Linear and nonlinear optical line shapes reveal details of excitonic structure in polymer semiconductors. We implement absorption, photoluminescence, and transient absorption spectroscopies in DPP-DTT, an electron push−pull copolymer, to explore the relationship between their spectral line shapes and chain conformation, deduced from resonance Raman spectroscopy and from ab initio calculations. The viscosity of precursor polymer solutions before film casting displays a transition that suggests gel formation above a critical concentration. Upon crossing this viscosity deflection concentration, the line shape analysis of the absorption spectra within a photophysical aggregate model reveals a gradual increase in interchain excitonic coupling. Catechol is a widespread atmospheric dihydroxybenzene present in vehicle emissions, biomass burning, and combustion pollution plumes. Although the daytime reactivity of catechol is controlled by ozone (O3) and hydroxyl radicals (HO), the action of nitrate radicals (NO3) on the surface of aqueous atmospheric particles should become significant at night. This work simulates nighttime interfacial chemistry between hydrated catechol and adsorbed NO3 to form 4-nitrocatechol during experiments lasting ≤1 μs. Surface-sensitive online electrospray ionization mass spectrometry (OESI-MS) examines the reaction on the water surface under variable ratios of [NO2] and [O3]. Per- and polyfluoroalkyl substances (PFAS) are anthropogenic compounds developed for various applications; some are connected to adverse health impacts including immunosuppression and higher susceptibility to some cancers. Current PFAS remediation treatments from aqueous sources include granular activated carbon (GAC) adsorption, membrane separation, and anion- exchange resin (AER) removal. Each has specific disadvantages, hence the need for a new and efficient technology. Herein, acrylamide-based hydrogel composites were synthesized with powdered activated carbon (PAC) and characterized to determine their affinity for PFAS. Metabolic pathways are a human-defined grouping of life sustaining biochemical reactions, metabolites being both the reactants and products of these reactions. But many public datasets include identified metabolites whose pathway involvement is unknown, hindering metabolic interpretation. To address these shortcomings, various machine learning models, including those trained on data from the Kyoto Encyclopedia of Genes and Genomes (KEGG), have been developed to predict the pathway involvement of metabolites based on their chemical descriptions; however, these prior models are based on old metabolite KEGG-based datasets, including one benchmark dataset that is invalid due to the presence of over 1500 duplicate entries. …

Joint orientation measurements collected by KGS personnel at rock exposures from 2009 to 2024. New joint data was collected using a Brunton compass. This data set is a compilation of joints measured by Steve Martin. There are three worksheets related to data structure. The joint location and joint measurement worksheets show a 1-to-many relationship. The 1to1 worksheet shows a 1-to-1 relationship between joint locations and measurements.

This dataset provides the geographical locations of karst groundwater basins, groundwater flowpaths, and dye injection/recovery points in Kentucky. Dye tracing is a common method used for understanding groundwater movement. Dye is poured (or injected) into a sinking stream, well, sinkhole, or body of water, where it is carried down-gradient by flowing water. The locations at which dye is recovered (typically at artesian springs) indicates a connection from injection to recovery site. This connection is refered to as a groundwater flowpath. Conducting multiple dye traces across an area allows for the interpretation of regional groundwater flow by assessing where flowpaths either …

BACKGROUND: Recent preclinical studies have investigated the atypical kappa-opioid receptor (KOR) agonist, nalfurafine, as a co-formulary with mu-opioid receptor (MOR) agonists as a potential deterrent for misuse. However, no study has investigated effects of nalfurafine combined with a MOR agonist using an oral route of administration. The objective of the current study was to measure behavioral effects of orally administered oxycodone and nalfurafine, alone and combined, in rhesus monkeys using a quantitative behavioral observation procedure.
METHODS: Adult male rhesus monkeys (N=5) were orally administered vehicle, oxycodone (0.56-1.8mg/kg), nalfurafine (0.001-0.0056mg/kg), or mixtures (1.0mg/kg oxycodone/0.001-0.0056mg/kg nalfurafine) in a Jell-O vehicle at multiple …

The aviation industry is a linchpin of Kentucky’s economy. Major firms such as UPS, Amazon, and DHL have major hubs at Louisville Muhammad Ali International Airport (SDF) and Cincinnati/Northern Kentucky International Airport (CVG). As of December 2022, 4,570 aircraft mechanics and service technicians (AMTs) were employed in the state. Demand in this sector is poised to grow. As such, it is critical to understand what resources Kentucky has available to train the next generation of professionals and the number of people the aviation industry is likely to employ statewide over the next decade. Platelets play many roles in the vasculature ensuring proper hemostasis and maintaining integrity. These roles are facilitated, in part, by cargo molecules released from platelet granules via Soluble NSF Attachment Protein Receptor (SNARE) mediated membrane fusion, which is controlled by several protein-protein interactions. Chaperones have been characterized for t-SNAREs (i.e. Munc18b for Syntaxin-11), but none have been clearly identified for v-SNAREs. α- Synuclein has been proposed as a v-SNARE chaperone which may affect SNARE-complex assembly, fusion pore opening, and thus secretion. This paper presents versatile protocols to prepare primary human Schwann cell (hSC) cultures from mature peripheral nervous system tissues, including fascicles from long spinal nerves, nerve roots, and ganglia. This protocol starts with a description of nerve tissue procurement, handling, and dissection to obtain tissue sections suitable for hSC isolation and culturing. A description follows on how to disintegrate the nerve tissue by delayed enzymatic dissociation, plate the initial cell suspensions on a two-dimensional substrate, and culture the primary hSCs. Each section contains detailed procedures, technical notes, and background information to aid investigators in understanding and managing all steps. Some …

Metastasis of lung adenocarcinoma (LUAD) is a major cause of death in patients. Aryl hydrocarbon receptor (AHR), an important transcription factor, is involved in the initiation and progression of lung cancer. Polo-like kinase 1 (PLK1), a serine/threonine kinase, acts as an oncogene promoting the malignancy of multiple cancer types. However, the interaction between these two factors and their significance in lung cancer remain to be determined. In this study, we demonstrate that PLK1 phosphorylates AHR at S489 in LUAD, leading to epithelial-mesenchymal transition (EMT) and metastatic events. Rates of tobacco and alcohol use in women are rising, and women are more vulnerable than men to escalating tobacco and alcohol use. Many women use hormonal birth control, with the oral contraceptive pill being the most prevalent. Oral contraceptives contain both a progestin (synthetic progesterone) and a synthetic estrogen (ethinyl estradiol; EE) and are contraindicated for women over 35 years who smoke. Despite this, no studies have examined how synthetic contraceptive hormones impact this pattern of polysubstance use in females. BACKGROUND: Research conducted in urban areas has highlighted the impact of housing instability on people who inject drugs (PWID), revealing that it exacerbates vulnerability to drug-related harms and impedes syringe service program (SSP) use. However, few studies have explored the effects of houselessness on SSP use among rural PWID. This study examines the relationship between houselessness and SSP utilization among PWID in eight rural areas across 10 states.

Generative AI tools such as ChatGPT have potential as student research tools, yet they generally do not cite sources or may invent them, presenting a challenge of information disconnected from any source. This presentation applied pieces of the Association of College and Research Libraries’ Framework for Information Literacy to analyze this challenge and suggest opportunities to use discussions around the use of generative AI to make students more reflective researchers and consumers of information.

A tricritical point as a crossover between (stationary finite-wavelength) type-Is and (stationary longwave) type-IIs bifurcations is identified in the study of diffusive-thermal (Turing) instability of flames propagating in a Hele-Shaw channel in a direction transverse to a shear flow. Three regimes exhibiting different scaling laws are identified in the neighbourhood of the tricritical point. For these three regimes, sixth-order partial differential equations are obtained governing the weakly nonlinear evolution of unstable solutions near the onset of instability. These sixth-order PDES may be regarded as the substitute for the classical fourth-order Kuramoto–Sivashinsky equation which is not applicable near the …

The multiexciton quintet state, 5 TT, generated as a singlet fission intermediate in pairs of molecular chromophores, is a promising candidate as a qubit or qudit in future quantum information science schemes. In this work, we synthesize a pyrene-bridged parallel tetracene dimer, TPT, with an optimized interchromophore coupling strength to prevent the dissociation of 5 TT to two decorrelated triplet (T1) states, which would contaminate the spin-state mixture. Long-lived and strongly spin-polarized pure 5 TT state population is observed via transient absorption spectroscopy and transient/pulsed electron paramagnetic resonance spectroscopy, and its lifetime is estimated to be >35 ms, with the …

Current development towards implementation of the future smart grid includes advanced controller and power hardware-in-the-loop (CHIL/PHIL) testing of new technology. The impact of new loads, distributed energy resources (DER) equipment, and controls spans two fields, both electric distribution power systems modeling, typically completed in the phasor domain, and electromagnetic transient (EMT) analysis across the frequency domain. Inflammasome activation is a critical defense mechanism against bacterial infection. Previous studies suggest that inflammasome activation protects against Salmonella oral infection. Here we find inflammasome activation plays a critical role in the pathogenesis of Salmonella systemic infection. We show that in a systemic infection model by i.p. injection of Salmonella, deficiency of caspase-1 or gasdermin-D prolonged survival time, reduced plasma concentrations of the proinflammatory cytokines IL-1β, IL-6 and TNFα. These deficiencies also protected against coagulopathy during Salmonella infection as evidenced by diminished prolongation of prothrombin time and increase in plasma thrombin-antithrombin complex concentrations in the caspase-1 or gasdermin-D deficient mice. …

Big Bone Lick is the birthplace of vertebrate paleontology in the Western Hemisphere and has a long and celebrated history in the exploration of the American colonial frontier and of the early United States. Notable European scientists of the 18th century such as Buffon, Cuvier, and Hunter discussed the fossils found there. Prominent Americans of the time, such as Boone, Washington, Franklin, and Jefferson are also part of the site’s history. It is the type locality for several extinct late Pleistocene megafaunal mammals, most notably the iconic American Mastodon, who were attracted to the area by salt licks dictated by …

Battery energy storage systems (BESS) that can be utilized for demand response (DR) and load shifting are limited in adoption by high capital cost. Large residential loads such as electric water heaters (EWH) and heating, ventilation and air-conditioning (HVAC) may be controlled using distributed energy resource management systems (DERMS) to perform functions like batteries, such as reducing cost and decreasing energy storage capacity requirements when implemented at the aggregate level. During fiscal year 2023, with financial support (Grant #M3Da-2022-04) from the Kentucky Office of Highway Safety, the Kentucky Transportation Center (KTC)-continued its work with traffic database officials to facilitate successful attainment of the remaining goals from the 2017-21 Traffic Records Strategic Plan (TRSP), which goals are in the 2022-26 TRSP Transportation Records Improvement Plan (TRIP). For most databases, some new goals were developed; this required further refinement of some projects or strategies to improve database performance. Introduction: Thrombin, the enzyme which converts fibrinogen into a fibrin clot, is produced by the prothrombinase complex, composed of factor Xa (FXa) and factor Va (FVa). Down-regulation of this process is critical, as excess thrombin can lead to life-threatening thrombotic events. FXa and FVa are inhibited by the anticoagulants tissue factor pathway inhibitor alpha (TFPIα) and activated protein C (APC), respectively, and their common cofactor protein S (PS). However, prothrombinase is resistant to either of these inhibitory systems in isolation. Antithrombotic therapy is a delicate balance between the benefits of preventing a thrombotic event and the risks of inducing a major bleed. Traditional approaches have included antiplatelet and anticoagulant medications, require careful dosing and monitoring, and all carry some risk of bleeding. In recent years, several new targets have been identified, both in the platelet and coagulation systems, which may mitigate this bleeding risk. People with dementia have an increase in brain inflammation, caused in part by innate and adaptive immune cells. However, it remains unknown whether dementia-associated diseases alter neuro-immune reflex arcs to impact the systemic immune system. We examined peripheral immune cells from a community-based cohort of older adults to test if systemic inflammatory cytokine signatures associated with early stages of cognitive impairment. Human peripheral blood mononuclear cells were cultured with monocyte or T-cell-targeted stimuli, and multiplex assays quantitated cytokines in the conditioned media. Following T-cell-targeted stimulation, cells from women with cognitive impairment produced lower amounts of TH17 cytokines compared with cells …

CD4+ T cells, particularly IL-17-secreting helper CD4+ T cells, play a central role in the inflammatory processes underlying autoimmune disorders. Eukaryotic Elongation Factor 2 Kinase (eEF2K) is pivotal in CD8+ T cells and has important implications in vascular dysfunction and inflammation-related diseases such as hypertension. However, its specific immunological role in CD4+ T cell activities and related inflammatory diseases remains elusive. Our investigation has uncovered that the deficiency of eEF2K disrupts the survival and proliferation of CD4+ T cells, impairs their ability to secrete cytokines. Genome instability in cancer cells causes not only point mutations but also structural variations of the genome, including copy number variations (CNVs). It has recently been proposed that CNVs arise in cancer to adapt to a given microenvironment to survive. However, how CNV influences cellular resistance against ionizing radiation remains unknown. PRMT5 (protein arginine methyltransferase 5) and APE1 (apurinic/apyrimidinic endonuclease 1), which enhance repair of DNA double-strand breaks and oxidative DNA damage, are closely localized in the chromosome 14 of the human genome. Toxoplasma gondii establishes chronic infection by forming tissue cysts, and this chronic infection is one of the most common parasitic infections in humans. Our recent studies revealed that whereas CD8 + T cells of genetically resistant BALB/c mice have the capability to remove the tissue cysts of the parasite through their perforin-mediated activities, small portions of the cysts are capable of persisting in the presence of the anti-cyst CD8+ T cells.
