Mulching practice offers farmers an opportunity to minimize the effects of drought, water loss, and soil erosion on crop production. Plastic film is widely used as a mulching material; however, contamination of arable lands by residual plastic has become a serious concern. Synthetic biodegradable mulch films and sprays may offer a more sustainable alternative to plastic films, however current evidence on the factors that influence the suitability of these products for agricultural applications is fragmented, making it unclear under what conditions these products meet agronomic, environmental, and societal needs. We address this gap by conducting a systematic review of studies …

Mechanistic investigation of host-microbe interactions in the human gut are hindered by difficulty of co-culturing microbes with intestinal epithelial cells. On one hand the gut bacteria are a mix of facultative, aerotolerant or obligate anaerobes, while the intestinal epithelium requires oxygen for growth and function. Thus, a coculture system that can recreate these contrasting oxygen requirements is critical step towards our understanding microbial-host interactions in the human gut. Here, we demonstrate Intestinal Organoid Physoxic Coculture (IOPC) system, a simple and cost-effective method for coculturing anaerobic intestinal bacteria with human intestinal organoids (HIOs). Using commensal anaerobes with varying degrees of oxygen …

Pollinator Communities And Their Ecosystem Services At Conservation Grasslands And Adjacent Croplands, Araceli Gomez Villegas
Pollinator Communities And Their Ecosystem Services At Conservation Grasslands And Adjacent Croplands, Araceli Gomez Villegas
Department of Entomology: Dissertations, Theses, and Student Research
Pollinators are intrinsically linked to the success of unmanaged and managed ecosystems by providing pollination services that aid in the reproduction of wildflowers and many crops. Land use change, habitat loss, fragmentation, and related landscape-level phenomena (for example, increased pesticide exposure) threaten pollinators and have been associated with population declines. In the Midwestern region of the United States, land conversion of native prairies and grasslands to row-crop agriculture has been one of the largest contributors to pollinator habitat loss. Conservation programs, such as the Conservation Reserve Program, have worked towards removing environmentally sensitive lands from agriculture production and enrolling them …

A Dietary Variety: Analyzing The Impacts Of Diverse Vs Worm-Exclusive Diets On The Blanding’S Turtle (Emydoidea Blandingii), Caitlin F. Copenhaver
Honors Program: Senior Projects (Public)
The endangered Blanding’s turtle (Emydoidea blandingii) is a species with a generally diverse diet in their freshwater wetland habitats; they consume prey such as crayfish and snails. In captivity, these options often become more limited due to logistical reasons. As ex situ conservation efforts such as captive rearing and head-starting are becoming more popular for this species, it will be important to gain more insight on how these more limited diets might affect these turtles. This study describes a comparison between diets in six captive Blanding’s turtles from September to November of 2023, with three turtles receiving only …

The ability of tumour cells to thrive in harsh microenvironments depends on the utilization of nutrients available in the milieu. Here we show that pancreatic cancer-associated fibroblasts (CAFs) regulate tumour cell metabolism through the secretion of acetate, which can be blocked by silencing ATP citrate lyase (ACLY) in CAFs. We further show that acetyl-CoA synthetase short-chain family member 2 (ACSS2) channels the exogenous acetate to regulate the dynamic cancer epigenome and transcriptome, thereby facilitating cancer cell survival in an acidic microenvironment. Comparative H3K27ac ChIP–seq and RNA–seq analyses revealed alterations in polyamine homeostasis through regulation of SAT1 gene expression and enrichment …

Homogentisate Phytyltransferase (HPT) catalyzes condensation of homogentisate (HGA) and phytyl diphosphate (PDP) to produce tocopherols, but can also synthesize tocotrienols using geranylgeranyl diphosphate (GGDP) in plants engineered for deregulated HGA synthesis. In contrast to prior tocotrienol biofortification efforts, engineering enhanced tocopherol concentrations in green oilseeds has proven more challenging due to the integral role of chlorophyll metabolism in supplying the PDP substrate. This study show that RNAi suppression of CHLSYN coupled with HPT overexpression increases tocopherol concentrations by >two-fold in Arabidopsis seeds. We obtained additional increases in seed tocopherol concentrations by engineering increased HGA production via overexpression of …

Introduction: Early childhood educators (ECEs) play a critical role in supporting the development of young children’s executive functions (EF). EF, in turn, underpins lifelong resilience and well-being. Unfortunately, many ECEs report adverse childhood experiences (ACEs) that may compound high stress levels associated with an emotionally and physically demanding profession. ACEs have well-established negative implications for adult well-being and may dampen ECEs’ capacities to engage in emotionally responsive interactions with children. However, many individuals who experience ACEs also report post-traumatic growth experiences that foster empathy, self-determination, and resilience. Such post-traumatic growth may equip teachers with skills to engage in responsive …

The Impact Of Iron Supplementation On The Preterm Neonatal Gut Microbiome: A Pilot Study, Matthew Van Ormer, Maranda Thompson, Melissa K. Thoene, Jean-Jack Riethoven, Sathish Kumar Natarajan, Ann Anderson-Berry
Department of Nutrition and Health Sciences: Faculty Publications
Objective The gastrointestinal microbiome in preterm infants exhibits significant influence on optimal outcomes–with dysbiosis shown to substantially increase the risk of the life-threatening necrotizing enterocolitis. Iron is a vital nutrient especially during the perinatal window of rapid hemoglobin production, tissue growth, and foundational neurodevelopment. However, excess colonic iron exhibits potent oxidation capacity and alters the gut microbiome–potentially facilitating the proliferation of pathological bacterial strains. Breastfed preterm infants routinely receive iron supplementation starting 14 days after delivery and are highly vulnerable to morbidities associated with gastrointestinal dysbiosis. Therefore, we set out to determine if routine iron supplementation alters the preterm gut …

Introduction Food security and nutrition have been severely impacted during the COVID-19 pandemic, particularly in low- and middle-income countries (LMICs). We aimed to quantify the impacts of the pandemic on food security and diet diversity within Chilanga District in Zambia and identify target areas for high-impact social protection and safety net programs.
Methods We conducted a cross-sectional study in Chilanga district immediately after the Omicron variant surge in February 2022. Diet quality and food security were assessed based on a household diet questionnaire and a Minimum Dietary Diversity-Women (MDD-W) score was calculated. A paired t-test was used to determine whether …

Disruption Of The Sorghum Circadian Clock Impacts Sorghum-Sugarcane Aphid Interaction Dynamics And Aphid Feeding Behavior, Kumar Shrestha, Prince Zogli, Lise Pingault, Sajjan Grover, Juan Betancurt Cardona, Joe Louis
Department of Entomology: Faculty Publications
Circadian clocks play a pivotal role in orchestrating metabolic rhythms in response to environmental stress. Sorghum bicolor (sorghum), a multipurpose cereal crop, suffers severe growth and yield reduction due to feeding by sugarcane aphids (SCA; Melanaphis sacchari), which are phloem-feeding pests. Previously, it has been shown that sorghum utilizes a multitude of defense mechanisms to curb SCA colonization. However, our understanding of the impact of circadian clock on sorghum-aphid interaction dynamics is limited. To explore this, a time-series transcriptomics was conducted on sorghum plants in disrupted circadian rhythm with and without SCA infestation. Transcriptomic analysis revealed a total of …

Conservation is increasingly focused on preventing losses in species’ populations before they occur. Tracking changes in demographic parameters that can impact a population’s resilience in response to drivers of global change can support early conservation efforts. We assessed trends in population productivity (late summer juveniles per 100 females) relative to drivers of global change in 40 pronghorn (Antilocapra americana) herds across sagebrush (Artemisia spp.) steppe in Wyoming. Pronghorn are an iconic rangeland species that have been exposed to increasing levels of anthropogenic, climatic, and land-use change. Using data collected across the state of Wyoming, we (1) assessed …

Foraging Rates From Metabarcoding: Predators Have Reduced Functional Responses In Wild, Diverse Prey Communities, Stella F. Uiterwaal, John Delong
School of Biological Sciences: Faculty Publications
Functional responses describe foraging rates across prey densities and underlie many fundamental ecological processes. Most functional response knowledge comes from simplified lab experiments, but we do not know whether these experiments accurately represent foraging in nature. In addition, the difficulty of conducting multispecies functional response experiments means that it is unclear whether interaction strengths are weakened in the presence of multiple prey types. We developed a novel method to estimate wild predators' foraging rates from metabarcoding data and use this method to present functional responses for wild wolf spiders foraging on 27 prey families. These field functional responses were considerably …

Center for Plant Science Innovation: Faculty Publications
Plant species utilize a variety of regulatory mechanisms to ensure sustainable productivity. Within this intricate framework, numerous non-coding RNAs (ncRNAs) play a crucial regulatory role in plant biology, surpassing the essential functions of RNA molecules as messengers, ribosomal, and transfer RNAs. ncRNAs represent an emerging class of regulators, operating directly in the form of small interfering RNAs (siRNAs), microRNAs (miRNAs), long noncoding RNAs (lncRNAs), and circular RNAs (circRNAs). These ncRNAs exert control at various levels, including transcription, posttranscription, translation, and epigenetic. Furthermore, they interact with each other, contributing to a variety of biological processes and mechanisms associated with stress resilience. …

Embracing Diversity In Agricultural Economics, Timothy Meyer
Cornhusker Economics
To steal an overused cliché, “There’s room in the tent for everyone.” Over the 2023 academic year, I have reiterated this message to all my students, with one addition. Not only is there room for everyone, but all are invited AND welcome. Food is something we all have in common, no matter the background. I think this is why producers in the state of Nebraska feel as strongly as they do about the food they produce; it is life-giving and should be taken seriously. Nebraska Agriculture is part of what makes our state great, and that is not a secret …

Predicting Sedentarism And Its Impact On Caloric Requirements, Jacob Michels, John C. Beghin
Yeutter Institute International Trade Policy Review
A recent study conducted by PhD candidate Jacob Michels and Agricultural economist John Beghin delves into the question of whether global estimates of food insecure populations need a reevaluation of their methodology to account for increasing sedentarism. This reevaluation is prompted by the increasing prevalence of sedentary lifestyles worldwide, which calls for a redefinition of caloric thresholds indicating the onset of food insecurity. In this short article, we provide a nontechnical summary of their investigation recently published in Michels and Beghin (2024).

Gut microbiomes profoundly influence insect health and mediate interactions between plant hosts and their environments. Insects, including aphids, harbour diverse obligate symbionts that synthesize essential nutrients and facultative symbionts that enhance host fitness in specific ecological contexts. Sorghum (Sorghum bicolor) is a significant cereal crop cultivated worldwide that has been negatively affected by the presence of an invasive piercing-sucking insect pest, the sugarcane aphid (SCA; Melanaphis sacchari). We previously identified SC265 and SC1345 as the resistant and susceptible sorghum lines, respectively, among the founder nested association mapping (NAM) population. Here, using these resistant and susceptible lines, we …

Previously, we described the RNA recombinants accumulating in tissues infected with the bromoviruses BMV (Brome mosaic virus) and CCMV (Cowpea chlorotic mottle virus). In this work, we characterize the recombinants encapsidated inside the purified virion particles of BMV and CCMV. By using a tool called the Viral Recombination Mapper (ViReMa) that detects recombination junctions, we analyzed a high number of high-throughput sequencing (HTS) short RNA sequence reads. Over 28% of BMV or CCMV RNA reads did not perfectly map to the viral genomes. ViReMa identified 1.40% and 1.83% of these unmapped reads as the RNA recombinants, respectively, in BMV and …

Piglet mortality during lactation is a significant concern in swine production, influenced by complex interactions involving sow, piglet, environmental, and management factors. While crushing by the sow may be the ultimate cause of piglet mortality, there are many factors influencing the outcome, including parity, thermal stress, and animal housing systems. New farrowing systems are continuously being developed; however, it is difficult for producers to make decisions without any scientific basis. This study aimed to assess the impact of different farrowing pen layouts on piglet performance, considering parity and season. A total of 546 sows and 9123 piglets were monitored across …

Emerging evidence indicates that antibiotic-induced dysbiosis can play an etiological role in the pathogenesis of neuropsychiatric disorders. However, most of this evidence comes from rodent models. The objective of this study was to evaluate if antibiotic-induced gut dysbiosis can elicit changes in gut metabolites and behavior indicative of gut-brain axis disruption in common marmosets (Callithrix jacchus) – a nonhuman primate model often used to study sociability and stress. We were able to successfully induce dysbiosis in marmosets using a custom antibiotic cocktail (vancomycin, enrofloxacin and neomycin) administered orally for 28 days. This gut dysbiosis altered gut metabolite profiles, …

Phenethyl isothiocyanate (PEITC) is a secondary metabolic product yielded upon the hydrolysis of gluconasturtiin and it is highly accumulated in the flowers of watercress. The aim of the current study was to assess the role of a naturally derived PEITC-enriched extract in the induction of oxidative stress and to evaluate its anti-melanoma potency through the regulation of its metabolism with the concurrent production of the N-acetyl cysteine conjugated by-product. For this purpose, an in vitro melanoma model was utilized consisting of human primary (A375) cells as well as metastatic (COLO-679) malignant melanoma cells together with non-tumorigenic immortalized keratinocytes (HaCaT). …

Dry bean is a nutrient-dense food targeted in biofortification programs to increase seed iron and zinc levels. The underlying assumption of breeding for higher mineral content is that enhanced iron and zinc levels will deliver health benefits to the consumers of these biofortified foods. This study characterized a diversity panel of 275 genotypes comprising the Yellow Bean Collection (YBC) for seed Fe and Zn concentration, Fe bioavailability (FeBio), and seed yield across 2 years in two field locations. The genetic architecture of each trait was elucidated via genome-wide association studies (GWAS) and the efficacy of genomic prediction (GP) was assessed. …
