The Asian clam Corbicula fluminea (Family: Cyneridae) has aggressively invaded freshwater habitats worldwide, resulting in dramatic ecological changes and declines of native bivalves such as freshwater mussels (Family: Unionidae), one of the most imperiled faunal groups. Despite increases in our knowledge of invasive C. fluminea biology, little is known of how intrinsic and extrinsic factors, including co-occurring native species, influence its microbiome. We investigated the gut bacterial microbiome across genetically differentiated populations of C. fluminea in the Tennessee and Mobile River Basins in the Southeastern United States and compared them to those of six co-occurring species of native freshwater mussels. …

Vitamin E acetate, which is used as a diluent of tetrahydrocannabinol (THC), has been reported as the primary causative agent of e-cigarette, or vaping, product use-associated lung injury (EVALI). Here, we employ in vitro assays, docking, and molecular dynamics (MD) computer simulations to investigate the interaction of vitamin E with the membrane-bound cannabinoid 2 receptor (CB2R), and its role in modulating the binding affinity of THC to CB2R. From the MD simulations, we determined that vitamin E interacts with both CB2R and membrane phospholipids. Notably, the synchronized effect of these interactions likely facilitates vitamin E acting as a lipid modulator …

Docosanol is the only US Food and Drug Administration (FDA) approved over-the-counter topical product for treating recurrent oral-facial herpes simplex labialis. Validated analytical methods for docosanol are required to demonstrate the bioequivalence of docosanol topical products. A gas chromatography/selected ion monitoring mode mass spectrometry (GC/SIM-MS) method was developed and validated for docosanol determination in biological samples. Docosanol and isopropyl palmitate (internal standard) were separated on a high-polarity GC capillary column with (88% cyanopropy)aryl-polysiloxane employed as the stationary phase. The ions of m/z 83 and 256 were selected to monitor docosanol and isopropyl palmitate, respectively; the total run time was 20 …

Oxygen levels in vivo are autonomously regulated by a supply–demand balance, which can be altered in disease states. However, the oxygen levels of in vitro cell culture systems, particularly microscale cell culture, are typically dominated by either supply or demand. Further, the oxygen microenvironment in these systems is rarely monitored or reported. Here, a method to establish and dynamically monitor autonomously regulated oxygen microenvironments (AROM) using an oil overlay in an open microscale cell culture system is presented. Using this method, the oxygen microenvironment is dynamically regulated via the supply–demand balance of the system. Numerical simulation and experimental validation of …

The use of pulse wave analysis may assist cardiologists in diagnosing patients with vascular diseases. However, it is not common in clinical practice to interpret and analyze pulse wave data and utilize them to detect the abnormalities of the signal. This paper presents a novel approach to the clinical application of pulse waveform analysis using the wavelet technique by decomposing the normal and pathology signal into many levels. The discrete wavelet transform (DWT) decomposes the carotid arterial pulse wave (CAPW) signal, and the continuous wavelet transform (CWT) creates images of the decomposed signal. The wavelet analysis technique in this work …

Enid Lake is one of the largest reservoirs located in Yazoo River Basin, the largest basin in the state of Mississippi. The lake was impounded by Enid Dam on the Yocona River in Yalobusha County and covers an area of 30 square kilometers. It provides significant natural and recreational resources. The soils in this region are highly erodible, resulting in a large amount of fine-grained cohesive sediment discharged into the lake. In this study, a 3D numerical model was developed to simulate the free surface hydrodynamics and transportation of cohesive sediment with a median diameter of 0.0025 to 0.003 mm …

Synthesis Of Alpha Domain Of Metallothionein For Hydrogen Production, Aditya Surakanti
Honors Theses
The world relies on nonrenewable resources for a significant portion of its usable energy, the most common being fossil fuels. Fossil fuels release many unwanted byproducts during their extractions and conversion processes. As a result, this excessive use leads to harsh consequences upon the environment, including rapid global warming, release of numerous water and air pollutants, and it could possibly lead to an increased risk of death in those who are exposed to these toxins. In response to this scenario, the goal of my project is to design an artificial hydrogen catalyst, which can absorb sunlight and oxidize H2O into …

Research on the microbiomes of animals has increased substantially within the past decades. More recently, microbial analyses of aquatic invertebrates have become of increased interest. The storage method used while collecting aquatic invertebrates has not been standardized throughout the scientific community, and the effects of common storage methods on the microbial composition of the organism is unknown. Using crayfish and dragonfly nymphs collected from a natural pond and crayfish maintained in an aquarium, the effects of two common storage methods, preserving in 95% ethanol and freezing at −20◦C, on the invertebrate bacterial microbiome was evaluated. We found that the bacterial …

Misophonia is a disorder of decreased tolerance to specific sounds or their associated stimuli that has been characterized using different language and methodologies. The absence of a common understanding or foundational definition of misophonia hinders progress in research to understand the disorder and develop effective treatments for individuals suffering from misophonia. From June 2020 through January 2021, the authors conducted a study to determine whether a committee of experts with diverse expertise related to misophonia could develop a consensus definition of misophonia. An expert committee used a modified Delphi method to evaluate candidate definitional statements that were identified through a …

Dairy cattle with a high milk yield are susceptible to many infectious diseases, such as mastitis. Subclinical mastitis (SCM) is the most prevalent form of mastitis that predominantly affects animal health, and causes adverse effects on the quality and quantity of milk. In dairy animals, subclinical mastitis often remains undetected, as no gross changes in udder characteristics are visible. In the present study, 135 Holstein Friesian dairy animals were selected and screened as healthy (n = 25) and mastitic (n = 110) based on diagnostic tests such as the California mastitis test, pH, electrical conductivity, and somatic cell count. Physalis angulata L. belongs to the family Solanaceae and is distributed throughout the tropical and subtropical regions. Physalis angulata leaf and fruit extracts were assessed for in vitro anticancer, antioxidant activity, and total phenolic and flavonoid content. The GC-MS technique investigated the chemical composition and structure of bioactive chemicals reported in extracts. The anticancer activity results revealed a decrease in the percentage of anticancer cells’ viability in a concentration-and time-dependent way. We also noticed morphological alterations in the cells, which we believe are related to Physalis angulata extracts. Under light microscopy, we observed that as the concentration of ethanolic extract …

Exploring Rural-Urban Differences In Polygenic Associations For Health Among Older Adults In The United States, Trent Davidson, Jason D. Boardman, Lori M. Hunter
Journal of Rural Social Sciences
A complex combination of genes and environment influence health and, as a result, both genes and environment can play a role in shaping health disparities. We consider distinctions in these influences across rural and urban settings, expanding upon work that shows lower genetic associations in rural compared to urban places by studying an older age group and examining more than the typical outcomes of alcohol/substance abuse. Using a sample of 14,994 adults from the 1992 through 2016 waves of the Health andRetirement Study, our results suggest genetic associations for BMI and heart conditions are significantly lower in rural compared to …

Aquilaria sinensis (Lour.) Spreng is known for its resinous secretion (agarwood), often secreted in defense against injuries. We investigated the effects of A. sinensis flower extract (AF) on peroxisome proliferator-activated receptors alpha and gamma (PPARα and PPARγ), liver X receptor (LXR), glucose uptake, and lipid accumulation (adipogenesis). Activation of PPARα, PPARγ and LXR was determined in hepatic (HepG2) cells by reporter gene assays. Glucose uptake was determined in differentiated muscle (C2C12) cells using 2-NBDG (2-deoxy-2-[(7-nitro-2,1,3-benzoxadiazol-4-yl)amino]-D-glucose). Adipogenesis was determined in adipocytes (3T3-L1 cells) by Oil red O staining. At a concentration of 50 µg/mL, AF caused 12.2-fold activation of PPARα and …
