Structural Determinants For The Interactions Of Chemically Modified Nucleic Acids With The Stabilin‑2 Clearance Receptor, Hans Gaus, Colton M. Miller, Punit P. Seth, Edward N. Harris
Department of Biochemistry: Faculty Publications
The Stabilin receptors are systemic clearance receptors for some classes of chemically modified nucleic acid therapeutics. In this study, the recombinant human secreted ecto-domain of the small isoform of Stabilin-2 (s190) was purified from cell culture and evaluated for direct binding with a multitude of antisense oligonucleotides (ASOs) using a fluorescence polarizationbased assay. The tested ASOs varied in their backbone composition, modification of the ribose 2′ position, overall length of the oligo, and sequence of the nucleotide bases. A fully phosphorothioate (PS) ASO with a 5−10−5 pattern of flanking 2′-O-methoxyethyl modifications was then used to test the effects …

Triclosan is frequently used for its antimicrobial properties and has been detected in human serum, urine, and breast milk. Animal and molecular studies have shown that triclosan exerts a wide range of adverse health effects at both high (ppm) and low (ppb) concentrations. Since triclosan is of growing concern to human and environmental health, there is a need to improve extraction procedures and to study additional effects from triclosan exposure. In this study, we have improved triclosan extraction from breast milk by using salt (MgSO4) to reduce emulsion formation and increase water polarity and water (~80%) to enhance the overall …
High-Resolution Water Footprints Of Production Of The United States
High-Resolution Water Footprints Of Production Of The United States
Daugherty Water for Food Global Institute: Faculty Publications
The United States is the largest producer of goods and services in the world. Rainfall, surface water supplies, and groundwater aquifers represent a fundamental input to economic production. Despite the importance of water resources to economic activity, we do not have consistent information on water use for specific locations and economic sectors. A national, spatially detailed database of water use by sector would provide insight into U.S. utilization and dependence on water resources for economic production. To this end, we calculate the water footprint of over 500 food, energy, mining, services, and manufacturing industries and goods produced in the United …

How asymmetries in motor behavior become established normally or atypically in mammals remains unclear. An established model for motor asymmetry that is conserved across mammals can be obtained by experimentally inducing asymmetric striatal dopamine activity. However, the factors that can cause motor asymmetries in the absence of experimental manipulations to the brain remain unknown. Here, we show that mice with inner ear dysfunction display a robust left or right rotational preference, and this motor preference reflects an atypical asymmetry in cortico-striatal neurotransmission. By unilaterally targeting striatal activity with an antagonist of extracellular signal-regulated kinase (ERK), a downstream integrator of striatal …

Rapid Identification And Typing Of Mycobacterium Avium Complex Using Matrix Assisted Laser Desorption/Ionization Time-Of-Flight Mass Spectrometry (Maldi-Tof) Library And Biomarker Based Approaches, Claudia Antonika, John Dustin Loy, Raul Barletta, Michael Collins
UCARE: Research Products
Bacteria associated with the Mycobacterium avium complex (MAC) cause significant problems in animal agriculture and cause large economic losses and morbidity of livestock. Two clinically important subspecies of M. avium are M.avium ssp paratuberculosis (MAP) and M. avium ssp hominissuis (MAH). MAP is the etiologic agent of Johne’s disease, a chronic and fatal enteritis in ruminants, that has been linked to Crohn’s disease in humans.1 MAH has zoonotic potential as it is one of the leading causes of secondary infections in AIDS patients. Identification of M. avium in clinical samples is challenging as they have overlapping host ranges and clinical …

Department of Sociology: Faculty Publications
In this paper, we describe the long-term evolution of societies of secondary users in dynamic spectrum access networks. Such an understanding is important to help us anticipate future trends in the organization of large-scale distributed networked deployments. Such deployments are expected to arise in support of a wide variety of applications, including vehicular networks and the Internet of Things. Two new biologically-inspired spectrum access strategies are presented here, and compared with a random access baseline strategy. The proposed strategies embody a range of plausible assumptions concerning the sensing capabilities and social characteristics of individual secondary users. Considering these strategies as …

Department of Animal Science: Faculty Publications
Intrauterine growth restriction (IUGR) is linked to metabolic dysfunction in offspring, but the mediating mechanisms are still under investigation (Barker et al., 1993). IUGR fetuses adapt to their poor intrauterine environment by repartitioning nutrients to organs critical for survival (i.e., brain, heart) at the expense of tissues such as muscle (Yates et al., 2012c). These developmental adaptations help the fetus to survive in utero but have lifelong consequences in offspring; persistent reduction of highly metabolic muscle mass is detrimental to glucose homeostasis (DeFronzo et al., 1981). Glucose metabolism is regulated primarily by insulin, and nutrient depravation is associated with impaired …

Taxonomy is a scientific discipline that has provided the universal naming and classification system of biodiversity for centuries and continues effectively to accommodate new knowledge. A recent publication by Garnett and Christidis expressed concerns regarding the difficulty that taxonomic changes represent for conservation efforts and proposed the establishment of a system to govern taxonomic changes. Their proposal to “restrict the freedom of taxonomic action” through governing subcommittees that would “review taxonomic papers for compliance” and their assertion that “the scientific community's failure to govern taxonomy threatens the effectiveness of global efforts to halt biodiversity loss, damages the credibility of science, …

University of Nebraska State Museum: Mammalogy Papers
The bat fauna of the Caribbean island of Sint Eustatius consists of five documented species—Monophyllus plethodon, Brachyphylla cavernarum, Artibeus jamaicensis, Ardops nichollsi, and Molossus molossus—and one provisional species—Tadarida brasiliensis. The Insular Single-leaf Bat, M. plethodon, is reported in the scientific literature for the first time from Sint Eustatius based on material presented herein. The bat fauna of the island is considered to be unbalanced because only three species, which are the environmental generalists, are abundant, whereas the more specialized species are rare or absent from the fauna. It is our hypothesis …

Sexual Assault In The Political Sphere, Robert Larsen
Honors Program: Senior Projects (Public)
This project sought to analyze how sexual assault in the political sphere is perceived and treated in contemporary society in the United States of America. The thesis analyzed eight cases of sexual misconduct, including six from the past thirty years. In each case, the reaction of party and social leaders, of the politician’s constituents and of the politician himself were looked at, as well as the consequences the politician faced. The results were then analyzed side-by-side to discover similarities and differences between ho cases of sexual assault allegations were treated and in terms of what happened to the politician after …
Preliminary Analysis Of Hieroglyph And Iconography Placement On Freestanding Monuments At Copán, Honduras, Elizabeth Koenen
Preliminary Analysis Of Hieroglyph And Iconography Placement On Freestanding Monuments At Copán, Honduras, Elizabeth Koenen
Honors Program: Senior Projects (Public)
This paper analyzes the placement of hieroglyphs and iconography on freestanding monuments at the ancient Maya site of Copán, Honduras. Preliminary spatial analysis using Geographic Information Systems (GIS) highlight two potentially important findings. First, stelae in the main civic-ceremonial precinct (Principal Group), while erected in the most centralized and public location in the city, are not always placed to allow for public viewing of their fronts. Second, differences may exist in the number of logographic and syllabic glyphs used on a object depending on the type of object and its location. Further research and data collection are needed in order …

Sexism in American culture comes as no surprise. In 2017, the film industry saw an increasing number of powerful men within its ranks exposed and exiled for allegations of sexually predatory behavior. Many see this purging as a sign of changing times, but this view is optimistic: the tides of change ebb in and out, and this problem far exceeds the movie industry. Well before the onslaught of articles on the likes of Harvey Weinstein, my classmates and I resolved to craft our senior thesis film with a crew made up entirely of women: we simply saw the pool of …

An Analysis Of The Social And Political Barriers To Wind Energy Development In Lancaster County, Nebraska, Corrin Bemis
Honors Program: Senior Projects (Public)
With the increasing threat of climate change beginning to impact cities and regions in the United States, many states are turning to renewable energy sources as a way to mitigate their carbon footprint. While Nebraska has the potential to be a leader in wind energy development within the next few years, some restrictions are still put in place that slow down the expansion of wind farms. Although the state is ranked 4th in the nation for wind energy potential, Nebraska is ranked 20th for actual wind energy development. This paper will define the social and political barriers that …
Parallels Of Morality: Wilde And Nietzsche’S Challenge To Social Obligation, Amzie A. Dunekacke
Parallels Of Morality: Wilde And Nietzsche’S Challenge To Social Obligation, Amzie A. Dunekacke
Honors Program: Senior Projects (Public)
This thesis explores Irish author Oscar Wilde’s The Picture of Dorian Gray in relation to a selection of texts by the German philosopher Friedrich Nietzsche. To demonstrate the similarities between Wilde and Nietzsche’s challenges to European morality, this work considers these themes, which are present in the ideologies of both Wilde and Nietzsche: the body and sensual pleasure, social construction, and the hypocrisy of altruism. Both radical thinkers castigate Platonic notions of the body as ignoble and weak, and they mock European propriety’s shyness of the body. In addition, Wilde and Nietzsche offer similar criticisms of social laws, adopting a …

Olfactory Learning Capabilities Of Paraphrynus Laevifrons, John Perez
Honors Program: Senior Projects (Public)
Amblypygids, a species of nocturnal arachnids from the tropics and subtropics are incredibly intelligent and are able to not only navigate through difficult tropical terrain but also, it is believed, possess olfactory learning capabilities that aid in navigation and recollection of environments. It is hypothesized that through sensitive olfactory receptors on their antenniform legs and highly developed mushroom bodies, Amblypygi can not only learn smells but also learn to associate smells with certain stimuli such as a crevice to take refuge in. To test this hypothesis, Paraphrynus laevifrons, a species of amplypygid was subjected to four different learning treatment groups …

Copy Number Variation In The Porcine Genome Detected From Whole-Genome Sequence, Rebecca Anderson
Copy Number Variation In The Porcine Genome Detected From Whole-Genome Sequence, Rebecca Anderson
Honors Program: Senior Projects (Public)
Copy number variations (CNVs) are large insertions, deletions, and duplications in the genome that vary between individuals in a species. These variations are known to impact a broad range of phenotypes from molecular-level traits to higher-order clinical phenotypes. CNVs have been linked to complex traits in humans such as autism, attention deficit hyperactivity disorder, nervous system disorders, and early-onset extreme obesity. In this study, whole-genome sequence was obtained from 72 founders of an intensely phenotyped experimental swine herd at the U.S. Meat Animal Research Center (USMARC) in Clay Center, Nebraska. This included 24 boars (12 Duroc and 12 Landrace) and …
South Dakota V. Wayfair And Tax Modernization In Nebraska, Adam B. Thimmesch
South Dakota V. Wayfair And Tax Modernization In Nebraska, Adam B. Thimmesch
Nebraska Law Bulletin (Selected Issues)
The United States Supreme Court is poised to make a significant change to how it regulates states and their exercise of taxing autonomy under our federal structure. The Court’s current position has severely limited Nebraska’s ability to conform its tax system to the modern economy and undercuts its ability to raise funds to support its operations. Nebraska is also otherwise at an important point in its history. Changes in the farm economy and ongoing budget issues have undercut the state’s ability to provide the public services necessary for a state to compete and thrive in the modern world. The State …
The Victorian Body, Peter J. Capuano
The Victorian Body, Peter J. Capuano
Department of English: Faculty Publications
The nineteenth century is extremely important for the study of embodiment because it is the period in which the modern body, as we currently understand it, was most thoroughly explored. This was the era when modern medical models of the body were developed and disseminated, when modern political relations to the body were instantiated, and when modern identities in relation to class, race, and gender were inscribed. While questions about the distinctions between personhood and the body were studied by the ancients, nineteenth-century developments in technology, economics, medicine, and science rendered such categories newly important for Britons who were the …

Synchrony — An Emergent Property Of Recreational Fisheries, Mark A. Kaemingk, Christopher J. Chizinski, Keith Hurley, Kevin L. Pope
Nebraska Cooperative Fish and Wildlife Research Unit: Staff Publications
1. Recreational fisheries are traditionally managed at local scales, but more effective management could be achieved using a cross-scale approach. To do this, we must first understand how local processes scale up to influence landscape patterns between anglers and resources. We highlight how population-based synchrony methods, used in conjunction with a complex-adaptive-systems framework, can reveal emergent spatial properties within social-ecological systems such as recreational fisheries.
2. Herein, we quantified the level of spatial synchrony in angler behaviour, defined the relationship between angler synchrony and distance among waterbodies, and highlighted social-ecological attributes contributing to these patterns. We leveraged a 111 waterbody-year …
