Natural And Experimental Infection With An Attaching And Effacing Strain Of Escherichia Coli In Calvest, Rodney A. Moxley, David H. Francis

Gnotobiotic calves were inoculated with an 05:K4:H-, urease-positive strain of Escherichia coli isolated from a 2-day-old calf with diarrhea. The calves developed elevated temperatures and passed loose mucoid feces, with or without blood. The E. coli strain was negative for heat-stable and heat-labile enterotoxins but produced high levels of Shiga-like toxin. Bacteria attached diffusely to the epithelium of the large intestine and multifocally to the epithelium of the ileum. The duodenum and jejunum were not affected. At the sites of bacterial attachment, microvilli were effaced, enterocytes were degenerate, and necrosis and exfoliation had occurred. These results confirm a …

Purpose. The primary purpose of this study was to assess the perceptions of Nebraska vocational agriculture instructors with regard to the Cooperatives--Serving Our Community instructional unit. Method. A mailed questionnaire procedure was used to obtain information from 108 of the possible 127 Nebraska vocational agriculture instructors. A total of 78 observations were valid. The dependent variables were instructors' perceptions of instructional unit quality, value, and in-service training. Independent variables included Nebraska Vocational Agriculture Association districts, years of experience as a vocational agriculture instructor, total number of students enrolled in local vocational agriculture programs, instructional unit use-rate and participation mode at …

Production Of Acquired Immunodeficiency Syndrome-Associated Retrovirus In Human And Nonhuman Cells Transfected With An Infectious Molecular Clone, Akio Adachi, Howard E. Gendelman, Scott Koenig, Thomas Folks, Ronald Willey, Arnold Rabson, Malcom A. Martin

We constructed an infectious molecular clone of acquired immunodeficiency syndrome-associated retrovirus. Upon transfection, this clone directed the production of infectious virus particles in a wide variety of cells in addition to human T4 cells. The progeny, infectious virions, were synthesized in mouse, mink, monkey, and several human non-T cell lines, indicating the absence of any intracellular obstacle to viral RNA or protein production or assembly. During the course of these studies, a human colon carcinoma cell line, exquisitely sensitive to DNA transfection, was identified.

Within the last decade a new etiologic agent has been identified as a possible cause of diarrhea in humans and domestic animals—that being the protozoan parasite Cryptosporidium. The purpose of this study was to determine the incidence and distribution of Cryptosporidium in the state of Nebraska, to determine if Cryptosporidium occurs more frequently in association with other enteropathogens and to determine about the relation of scouring in the calves to infection with Cryptosporidium. A total of 620 fecal samples from 334 dairy calves from 71 herds in Nebraska were examined for cyptosporidial oocysts using the Sheather’s sugar flotation …

The economics of the newspaper industry occupies a special place in industrial organization. Unlike the vast majority of enterprises, this business is intimately linked to the protection of freedom of expression. Constitutional democracies generally have monitored it with special care. In the United States the Newspaper Preservation Act of 1970 provides some antitrust immunity for segments of the industry. Ignoring a captive regulation (pro-producer, wealth-maximizing) rationale for the act, we may presume that the United States Congress deems daily newspaper viability as an objective superior to the conventional reasons for antitrust. Dental practitioners treating children are often faced with the question of when to initiate orthodontic treatment. Preventive and interceptive orthodontics encourage treatment of young patients to guide developing malocclusions into functional and aesthetic harmony. Success in early orthodontic treatment depends on accurate diagnosis. Dimensional growth of the arches anterior to the first permanent molar is essentially complete by the age of eight, years of age. 24 Therefore, two important factors in dentition alignment may be the arch length anterior to the first permanent molars and the mesiodistal crown diameter (MDCD) of the permanent teeth in the arch.

Restriction Endonuclease Activity Induced By Pbcv-1 Virus Infection Of A Chlorella-Like Green Alga, Yuannan Xia, Dwight Burbank, Lothar Uher

An enzyme was isolated from a eucaryotic, Chlorella-like green alga infected with the virus PBCV-1 which exhibits type I1 restriction endonuclease activity. The enzyme recognized the sequence GATC and cleaved DNA 5' to the G. Methylation of deoxyadenosine in the GATC sequence inhibited enzyme activity. In vitro the enzyme cleaved host Chlorella nuclear DNA but not viral DNA because host DNA contains GATC and PBCV-1 DNA contains GmATC sequences. PBCV-1 DNA is probably methylated in vivo by the PBCV-1-induced methyltransferase described elsewhere (Y. Xia and J. L. Van Etten, Mol. Cell. Biol. 6:144-1445). Dna Methyltransferase Induced By Pbcv-1 Virus Infection Of A Chlorella-Like Green Alga, Yuannan Xia, James L. Van Etten

A DNA methyltransferase was isolated from a eucaryotic, Chlorella-like green alga infected with the virus PBCV-1. The enzyme recognized the sequence GATC and methylated deoxyadenosine solely in GATC sequences. Host DNA, which contains GATC sequences, but not PBCV-1 DNA, which contains GmATC sequences, was a good substrate for the enzyme in vitro. The DNA methyltransferase activity was first detected about 1 h after viral infection; PBCV-1 DNA synthesis and host DNA degradation also began at about this time. The appearance of the DNA methyltransferase activity required de novo protein synthesis, and the enzyme was probably virus encoded. Methylation of …

This research was undertaken with the specific intent to develop a personality profile of licensed nurses (RN's and LPN's) which would provide a basis for recommendations of techniques to be utilized in continuing education programs for nurses. The instrument selected to measure the psychological type preference, and thus create the personality profile, was the Myers-Briggs Type Indicator (MBTI) Carl Jung's theory pertaining to personality, known as type theory, states that individuals perceive and respond to information differently according to their psychological type preference which is measured by the MBTI. Galactose-fermenting (galactose-posirive) strains of Streptococcus tbermopbilus, alone and combined with galactosepositive and galactose-negative strains of Lactobacillus bulgaricus, were used as starter cultures in the manufacture of Swiss and Mozzarella cheese and were paired with Streptococcus lactis (also galactose-positive) in short-method Cheddar cheese manufacture. Experimental Swiss cheese made with the galactosepositive Streptococcus tbermopbilus starter alone contained a large amount of galactose (ca. 26 to 28 /amol/g of curd) 28 h after hooping compared with control Swiss (< 2/amol/g) made with a nongalactose fermenting strain of Streptococcus tbermopbilus and a galactosepositive strain of Lactobacillus bulgaricus. We surveyed the IRRI farm to determine the extent of parasitization of yellow stemborer egg masses. The egg masses were randomly collected at weekly intervals from July to October 1984 from rice fields, 15-20 days after transplanting, and brought to the laboratory for collection of emerging parasites. Three species of hymenopterous parasitoids—Tetrasticus schoenobii, Telenomus rowani, and Trichogramma japonicum—were found from 700 egg masses.

We introduced a method to calculate percent parasitism as based on parasite biology and behaviour. Among the three species, the combination of T. rowani and T. japonicum was the highest multiparasitization of yellow stem borer egg …

Visna lentiviruses have a natural tropism for cells of the macrophage lineage of sheep and goats, but virus replication in these cells in vivo is restricted so that only small quantities of virus are produced. One restricting factor suggested in previous studies is that virus replication is dependent on the maturity of the cells: the more mature the cell, the less restrictive the replication of the virus. The Sun Refinery at Tulsa presented a new and special type of problem that I had never faced before. The refinery is just across the Arkansas River from downtown Tulsa. Secondary Toxicity Of Coyotes Killed By 1080 Single-Dose Baits, Richard J. Burns, Guy E. Connolly, Iwao Okuno

Carcasses and viscera of coyotes poisoned by Compound 1080 (sodium fluoroacetate) in single-dose tallow baits (SDBs) were fed to 3 coyotes, 3 domestic dogs, 4 striped skunks, and 15 black-billed magpies to determine if these species would be poisoned secondarily. Test subjects received no food other than tissues from poisoned coyotes for periods of 14 to 35 days. Total amounts of contaminated coyote tissues consumed by dogs, coyotes, skunks, and magpies, respectively, averaged 67, 152, 117, and 371% of body weight. Except for one skunk that refused to eat, no mortalities occurred and no evidence of poisoning was seen.

During the summer months of 1979, public agencies in Ventura County received complaints which pointed to a rodent infestation of campground areas along the north coastal strip. Investigations revealed a widespread infestation of oceanfront riprap by roof rats (Rattus rattus). Visual surveillance, trapping, baiting and population-estimating techniques and results are described and discussed. Implementation of integrated pest management practices resulted in the reduction of rat populations to a no-complaint level and provided a framework for a long-term maintenance program.

In 1984 studies were initiated to identify the bird species causing damage to pistachio nuts, quantify the losses, and explore possible methods for alleviating bird losses. The first year's field observations were made to identify and determine the level of activity of the various depredating bird species in the orchard. A combination Purivox® Double-John carousel propane exploder and CO2 pop-up scarecrow operated in synchrony was evaluated in five ripening sunflower fields in 1981 and 1982, respectively, near Devils Lake and Westhope, North Dakota. In each field, this treatment was evaluated in an alternating off-on sequence of 5-day intervals for a period of 20 days. In 1981, devices were deployed at one unit per 8 to 10 acres and in 1982 at one unit per 4 to 6 acres. Denial of re-entry (batproofing) through structural modification is widely accepted as the most effective and ecologically sound method for eliminating commensal bats from structures. Such methods are clearly superior to lethal measures which have only questionable efficacy and may exacerbate bat/human interactions. However, since bats are able to enter small and obscure openings, conventional batproofing of all such openings is often not practical or economical. Salmon poisoning disease (SPD) was tested as a potential method of lethal control for coyotes (Canis latrans). Fresh fish containing the agents for SPD was fed to 72 captive adult coyotes. Coho (Oncorhunchus kisutch) and steel head salmon (Salmo gairdneri) from Oregon hatcheries were the principal species of fish used. Coyotes that ate the fish developed observable signs of SPD in a mean of 8 days. The overall rate of mortality was 50%, and death occurred in a mean of 20 days from consuming fish. Conservation-tillage farming systems have changed agriculture and brought new challenges to the wildlife damage field. Associated minor problems may result from the presence of rattlesnakes (Crotalus viridis) or badgers (Taxidea taxus), but sprout-pulling damage by birds has not been reported. Rodents dig and consume newly planted corn. At least 14 small mammal species have been captured in notillage cornfields, some species throughout fields and others primarily at edges. Deer mice (Peromyscus maniculatus) generally have been the most abundant, comprising 71 to 93% of total captures. Small mammal populations in no-tillage fields are generally no …

The Townsend pocket gopher, (Thomomys townsendi), poses a serious economic threat to alfalfa production in Diamond Valley, Nevada. Many control methods have been practiced with only limited or seasonal success. Application of strychnine-treated alfalfa hay has been an effective control method; however, the relationship between strychnine concentration and pocket gopher kill has not been examined. This study was conducted to evaluate the effectiveness of three concentrations (0.5%, 1.0% and 1.7%) of strychnine-treated alfalfa hay baits.
