Lactic Acid Bacterial Surface Display Of Scytovirin Inhibitors For Anti-Ebolavirus Infection, Joshua Wiggins, Ngan Nguyen, Wenzhong Wei, Leah Liu Wang, Haley Hollingsead Olson, Shi-Hua Xiang

Nebraska Center for Virology: Faculty Publications

Scytovirin (SVN) is a lectin from cyanobacteria which has a strong inhibitory activity against Ebola virus infection. We engineered scytovirin as the inhibitor for surface display of lactic acid bacteria to block Ebola virus infection. Two different bacterial strains (Lactobacillus casei and Lactococcus lactis) were successfully engineered for scytovirin expression on the bacterial surface. These bacteria were found to be effective at neutralizing pseudotyped Ebolavirus in a cell-based assay. This approach can be utilized for prophylactic prevention, as well as for treatment. Since lactic acid bacteria can colonize the human body, a long-term efficacy could be achieved. Furthermore, …

Pancreatic ductal adenocarcinoma (PDAC) presents a critical global health challenge, and early detection is crucial for improving the 5-year survival rate. Recent medical imaging and computational algorithm advances offer potential solutions for early diagnosis. Deep learning, particularly in the form of convolutional neural networks (CNNs), has demonstrated success in medical image analysis tasks, including classification and segmentation. However, the limited availability of clinical data for training purposes continues to represent a significant obstacle. Data augmentation, generative adversarial networks (GANs), and cross-validation are potential techniques to address this limitation and improve model performance, but effective solutions are still rare for 3D …

Department of Entomology: Faculty Publications

BACKGROUND: Highly mobile stored product insects may be able to readily orient in response to food cues and pheromones to attack durable commodities at each link of the postharvest supply chain. A 0.4% deltamethrin-incorporated long-lasting insecticide-incorporated netting (LLIN) is a successful novel preventative integrated pest management (IPM) tactic to intercept dispersing insects after harvest. However, it is unknown whether exposure to LLIN may affect olfaction and orientation to important semiochemicals by immature stored product dermestids, therefore the aim of this study was to assess whether exposure to LLIN disrupts the normal olfactory and chemotactic behavior of warehouse beetle, Trogoderma variabile …

Department of Civil and Environmental Engineering: Faculty Publications

Per and polyfluoroalkyl substances (PFAS) present a unique challenge to remediation techniques because their strong carbon−fluorine bonds make them difficult to degrade. This review explores the use of in silico enzymatic design as a potential PFAS degradation technique. The scope of the enzymes included is based on currently known PFAS degradation techniques, including chemical redox systems that have been studied for perfluorooctanesulfonic acid (PFOS) and perfluorooctanoic acid (PFOA) defluorination, such as those that incorporate hydrated electrons, sulfate, peroxide, and metal catalysts. Bioremediation techniques are also discussed, namely the laccase and horseradish peroxidase systems. The redox potential of known reactants and …

Bovine respiratory disease (BRD) is a leading cause of disease in feedlot and stocker calves with Mannheimia haemolytica (MH) as one of the most common etiologies. One of the most effective means of controlling BRD is through metaphylaxis, which involves administering antimicrobials to all animals at high risk of developing BRD. However, increasing prevalence of multidrug resistant (MDR) MH may reduce efficacy of metaphylaxis due to decreased susceptibility to drugs used for metaphylaxis. Primarily, this study aimed to determine the effect of tulathromycin metaphylaxis and subsequent BRD treatment on antimicrobial resistance (AMR) in MH isolated from stocker calves. Secondary objectives …

Radiofrequency Inactivation Of Salmonella In Black Pepper And Dried Basil Leaves Using In-Package Steaming, Surabhi Wason, Rossana Villa-Rojas, Jeyamkondan Subbiah

Department of Food Science and Technology: Faculty Publications

Radiofrequency (RF) heating has been extensively studied for pasteurizing low‐moisture foods. Currently, bulk foods are treated with radiofrequency; potential cross‐contamination may occur during packaging of pasteurized products. As an alternative, in‐package RF processing was evaluated for Salmonella inactivation on black peppercorns and dried basil leaves and prevention of cross‐contamination during storage postprocessing. Inpackage steaming refers to the process in which the samples were heated in a steam vent package to generate and retain steam during the treatment. This treatment achieved good heating uniformity which could be because of the circulation of steam within the package. One‐way steam vent allowed the …

Analyzing Major Elements Of Crash Injury Severity Involving Priority-I Detriments Of Vision-Zero Plan, Muhammad Umer Farooq, Cole Madson, Aemal Khattak

Mid-America Transportation Center: Final Reports and Technical Briefs

Road traffic crashes result in significant economic losses for individuals, their families, and entire nations. These losses stem from the expenses associated with injury treatment, as well as the productivity lost due to fatalities or disabilities caused by these injuries. The 2030 Agenda for Sustainable Development set an ambitious target of halving the global number of fatalities and injuries from road traffic crashes by 2020 and achieving zero deaths by 2030, commonly referred to as the 'Target Zero Plan.' The Target Zero Plan prioritizes traffic safety issues into three distinct levels. The three priority levels are determined based on the …

Integrated Flood Risk Assessment Of Properties And Associated Population At County Scale For Nebraska, Usa, Shivendra Srivastava, Tirthankar Roy

Department of Civil and Environmental Engineering: Faculty Publications

Risk assessment of properties and associated population was conducted for the state of Nebraska, leveraging only open-source datasets. The flood risk framework consisted of interactions among drivers, i.e. hazard, exposure, vulnerability, and response, to assess the risks related to properties and associated populations. To quantify hazard on a county scale, we considered properties at risk of flooding based on a flood score (a higher score represents a greater chance of flooding). Exposure was quantified by considering population density at the county level. We quantified vulnerability under four categories: social, ecological, economic, and health. Response, a relatively newer component in flood …

Factors Affecting Forage Quality And The Subsequent Response In Production And Energy Metabolism In Lactating Jersey Cows, Kassidy Kate Buse Nov 2023

Factors Affecting Forage Quality And The Subsequent Response In Production And Energy Metabolism In Lactating Jersey Cows, Kassidy Kate Buse

Dissertations and Doctoral Documents, University of Nebraska-Lincoln, 2023–

Forage quality is the potential of a forage to produce a production response in animals. Because of how essential they are in dairy cattle rations, forages can have major influence on the behavior and production of dairy cattle. In experiment 1, the effect of forage fragility on the rumen environment and digestibility was evaluated. Increasing the inclusion of a less fragile forage led to greater rumen retention time, which resulted in lower rumen pH and greater NDF digestibility. Experiment 2 evaluated the effects of diets high in either corn silage or alfalfa haylage supplemented with either corn- or soy-based protein …
