Impacts Of Trogocytosis-Mediated Intracellular Signaling On Cd4+ T Cell Effector Cytokine Production And Differentiation, Steven James Reed Jan 2019

Impacts Of Trogocytosis-Mediated Intracellular Signaling On Cd4+ T Cell Effector Cytokine Production And Differentiation, Steven James Reed

Graduate Student Theses, Dissertations, & Professional Papers

Trogocytosis is the direct intercellular transfer of membrane and membrane associated molecules. Unlike other passive-membrane transfer events, trogocytosed molecules may remain fully functional and become re-expressed on the surface of the trogocytosis-positive (trog+) recipient. This phenomenon commonly occurs between various cell types, including those of the immune system. CD4+ T cell trogocytosis occurs during their activation by antigen presenting cells (APC). Consequently, the acquired molecules include ligands for signaling receptors on the T cell.

The impacts of CD4+ trogocytosis on the immune response are largely unknown. While it has been demonstrated that trog+ cells can present trogocytosed peptide:MHC (pMHC), and …

Prevention Of Crystalline Silica-Induced Inflammation By The Anti-Malarial Hydroxychloroquine, Rachel Dawn Burmeister Jan 2019

Prevention Of Crystalline Silica-Induced Inflammation By The Anti-Malarial Hydroxychloroquine, Rachel Dawn Burmeister

Graduate Student Theses, Dissertations, & Professional Papers

Exposure to inhaled crystalline silica (cSiO2) is common in occupations where there is cutting, milling, or grinding of cSiO2 containing material. The Occupational Safety and Health Administration estimates that over 2 million workers may be exposed to inhaled cSiO2 in the United States. Inhalation of cSiO2 causes acute and chronic inflammation and may lead pulmonary diseases such as silicosis, as well as an increased risk of developing autoimmune diseases.Unfortunately, treatment of cSiO2-induced lung diseases is limited and primarily focused on supportive care.

Inflammation caused by cSiO2 begins when, cSiO2 particles are phagocytized by alveolar macrophages. Interaction between the particle and …

State Regulated Relationships: Mothers' Experiences Of Partner Incarceration, Hannah Brianne Fields Jan 2019

State Regulated Relationships: Mothers' Experiences Of Partner Incarceration, Hannah Brianne Fields

Graduate Student Theses, Dissertations, & Professional Papers

The effects of incarceration on families have been studied in-depth, but little research evaluates the effects on women parenting children after the incarceration of their romantic partner. This research evaluates how mothers manage to keep their families intact throughout the duration of their partner’s incarceration. I approached this question using a geography theory of care developed by Sophie Bowlby and Linda McKie. This theory states that the quality of care is dependent on the space in which it is provided, the social expectations within the caring environment, and the amount of time required to provide or receive care. Using this …

Statistical Modeling Of Influenza-Like-Illness In Montana Using Spatial And Temporal Methods, Benjamin A. Stark Jan 2019

Statistical Modeling Of Influenza-Like-Illness In Montana Using Spatial And Temporal Methods, Benjamin A. Stark

Graduate Student Theses, Dissertations, & Professional Papers

Studying air pollution and public health has been a historically important question in science. It has long been hypothesized that severe air pollution conditions lead to negative implications in basic human health. Primarily, areas thats are prone to severe degrees of human pollution are the focus of such studies. Such research relating to less populated areas are scarce, and this scarcity raises the question of how such pollution dynamics (human-made and natural) influence human health in more rural areas.

The aim of this study is to explore this hole in research; in particular we explore possible links between air pollution …

Measuring Transmissibility Of Sin Nombre Hantavirus In Deermice, Angela Luis May 2018

Measuring Transmissibility Of Sin Nombre Hantavirus In Deermice, Angela Luis

University Grant Program Reports

The purpose of this UGP proposal was to develop methods to measure transmissibility of SNV in deermice. Transmissibility is the probability of transmission of the virus to a susceptible mouse, given contact with an infected mouse, and is a function of each animal’s physiological stress level and immunity. If an animal is more stressed, its immunity will be depressed, which will increase susceptibility (of uninfected mice) and infectiousness (of infected mice). Therefore, in order to measure transmissibility, we need to develop methods to measure stress levels, immune function, and viral loads in deermice. With this UGP seed grant, we developed …

Chasing Plenty: A Documentary On Food Security In Montana, Kurt Swimley, Colin Brust, Aspen Anderson, John Potenberg, Nathaniel Smith

University of Montana Conference on Undergraduate Research (UMCUR)

Chasing Plenty: A Documentary on Food Security in Montana Our GLI group aims to examine the following question: how will climate change impact food security? To address this question we are conducting interviews in Montana. Despite being an agricultural state, many Montanans struggle with food insecurity, and are left without plentiful or healthy options for food. We aim to research this issue by interviewing local community members involved in food distribution. The interview data will be compiled in a short documentary discussing current issues in food security, how these problems are likely to evolve in the future, and propose solutions …

Polystyrene Surface-Absorbed Trehalose Diester As A Means For Performing Pulldown Assays, Alexander S. Riffey, Roman Schoener, Robert Child

University of Montana Conference on Undergraduate Research (UMCUR)

In 2016, over 10.4 million people contracted Tuberculosis (TB) infections for the first time, resulting in the deaths of 1.7 million people worldwide. Despite the existence of a vaccine, TB remains the number one cause of death from a single infectious disease according to the 2017 report from the World Health Organization. As a result, research and development of a new TB vaccine is ongoing, as it is suspected the currently available formulation does not contain an adjuvant which produces the necessary TH-17 response. In the search for new drugs to combat this epidemic, knowledge of how a compound triggers …

Streamlined Preparation Of Genomic Dna In Agarose Plugs For Pulsed-Field Gel Electrophoresis, Linda D. Hicks, Charlotte M. Van Der Graaf, Jacob Childress, Emily Cook, Karen Schmidt, Frank Rosenzweig, Eugene Kroll Mar 2018

Streamlined Preparation Of Genomic Dna In Agarose Plugs For Pulsed-Field Gel Electrophoresis, Linda D. Hicks, Charlotte M. Van Der Graaf, Jacob Childress, Emily Cook, Karen Schmidt, Frank Rosenzweig, Eugene Kroll

Biological Sciences Faculty Publications

Genome analysis using pulsed-field gel electrophoresis (PFGE) has been used in applications ranging from typing bacterial strains to radiobiology to cancer research. While methods for running PFGE have been significantly improved since its invention, the method for preparing chromosomal DNA itself has remained essentially unchanged. This limits the applicability of PFGE, especially when analyses require many samples. We have streamlined sample preparation for routine applications of PFGE through the use of deep-well 48-well plates. Besides saving time, our protocol has the added advantage of reducing the volume of expensive reagents. Our improved protocol enables us to reduce throughput time and …

Identification Of Novel Mites (Miniature Inverted-Repeat Transposable Elements) In Coxiella Burnetii: Implications For Protein And Small Rna Evolution, Shaun Wachter, Rahul Raghavan, Jenny Wachter, Michael F. Minnick Jan 2018

Identification Of Novel Mites (Miniature Inverted-Repeat Transposable Elements) In Coxiella Burnetii: Implications For Protein And Small Rna Evolution, Shaun Wachter, Rahul Raghavan, Jenny Wachter, Michael F. Minnick

Biological Sciences Faculty Publications

Background: Coxiella burnetii is a Gram-negative gammaproteobacterium and zoonotic agent of Q fever. C. burnetii’s genome contains an abundance of pseudogenes and numerous selfish genetic elements. MITEs (miniature invertedrepeat transposable elements) are non-autonomous transposons that occur in all domains of life and are thought to be insertion sequences (ISs) that have lost their transposase function. Like most transposable elements (TEs), MITEs are thought to play an active role in evolution by altering gene function and expression through insertion and deletion activities. However, information regarding bacterial MITEs is limited.

Results: We describe two MITE families discovered during research on small non-coding …

An Interdisciplinary Approach To The Target Elucidation Of Novel Antibiotic 31g12, Larissa A. Walker Jan 2018

An Interdisciplinary Approach To The Target Elucidation Of Novel Antibiotic 31g12, Larissa A. Walker

Graduate Student Theses, Dissertations, & Professional Papers

Staphylococcus aureus is a Gram-positive bacterial pathogen responsible for nosocomial and community-acquired infections that can quickly acquire antibiotic resistance. We have identified a novel triazole antimicrobial 31G12 based on the natural product core of nonactin isolated from the fermentation of Streptomyces griseus, that is active against many Gram-positive bacteria as well as antibiotic resistant methicillin-resistant S. aureus and vancomycin-resistant Enterococcus. The synthesis and characterization indicate that 31G12 exists as a mixture of two rotamers at room temperature and displays bacteriostatic activity against S. aureus with moderate mammalian cell toxicity. We have currently identified potential protein targets of 31G12 in …

Examining Injury Data Reporting Practices Among Wildland Firefighters, Erin M. Boggs Jan 2018

Examining Injury Data Reporting Practices Among Wildland Firefighters, Erin M. Boggs

Graduate Student Theses, Dissertations, & Professional Papers

Wildland Firefighter (WLFF) Crews lack an injury surveillance program to collect information on non-traumatic and traumatic injuries. Tactical athletes including: police, military and structural firefighters all have a variety of collection systems in place for injury data collection. The lack of injury data prevents WLFF crews from identifying high-risk environments, implementing injury prevention programs, and improving the overall quality of care on the job. Over the last several decades the recent rise in wildfire occurrences and wildfire suppression costs creates a need for healthy and strong individuals in the field. The cost of firefighter salaries and medical care can be …

Ecology, Epidemiology, And Evolutionary Genetics Of Canine Distemper Virus Spillover In African Lions, Julie Kay Weckworth Jan 2018

Ecology, Epidemiology, And Evolutionary Genetics Of Canine Distemper Virus Spillover In African Lions, Julie Kay Weckworth

Graduate Student Theses, Dissertations, & Professional Papers

The impact of emerging infectious diseases (EIDs) on the health and persistence of wildlife populations is an increasing conservation concern. Large carnivores are particularly vulnerable to EID impacts because they often occur in small, isolated populations with demographic and genetic challenges to long-term persistence. Ecological forces that isolate carnivore populations, e.g. agricultural intensification, simultaneously increase the probability of disease exposure from domestic species and can amplify population susceptibility to infection.

Canine distemper virus emerged as a conservation threat to African lions when an explosive epizootic caused the death or disappearance of a third of the Serengeti lion population in 1994. …

Bottom-Up And Top-Down Controls On Food Webs In Headwater Streams, Miriam O. Bayer Jan 2018

Bottom-Up And Top-Down Controls On Food Webs In Headwater Streams, Miriam O. Bayer

Graduate Student Theses, Dissertations, & Professional Papers

Headwater streams account for 70% of stream channel length in the USA and are important as hotspots of nutrient uptake and native biodiversity. Biofilm, the mixed auto- and heterotrophic microbial community covering stream substrates, is where the majority of nutrient processing occurs, and forms the base of stream food webs, particularly in heavily shaded, oligotrophic streams. Both bottom-up (e.g., nutrients, light) and top-down (i.e., consumption) processes are known to affect periphyton, the autotrophic component of biofilm, but little is known about what controls the biofilm community as a whole. Top-down effects are common in streams, where fish are often the …
