasslands. I use stable isotopes from a variety of Quaternary grazing mammals to measure grassland changes following the Last Glacial Maximum (LGM) in Nebraska and to develop a climate proxy to compare with mammalian body size. Results indicate that latest Pleistocene equids and mammoths have δ13C values indicative of a C3 or mixed C3/C4 diet. However, Holocene bison have greatly elevated δ13 …

Quantitative Risk Assessment Of Uk Food Products Cross-Contaminated With Allergens, Benjamin C. Remington, Joseph L. Baumert, Marty W. Blom, Geert F. Houben, Steve L. Taylor, Astrid G. Kruizinga
Department of Food Science and Technology: Faculty Publications
Allergens in food pose a risk to allergic consumers, especially if they are present in food without declaration or warning. While there is EU regulation for allergens present as an ingredient, this is not the case for unintended allergen presence (UAP). Food companies use precautionary “may contain” labels to inform allergic individuals of a potential risk from UAPs. However, the use or absence of precautionary label has a limited correlation with the level of UAP and consequently the risk of an unexpected allergic reaction. Allergen risk assessment using probabilistic techniques enables estimation of the residual risk after the consumption of …

Development Modeling Of Lucilia Sericata (Diptera: Calliphoridae), Amanda Roe, Leon G. Higley
School of Natural Resources: Faculty Publications
The relationship between insect development and temperature has been well established and has a wide range of uses, including the use of blow flies for postmortem (PMI) interval estimations in death investigations. To use insects in estimating PMI, we must be able to determine the insect age at the time of discovery and backtrack to time of oviposition. Unfortunately, existing development models of forensically important insects are only linear approximations and do not take into account the curvilinear properties experienced at extreme temperatures. A series of experiments were conducted with Lucilia sericata, a forensically important blow fly species, that met …
Tallgrass Prairie Vegetation Response To Spring Fires And Bison Grazing, Stephen L. Winter, Brady W. Allred, Karen R. Hickman, Samuel D. Fuhlendorf
Tallgrass Prairie Vegetation Response To Spring Fires And Bison Grazing, Stephen L. Winter, Brady W. Allred, Karen R. Hickman, Samuel D. Fuhlendorf
School of Natural Resources: Faculty Publications
Spring fires in tallgrass prairie can create environmental conditions conducive to plant growth in the subsequent growing season. Following fires, burned areas can also be attractive to grazing animals such as bison (Bison bison). Sustained grazing activity within recently burned areas can alter vegetation structure relative to nearby landscape patches that haven’t burned recently. In 2007, we collected data on bison grazing activity, vegetation structure, and the growth and reproduction of a perennial forb, Arnoglossum plantagineum, in Oklahoma tallgrass prairie. We compared these variables in landscape patches that had burned in the spring of 2007 to measurements …

Temporal Patterns Of Nutrition Dependence In Secondary Sexual Traits And Their Varying Impacts On Male Mating Success, Malcolm F. Rosenthal, Eileen A. Hebets
Temporal Patterns Of Nutrition Dependence In Secondary Sexual Traits And Their Varying Impacts On Male Mating Success, Malcolm F. Rosenthal, Eileen A. Hebets
Eileen Hebets Publications
Variation in the quantity of nutrients ingested over an individual’s lifetime is likely to differentially affect distinct male secondary sexual traits and courtship signals, potentially providing females with information about a male’s past and present foraging history. We hypothesize that female choice is thus influenced by a male’s lifetime foraging history. To test this, we manipulated the quantity of nutrients (i.e. prey items) available to male wolf spiders, Schizocosa stridulans, using a fully crossed 2 × 2 design with low versus high prey quantity across juvenile and adult life stages, and assessed the impact of these diet treatments on male …

The aim of this study was to evaluate the carcass and meat characteristics of eight muscles from bulls with distinct growth paths. A total of 40 Alentejana male calves were allocated to two distinct feeding regimes. In the continuous growth (CG) system, the animals were fed concentrates plus hay and were slaughtered at 18 months of age. On the other hand, in the discontinuous growth (DG) system, the animals were fed hay until 15 months of age; the cattle were then fed the same diet provided to the CG group from 15 to 24 months of age. The DG reduced …
Ectoparasitism Shortens The Breeding Season In A Colonial Bird, Charles R. Brown, Mary Bomberger Brown
Ectoparasitism Shortens The Breeding Season In A Colonial Bird, Charles R. Brown, Mary Bomberger Brown
School of Natural Resources: Faculty Publications
When blood-feeding parasites increase seasonally, their deleterious effects may prevent some host species, especially those living in large groups where parasites are numerous, from reproducing later in the summer. Yet the role of parasites in regulating the length of a host’s breeding season—and thus the host’s opportunity for multiple brooding—has not been systematically investigated. The highly colonial cliff swallow (Petrochelidon pyrrhonota), a temperate-latitude migratory songbird in the western Great Plains, USA, typically has a relatively short (eight to nine week) breeding season, with birds rarely nesting late in the summer. Colonies at which ectoparasitic swallow bugs (Oeciacus vicarius) were experimentally …
Proline Metabolism Increases Katg Expression And Oxidative Stress Resistance In Escherichia Coli, Lu Zhang, James R. Alfano, Donald F. Becker
Proline Metabolism Increases Katg Expression And Oxidative Stress Resistance In Escherichia Coli, Lu Zhang, James R. Alfano, Donald F. Becker
Department of Biochemistry: Faculty Publications
The oxidation of L-proline to glutamate in Gram-negative bacteria is catalyzed by the proline utilization A (PutA) flavoenzyme, which contains proline dehydrogenase (PRODH) and _1-pyrroline-5-carboxylate (P5C) dehydrogenase domains in a single polypeptide. Previous studies have suggested that aside from providing energy, proline metabolism influences oxidative stress resistance in different organisms. To explore this potential role and the mechanism, we characterized the oxidative stress resistance of wild-type and putA mutant strains of Escherichia coli. Initial stress assays revealed that the putA mutant strain was significantly more sensitive to oxidative stress than the parental wild-type strain. Expression of PutA in the …

Concomitant Uptake Of Antimicrobials And Salmonella In Soil And Into Lettuce Following Wastewater Irrigation, J. Brett Sallach, Yuping Zhang, Laurie Hodges, Daniel D. Snow, Xu Li, Shannon L. Bartelt-Hunt
Department of Civil and Environmental Engineering: Faculty Publications
The use of wastewater for irrigation may introduce antimicrobials and human pathogens into the food supply through vegetative uptake. The objective of this study was to investigate the uptake of three antimicrobials and Salmonella in two lettuce cultivars. After repeated subirrigation with synthetic wastewater, lettuce leaves and soil were collected at three sequential harvests. The internalization frequency of Salmonella in lettuce was low. A soil horizon-influenced Salmonella concentration gradient was determined with concentrations in bottom soil 2 log CFU/g higher than in top soil. Lincomycin and sulfamethoxazole were recovered from lettuce leaves at concentrations as high as 822 ng/g and …
