Heat stress occurring during rice (Oryza sativa) grain development reduces grain quality, which often manifests as increased grain chalkiness. Although the impact of heat stress on grain yield is well-studied, the genetic basis of rice grain quality under heat stress is less explored as quantifying grain quality is less tractable than grain yield. To address this, we used an image-based colorimetric assay (Red, R; and Green, G) for genome-wide association analysis to identify genetic loci underlying the phenotypic variation in rice grains exposed to heat stress. We found the R to G pixel ratio (RG) derived from mature …

Wheat streak mosaic virus (WSMV) is a mite-vectored virus with substantial economic impact on wheat production. One of the effective sources of resistance to WSMV, Wsm1, is carried on a translocation from Thinopyrum intermedium. The original whole arm form of this translocation (T4DL·4JsS) was highly effective against WSMV but carried a substantial yield penalty in the absence of the virus. Shorter recombinants of the Th. intermedium translocation are now available. This study characterized the agronomic performance of near-isogenic sib-pair families in 10 yield trials using one of these shortened T4DL·4DS-4JsS translocations, named “rec213,” in the absence …

Compared with most flowers where the showy part comprises specialized leaves (petals) directly subtending the reproductive structures, most Zingiberaceae species produce showy ‘‘flowers’’ through modifications of leaves (bracts) subtending the true flowers throughout an inflorescence. Curcuma alismatifolia, belonging to the Zingiberaceae family, a plant species originating from Southeast Asia, has become increasingly popular in the flower market worldwide because of its varied and esthetically pleasing bracts produced in different cultivars. Here, we present the chromosome-scale genome assembly of C. alismatifolia ‘‘Chiang Mai Pink’’ and explore the underlying mechanisms of bract pigmentation. Comparative genomic analysis revealed C. alismatifolia contains a …

Science is increasingly a collaborative pursuit. Although the modern scientific enterprise owes much to individuals working at the core of their field, humanity is increasingly confronted by highly complex problems that require the integration of a variety of disciplinary and methodological expertise. In 2016, the U.S. National Science Foundation launched an initiative prioritizing support for convergence research as a means of “solving vexing research problems, in particular, complex problems focusing on societal needs.” We discuss our understanding of the objectives of convergence research and describe in detail the conditions and processes likely to generate successful convergence research. We use our …

Comparative Analysis Of Untargetedmetabolomics In Tolerant And Sensitive Genotypes Of Common Bean (Phaseolus Vulgaris L.) Seeds Exposed To Terminal Drought Stress, Mayavan Subramani, Carlos Urrea, Venu Kalavacharla
Panhandle Research and Extension Center
Many environmental stresses can affect the accumulation of metabolites in plants, including drought. In the present study, we found a great deal of variability in the seed metabolic profiles of the tolerant (Matterhorn, SB-DT2 and SB-DT3) common bean genotypes in comparison to the sensitive genotypes (Sawtooth, Merlot and Stampede) using ultrahigh performance liquid chromatographytandem mass spectrometry (UPLC-MS). The genotypes were grown in the field and subjected to drought stress after flowering (terminal drought stress). We aimed to investigate the accumulation of genotype-specific metabolites and related pathways under terminal drought stress by comparing tolerant and sensitive genotypes within a race. A …

Harold W. Manter Laboratory of Parasitology: Library Materials
Morphological traits from blood stages have been the gold standard for determining haemosporidian parasite species. However, the status of some taxa and the value of such traits in parasites from reptiles remain contentious. The scarce sampling of these species worsens the situation, and several taxa lack molecular data. A survey was performed in the Magdalena Department in Colombia, where 16 species of reptiles were captured. A peculiar haemosporidian parasite was found in the Turnip-tailed gecko Thecadactylus rapicauda. This haemosporidian does not show malarial pigment in blood stages under light microscopy; thus, it fits the Garnia genus’s characters belonging to …

Nebraska Game and Parks Commission: Publications
The primary goal in the development of at-risk species conservation assessments is to compile biological and ecological information that may assist conservation practitioners in making decisions regarding the conservation of species of interest. The Nebraska Natural Legacy Project recognizes the timber rattlesnake (Crotalus horridus) as a Tier 1 at-risk species. Provided here are general management recommendations regarding timber rattlesnakes. Conservation practitioners will need to use their professional judgment for management decisions based on objectives, location, and site-specific conditions. Based on the body of literature and available data, this species conservation assessment provides an overview of our current knowledge of timber …
Glutathione And Glutaredoxin In Redox Regulation And Cell Signaling Of The Lens, Marjorie F. Lou
Glutathione And Glutaredoxin In Redox Regulation And Cell Signaling Of The Lens, Marjorie F. Lou
School of Veterinary and Biomedical Sciences: Faculty Publications
The ocular lens has a very high content of the antioxidant glutathione (GSH) and the enzymes that can recycle its oxidized form, glutathione disulfide (GSSG), for further use. It can be synthesized in the lens and, in part, transported from the neighboring anterior aqueous humor and posterior vitreous body. GSH is known to protect the thiols of the structural lens crystallin proteins from oxidation by reactive oxygen species (ROS) so the lens can maintain its transparency for proper visual function. Age-related lens opacity or senile cataract is the major visual impairment in the general population, and its cause is closely …

Landscape Change, Scale, And Human Response To Change In The Great Plains, Kate Bird
Landscape Change, Scale, And Human Response To Change In The Great Plains, Kate Bird
School of Natural Resources: Dissertations, Theses, and Student Research
Great Plains landscapes are undergoing changes at multiple spatial and temporal scales due to processes ranging from woody encroachment to climate change. These changes may fundamentally alter the agroecosystems of the Great Plains such that the provisioning of ecosystem services including biodiversity and livestock production is affected. Improving our understanding of the effects of landscape change at multiple scales and how humans perceive and respond to these changes is important for facilitating research and management that enhances the resilience of these agroecosystems. As such, I first applied discontinuity theory and graph theory to evaluate the functional connectivity of the Central …

Intestinal Parasite Prevalence In Fecal Samples In Nebraska Dog Parks, Kristianna Muller
Honors Program: Senior Projects (Public)
From February to April 2022, fecal samples were collected from eight dog parks in the Omaha and Lincoln, NE areas. A total of 160 samples were collected and assessed for the presence of intestinal parasites. The zinc sulfate double centrifugal flotation technique was used to determine infection. Out of the 160 samples, ten were positive for parasite contamination (6.25% prevalence). The most observed parasites were hookworms, found in 3.75% of all samples (6/160). Cystoisospora, Giardia, Toxocara canis, and Toxascaris leonina were all found only once, for a prevalence of 0.625% each. Compared to other studies, our infection …

The Role Of Intertemporal Preferences, Active Consideration Of Health Outcomes, And Simple Health Prompts On The Nutritional Quality Of Food Choices, Olivier Tuyizere
The Role Of Intertemporal Preferences, Active Consideration Of Health Outcomes, And Simple Health Prompts On The Nutritional Quality Of Food Choices, Olivier Tuyizere
Department of Agricultural Economics: Dissertations, Theses, and Student Research
This thesis consists of three chapters. The first chapter introduces the thesis by highlighting a brief review of intertemporal preferences, active consideration of health outcomes, and health prompts during food choices. The introduction paves the way for the following two chapters, which are related, but stand-alone papers.
In the second chapter, we explore a novel question: how does actively considering health outcomes (both current and future) during decision-making affect the nutritional quality of food choices? We explore this question with an online experiment on food choices. Our findings show that active consideration of health outcomes leads to choosing products with …

Engaging Children With Animals Increases Their Awareness Of Their Footprint On Earth, Brett Foster
Engaging Children With Animals Increases Their Awareness Of Their Footprint On Earth, Brett Foster
Honors Program: Senior Projects (Public)
Teachers should reserve time in their classrooms to connect curriculum to animals and environmental topics. Connecting core subjects like writing, reading, or even math to animals builds student investment in learning as they can connect the topics of animals to their lives. Animals are all around us and are a topic that students naturally spend a lot of time with and can easily identify in the world around them. There are several benefits of incorporating animals into the curriculum, including an increased sense of empathy, an awareness of their environmental impact, and an investment in their community.
I have focused …
Seasonal Habitat Selection By American White Pelicans, Frederick L. Cunningham, Guiming Wang, D. Tommy King
Seasonal Habitat Selection By American White Pelicans, Frederick L. Cunningham, Guiming Wang, D. Tommy King
United States Department of Agriculture Wildlife Services: Staff Publications
Resource utilization strategies of avian migrants are a major concern for conservation and management. Understanding seasonal habitat selection by migratory birds helps us explain the ongoing continental declines of migratory bird populations. Our objective was to compare the secondorder and third-order habitat selection by the American White Pelican (Pelecanus erythrorhynchos; hereafter pelican) between the breeding and non-breeding grounds. We tested the Lack hypothesis that habitat selection by migratory birds is stronger on the breeding grounds than on the nonbreeding grounds. We used random-effect Dirichlet-multinomial models to estimate the second-order habitat selection between the seasons with the GPS locations …

Effects Of Early‑Life Experience On Innovation And Problem‑Solving In Captive Coyotes, Andrew C. Garcia, Mitchell A. Parsons, Julie K. Young
United States Department of Agriculture Wildlife Services: Staff Publications
Early-life experience often shapes behaviors like innovation and exploration. These behaviors are important to animals encountering novel food resources in diverse habitats, such as mesocarnivores in urban areas. To understand if early-life experiences impact later-life behavior, we examined how coyotes (Canis latrans) responded to a multi-access puzzle box at two life stages: pup (~ 7 weeks) and dispersal (~ 10 months). We first exposed pups, still living with their parents and littermates, to a baited puzzle box. At dispersal age, we again tested both these pups and an age-matched control group that was not exposed to the puzzle …

Bovine coronavirus (BCoV) has spilled over to many species, including humans, where the host range variant coronavirus OC43 is endemic. The balance of the opposing activities of the surface spike (S) and hemagglutinin-esterase (HE) glycoproteins controls BCoV avidity, which is critical for interspecies transmission and host adaptation. Here, 78 genomes were sequenced directly from clinical samples collected between 2013 and 2022 from cattle in 12 states, primarily in the Midwestern U.S. Relatively little genetic diversity was observed, with genomes having >98% nucleotide identity. Eleven isolates collected between 2020 and 2022 from four states (Nebraska, Colorado, California, and Wisconsin) contained a …

Microbiomes often form specific functional associations with their hosts. Correlations between microbiome membership and states of host health and disease abound in many systems. However, there are few systems that allow for in depth functional studies that include precise manipulation and interrogation of both microbiome composition and host function. Recently the nematode Caenorhabditis elegans - an excellent genetic model organism for studying many fields of biology, including neurobiology and behavior, development, cell biology, and innate immunity - has proven to be a robust system to probe microbiome interactions and their effect on host physiology.
