© 2019 ABRF. Methionine oxidation plays a critical role in many processes of biologic and biomedical importance, including cellular redox responses and stability of protein pharmaceuticals. Bottom-up methods for analysis of methionine oxidation can suffer from incomplete sequence coverage, as well as an inability to readily detect correlated oxidation between 2 or more methionines. However, the methodology for quantifying protein oxidation in top-down analyses is lacking. Previous work has shown that electron transfer dissociation (ETD)–based tandem mass spectrometry (MS/MS) fragmentation offers accurate and precise quantification of amino acid oxidation in peptides, even in complex samples. However, the ability of ETD-based …

Physical Activity And Cognitive Function Among Older Adults With An Elevated Gamma Gap, Emily Frith, Paul D. Loprinzi

Faculty and Student Publications

© 2018 The Author(s) Published by S. Karger AG, Basel. Objective: An elevated gamma gap is indicative of high serum concentrations of globulin proteins, some of which elicit acute inflammatory responses. An impaired cognitive function has been linked to central and peripheral inflammation, while exercise is associated with protective, anti-inflammatory benefits. In this study, we evaluated whether the gamma gap is associated with cognitive function among older adults and whether physical activity is favorably associated with cognitive function among those with an elevated gamma gap. Materials and Methods: Data from the 1999-2002 National Health and Nutrition Examination Survey (NHANES) were …

© 2019 Amend et al. Terrestrial fungi play critical roles in nutrient cycling and food webs and can shape macroorganism communities as parasites and mutualists. Although estimates for the number of fungal species on the planet range from 1.5 to over 5 million, likely fewer than 10% of fungi have been identified so far. To date, a relatively small percentage of described species are associated with marine environments, with ~1,100 species retrieved exclusively from the marine environment. Nevertheless, fungi have been found in nearly every marine habitat explored, from the surface of the ocean to kilometers below ocean sediments. Fungi …

© 2019 American Chemical Society and American Society of Pharmacognosy. Cranberry (Vaccinium macrocarpon) juice is traditionally used for the prevention of urinary tract infections. Human urine produced after cranberry juice consumption can prevent Escherichia coli adhesion, but the antiadhesive urinary metabolites responsible have not been conclusively identified. Adult female sows were therefore fed spray-dried cranberry powder (5 g/kg/day), and urine was collected via catheter. Urine fractions were tested for antiadhesion activity using a human red blood cell (A+) anti-hemagglutination assay with uropathogenic P-fimbriated E. coli. Components were isolated from fractions of interest using Sephadex LH-20 gel filtration chromatography followed by …

Predator Diet Affects Perceived Oviposition Site Quality For Mosquitoes, Brandon Mcdaniel Feb 2019

Predator Diet Affects Perceived Oviposition Site Quality For Mosquitoes, Brandon Mcdaniel

Honors Theses

Ovipositing freshwater organisms rely primarily on cues to determine site quality (Silberbush and Blaustein 2008) and to select an ideal habitat for their offspring (Resetarits 2014). Characteristics of high quality oviposition sites that yield the greatest fitness for offspring include the absence of predators and high resource availability that allows for optimal growth and development (Kershenbaum et al. 2012). It has been shown that predator presence is a large deterrent of oviposition due to threats of offspring survival, however, there is a little information about the predatory cues mosquitoes experience before ovipositing (Angelon and Petranka 2002). One hypothesis is that …

Purpose: Δ9-Tetrahydrocannabinol-valine-hemisuccinate, a hydrophilic prodrug of Δ9-tetrahydrocannabinol, synthesized with the aim of improving the ocular bioavailability of the parent molecule, was investigated in a lipid-based nanoparticle dosage form for ocular delivery. Methods: Solid lipid nanoparticles (SLNs) of Δ9-tetrahydrocannabinol-valine-hemisuccinate and Δ9-tetrahydrocannabinol, along with a nanoemulsion of Δ9-tetrahydrocannabinol-valine-hemisuccinate, were tested for glaucoma management in a normotensive rabbit model by using a multiple-dosing protocol. Marketed formulations of timolol maleate and pilocarpine HCl were also tested for their pharmacodynamic profile, post-single dose administration. Results: A peak intraocular pressure (IOP) drop of 30% from baseline was observed in rabbits treated with …

Malaria is a major global health threat, with nearly half the world's population at risk of infection. Given the recently described delayed clearance of parasites by artemisinincombined therapies, new antimalarials are needed to facilitate the global effort toward elimination and eradication. NPC1161 is an 8-aminoquinoline that is derived from primaquine with an improved therapeutic profile compared to the parent compound. The (R)-(-) enantiomer (NPC1161B) has a lower effective dose that results in decreased toxic side effects such as hemolysis compared to the (S)-(+)-enantiomer, making it a promising compound …

© 2019 by the authors. Licensee MDPI, Basel, Switzerland. Prostate cancer (PC) is one of the most common cancers in men. The global burden of this disease is rising. Its incidence and mortality rates are higher in African American (AA) men compared to white men and other ethnic groups. The treatment decisions for PC are based exclusively on histological architecture, prostate-specific antigen (PSA) levels, and local disease state. Despite advances in screening for and early detection of PC, a large percentage of men continue to be diagnosed with metastatic disease including about 20% of men affected with a high mortality …

© 2018, The Author(s), under exclusive licence to Springer Nature Limited. Giant tortoises are among the longest-lived vertebrate animals and, as such, provide an excellent model to study traits like longevity and age-related diseases. However, genomic and molecular evolutionary information on giant tortoises is scarce. Here, we describe a global analysis of the genomes of Lonesome George—the iconic last member of Chelonoidis abingdonii—and the Aldabra giant tortoise (Aldabrachelys gigantea). Comparison of these genomes with those of related species, using both unsupervised and supervised analyses, led us to detect lineage-specific variants affecting DNA repair genes, inflammatory mediators and genes related to …

© 2019 John Wiley & Sons Ltd Aim: Mesophotic coral ecosystems (MCEs) are unique communities that support a high proportion of depth-endemic species distinct from shallow-water coral reefs. However, there is currently little consensus on the boundaries between shallow and mesophotic coral reefs and between upper versus lower MCEs because studies of these communities are often site specific. Here, we examine the ecological evidence for community breaks, defined here as species loss, in fish and benthic taxa between shallow reefs and MCEs globally. Location: Global MCEs. Time period: 1973–2017. Major taxa studied: Macrophytes, Porifera, Scleractinia, Hydrozoa, Octocorallia, Antipatharia and teleost …

The goal of this study was to investigate the potential for a cannabidiol-rich cannabis extract (CRCE) to interact with the most common over-the-counter drug and the major known cause of drug-induced liver injury–acetaminophen (APAP)–in aged female CD-1 mice. Gavaging mice with 116 mg/kg of cannabidiol (CBD) [mouse equivalent dose (MED) of 10 mg/kg of CBD] in CRCE delivered with sesame oil for three consecutive days followed by intraperitoneally (i.p.) acetaminophen (APAP) administration (400 mg/kg) on day 4 resulted in overt toxicity with 37.5% mortality. No mortality was observed in mice treated with 290 mg/kg of …

Copyright 2019 Zachos and Platt Trace fossil zonation in the Hartselle Sandstone of Mississippian age (Chesterian: Visean-Serpukhovian) exposed on Fielder Ridge, Alabama is compared with modern macro-tidal flat ichnocoenoses on the Bay of Fundy at Lubec, Maine, and demonstrated to be analogous by sedimentologic and ichnotaxonomic criteria. The modern flat has minimal influence from either waves or freshwater influx, and can be divided into five distinct ichnocoenoses, characterized by surface traces (epichnia) and four sedimentologic facies defined by gross grain texture or hydrodynamic characteristics, but lacking significant surface traces. Several characteristics of tidal flat deposits in a fetch-limited, marine (i.e., …
