Phosphatase of regenerating liver 3 (PRL-3) is associated with cancer metastasis and has been shown to interact with the cyclin and CBS domain divalent metal cation transport mediator (CNNM) family of proteins to regulate the intracellular concentration of magnesium and other divalent metals. Despite PRL-3’s importance in cancer, factors that regulate PRL-3’s phosphatase activity and its interactions with CNNM proteins remain unknown. Here, we show that divalent metal ions, including magnesium, calcium, and manganese, have no impact on PRL-3’s structure, stability, phosphatase activity, or CNNM binding capacity, indicating that PRL-3 does not act as a metal sensor, despite its interaction …

To enable non-destructive metabolic characterizations on in vitro cancer cells and organotypic tumor models for therapeutic studies in an easy-to-access way, we report a highly portable optical spectroscopic assay for simultaneous measurement of glucose uptake and mitochondrial function on various cancer models with high sensitivity. Well-established breast cancer cell lines (MCF-7 and MDA-MB-231) were used to validate the optical spectroscopic assay for metabolic characterizations, while fresh tumor samples harvested from both animals and human cancer patients were used to test the feasibility of our optical metabolic assay for non-destructive measurement of key metabolic parameters on organotypic tumor slices. The Toxin Complex (Tc) superfamily consists of toxin translocases that contribute to the targeting, delivery, and cytotoxicity of certain pathogenic Gram-negative bacteria. Membrane receptor targeting is driven by the A-subunit (TcA), which comprises IgG-like receptor binding domains (RBDs) at the surface. To better understand XptA2, an insect specific TcA secreted by the symbiont X. nematophilus from the intestine of entomopathogenic nematodes, we determined structures by X-ray crystallography and cryo-EM. Contrary to a previous report, XptA2 is pentameric. Lung cancer heterogeneity is a major barrier to effective treatments and encompasses not only the malignant epithelial cell phenotypes and genetics but also the diverse tumor-associated cell types. Current techniques used to investigate the tumor microenvironment can be time-consuming, expensive, complicated to interpret, and often involves destruction of the sample. Here we use standard hematoxylin and eosinestained tumor sections and the HALO AI nuclear phenotyping software to characterize 6 distinct cell types (epithelial, mesenchymal, macrophage, neutrophil, lymphocyte, and plasma cells) in both murine lung cancer models and human lung cancer samples. Introduction: Vaccination is the most effective mechanism to prevent severe COVID-19. However, breakthrough infections and subsequent transmission of SARS-CoV-2 remain a significant problem. Intranasal vaccination has the potential to be more effective in preventing disease and limiting transmission between individuals as it induces potent responses at mucosal sites.

Methods: Utilizing a replication-deficient adenovirus serotype 5-vectored vaccine expressing the SARS-CoV-2 RBD (AdCOVID) in homozygous and heterozygous transgenic K18-hACE2, we investigated the impact of the route of administration on vaccine immunogenicity, SARS-CoV-2 transmission, and survival.

Background Previously, we showed that the sphingosine-1-phosphate (S1P) transporter spinster 2 (Spns2) mediates activation of microglia in response to amyloid β peptide (Aβ). Here, we investigated if Ponesimod, a functional S1P receptor 1 (S1PR1) antagonist, prevents Aβ-induced activation of glial cells and Alzheimer’s disease (AD) pathology.

Methods We used primary cultures of glial cells and the 5XFAD mouse model to determine the effect of Aβ and Ponesimod on glial activation, Aβ phagocytosis, cytokine levels and pro-inflammatory signaling pathways, AD pathology, and cognitive performance.

Missing data is a common issue in many biomedical studies. Under a paired design, some subjects may have missing values in either one or both of the conditions due to loss of follow-up, insufficient biological samples, etc. Such partially paired data complicate statistical comparison of the distribution of the variable of interest between the two conditions. In this article, we propose a general class of test statistics based on the difference in weighted sample means without imposing any distributional or model assumption. The shift of energy production towards renewable, yet at times inconsistent, resources like solar and wind have increased the need for better energy storage solutions. An emerging energy storage technology that is highly scalable and cost-effective is the redox flow battery comprised of redox-active organic materials. The biggest part of Bosnia and Herzegovina is covered with hilly and mountainous terrain, which, from the agricultural perspective, determines the manner of soil usage. Of all agricultural areas in Bosnia and Herzegovina, 55.49% are covered by grassland with that figure increasing to 79.14% in mountainous regions. The membership of AFGC is divided into three main sectors: private, public and industry. Leucaena leucocephala is a productive and sustainable forage tree legume for beef cattle production in northern Australia. Following a protracted period of research and development to overcome agronomic and social constraints, substantial adoption by graziers is now occurring (Mullen, these proceedings). Canada is the world's second largest country covering approximately 10 million km2 (McCartney & Horton, 1997). About 90% of Canada is uninhabited with 90 percent of Canadians living within 500 km of the American border. The forage resources used by livestock grazing and the production of forage crops covers over 36 million ha of Canada's land base (3.6%) and is divided into 72% native range (26 million ha), 11% cultivated pastures (4 million ha) and 17% forage crops (6 million ha) There are 25 million ha in grain and oilseed crops (McCartney & Horton, 1997).

The Chinese Grassland Society (CGS) was founded in 1979 in Beijing. Since 1991, it has become a national science society authorised by the State Ministry of Civil Affairs. The basic task of CGS is to unite the national grassland scientists and technicians for academic exchanges at home and abroad, to popularise grassland science, to train qualified personnel for the further development of grassland science and to accelerate the development of grassland science. In most of the world the priority for production-oriented research has been succeeded by the need for grassland research to focus on systems which satisfy requirements relating to the stability and protection of land, water and atmospheric resources and to biodiversity, in addition to production efficiency. This dictates not only a new approach to research, but also new approaches for the organisation of research, the training and development of research scientists and the activities of Grassland Societies and associated organisations.

The Tri-Societies (American Society of Agronomy, Crop Science Society of America and Soil Science Society of America), the largest life science professional societies in the United States (US) dedicated to the agronomic, crop and soil sciences, encompass approximately 18,000 members globally and include approximately 14,000 certified professionals who are a direct conduit to the farming communities across the US. The Tri-Societies' US-based federal science policy programmes have been active for decades. There is a Washington, DC-based Director of Science Policy, Congressional Science Fellows (CSF) and Science Policy Interns.

In this presentation, we describe the current web archiving program at the University of Kentucky Libraries; provide an overview of the "Wildcat Histories" activist student organization social media preservation project, funded by Project STAND; discuss the technical aspects of the "Wildcat Histories" project; and the project's current status and lessons learned.
