Transposon Mutagenesis In Mycobacterium Avium Subspecies Paratuberculosis, John P. Bannantine, Denise K. Zinniel, Raul G. Barletta
School of Veterinary and Biomedical Sciences: Faculty Publications
While transposon mutagenesis has been developed for Mycobacterium avium subspecies paratuberculosis (Map), relatively few laboratories have adopted this important genetic tool to examine gene function and essentiality. Here we describe the construction of a Map transposon library using the Himar1 mariner transposon, but concepts can also be applied to the Tn5367 transposon, which has also been used by our group. Delivery of the transposon is by a temperature-sensitive phagemid, ϕMycoMarT7, and plating transductants requires patience and specialized media due to length of incubation required to observe colonies. Several transposon mutants obtained from these libraries have been tested in vaccine and …

Establishing Enhanced Learning Outcomes In Science For First Generational Undergraduate College Students, Marianna Burks
Establishing Enhanced Learning Outcomes In Science For First Generational Undergraduate College Students, Marianna Burks
Department of Teaching, Learning, and Teacher Education: Theses and Other Student Research
First-generation students represent an increasing percentage of the demographic for college and university enrollment at both the undergraduate and graduate levels. Since 2001, results from the National Center for Education Statistics Longitudinal Study, indicates ‘higher percentages of first-generations college students attended public postsecondary institutions-76 vs. 72%’ (Redford & Hoyer, 2017). There is insufficient research, however, concerning how first-generation students’ needs are different, even unique, from those students who are second- generation and beyond attending college (National Center for Educational Statistics, 2017). Since this increase of students enrolling in post-secondary education, ‘understanding the needs of first-generation students is imperative more now …

Gaindroid: General Automated Incompatibility Notifier For Android Applications, Bruno Vieira Resende E Silva
School of Computing: Dissertations, Theses, and Student Research
With the ever-increasing popularity of mobile devices over the last decade, mobile apps and the frameworks upon which they are built frequently change. This rapid evolution leads to a confusing jumble of devices and applications utilizing differing features even within the same framework. For Android apps and devices, representing over 80% of the market share, mismatches between the version of the Android operating system installed on a device and the version of the app installed, can lead to several run-time crashes, providing a poor user experience.
This thesis presents GAINDroid, an analysis approach, backed with a classloader based program analyzer, …
Simulation Of Antimicrobial Resistance Gene Fate In Narrow Grass Hedges, Marzieh Khedmati
Simulation Of Antimicrobial Resistance Gene Fate In Narrow Grass Hedges, Marzieh Khedmati
Department of Civil and Environmental Engineering: Dissertations, Theses, and Student Research
Vegetative Filter Strips (VFS) are used for controlling the volume of runoff and decreasing the contaminants in runoff before entering the water bodies. Many studies investigated the role of VFS in sediment and nutrient removal, but little is known about their efficiency in the removal of emerging contaminants such as antimicrobial resistance genes (ARGs). VFSMOD was used to simulate the efficiency of VFS in this regard. The objectives of this study were to calibrate the VFSMOD with some experimental data and asses the efficiency of the model in simulating the filter behavior in removing ARGs. The tests were conducted in …

College of Education and Human Sciences: Dissertations, Theses, and Student Research
In 2018, the Nobel Peace Prize was awarded to Nadia Murad, a Yazidi woman who captured the attention of the world with her story of survival from the combatant and abusive hands of ISIS. Murad used her voice to speak out against wartime sexual assault and in doing so, has supported the wellbeing of women and girls around the world. A Goodwill Ambassador, her example and work has visibilized their trail of courage and lifted up the importance of work to support their wellbeing. This research, in albeit a very small way, seeks to follow Murad’s path. Though literature abounds …
Defining, Measuring, And Applying Soil Quality: An Unresolved Debate, Salvador Ramirez Ii
Defining, Measuring, And Applying Soil Quality: An Unresolved Debate, Salvador Ramirez Ii
Doctor of Plant Health Program: Dissertations and Student Research
Paradigm shifts occur periodically in the sciences. Proposed paradigm shifts offer opportunities to reexamine extant paradigms and evaluate new ones. Soil science experienced a controversy resembling a proposed paradigm shift in the late 1990s and early 2000s when it grappled with the idea of soil quality (SQ). The most contentious points in the SQ debates were (1) the word “quality” itself and designing a functional definition, (2) identifying appropriate soil quality indicators, (3) the integration of soil quality indicators, and (4) the interpretation of soil quality indicators. The latter three disagreements were rooted in the established, fundamental, soil science paradigm. …
A Descriptive Study Of Wild Bees (Hymenoptera: Apoidea: Apiformes) And Angiosperms In A Tallgrass Prairie Corridor Of Southeastern Nebraska, Katie Lamke
Department of Entomology: Dissertations, Theses, and Student Research
The presence of diverse bee communities in an ecosystem is vital for maintaining healthy plant communities, promoting habitat resilience, and supporting sustainable agricultural production and urbanization. Approximately 20,000 known species of bees exist worldwide and assist with the successful reproduction of nearly 80% of Earth’s flowering plants by providing pollination services. In the US, wild bee declines have led to increased monitoring efforts for bees but there remain critical data gaps in prairies of the Great Plains ecoregion. Specific to the Tallgrass prairie where only 1-3% remains in native vegetation, the Nebraska Wildlife Action Plan has identified the loss of …
Encapsulation Of Astaxanthin-Enriched Camelina Seed Oil Obtained By Ethanol-Modified Supercritical Carbon Dioxide Extraction, Liyang Xie
Department of Food Science and Technology: Dissertations, Theses, and Student Research
Astaxanthin is a high-value carotenoid widely used in the food, feed, nutraceutical, and pharmaceutical industries. Natural sources of astaxanthin are preferable to synthetic sources. However, current sources of natural astaxanthin are limited and fall short of global demand. In addition, effective extraction and stabilization strategies are needed before the utilization of natural astaxanthin in food applications. This work investigated 1) the feasibility of ethanol-modified supercritical carbon dioxide (SC-CO2) extraction of astaxanthin from engineered camelina seed, a potential source of natural astaxanthin, and 2) the potential of an emulsion-based system to protect astaxanthin after extraction. The physical properties of the emulsion …
Density And Mass Effect On The Development Of Phormia Regina, Brandon H. Strauss
Density And Mass Effect On The Development Of Phormia Regina, Brandon H. Strauss
School of Natural Resources: Dissertations, Theses, and Student Research
Forensic entomology is the application of the study of arthropods to the criminal justice system. This is primarily done through the development of a post mortem interval (PMI) based the insect evidence present. A practitioner must be able to determine the age of the insect through temperature data. One factor influencing the temperature dependent development is gregarious behavior. Current literature describes a faster development rate due to an increase in feeding efficiency and temperatures produced by this aggregate. However, there is very little literature defining a minimum number needed to induce this effect and little to none on it for …
Detection Of Heteroplasmic Single Nucleotide Polymorphisms Using Melt Curve Analysis And Dual Labeled Fluorescent Probes, Emily Jezewski
Detection Of Heteroplasmic Single Nucleotide Polymorphisms Using Melt Curve Analysis And Dual Labeled Fluorescent Probes, Emily Jezewski
Honors Program: Senior Projects (Public)
Plant mitochondrial genomes are strange – they are unusually large, consist of huge amounts of non-coding DNA, and contain of several overlapping regions throughout the genome. The genome is made of several different sized linear and circular molecules and different mitochondria within a cell will have different pieces of the genome. Even if multiple mitochondria contain the same region of the genome, these sequences can differ by single nucleotide polymorphisms (SNPS). This is known as heteroplasmy. Heteroplasmy has been documented in Arabidopsis thaliana using whole-genome sequencing data. While heteroplasmy is well-documented, its existence in the mitochondrial genome is unexpected and …

Modeling And Economic Analysis Of A Crop–Livestock Production System Incorporating Cereal Rye As A Forage, Eric R. Coufal
Department of Agricultural Economics: Dissertations, Theses, and Student Research
This thesis consists of two chapters using agent-based modeling for a crop-livestock production system incorporating human labor. The first chapter examines the principles used to develop a fundamental simulation pertaining to grazing cereal rye (Secale cereal L.) with calves. Within the software guidelines, the base model has the ability to capture diverse system interactions between livestock/plants and land management with human labor efficiency. AnyLogic incorporates agent-based modeling while combining with discrete event modeling and system dynamics. The purpose of the model was to find the economic returns of grazing cover crops relative to the area of Mead, Nebraska. In …
Field Obstacle Identification For Autonomous Tractor Applications, Caleb Lindhorst
Field Obstacle Identification For Autonomous Tractor Applications, Caleb Lindhorst
Department of Agricultural and Biological Systems Engineering: Dissertations, Theses, and Student Research
New technologies are being developed to meet the growing demand for agricultural products. Autonomous tractors are one of the many solutions to address this demand. Obstacle detection and avoidance is an important consideration for safe operation of any autonomous machine. Three field obstacles were chosen to be identified in this thesis work: tractors, round bales, and center pivots. Limited research work was found on the identification of center pivot detection.
Feasibility of using low cost LIDARs was considered for the detection of tractors, bales, and agricultural center pivots. Performance of LIDARs in different lighting conditions, different colors of obstacles, accuracy …

Nutrient Management Suggestions For Corn, Charles A. Shapiro, Richard Ferguson, Charles S. Wortmann, Bijesh Maharjan, Brian T. Krienke
Department of Agronomy and Horticulture: Faculty Publications
Fertilizer nutrient requirements for corn are based on expected yield and soil nutrient availability. The preplant nitrogen (N) recommendation equation, with adjustment for fertilizer cost and time of application, is retained from the previous edition of this publication. Suggestions for in-season nitrogen decisions are briefly outlined. The major change is providing a phosphorus (P) recommendation based on yield history with an implied intent to build and maintain soil test P above the critical level, which has not changed.
Plant Mitochondrial Genome Evolution And Structure Has Been Shaped By Double-Strand Break Repair And Recombination, Emily Wynn
School of Biological Sciences: Dissertations, Theses, and Student Research
Plant mitochondrial genomes are large but contain a small number of genes. These genes have very low mutation rates, but genomes rearrange and expand at significant rates. We propose that much of the apparent complexity of plant mitochondrial genomes can be explained by the interactions of double-strand break repair, recombination, and selection. One possible explanation for the disparity between the low mutation rates of genes and the high divergence of non-genes is that synonymous mutations in genes are not truly neutral. In some species, rps14 has been duplicated in the nucleus, allowing the mitochondrial copy to become a pseudogene. By …

English abstract: Understanding the factors shaping the niche of parasites and its expression over geographical space and through time continues to be a modern scientific challenge with the results of research in this area directly influencing both theoretical and applied biology. This is especially important for proactive management of zoonotic parasites such as Echinococcus multilocularis, the etiologic agent of alveolar echinococcosis. Echinococcus multilocularis has a Holarctic distribution; with its geographic range and prevalence increasing recently in areas of the western Palearctic, while its distribution dynamics are poorly understood in the Nearctic. In this paper, we use an ecological niche …
Regulation Of Vaccinia Virus Replication: A Story Of Viral Mimicry And A Novel Antagonistic Relationship Between Vaccinia Kinase And Pseudokinase, Annabel T. Olson
Regulation Of Vaccinia Virus Replication: A Story Of Viral Mimicry And A Novel Antagonistic Relationship Between Vaccinia Kinase And Pseudokinase, Annabel T. Olson
School of Biological Sciences: Dissertations, Theses, and Student Research
Poxviruses employ sophisticated signaling pathways that thwart cellular defense mechanisms and simultaneously ensure viral factors are modulated properly. Yet, our understanding of these complex signaling networks are incomplete. For example, the vaccinia B1 kinase plays a vital role in inactivating the cellular antiviral factor BAF, and is suggested to orchestrate other pathways. B1 is highly conserved among poxviruses and exhibits a remarkable degree of similarity to VRKs, a family of cellular kinases, suggesting that the viral enzyme has evolved to mimic VRK activity. Indeed, B1 and VRKs have been demonstrated to target a shared substrate, the DNA binding protein BAF, …
The Role Of Inflammatory Pathways In Development, Growth, And Metabolism Of Skeletal Muscle In Iugr Offspring: Blood Gene Expression Of Inflammatory Factors As Novel Biomarkers For Assessing Stress And Wellbeing In Exotic Species, Robert J. Posont
Department of Animal Science: Dissertations, Theses, and Student Research
Our first study identified the effects of maternal inflammation-induced intrauterine growth restriction (MI-IUGR) on growth and muscle glucose metabolism in offspring supplemented with curcumin. MI-IUGR lambs exhibited asymmetric growth restriction at birth and 30d of age, but normal glucose-stimulated insulin secretion. Hindlimb glucose oxidation was reduced by MI-IUGR and not improved by curcumin supplementation. Ex vivo muscle glucose oxidation was reduced by MI-IUGR but improved somewhat by curcumin. These finding indicate that fetal programming responses to MI contribute to neonatal growth and metabolic deficits. Neonatal curcumin supplementation had minimal effect on growth deficits but may improve glucose metabolism. …
Examining The Capacity Of Nebraska Rangelands For Cattle Production And Evaluating Drought Management Strategies, Kaitlyn L. Cumming
Examining The Capacity Of Nebraska Rangelands For Cattle Production And Evaluating Drought Management Strategies, Kaitlyn L. Cumming
Department of Agricultural Economics: Dissertations, Theses, and Student Research
This thesis has two chapters focusing on the grazing capacity of Nebraska rangelands, and drought management strategies. The first chapter conducts a gap analysis comparing forage supplied by perennial grasslands to the animal unit months (AUMs) required by the cattle. The purpose of this research was to quantify potential AUM supply (i.e., carrying capacity) of grazing lands dominated by perennial grasses on a regional basis in Nebraska to the AUM demand based on cattle inventories and standard production practices in each region of Nebraska. The results suggest that Nebraska is operating at 100% of potential carrying capacity. Harvest efficiency for …
Malleable Perspectives, Erik White
Malleable Perspectives, Erik White
School of Art, Art History, and Design: Theses and Student Creative Work
I attentively paint hastily formed figures, symbols, objects, and scenes that were modeled with never-dry clay onto stretched canvases of various sizes. I paint the malleable character of the clay by depicting its bumps, marks and dents, which emphasizes the fragility, impermanence, and the physical construction of the forms. Those visual attributes serve as a metaphor for the social construction of the concepts within the chosen imagery. To create my images I build clay up and physically move it around—in a similar way, ideas and concepts get built up, manipulated, and changed over time. These paintings serve as a cultural …
Lithic Analysis Of An Early Archaic Assemblage On The Great Plains: The Spring Creek Site (25ft31), Andrea Elizabeth Kruse
Lithic Analysis Of An Early Archaic Assemblage On The Great Plains: The Spring Creek Site (25ft31), Andrea Elizabeth Kruse
Department of Anthropology: Theses and Student Research
Early Archaic sites on the Great Plains are few in number and often little studied and poorly reported, as they are almost always found via salvage or compliance archaeology. Of those Early Archaic sites that have been studied, rarely has the recovered debitage been analyzed in detail nor have tools been fully evaluated for use-wear. This thesis describes the lithic assemblage from the Spring Creek (25FT31) site located in southwestern Nebraska. As one of two important early sites in the state, detailed lithic analysis will complement the thorough analysis of faunal remains conducted in the 2000s. This thesis presents the …
The Search For Fort Lisa In The Vicinity Of Omaha, Nebraska: A Gis Site Location Model, Brian C. Goodrich
The Search For Fort Lisa In The Vicinity Of Omaha, Nebraska: A Gis Site Location Model, Brian C. Goodrich
Department of Anthropology: Theses and Student Research
Fort Lisa was one of several important Euro-American fur trade sites in the vicinity of what is today Omaha, Nebraska. It, along with the other sites on that stretch of the Missouri River, were key locations both for trade with local tribes and as waypoints for those travelling to northern tribes in the early 19th Century. With the decline of the fur trade era, most of the sites that were once so central to life on the Missouri were abandoned and lost to memory. Archaeologists have rediscovered many of the sites along the Missouri River, including Fort Clark and …

Using Virtual Reality And Remotely Sensed Data To Explore Object Identity And Embodiment In A Virtual Mayan City, Cole F. Juckette
Department of Anthropology: Theses and Student Research
3D visualization, LiDAR (Light Detection and Ranging), and 3D modeling are not new concepts in archaeology, however when combined they represent a growing body of research that seeks to understand both how these tools can help us to study the people of the past, and the past itself. Recently, archaeologists have been creating large amounts of 3D digital assets because of new and more advanced technologies. Along with these digital assets has come a myriad of single object viewers—both web and desktop based. These platforms specifically focus on visualizing individual objects (i.e., artifacts or buildings). In contrast, 3DGIS and Virtual …
