Pre-eclampsia, a condition characterized by new-onset hypertension during pregnancy and one or more indices of organ damage, is a major driver of both short and long-term maternal health outcomes. Individuals with a history of pre-eclampsia are at increased risk for cardiovascular, renal, and other chronic diseases. Recent studies have linked pre-eclampsia history with increased risk of dementia later in life, underscoring the importance of new guidelines emphasizing blood pressure control to reduce the risk of cognitive decline. We present a multiphase study of the star-formation-driven outflow in the Virgo galaxy NGC 4383, combining ALMA CO(2-1) data with deep MeerKAT Hi imaging and MUSE spectroscopy obtained as part of the Multiphase Astrophysics to Unveil the Virgo Environment (MAUVE) program. Our previous work revealed a spectacular ionised outflow, but the effect of the outflow on the cold phase remained unclear. Our analysis shows that potentially outflowing molecular gas is detected only within the inner 1 kpc above the disc, where CO clouds exhibit disturbed kinematics and spatial correspondence with the ionisation cone. Purpose ASP9801, an oncolytic virus encoding interleukin-7 and interleukin-12, was assessed for safety, tolerability, pharmacokinetics, and antitumor activity in patients with advanced solid tumors in a Phase 1 study. Methods The study comprised Part 1 ASP9801 monotherapy dose escalation and Part 2 dose expansion with ASP9801 monotherapy or plus pembrolizumab. Each part was divided into Groups A (ASP9801 injection into cutaneous/subcutaneous lesions) and B (ASP9801 injection into visceral lesions). Primary objectives were safety and maximum tolerated dose and/or recommended Phase 2 dose expansion (RP2D); secondary objectives included antitumor activity and pharmacokinetics. The heart can adapt its performance in response to changing metabolic demands of the rest of the body. A central mechanism intrinsic to the heart is to modulate the function of the cardiac contractile proteins via posttranslational modifications. Although phosphorylation of the cardiac myosin motor-associated regulatory light chain (RLC) by cardiac myosin light chain kinase (cMLCK) has been recognized as a key signaling pathway to increase myocardial contractile function, little is known about its molecular mechanism of action. EGFR-mutant lung adenocarcinomas (LUADs) that are vulnerable to the EGFR antagonist osimertinib (Osi) eventually relapse, owing in part to the emergence of drug-tolerant persister (DTP) cells that arise through epigenetic mechanisms. Intratumoral DTP cells can herald a worse clinical outcome, but the way in which DTP cells influence LUAD progression remains unclear. Osi-resistant (OR) cells exhibit typical DTP cell features, including a propensity to undergo senescence and epithelial-mesenchymal transition (EMT), which can activate heightened secretory states. Every other year, the Federal Highway Administration (FHWA) requires that states submit a Local Highway Finance Report (FHWA-536) to the agency. This report summarizes data on local highway finances and is used by FHWA to evaluate relationships between and changes in revenues, expenditures and investment patterns, and financial trends. This report presents information submitted by Kentucky in its fiscal year (FY) 2023 FHWA-536. Total receipts from local, state, and federal sources used for road and street infrastructure equaled approximately $1.11 billion. Local sources of funding accounted for the largest share of total receipts (47%), followed by state government sources (45%), …

The Kentucky Transportation Cabinet (KYTC) launched its Digital Project Delivery (DPD) Initiative in September 2020 with the goal of expanding the use of digitally formatted plan and project information throughout the transportation asset life cycle, from project delivery through asset management. KYTC retained Kentucky Transportation Center (KTC) staff with expertise in construction and design to facilitate DPD implementation. This document summarizes DPD-related efforts undertaken by the Cabinet over the past five years. We establish resolvent estimates in spaces of bounded solenoidal functions for the Stokes operator in a bounded domain Ω in Rd under the assumptions that Ω is C1 for d≥3 and Lipschitz for d=2. As a corollary, it follows that the Stokes operator generates a uniformly bounded analytic semigroup in the spaces of bounded solenoidal functions in Ω. The smoothness conditions on Ω are sharp. The case of exterior domains with nonsmooth boundaries is also studied. We focus on two aspects of cyclic orbit codes: invariants under equivalence and quasi-optimality. Regarding the first aspect, we establish a connection between the cyclic orbit code generated by a subspace U of Fqn and the associated linear set LU×U. Relating the size of the linear set to the number of fractions formed by the elements of U allows us to derive new bounds on the parameters of the cyclic orbit code. In the second part, we study a particular family of (quasi-)optimal cyclic orbit codes. With the aid of these codes we establish the existence of quasi-optimal codes in …

This report examines trends in how Kentucky District Courts have adjudicated charges related to commercial driver’s license (CDL) and commercial motor vehicles (CMVs). Using court data, researchers evaluated historical patterns in charge filing volumes as well as conviction, dismissal, amendment, and diversion rates. Between 2002 and 2024, conviction rates declined and dismissals increased. Convictions peaked in the mid-70% range in the early 2000s, but fell to the mid-50% range by 2024. Speeding and other serious traffic violations — many of which are advanceable offenses — were dismissed or amended at rates that could have substantively affected the integrity of CDL …

Light reprocessed by dust grains emitting in the infrared enables the study of the physics at play in dusty embedded regions, where ultraviolet and optical wavelengths are attenuated. Infrared telescopes such as JWST have made it possible to study the earliest feedback phases, when stars are shielded by cocoons of gas and dust. Comprehending this phase is crucial for unravelling the effects of feedback from young stars that leads to their emergence and the dispersal of their host molecular clouds. Here we show that the transition from the embedded to the exposed phase of star formation is short (< 4 Myr) and sometimes almost absent (< 1 Myr) across a sample of 37 nearby star-forming galaxies covering a wide range of morphologies, from massive barred spirals to irregular dwarfs. The short duration of the dust-clearing timescales suggests a predominant role of pre-supernova feedback mechanisms in revealing newborn stars, confirming previous results on smaller samples and allowing, for the first time, a statistical analysis of their dependencies. We find that the timescales associated with mid-infrared emission at 21 μm, tracing a dust-embedded feedback phase, are controlled by a complex interplay between giant molecular cloud properties (masses and velocity dispersions) and galaxy morphology. We report relatively longer durations of the embedded phase of star formation in barred spiral galaxies, while this phase is significantly reduced in low-mass irregular dwarf galaxies. We discuss tentative trends with gas-phase metallicity, which may favor faster cloud dispersal at low metallicities.

We discuss the holographic correspondence between 3d “Chern-Simons gravity” and an ensemble of 2d Narain code CFTs. Starting from 3d abelian Chern-Simons theory, we construct an ensemble of boundary CFTs defined by gauging all possible maximal subgroups of the bulk one-form symmetry. Each maximal non-anomalous subgroup is isomorphic to a classical even self-dual error-correcting code over ℤp × ℤp, providing a way to define a boundary “code CFT.” The average over the ensemble of such theories is holographically dual to Chern-Simons gravity, a bulk theory summed over 3d topologies sharing the same boundary. Despite improvement over the past few decades, particularly for white, cisgender women, intersectional gender-based inequality remains prominent within anthropological archaeology and beyond. Building on critiques of the leaky pipeline metaphor laid out in the introduction to this themed issue, and drawing on Black, Indigenous, and Posthumanist Feminisms, we advocate for a metaphorical shift focused on care, inclusivity, and diversity—that of a garden. The garden metaphor provides a way to express and explore the complex and intertwined ways disciplinary norms, institutions, and individuals structure and shape experiences in archaeology. Parents have evolved strategies to reduce the risk of malnutrition in offspring, including the production of specialized nutritional secretions that are tailored to meet the unique needs of developing offspring. Studies in vertebrates, however, show abundant individual variation in nutritional secretions; the causes and consequences of this variation, and the extent to which such patterns can be generalized beyond vertebrates, remain unclear. Here, we investigated natural variation in nutritional secretions in an invertebrate species, the Western honey bee (Apis mellifera L.). Juvenile hormone (JH) is essential for maintaining juvenile status and promoting reproduction in insects, yet the transcriptional regulation of JH biosynthesis remains poorly understood. Previous transcriptomic studies of the Drosophila ring gland—which is composed of the corpus allatum (CA), prothoracic gland (PG), and corpora cardiaca (CC)—have treated the ring gland as a single tissue. As a result, these approaches lacked the resolution necessary to dissect JH biosynthesis and its regulation in a cell-type-specific manner. In humans, the left atria (LA) and the left ventricle (LV) play distinct physiological roles and express sarcomeric proteins with chamber-specific patterns. Despite these important differences, most multi-chamber descriptions of the heart assume uniform myocardial properties. To facilitate a more accurate representation of cardiac function, we measured and compared the contractile properties of isolated skinned human LA and LV muscle fibers at 37 °C. Our experimental measurements included the length-dependent activation (LDA) of force in the isometric steady state, the force response to small quick length changes, and tension redevelopment dynamics. Sarcopenia in alcohol-related liver disease (ALD) is of high clinical significance, but there are no effective treatments. Hyaluronan 35 kDa (HA35), a glycosaminoglycan polymer, modulates responses to Toll-like receptor 4 (TLR4), a lipopolysaccharide receptor, to improve hepatic and macrophage function in ALD. We evaluated skeletal muscle responses to HA35 in preclinical models of ALD. Responses to ethanol, lipopolysaccharide (LPS), and HA35 were studied in differentiated murine C2C12/human-induced pluripotent stem cell (hiPSC)-derived myotubes, and wild-type/HA receptor CD44 knockout (CD44—/—) mouse models of ALD (mALD). Climate change is altering temperature and precipitation regimes in Argentina, with potential consequences for regeneration and persistence of forest tree species, emphasizing the importance of ex situ seed conservation. We evaluated interannual variation in seed traits, desiccation tolerance, storage behavior, and longevity of Cedrela balansae C. DC. and C. fissilis Vell. (Meliaceae), two endangered native species of subtropical rainforests in Argentina. Both species produced desiccation-tolerant seeds, independently of collection year, seed traits, or climatic conditions. Depending on the species, seed traits and longevity varied across years and showed strong relationships with temperature and precipitation, particularly during seed development. Cedrela balansae …

Licochalcone A (LCA) is a characteristic compound in licorice Glycyrrhiza inflata and is widely utilized in pharmaceutical and cosmetic industries. However, the biosynthetic pathway and regulatory mechanisms of LCA remain poorly understood. In this study, we first found the accumulation of LCA is induced by methyl jasmonate (MeJA). Given that MYB transcriptional factors are well-documented as key regulators of flavonoid biosynthesis, we identified a total of 147 GiR2R3-MYB genes in G. inflata, which were classified into 28 subgroups. The chromosome distributions, sequence characteristics, gene structures, duplication events and cis-acting elements were also investigated. The fermentation of cigar tobacco leaves (CTLs) is a critical process driven by microbial activity, which directly influences the degradation of macromolecules and the development of distinctive aromas. However, mechanisms for the directional regulation of this process using defined microbial consortia remain underexplored. This study aimed to construct a synthetic microbial community (SynCom) to enhance the fermentation efficiency and quality of cigar tobacco leaves. Bacillus safensis and Bacillus velezensis with high extracellular cellulase, amylase and protease activities were screened out, and synthetic communities were constructed. The evolutionarily conserved mitogen-activated protein kinase (MAPK) cascades relay extracellular signals into cells, triggering a variety of cellular responses. We previously revealed NtMPK4 as a positive regulator of nicotine biosynthesis; however, its upstream regulation remains unclear. Here, we characterized a MAPK cascade, comprising NtMEKK1b, NtMPKK2a, and NtMPK4, that promotes nicotine biosynthesis. This signaling module transduces external cues, including jasmonate and pathogen elicitors such as flg22, into post-translational modifications that enhance transcriptional activity and pathway gene expression. NtMPKK2a physically interacts with and phosphorylates NtMPK4 in vivo, confirming its role as an upstream kinase. RNAi-mediated silencing of NtMPKK2a significantly reduced the expression …

Autism spectrum disorder (ASD) is a heterogeneous neurological condition with an unclear etiology and pathogenesis. In recent years, studies have identified changes in lipid metabolism, inflammation, mitochondrial dysfunction, and mitophagy in patients with ASD. However, the specific interactions between these molecular signatures and their clinical applications in ASD remain largely unexplored. Background: Prosthetic valve endocarditis (PVE) is a serious complication of valve replacement associated with significant morbidity and mortality. However, outcomes for medical vs surgical strategies remain incompletely characterized. Methods: We conducted a retrospective cohort study of adult patients with PVE, defined by modified Duke criteria, evaluated by a multidisciplinary endocarditis team at a tertiary-care centre between September 2021 and February 2024. Demographic, clinical, management, and outcome data were collected. Logistic regression analyses were performed to identify factors associated with in-hospital mortality. Aims: Biliary strictures are a known complication of hepatobiliary and pancreatic surgeries. This retrospective descriptive series highlights six consecutive patients with complete bile duct occlusion or disruption who required a multidisciplinary approach with a combination of interventional radiology (IR) and endoscopic techniques to either re-cannulate or create a neo-anastomosis in the biliary system. Background: The ACR Diagnostic Fluoroscopy Dose Index Registry (DIR-Fluoro) is expanding to include diagnostic fluoroscopy. Variations in dose reference points and overhead radiography events may introduce unique challenges for benchmarking. Purpose: To survey the technological status and dosimetric performance of fluoroscopes participating in the DIR-Fluoro pilot project, focusing on longitudinal stability and variability of fluoroscopic dose reporting accuracy across multiple institutions and vendors. Methods: Sixty-six fluoroscopic systems from nine institutions (24 facilities) were surveyed for facility type, fluoroscope type, image receptor type, age, dose reporting capabilities, and other key features. An emerging paradigm in public health focuses on enhancing nutrition in existing food staples to reduce chronic disease at the population scale, rather than relying on individuals to change their behavior. This paradigm leverages plant and animal breeding, production practices, and processing to enhance nutrition, whereby foods consumed by millions can be improved at low incremental cost. This article supports and operationalizes this paradigm, illustrating the potential to improve diets through a case study that increases the arabinoxylan fiber content of commodity wheat through classical plant breeding (a non-GMO technology). Lead halide perovskites (APbX3) have demonstrated exceptional opto-electronic properties, but their inherent toxicity and environmental hazards hindered their practical deployment in display technologies such as liquid crystal display (LCD) backlights. Herein, we report for the first time a facile, water-mediated synthesis of red-emitting CsMnBr3 thin films from an aqueous solution of CsBr and MnBr2 precursors at a low temperature of ∼50 °C. Genetic diseases such as ion channelopathies substantially burden human health. Existing treatments are limited and not genotype specific. Here, we report a 2-step high-throughput approach to rapidly identify drug candidates for repurposing as genotype-specific therapy. We first screened 1,680 medicines using a thallium-flux trafficking assay against Kv11.1 gene variants causing long QT syndrome (LQTS), an ion channelopathy associated with fatal cardiac arrhythmia. We identified evacetrapib as a suitable drug candidate that improves membrane trafficking and activates channels. We then used deep mutational scanning to prospectively identify all Kv11.1 missense variants in an LQTS hotspot region responsive to treatment with evacetrapib. …

Nitrogen dioxide (NO2) is an environmental air pollutant and a potent oxidant. Epidemiological studies have revealed a close association between chronic cough and long-term inhalation exposure to NO2. ATP can be released from airway/lung epithelial cells upon inhalation exposure to oxidants, and convincing evidence established in recent clinical studies indicated that extracellular ATP in the respiratory tract plays an important role in the pathogenesis of refractory chronic cough.
