Tumor-associated macrophages (TAMs) play a substantial role in sculpting the tumor microenvironment (TME) into an immunosuppressive bastion that undermines immune surveillance and enables tumor progression. They therefore represent a promising therapeutic target to make the TME more conducive for anti-tumor immunity and greatly improve treatment outcomes in cancers that are highly resistant to conventional forms of therapy. Several medications have been developed to modulate the activities of TAMs, showing tremendous potential, but these medications lack specificity for TAMs making their use susceptible to adverse side effects that could outweigh prospective benefits. Members of the Bcl-2 family are responsible for regulating the intrinsic pathway of apoptosis through Bcl-2 homology 3 (BH3) domain interactions of pro- and anti-apoptotic members. Apoptosis occurs when BH3-only pro-death members overwhelm the pro-survival ones, leading to oligomerization of the multi-BH domain pro-death protein Bax to signal death by releasing cytochrome c from mitochondria into the cytoplasm. This process is essential for proper embryonic tissue development as well as maintenance of mature tissues. While BH3-only proteins such as Bim and Bid bind to and activate Bax through their BH3 domains, other regulators of this mitochondrial permeabilizer may exist. Two …

Caulobacter crescentus is a model bacterium for its unusual life cycle. This cycle features three distinct phases- the stalked stage, pre-divisional or actively dividing stage, and the swarmer stage, characterized by the production of a flagellum and multiple pili. Pili are necessary for cell attachment to surfaces and their reuptake into the cell can help drive the cell cycle forward. The gene that encodes the pilus subunit, pilA, is regulated by four binding sites for the cell cycle regulator CtrA. The Single Frequency Acoustic Attenuation System (SFAAS) has been developed at the National Center for Physical Acoustics (NCPA). This system uses acoustic methods to monitor fine suspended sediment concentration in streams. Site-specific calibrations are necessary for this system, and previous research has indicated that algal concentration or dissolved oxygen concentration of water may affect collected data. Temperature, dissolved oxygen concentration, and fluorescence, used as a proxy for algal concentration, were monitored during this system’s use, and calibrations were derived to account for the effect of each of these variables on acoustic measurements. Drag queens are the most indicative symbols of this boom, traditional practitioners of gender-bending performances that blend music, dance, makeup, and fashion to create breathtaking performances. To this end, this thesis asks the following question: How is local drag performance culture generated in Rio de Janeiro in the context of this 21st century gay boom? There is little comparison between the two of which works: the threat of damnation through vice, or the promise of salvation through virtue. This study examines how each of these affect donation behavior. There are two studies within the present research, each with 149 participants randomly assigned into three categories: control, vice, and virtue. Biogeographic Patterns In Phyllosphere Bacterial Communities On Southern Magnolia Trees In A Small Town, Elizabeth M. Eckard, Venkat K. Kota, Kurre T. Luber

The phyllosphere, consisting of aerial plant surfaces colonized by microorganisms, remains a relatively understudied ecosystem in terms of bacterial biogeography, particularly at intermediate spatial scales. This study characterized the bacterial community on the leaves of 89 Magnolia grandiflora trees throughout a small town, encompassing an area of approximately 60 km2. Sequencing of the 16S ribosomal RNA gene revealed the dominant bacterial phyla to be Alphaproteobacteria, Bacteroidetes, and Acidobacteria, consistent with other studies of the phyllosphere. Moral convictions have been shown to impact learning about science topics, but there exists a gap in literature about whether education on the mechanisms of DNA technologies, such as the recently developed CRISPR-Cas9 system, can affect moral convictions and emotions regarding its use on humans. To investigate this relationship, we examined the results of how three-hundred and fifty-three introductory psychology students at the University of Mississippi rated their moral convictions and emotions on the subject before and after engaging with instructional material. The red alga Botryocladia occidentalis presents three distinct fractions of sulfated polysaccharides. The elucidation of structures of various B. occidentalis fractions has been explored in prior research. In this study, the first fraction with the least sulfation underwent NMR structure elucidation. Additionally, this sulfated polysaccharide underwent analysis following chemical modifications. Findings have unveiled the presence of both 4-linked α- and 3-linked β-galactose units with the following chemical characteristics: over half of the total galactose units are unsulfated, the α-units are primarily 3,6-anhydrogalactose units either 2-O-methylated or 2-O-sulfated, and the β-galactose units can be 4-O-sulfated or 2,4-O-disulfated. Cyanobacteria are an abundant and diverse group of photosynthetic prokaryotes with a long evolutionary history, which presents challenges for comprehensive taxonomic classification. Phylogenies were traditionally constructed based on the highly conserved 16S rRNA gene, but research has increasingly relied upon whole genome sequencing to elucidate evolutionary relationships, despite increased cost and time. In this thesis, publicly available genome sequences of various cyanobacterial strains were utilized to determine if the whole rRNA region, which includes the 16S, 23S, and 5S genes as well as the spacer regions between, could provide accurate, yet cost/time efficient representations of their evolutionary relationships. Self Determination Theory (SDT) theorizes that when three core psychological needs are met (i.e., competence, relatedness, and autonomy) then humans reach their highest level of optimization (Deci & Ryan, 2000). One common way people tend to satisfy these needs is through social relationships, typically ones where their efforts can be reciprocated; however, not all relationships are reciprocal. Functional alcohol tolerance is an adaptation made by the nervous system to high doses of alcohol, and is seen by a significant reduction in sensitivity to this drug. This tolerance is a significant contributing factor in the progression to Alcohol Use Disorders. A major goal of alcohol research is to understand how alcohol induces this tolerance. Dunc13 is a presynaptic active zone protein essential in synaptic vesicle docking and priming. Alcohol binds to the C1 domain of Dunc13 at physiological concentrations, which inhibits Dunc13 from binding Diacylglycerol. Treatment-resistant depression (TRD) is a clinical diagnosis for patients suffering from major depressive disorder (MDD). An inadequate response to the dosage or duration of antidepressant medication can lead to the diagnosis of TRD. This diagnosis is common in up to 50% of patients with MDD following treatment. Microdosing of classical psychedelics such as lysergic acid diethylamide (LSD) has been shown to have positive cognitive and therapeutic effects on patients with TRD. This thesis examines the linguistic representation of African peoples and cultures in French popular culture, specifically as this pertains to immigration. The foundational research question of this project is: how has the representation of Africans in French popular culture evolved since the colonial period? Despite the opioid crisis taking headlines, the abuse of cocaine and amphetamines remains rampant and laboratory instruments must be able to effectively and reliably handle the overwhelming case load. In this study, an Agilent 6475 Triple Quadrupole (QQQ) LCMS, recently acquired by the Tarrant County Medical Examiner’s Office (TCMEO) in Fort Worth, Texas, was validated for identifying and quantifying CocAmps. Results show that the instrument was fit-for-purpose and that the instrument passed all validation tests. Reproducibility was ≤ 20% RSD for 12 QC replicates, 6 of blood and 6 of urine. Acute exercise has been shown to differentially impact memory, dependent on whether the task is considered prefrontal cortex- or hippocampal-dependent. More specifically, acute exercise has been shown to impair PFC-dependent memory while enhancing hippocampal-dependent memory. No study has compared whether exercise differentially impacts separate types of cognition, so this experiment was designed to determine the relationship between high-intensity acute exercise and both prefrontal cortex-dependent and hippocampal-dependent memory. This study consisted of five total visits for 55 participants. In the first visit, participants engaged in a maximal exercise test, where their heart rate reserve was obtained.
